Gas exchange characteristics and water relations in two cultivars of Hibiscus esculentus under waterlogging

Thirty-day-old plants of two okra cultivars, Sabzpari and Chinese-red, were subjected for 30 d to normal watering or continuous flooding. Continuous flooding did not cause any adverse effect on shoot fresh and dry biomass. Leaf water potential and pressure potential of both cultivars increased significantly due to waterlogging, but there was a slight increase in leaf osmotic potential. Chlorophyll a and b contents decreased significantly and chlorophyll a/b ratio increased. Waterlogging caused a significant reduction in net photosynthetic rate, water use efficiency and intrinsic water use efficiency, but stomatal conductance and intercellular CO2/ambient CO2 ratio remained unchanged.

Characterization of Transgenic Rice Plants Expressing an Arabidopsis FAD7

Fatty acid ω-3 desaturase (FAD) is the key enzyme catalyzing the formation of trienoic fatty acids. We utilized an Arabidopsis FAD7 gene and the seven independent transgenic rice plants harbouring 1 to 3 copies of this gene were generated. The expression of FAD7 mRNA was different among independent transgenic lines regardless of the copy number. The total linolenic acid (18:3) contents reduced by about 7 - 32 % in transgenic rice plants but the linoleic acid (18:2) content increased accordingly. With or without wounding treatments, the jasmonate content was higher in transgenic lines than in wild-type rice plant. The transgenic lines overproducing jasmonate also showed increased expression of PR1b mRNA and allene oxide synthase inresponse to wounding.

Microspore development during in vitro androgenesis in triticale

Microspore division was monitored in three triticale (× Triticosecale Wittmack) genotypes over 21 d of in vitro anther culture, on two media differing in their 2,4-dichlorophenoxyacetic acid content. After low temperature (4 °C) pre-treatment for two weeks, all the microspores were still alive, but they began to die from day one of culture. Both genotype and culture medium affected the number of microspores that aborted over time (82 - 97 % by day 21), the number of microspores that underwent the first symmetrical division (> 82 % over all), the number of microspores that attained four or more nuclei, and the number of divisions per 100 alive microspores after 21 d of culture.

Structure and development of the secretory cavities of Myrtus communis leaves

The structure and development of Myrtus communis L. secretory cavities has been studied in young and expanded leaves, using light and scanning electron microscope. Secretory cavities are continuously formed during leaf development, but in mature leaves the rhythm of their appearance shows steep decrease. Each secretory cavity is developed from a single epidermal cell, which undergoes a periclinal division followed by anticlinal and several oblique cell divisions. The lumen of the secretory cavity is initiated by cell wall separation, i.e., schizogenously. The secretory cells line the cavity, where the secreted material is collected. Secretory cavities are covered by modified epidermal cells, which do not seem to form any special aperture. Essential oils seem to be discharged after mechanical treatment of the leaf.

Importance of glucose-6-phosphate dehydrogenase in taxol biosynthesis in Taxus chinensis cultures

The roles of glucose-6-phosphate dehydrogenase (G6PDH) in paclitaxel production were investigated in cell suspension cultures of Taxus chinensis. In the normal cultures, the trend of G6PDH activity was similar to that of cell growth. Addition of glutamate increased G6PDH activity, while dehydroepiandrosterone (DHEA) decreased G6PDH activity. In elicitor-treated cultures, cell growth was depressed, while G6PDH activity and taxol production were enhanced compared with the control. Glutamate recovered the depression of cell growth, and resulted in further increase in G6PDH activity and taxol production. Contrarily, DHEA exacerbated the depression of cell growth, and decreased G6PDH activity and taxol production induced by fungal elicítor. The results indicated that G6PDH played a critic role of taxol production by affecting cell viability.

