Histomorphological changes in shoot apices of Lactuca sativa treated with gibberellic acidO. Lee, N. SugiyamaBiologia plantarum 55:479-484, 2011 | DOI: 10.1007/s10535-011-0113-8 Lettuce plants were treated with gibberellic acid (GA3) and uniconazole (UZ; a gibberellin synthesis inhibitor) to investigate the influence of GA3 on cell division frequency in the shoot apical meristem (SAM) during stem elongation and flower initiation in lettuce (Lactuca sativa) grown in a greenhouse. GA3 (0.1 mM) was sprayed on the surface of outer leaves and uniconazole solution (0.86 mM) was applied to the soil. GA3 increased cell division frequency in the peripheral zone and the rib meristem of shoot apices, and this was associated with the stimulation of stem elongation. UZ treatment decreased cell division frequency in the peripheral zone, rib meristem and subapical pith, and this was associated with restricted stem elongation. Treatment with UZ and GA3 together induced minor stem elongation. Flower induction occurred 3 d earlier in the GA3 and UZ+GA3 treatments than in the control, while the UZ treatment delayed flower initiation for more than 9 d relative to the control. |
Comparison of genomic SSR and EST-SSR markers for estimating genetic diversity in cucumberJ. Hu, L. Wang, J. LiBiologia plantarum 55:577-580, 2011 | DOI: 10.1007/s10535-011-0129-0 Thirteen genomic microsatellite (gSSR) and sixteen expressed sequence tag (EST)-SSR (eSSR) markers were compared to estimate genetic diversity among 29 cucumber (Cucumis sativus L.) accessions. gSSR markers detected mean 4.46 alleles with a mean polymorphic information content (PIC) of 0.664, against eSSR markers with mean 3.38 alleles and a mean PIC of 0.397. gSSRs amplified more null alleles than eSSRs. Genetic diversity within the accession set was estimated by construction of dendrograms using gSSR or eSSR data. There was a clear consistency between gSSR and eSSR trees in terms of positioning of most cucumber germplasms. gSSR markers could separate various types of cucumber germplasms on the whole, although clustering of some accessions was not based on their geographical origins in eSSR tree. eSSR markers identified an independent sub-cluster containing five accessions resistant to downy mildew, suggesting a probable relationship between eSSRs and disease-resistance trait in cucumber. The Mantel test between gSSR and eSSR matrices revealed a good fit correlation (r = 0.836). The general dendrogram constructed using the combined data of gSSRs and eSSRs was similar to those obtained separately with each marker. |
Effect of low boron supply in turnip plants under drought stressR. Hajiboland, F. FarhanghiBiologia plantarum 55:775, 2011 | DOI: 10.1007/s10535-011-0186-4 Turnip (Brassica rapa L.) plants were grown in Perlite with low (< 2.5 μM) or adequate (25 μM) boron supply under well-watered and drought conditions for 12 weeks. Dry mass of leaves and roots was reduced under drought by about 61 and 56 % in plants supplied adequately with B, while up to 84 and 74 % under B starvation. Drought reduced B content by about 70 and 82 % for B-sufficient and B-deficient plants, respectively. According to the chlorophyll fluorescence parameters, the photosynthesis processes conserved their normal activities under low B supply in well-watered plants, while a serious damage to photosystem 2 occurred under drought stress. Stomatal limitation was the most important cause for a 17 % lower net photosynthetic rate (PN) of drought stressed B-sufficient plants. In B-deficient plants, however, both stomatal and non-stomatal limitations were involved in 53 % reduction of PN. Low B supply reduced strongly leaf water potential. |

MorabitoBiologia plantarum 54:191-194, 2010 | DOI: 10.1007/s10535-010-0033-z The effects of cadmium and zinc on growth and mineral distribution in Populus tremula × P. alba genotype 717-1B4 were investigated. Exposure to 360 mg(Cd) kg-1(soil) resulted in accumulation of Cd in all organs and inhibition of primary and secondary growth as well as of the net photosynthetic rate. No growth inhibition occurred under zinc exposure. Cd was mainly stored in the woody parts of stem, whereas zinc was preferentially localized in the leaves. Cd treatment also altered distribution of Zn2+, Ca2+, Mg2+, K+, and Fe2+ in different organs. |
Effect of zinc and cadmium on physiological and production characteristics in Matricaria recutitaM. Kummerová, Š. Zezulka, K. Kráľová, E. MasarovičováBiologia plantarum 54:308-314, 2010 | DOI: 10.1007/s10535-010-0053-8 Effects of zinc (12-180 μM) alone and in mixtures with 12 μM Cd on metal accumulation, dry masses of roots and shoots, root respiration rate, variable to maximum fluorescence ratio (FV/FM), and content of photosynthetic pigments were studied in hydroponically cultivated chamomile (Matricaria recutita) plants. The content of Zn in roots and shoots increased with the increasing external Zn concentration and its accumulation in the roots was higher than that in the shoots. While at lower Zn concentrations (12 and 60 μM) the presence of 12 μM Cd decreased Zn accumulation in the roots, treatment with 120 and 180 μM Zn together with 12 μM Cd caused enhancement of Zn content in the root. Presence of Zn (12-120 μM) decreased Cd accumulation in roots. On the other hand, Cd content in the shoots of plants treated with Zn + Cd exceeded that in the plants treated only with 12 μM Cd. Only higher Zn concentrations (120 and 180 μM) and Zn + Cd mixtures negatively influenced dry mass, chlorophyll (Chl) and carotenoid content, FV/FM and root respiration rate. Chl b was reduced to a higher extent than Chl a. |
Strategies of cadmium and zinc resistance in willow by regulation of net accumulationT. Landberg, P. Jensén, M. GregerBiologia plantarum 55:133-140, 2011 | DOI: 10.1007/s10535-011-0018-6 This work was performed to find out if metal resistant clones of Salix viminalis L. are capable to achieve high resistance to the metals by regulating their net accumulation. Salix clones with low or high resistance in combination with low or high accumulation capacity of either Zn or Cd were cultivated from cuttings in nutrient solution. The investigation included leakage and uptake experiments using 65Zn or 109Cd and analysis of root cation exchange capacity (CEC). Some plants were pre-treated with unlabeled 0.5 μM Cd or 2.5 μM Zn 24 h prior to the experiments to induce possible tolerance mechanisms. To find out if the regulation was a metabolic process, experiments were also performed with 2,4-dinitrophenol (DNP). Clones with high resistance and low Cd accumulation had higher efflux of Cd compared to the other clones, in both untreated and Cd pre-treated plants. This indicates a constitutive property to lower Cd accumulation by high Cd leakage. Pre-treatment with 0.5 μM Cd diminished the Cd net uptake to a level near zero in all clones, likely to be due to decreased the Cd uptake. In contrast, resistant clones with high Cd accumulation had the highest root CEC, which may be used to bind up Cd in the free space. No clear regulation of Zn net uptake was found in Zn-resistant clones. Pre-treatment with Zn decreased the uptake of Zn into the free space in Zn-resistant clones. The resistant high-accumulating clones, however, showed the highest leakage of Zn in both untreated and pre-treated plants, a constitutive process not related to high accumulation. Neither the influx nor the efflux of Cd or Zn was affected by DNP indicating passive transport across the plasma membrane. |

Developmental stages of apices were evaluated based on the morphology of shoot apical meristem (SAM) from various collection dates. Besides, the frequency of each stage was calculated within apices populations after full blooming (DAFB). Prior to doming of apex, three marked phases were found based on SAM morphology: 1) narrow appearance (vegetative phase), 2) broadened form (transition phase), and 3) prominent shape (commitment phase). A furrow region was formed at the base of leaf primordium during the bract initiation, while significant broadening of SAM was observed. Cell division patterns manifested in modification of anisotropic clusters from isotropic cellular packets, as a result of which profound morphological changes of apices occurred. Based on these findings, we propose that the structural alterations prior to doming may be taken into account for determination of the initial development and reproduction signs in apple trees. |