Propagation of Haemaria discolor via in vitro seed germination

In vitro propagation protocol for Haemaria discolor (Ker) Lindl. var. dawsoniana by artificial cross-pollination and asymbiotic germination of seeds has been developed. Fruit set (100 %) was obtained when the pollinia and ovules of various aged flowers were used for pollination. In vitro germination of seeds obtained from capsules of various ages was achieved on half-strength Murashige and Skoog's (MS) medium supplemented with 3 % sucrose and 0.85 % agar. The germinated seedlings were cultured on half-strength MS medium with 0.2 % activated charcoal, 8 % banana homogenate, 0.1 mg dm-3 1-phenyl-3-(1,2,3-thiadiazol-5-yl)urea (TDZ) and 1 mg dm-3 α-naphthaleneacetic acid (NAA). Ninety-six percent of plantlets survived after hardening in greenhouse.

Micropropagation of Lavandula dentata from axillary buds of field-grown adult plants

Axillary buds from adult field-grown plants of Lavandula dentata L. were used to evaluate the effect of growth regulators and culture media on the in vitro shoot proliferation and growth. The highest multiplication rate was obtained using Murashige and Skoog (MS) medium supplemented with a combination of 2.2 μM of benzyladenine and 2.5 μM indole-3-butyric acid. The best condition for rooting was MS medium plus 2.5 μM naphthaleneacetic acid. Rooted plantlets were successfully transferred to soil. Short-term culture derived plants (6 month) exhibited a normal development, but a low frequency of not heritable morphological changes were detected in long term culture derived plants (more than 1 year).

Protein analysis of dwarfed transgenic rice plants overexpressing GA2-oxidase gene

Using 2-D electrophoresis, we analyzed proteins from transgenic rice overexpressing gibberellin acid (GA) catabolic enzyme, GA2-oxidase. These results indicate eight specific proteins differentially expressed in the transformed rice stems of T1 generation, but non in case of T2 generation. Proteins isolated from different stages of leaves of T1 generation showed no significant differences, except one-month-old leaf, where five differentially expressed proteins are visible.

In vitro Regeneration and Transformation of Blackstonia perfoliata

In vitro root culture of yellow wort (Blackstonia perfoliata (L.) Huds.) was initiated on Murashige and Skoog (MS) medium. In the presence of benzylaminopurine (BAP) numerous adventitious buds formed, which developed into shoots. Presence of indole-3-butyric acid (IBA) in media significantly decreased number of buds, but increased development of lateral roots. On hormone-free medium shoots successfully rooted and developed flowers and viable seeds that formed another generation. Shoot cultures of B. perfoliata inoculated with suspension of Agrobacterium rhizogenes strain A4M70GUS developed hairy roots at 3 weeks and they were cultured on hormone-free MS medium. Spontaneous shoot regeneration occurred in 3 clones.

Somatic embryogenesis from ovaries, developing ovules and immature zygotic embryos, and improved embryo development of Castanea sativa

Somatic embryogenesis of European chestnut (Castanea sativa Mill.) was obtained using juvenile tissue cultured on P24 medium with 5 μM 2,4-dichlorophenoxyacetic acid plus 0.5 μM 6-benzylaminopurine (BA) for three weeks and then cultured on 0.89 μM BA. Induction frequency with ovaries ranged from 2.0 to 19.1 % and was observed in tissue collected 2 to 8 weeks postanthesis, ovules used as a starting tissue gained 0.8 to 7.8 %, 3 to 9 weeks postanthesis. Zygotic embryos collected 5 to 10 weeks postanthesis formed 10.5 to 57.1 % somatic embryos, respectively. The culture lines were maintained via secondary embryogenesis on P24 medium with 0.89 μM BA. Development and maturation were stimulated on P24 medium with increased agar concentration (1.1 %). Five plantlets were transferred to substrate and acclimatized successfully in greenhouse.