High frequency plant regeneration from the mature seeds of Garcinia indicaM. Baskaran, S. KrishnanBiologia plantarum 55:554-558, 2011 | DOI: 10.1007/s10535-011-0124-5 A high frequency plant regeneration system was developed for the production of high yielding elite clones of Garcinia indica via direct organogenesis. A maximum number of 86.2 shoot buds per explant were induced from the mature seed segments cultured on Woody plant medium (WPM) supplemented with 2.0 mg dm-3 N6-benzyladenine and 1 mg dm-3 indole-3-acetic acid. Rooting was achieved on half-strength WPM medium supplemented with 3 mg dm-3 indole-3-butyric acid. Rooted plantlets were acclimatized and transferred to greenhouse for further growth. The highest survival rate of 95 % was recorded using a mixture of garden soil and sand. Histological studies clearly revealed multiple shoot formation from mature seed segments. |
Hormonal control of the outgrowth of axillary buds in Alstroemeria cultured in vitroP. Pumisutapon, R. G. F. Visser, G. -J. De KlerkBiologia plantarum 55:664, 2011 | DOI: 10.1007/s10535-011-0165-9 We study apical dominance in Alstroemeria, a plant with an architecture very different from the model species used in research on apical dominance. The standard explant was a rhizome with a tip and two vertically growing shoots from which the larger part had been excised leaving ca. 1 cm stem. The axillary buds that resumed growth were located at this 1-cm stem just above the rhizome. They were released by removal of the rhizome tip and the shoot tips. Replacement of excised tips by lanolin with indole-3-butyric acid (IBA) restored apical dominance. The auxin transport inhibitors 2,3,5-triiodobenzoic acid (TIBA) and N-1-napthylphthalamic acid (NPA) reduced apical dominance. 6-Benzylaminopurine (BAP) enhanced axillary bud outgrowth but the highest concentrations (> 9 μM) caused fasciation. Thidiazuron (TDZ) did not show improvement relative to BAP. Even though the architecture of Alstroemeria and the model species are very different, their hormonal mechanisms in apical dominance are for the greater part very similar. |

TianBiologia plantarum 55:752, 2011 | DOI: 10.1007/s10535-011-0181-9 Microtubule cytoskeleton organization during microspore mother cell (MMC) meiosis in Allium cepa L. and microsporogenesis in Nicotiana tabacum L. was examined. The MMC microtubules (MTs) were short and well dispersed in the cytoplasm of both taxa. As the MMCs of both species entered metaphase of meiosis I, the MTs constructed a spindle that facilitated the chromosomes to orient in the meridian plane. At anaphase of meiosis I, the spindle MTs differentiated into two types: one MT type became short, pulled the chromosomes toward the two poles, and was designated as centromere MTs; the second type of MT connected the two poles, and was designated as pole MTs. In A. cepa, where successive cytokinesis was observed, pole MTs assumed a tubbish shape. Some new short MTs aggregated in the meridian plane and constricted to form a phragmoplast, which developed into a cell plate, divided the cytoplasm into two parts and produced a dyad. However, in tobacco, a phragmoplast was not generated in anaphase of meiosis I and II and cytokinesis did not occur. The spindle MTs depolymerized and reorganized the radial arrangement of MTs from the nucleate surface to the periplasm during anaphase. Following telophase of meiosis II, the cytoplasm produced centripetal furrows, which met in the center of the cell and divided it into four parts, serving as a form of cytokinesis. In this process, MTs appeared to bear no relationship to cytokinesis. |
The protection of wheat plasma membrane under cold stress by glycine betaine overproductionX. -Y. Zhang, C. Liang, G. -P. Wang, Y. Luo, W. WangBiologia plantarum 54:83-88, 2010 | DOI: 10.1007/s10535-010-0012-4 We aimed to study the protection of wheat plasma membrane (PM) under cold stress (0-2 °C) by the overaccumulation of glycine betaine (GB). For this, we used wild-type winter wheat (Triticum aestivum L.) cv. Shi 4185 (WT) and 3 transgenic lines (T1, T4, and T6) expressing the BADH gene isolated from Atriplex hortensis L. Under cold stress, the transgenic lines with higher GB content maintained better membrane integrity and higher plasma membrane H+-ATPase activity than WT. In these transgenic lines, ROS production and membrane lipid peroxidation were lower, while antioxidative enzyme activities and compatible solute contents were higher in comparison with WT. This may be attributable to their enhanced cold-stress tolerance mediated by GB overproduction. |
DIANTHIN, a negative selection marker in tobacco, is non-toxic in transgenic rice and confers sheath blight resistanceJ. M. Shah, K. VeluthambiBiologia plantarum 54:443-450, 2010 | DOI: 10.1007/s10535-010-0080-5 The DIANTHIN gene encoding a ribosome-inactivating protein (RIP) from Dianthus caryophyllus L. was tested for negative selection in tobacco and rice. Tobacco leaf discs and scutellum-derived callus of rice were transformed with Agrobacterium tumefaciens strain LBA4404 (pSB1, pJAS1). pJAS1 harbors the DIANTHIN gene under the control of the CaMV 35S promoter. Tobacco transformation efficiency, in comparison to pCAMBIA1301, was reduced by 87 % in pJAS1-transformed leaf discs. The DIANTHIN gene proved to be completely toxic to tobacco as all the recovered hygromycin-resistant transgenic plants harbored truncated T-DNAs with deletions of the DIANTHIN gene. Transformation of the DIANTHIN gene under a Mungbean yellow mosaic virus (MYMV)-inducible promoter did not cause any toxicity in tobacco as reflected by the recovery of transgenic tobacco plants with the complete DIANTHIN gene. Transformation efficiency of pJAS1 did not decline in rice. Interestingly, all transgenic rice plants harbored the complete DIANTHIN gene and expressed the gene. The T1 transgenic lines showed reduction of sheath blight symptom in the range of 29 to 42 %. The difference in the sensitivity to DIANTHIN between tobacco and rice provides a new direction to study the mechanisms underlying RIP toxicity in plants. |
Functional characterization of the apple MdSAMDC2 gene by ectopic promoter analysis and over-expression in tobaccoL. -L. Zhao, L. -Q. Song, C. -X. You, T. Moriguchi, Y. -J. HaoBiologia plantarum 54:631-638, 2010 | DOI: 10.1007/s10535-010-0113-0 The expression of MdSAMDC2 gene, which encodes S-adenosylmethionine decarboxylase (SAMDC) in apple, was up-regulated by low temperature, salt and drought stresses. To identify its in vivo biological functions in the responses to stresses, the promoter region of MdSAMDC2 was isolated and characterized by analyzing the cis-acting regulatory elements and GUS reporter gene by an Agrobacterium-mediated transformation. GUS activity was enhanced upon salt and cold stresses, indicating that MdSAMDC2 promoter region controls gene transcription under stresses. In parallel, several lines of the transgenic tobacco plants over-expressing MdSAMDC2 were obtained. The contents of three polyamines greatly increased in the transgenic lines compared with the non-transgenic WT control. Upon exposed to low temperature (4 °C), salt (150 and 250 mM NaCl) and osmotic (20 % polyethylene glycol) stresses, transgenic plants produced more free polyamines and more active antioxidative enzymes such as superoxide dismutase and catalase than the WT control. Meanwhile, malondialdehyde content, an indicator for membrane lipid peroxidation, decreased in transgenic plants relative to the WT control. Thus, over-expression of MdSAMDC2 in tobacco conferred tolerance to stresses. |