Water stress-induced oxidative damage and antioxidant responses in micropropagated banana plantlets

Oxidative injury and antioxidant responses were investigated in two banana genotypes (Musa AAA 'Berangan' and Musa AA 'Mas') subjected to 40 % PEG-induced water stress. PEG treatment resulted in oxidative injury, as expressed in increased lipid peroxidation and reduced membrane stability index, in both cultivars; however, greater oxidative injury was detected in 'Mas'. Under PEG treatment, catalase activity and glutathione reductase activity were enhanced in both cultivars, but were higher in 'Mas'. Ascorbate peroxidase activity was enhanced in 'Berangan' under water stress, but was unaffected in 'Mas'. Meanwhile, superoxide dismutase activity was inhibited in both cultivars under water stress, but higher activity was detected in 'Berangan'. Higher ascorbate peroxidase and superoxide dismutase activities were associated with greater protection against water stress-induced oxidative injury.

Use of a simple semiquantitative method for appraisal of green fluorescent protein gene expression in transgenic tobacco plants

We have applied a simple method for evaluation of gfp gene expression in plants using a CCD camera and computerized processing of images. Transgenic tobacco plants were obtained by Agrobacterium tumefaciens-mediated transfer of plasmid T-DNA bearing a m-gfp5-ER sequence governed by the 35S promoter together with the nptII selectable marker gene. Presence of the gfp gene in plants was confirmed by a polymerase chain reaction method. Mean brightness values measured using image analysis software showed differences between transgenic and control plants and suggest the possibility of rapid selection of transgenic individuals among regenerants and their progenies.

Mapping of esterase loci in Aegilops uniaristata and homoeologous group 3 chromosomes of wheat

This study was planned to identify the chromosomal location of esterase loci in wheat (Triticum aestivum), in comparison to Aegilops uniaristata, using wheat Ae. uniaristata disomic addition and translocation lines. Two loci (Est-N1 and Est-N8) were identified on 3N chromosome of Ae. uniaristata and their probable homoeoloci were, for the first time, mapped close to three RFLP probes (Xpsr56, Xpsr394, and Xpsr1196) on homoeologous group 3 wheat chromosomes.

Optimalization of the peroxidase production by tissue cultures of horseradish in vitro

Tissue cultures of Armoracia rusticana L., both transformed with Agrobacterium rhizogenes and nontransformed, were screened for peroxidase activity. Most of the derived and tested strains exhibited 20 times higher activity [from 99 to 723 U g-1(d.m.)] than the root of the intact plant [(30 U g-1 (d.m.)]. The highest peroxidase activity was found in tumour culture growing on the medium without growth regulators. The influence of the addition of sugars and heavy metal ions in the medium on peroxidase production was tested. Increase in peroxidase activity was observed after cultivation of horseradish culture with cadmium, cobalt, nickel or lead ions.

Random amplified DNA polymorphism of Nicotiana tabacum L. cultivars

The polymorphism, similarities and relationships among Nicotiana tabacum L. cultivars were assessed with RAPD analyses. One hundred and forty-nine bands were detected, of which 94 were polymorphic (63.1 %). A primer distinguishing all of the tested cultivars was found. High similarity between cultivars was revealed, and cultivar relationships were estimated through cluster analysis (UPGMA) based on RAPD data.

Oxidations of various substrates and effects of the inhibitors on purified mitochondria isolated from Kalanchoë pinnata

Kalanchoë pinnata mitochondria readily oxidized succinate, malate, NADH, and NADPH at high rates and coupling. The highest respiration rates usually were observed in the presence of succinate. The high rate of malate oxidation was observed at pH 6.8 with thiamine pyrophosphate where both malic enzyme (ME) and pyruvate dehydrogenase were activated. In CAM phase III of K. pinnata mitochondria, both ME and malate dehydrogenase (MDH) simultaneously contributed to metabolism of malate. However, ME played a main function: malate was oxidized via ME to produce pyruvate and CO2 rather than via MDH to produce oxalacetate (OAA). Cooperative oxidation of two or three substrates was accompanied with the dramatic increase in the total respiration rates. Our results showed that the alternative (Alt) pathway was more active in malate oxidation at pH 6.8 with CoA and NAD+ where ME operated and was stimulated, indicating that both ME and Alt pathway were related to malate decarboxylation during the light. In K. pinnata mitochondria, NADH and NADPH oxidations were more sensitive with KCN than that with succinate and malate oxidations, suggesting that these oxidations were engaged to cytochrome pathway rather than to Alt pathway and these capacities would be desirable to supply enough energy for cytosol pyruvate orthophosphate dikinase activity.