Acetylsalicylic acid ameliorates negative effects of NaCl or osmotic stress in Solanum stoloniferum in vitroF. Daneshmand, M. J. Arvin, K. M. KalantariBiologia plantarum 54:781-784, 2010 | DOI: 10.1007/s10535-010-0142-8 The role of acetylsalicylic acid (0, 1 and 10 μM) pre-treatment in amelioration of salt and osmotic stress in a wild species of potato (Solanum stoloniferum) was investigated. We compared the effects of iso-osmotic concentrations of polyethylene glycol 6000 (15 %) and NaCl (80 mM) on the physiological responses of this species in explants grown in the liquid Murashige and Skoog medium. Both salt and drought reduced shoot growth parameters, photosynthetic pigment contents and increased lipid peroxidation, electrolyte leakage, H2O2 content and lipoxygenase activity. The effect of NaCl was more severe than that of polyethylene glycol. Salinity also increased Na+ content and decreased K+ content and K+/Na+ ratio. Under both stresses, the activities of superoxide dismutase, guaiacol peroxidase, ascorbate peroxidase, catalase and glutathione reductase enzymes were increased. Acetylsalicylic acid pre-treatment alleviated the adverse effects of both stresses on all parameters measured. |

YeoBiologia plantarum 53:329-333, 2009 | DOI: 10.1007/s10535-009-0060-9 Long-term (30 d) effects of 100, 200, 300, and 400 mM NaCl on photosystem 2 (PS 2)-mediated electron transport activity and content of D1 protein in the thylakoid membranes of chrysanthemum (Dendranthema grandiflorum) cultured in vitro at low irradiance 20 µmol(photon) m-2 s-1 were investigated. 100 mM NaCl increased contents of chlorophylls (Chl) a and b, carotenoids (Car; xanthophylls + carotenes), and the ratio of Chl a/b, and Car/Chl a+b. However, further increase in NaCl concentration led to the significant reduction in the contents of Chl a, and Chl b, and increase in the ratio of Chl a/b and Car/Chl a+b. NaCl treatment decreased the PS 2-mediated electron transport activity and contents of various thylakoid membrane polypeptides including D1 protein. |
Responses to drought stress in two poplar species originating from different altitudesF. Yang, X. Xu, X. Xiao, C. LiBiologia plantarum 53:511-516, 2009 | DOI: 10.1007/s10535-009-0092-1 Cuttings of Populus kangdingensis and Populus cathayana, originating from high and low altitudes in the eastern Himalaya, respectively, were examined during one growing season in a greenhouse to determine their responses to drought stress (soil moisture decreased from 100 to 55 or 25 % field capacity). Compared to control plants grown under 100 % field capacity, those poplars grown under 55 and 25 % field capacity possessed lower increases in height and stem diameter, and higher contents of soluble sugars, free proline, malondialdehyde (MDA) and hydrogen peroxide, and higher activities of catalase (CAT), superoxide dismutase (SOD), peroxidase (POD), ascorbate peroxidase (APX) and glutathione reductase (GR). Compared with P. cathayana with greater leaf area, P. kangdingensis with greater root/shoot ratio exhibited lower MDA and H2O2 contents, higher soluble sugar and free proline contents, and higher activities of CAT, SOD, POD, APX and GR. These results suggested that P. kangdingensis was more drought tolerant than P. cathayana. |

MegličBiologia plantarum 54:54-60, 2010 | DOI: 10.1007/s10535-010-0008-0 Three genetic similarity coefficients were estimated and compared for their usefulness: simple matching (S SM), Jaccard's (S J) and Dice's (S D), all based on dominant markers data from individuals representing predominantly self-pollinating species. AFLP markers were used to analyze 139 Phaseolus vulgaris L. (common bean) and 67 Lactuca saligna L. (least lettuce) accessions, and RAPD markers were used to analyze 110 Triticum dicoccoides Koern. (wild emmer wheat) accessions. Similar discriminating structure and power based on the three genetic similarity coefficients was found for each of the three species. This discriminating power was high for both P. vulgaris and L. saligna but moderate for T. dicoccoides. With closely related individuals, as in our study, the absence of a band in two individuals should be due to an identical cause inherited from the same ancestor. Accordingly we propose the use of S SM, which alone out of the three examined coefficients involved shared absence of DNA bands, as contributing to genetic similarity. When RAPDs are employed, inferences about population structure and nucleotide divergence should be made with prudence as the nature of genetic variation uncovered by RAPDs is often unclear. |
Effects of exogenous spermine on sweet sorghum during germination under salinityY. Y. Chai, C. D. Jiang, L. Shi, T. S. Shi, W. B. NaCl treatment resulted in the inhibition of growth and increased the content of free proline, soluble protein and malondialdehyde (MDA). Additionally, it also enhanced the activity of catalase (CAT), peroxidase (POX) in both shoots and roots, while decreased that of glutathione reductase (GR). When exogenous spermine was added to the test solution, the growth of sweet sorghum seedlings was improved, and a smaller increase in the free proline and MDA contents was observed. The addition of spermine also partially increased the activities of POX and GR, but had no effects on soluble protein content or the activity of CAT. |

AmâncioBiologia plantarum 54:237-244, 2010 | DOI: 10.1007/s10535-010-0042-y Plantain (Musa ABB CEMSA 3/4) plantlets were micropropagated in temporary immersion bioreactors (TIB) or in gelled medium (GM). After ex vitro transfer ROS accumulation was determined by infiltrating leaves with nitroblue tetrazolium (NBT) and 3,3'-diaminobenzidine (DAB). Stomatal cells were more stained with NBT and DAB in GM plants than in TIB plants, but the difference disappeared at the end of acclimatization. At the end of the in vitro phase, GM plantlets showed higher activities of ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR), monodehydroascorbate reductase (MDHAR) and glutathione reductase (GR), while activities of catalase (CAT), superoxide dismutase (SOD) and glutathione transferase (GT) were higher in TIB grown plantlets. At the end of acclimatization GT, SOD, CAT and MDHAR stabilized at low values of activity in plantlets derived from both treatments. Concerning the correspondent genes, GM plantlets showed higher expression of all transcripts with the exception of CuZnSOD. The immunobloting of peroxiredoxins (PRXs) showed that chloroplast-located PRXs were expressed at higher levels in TIB plantlets, some showing polymerization. In conclusion, TIB grown plantlets had an improved anti-oxidative response when compared with GM. |
Simple method for isolation of oxygen evolving photosystem 2 core complex free of cytochrome f contaminationR. K. MishraBiologia plantarum 54:520-524, 2010 | DOI: 10.1007/s10535-010-0091-2 This communication presents a simple method for isolation of oxygen evolving photosystem 2 (PS 2) core complex by solubilisation of PS 2 membranes with the nonionic detergent octyl-β-D-thioglucopyranoside (OTG). This complex was free of cytochrome (Cyt) f contamination and also lacked the 22 and 10 kDa proteins that may not be directly required for primary photochemistry of the PS 2 complex and water oxidation. OTG could also remove the Cyt f contamination from the PS 2-core complex isolated using octyl-β-D-glucopyranoside (OGP). The Cyt f contamination in the PS 2 complexes could potentially interfere with spectrophotometric determinations of redox states of Cyt b559 and its stoichiometry in the PS 2 complex. |
The effect of 2,4-dichlorophenol and pentachlorophenol on antioxidant system in the leaves of Phalaris arudinaceaJ. Michałowicz, H. Urbanek, B. Bukowska, W. DudaBiologia plantarum 54:597-600, 2010 | DOI: 10.1007/s10535-010-0108-x The purpose of this work was to evaluate the effect of 2,4-dichlorophenol (2,4-DCP) and pentachlorophenol (PCP) on the activity of antioxidative system and lipid peroxidation in the leaves of reed canary grass (Phalaris arudinacea). The activity of catalase (CAT), ascorbate peroxidase (APX), guaiacol peroxidase (GPX), glutathione reductase (GR) and glutathione S-transferase (GST) as well as the content of glutathione, ascorbate and phenolic compounds were determined. An induced-increase in the APX, CAT, GPX and GR activities was stronger for PCP, while a significant increase in the GST activity was noted only for 2,4-DCP. Both compounds increased the content of phenolic compounds, oxidized and reduced glutathione as well as the content of ascorbic acid. PCP induced stronger increase in lipid peroxidation than 2,4-DCP. The observed changes revealed that chlorophenols induce oxidative stress and oxidative damage in the leaves of reed canary grass. |