Effect of arsenic on some physiological parameters in bean plants

The objective of the study was to investigate the effect of different arsenic concentrations on some physiological parameters of bean (Phaseolus vulgaris L.) cultivars Plovdiv 10 and Prelom in the early growth phases. Seedlings, grown in sand with Hoagland-Arnon nutrient solution in a climatic box, were treated with 0, 2, 5 mg(As) dm-3 as Na3AsO4 (pH 5.5). After 5 d of As treatment, the changes in leaf gas-exchange, water potential, chlorophyll and protein contents, peroxidase activity and lipid peroxidation in roots were recorded. Physiological analysis showed a minor negative effect of arsenic at concentration 2 mg(As) dm-3, but at the higher dosage of 5 mg(As) dm-3 growth, leaf gas-exchange, water potential, protein content and biomass accumulation were reduced in both cultivars. The peroxidase activity and lipid peroxidation increased considerably at 5 mg(As) dm-3, which is a typical reaction of the plants to a presence of oxidative stress.

The effects of ozone on growth and stomatal response in the F2 generation of hybrid poplar (Populus trichocarpa × Populus deltoides)

Thirty-six F2 hybrid poplar (Populus trichocarpa × P. deltoides) clones were fumigated with ozone to record its effects on growth, correlate them with stomatal response and screen for ozone sensitivity. Fumigation was applied for 6 to 9 h each day for approximately 3 months at ozone concentrations of 85 to 128 μg g-1 using open-top chambers. Height, diameter, number of leaves, stomatal conductance, transpiration rate, total biomass, biomass components and root/shoot ratios were reduced by ozone stress. Percent of leaf fall in ozone-treated plants was nearly three times higher than in control plants exposed to charcoal-filtered air. Leaf senescence, because of ozone exposure, did not appear to be associated with reduced biomass production. Some clones had a high percentage of leaf-fall with ozone exposure, but were able to maintain total biomass production near that of the control. Their response may be an example of an ability to adjust or compensate for ozone damage. There was no significant or consistent relationship between stomatal conductance and total biomass or the change in stomatal conductance as a result of ozone exposure and the change in total biomass. Taken together, these results suggest that effects of ozone on poplar growth cannot be solely correlated to changes in stomatal conductance, more physiological and biochemical parameters should be examined.

Production of saponins from Panax ginseng suspension and adventitious root cultures

Biomass growth and ginsenoside production in cell suspension and adventitious roots of Panax ginseng C.A. Meyer cultures cultivated both in Erlenmayer flasks and a 3 dm3 bioreactor were studied. The maximum content of ginsenosides was found in the suspension culture cultivated in the bioreactor (4.34 % dry mass), however the saponin content was limited to two major ginsenosides, Rb1 and Rg1. The production of ginsenosides in adventitious roots was lower (1.45 or 1.72 % dry mass), nevertheless, the full range of ginsenosides was detected.