A putative LAR gene (DkLAR) was isolated by rapid amplification of cDNA ends from young fruits. The full-length cDNA of DkLAR gene was 1 356 bp long and encoded an open reading frame of 349 residues. The deduced DkLAR protein was closely related to the homolog in other plant species. The expression of the DkLAR gene in Chinese pollination-constant non-astringent (PCNA) genotype was coincident with the tannin cell development, but was not in Japanese PCNA and Chinese pollination-variant astringent (PCA) genotypes. |

HuBiologia plantarum 54:761-765, 2010 | DOI: 10.1007/s10535-010-0137-5 Leaf gas exchange, chlorophyll fluorescence and activities of antioxidant enzymes were studied in two pepper (Capsicum annuum L.) cultivars grown under drought (D) and heat (H), as well as under drought and heat in combination (HD). The drought-tolerant cv. Zhengjiao 13 exhibited greater net photosynthetic rate (PN) and cytochrome respiratory pathway activity (RSHAM), and lower contents of superoxide radical and hydrogen peroxide, as compared to the drought-sensitive cv. Longkouzaojiao. The PN and RSHAM decreased and ROS production increased under D and HD in both cultivars. As compared to the Longkouzaojiao, Zhengjiao 13 retained higher non-photochemical quenching (NPQ), photorespiration rate (RL), and alternative respiratory pathways (RKCN) under D and HD. Drought increased the superoxide dismutase (SOD) and ascorbate peroxidase (APX) activities in the cytosol, chloroplasts and mitochondria in the two cultivars. Although SOD and APX activities decreased in Longkouzaojiao under HD, SOD activity increased in Zhengjiao 13. There was no H-induced reactive oxygen species production due to increase of RL, NPQ, RSHAM, RKCN and activities of SOD and APX. However, H slightly decreased the PN. The results indicated that HD was more detrimental than either stress alone. |

Our results indicate that the cassava (Manihot esculenta Crantz) genome possesses two genes encoding for putative ATP/ADP translocases, which we have designated as MeAATP1 and MeAATP2. Their deduced products are 92 % identical, and phylogenetic reconstructions of plant AATP sequences suggest that MeAATP1 and MeAATP2 are the result of a relatively recent duplication event. Both genes were found to be expressed in a wide range of plant organs via RT-PCR, including young and mature leaves, fibrous and tuberous roots, and green stems. Neither MeAATP1 nor MeAATP2 showed evidence of increased transcription in the presence of exogenous sucrose. Interestingly, the transcriptional activity of MeAATP1 in leaves appeared to be upregulated after wounding, potentially indicating its involvement in the wound response mechanism of cassava. |

Three different strains of Agrobacterium (EHA105, AGL1 and LBA4404) harboring the binary vector pCAMBIA1301 containing β-glucuronidase (GUS) and hygromycin phosphotransferase (hpt) genes under the control of a CaMV35S promoter were used. The highest number of transgenic plants was obtained from cotyledonary node-derived calli of genotype Pusa-256. A highly efficient rooting was achieved on Murashige and Skoog medium supplemented with indole-3-butyric acid. The stable integration of the gene was confirmed by molecular analyses of the transformed plants. Inheritance of GUS and hpt gene was followed through two generations and they showed the expected 3:1 inheritance. |

Somatic embryos were induced from immature cotyledons on Murashige and Skoog's (MS) medium supplemented with different concentrations of 2,4-dichlorophenoxyacetic acid (2,4-D), 2,4,5-trichlorophenoxyacetic acid (2,4,5-T), α-naphthaleneacetic acid (NAA) and picloram alone or in combination with 0.5 - 2.0 mg dm-3 N6-benzylaminopurine (BA) or kinetin (KIN). NAA was better for somatic embryo induction compared to other auxins. The well formed, cotyledonary shaped embryos germinated into plantlets with 36.6 % frequency on MS medium supplemented with 2.0 mg dm-3 BA + 0.5 mg dm-3 abscisic acid (ABA). The frequency of embryogenesis and plantlet regeneration was higher in cv. ICCV-10 as compared to cv. Annigeri. Regenerated plants were transferred to soil (40 % survival) and grown to maturity. Histological studies of explants at various developmental stages of somatic embryogenesis reveled that somatic embryos developed directly from the cotyledon cells and they were single cell origin. |
Concurrent occurrence of α-amylase inhibitor and stimulator in red kidney bean seed: physiological implicationsH. Alizadeh, D. W. M. Leung, A. L. J. ColeBiologia plantarum 54:195-197, 2010 | DOI: 10.1007/s10535-010-0034-y It is hypothesized that since protein α-amylase inhibitor (α-AI) and stimulator might be present together in red kidney bean (Phaseolus vulgaris L.) seeds, their in vitro interactions might influence their detection and quantification. Assay of α-AI using extracts from the embryonic axes revealed an unexpected finding in that the extracts stimulated rather than inhibited α-amylase activity. The cotyledon extracts exhibited inhibitory or enhancement effect on α-amylase activity depending on whether prior to the α-amylase assay they had been boiled for 10 min or not. Phytohemagglutinin (PHA-L in particular) is implicated in the present study as a stimulator of α-amylase activity co-extracted with α-AI from red kidney bean cotyledons. The importance of these findings is discussed in relation to the possible widespread occurrence of protein α-amylase stimulator in seeds and other plant parts. |

Further, these treatments decreased relative water content, chlorophyll content, 14CO2-fixation, activities of phosphoenolpyruvate carboxylase and ribulose-1,5-bisphosphate carboxylase and abscisic acid content while increased malondialdehyde, hydrogen peroxide and free proline contents. Chloroplast and root ultrastructure was also changed. Pretreatment of seeds with SA (500 μM) for 20 h resulted in amelioration of these effects. |

Activities of pyrroline-5-carboxylate reductase (P5C reductase) and pyrroline-5-carboxylate synthetase (P5C synthetase), two enzymes involved in the Pro biosynthetic pathway were also estimated. Water stress resulted in a reduction in the leaf dry mass and chlorophyll content along with a gradual accumulation of Pro. RT-PCR results show higher expression of Δ1-pyrroline-5-carboxylate synthetase (p5cs) gene in correlation with up-regulated Pro accumulation in cv. A1. P5C reductase was found to be the Pro synthesis rate limiting whereas P5C synthetase did not show any specific response to the drought stress in both cultivars. |
Effect of temperature on water transport through aquaporinsI. F. Ionenko, A. V. Anisimov, N. R. DautovaBiologia plantarum 54:488-494, 2010 | DOI: 10.1007/s10535-010-0086-z The mean effective water self-diffusion coefficient in maize root segments under the effect of aquaporin blocker (mercuric chloride, 0.1 mM) was measured using the spin-echo NMR method with pulsed magnetic field gradient within the temperature range from 10 to 35 °C. HgCl2 caused the reduction in water diffusion by 30 % as compared to the control samples. Temperature dependences of water self-diffusion coefficients showed two linear regions with different values of Q10 and activation energy, Ea. As the temperature reduced from 20 to 10 °C, Ea values calculated from the Arrhenius plots were close to those of bulk water (20 ± 3 kJ mol-1) and slightly changed for the sample pretreated HgCl2. Within the temperature range from 25 to 35 °C the slope of temperature dependences became steeper and Ea values were 31 ± 3 kJ mol-1 for the control and 40 ± 4 kJ mol-1 for the treated sample. In the vicinity of 20 °C, the temperature dependence of water diffusion via the mercury-sensitive water channels showed extreme value. In the region, the specific area of the mercury-sensitive aquaporins was 0.004 % of the total cell surface area. The data indicate that water transfer via aquaporins is sensitive to temperature, and the contributions of the transmembrane pathways (aquaporins, lipid bilayer) differ in different temperature ranges. |