Role of enzymes of sucrose-starch conversion in seed sink strength in mung bean

Changes in the activities of sucrose synthase (SuSy), ADP-glucose pyrophosphorylase (AGPase), UDP-glucose pyrophosphorylase (UGPase), alkaline inorganic pyrophosphatase, 3-phosphoglycerate (3-PGA) phosphatase and amylases were monitored in relation to accumulation of starch in developing pods of mung bean (Vigna radiata L.). With the advancement in the seed development, the contents of starch rose with a concomitant fall in the branch of inflorescence and podwall after 10 d after flowering. The activity of UDPase in all the three pod tissues remained higher than the activity of AGPase showing it to be an important enzyme controlling carbon flux. The activity of alkaline inorganic pyrophosphatase in developing seed in contrast to 3-PGA phosphatase correlated with starch accumulation rate. Activity of β-amylase increased in all the pod tissues till maturity. It appears that the cooperative action of SuSy, UGPase and AGPase controls the efficient partitioning of sucrose into ADP glucose and thereby regulate the seed sink strength of the mung bean.

Detection of Intra-Clonal Genetic Variability in Vegetatively Propagated Tea Using RAPD Markers

The role of random amplified polymorphic DNA (RAPD) markers in detecting intra-clonal genetic variability in vegetatively propagated UPASI-9 clone of tea (Camellia sinensis) was studied. Twenty five decamer primers were used, of which three did not amplify, three gave single bands and the rest of nineteen primers generated upto twelve bands (an average of 6.3 bands per primer). Twenty one primers exhibiting amplified products gave monomorphic banding patterns. Only one primer (OPE-17) gave a unique extra band of similar size in four plants.

Semi-Automated Simple Sequence Repeat Analysis Reveals Narrow Genetic Base in Indian Potato Cultivars

A study was conducted to generate fingerprints of thirty-two Indian potato cultivars using capillary electrophoresis based semi-automated simple sequence repeat analysis. Five fluorescent-tagged primer pairs (STPOACUTR, LEGAST1, STPOAC58, STM0030 and STM0031) used in this study yielded 43 alleles at 16 different loci, showing multi-loci resolving character. The estimated similarity between the cultivars was in the range of 0.72 to 0.98 indicating narrow genetic relationship. None of the primer set alone could differentiate all 32 cultivars included in this study. However, two primer sets STM-0031 and STPOAC58 amplifying 12 and 9 polymorphic alleles, respectively, could together distinguish all of them. The results indicated usefulness of semi-automated capillary electrophoresis in quick and reproducible SSR genotyping of potato cultivars.

Water-use efficiency and carbon isotope discrimination of Acacia ampliceps and Eucalyptus camaldulensis at different soil moisture regimes under semi-arid conditions

Acacia ampliceps Maslin and Eucalyptus camaldulensis Dehnh. were grown for one year in lysimeters at three soil moisture regimes: 100 % (well-watered), 75 % (medium-watered) and 50 % (low-watered) of total plant available water. Biomass yield of both species increased with increase in soil moisture. Water-use efficiency (WUE) of E. camaldulensis decreased and that of A. ampliceps increased markedly with decrease in available soil moisture. A. ampliceps showed 4 - 5 times more biomass yield than E. camaldulensis grown at similar soil moisture. A. ampliceps showed almost 5, 9 and 12 times higher WUE than E. camaldulensis under low-, medium- and well-watered treatments, respectively. Significant negative correlation of δ13C with WUE (r = -0.99) was observed in A. ampliceps. In contrast, δ13C of E. camaldulensis showed a significant positive correlation with WUE (r = 0.82).

Changes in protein profiles associated with somatic embryogenesis in peanut

The somatic embryogenesis potential of zygotic embryo axes of peanut (Arachis hypogaea L. cv. DRG-12) at different stages of development was evaluated by culturing on MS medium with 18.1 μM 2,4-dichlorophenoxyacetic acid (2,4-D). A 100 % frequency with 18.3 somatic embryos per explant was observed from 4 mm long immature zygotic embryo axes collected 31 - 40 d after pollination. Medium supplemented with 16.6 μM picloram resulted in slow development of somatic embryos whereas in the presence of 21.5 μM α-naphthaleneacetic acid (NAA), the explants underwent maturation with induction of roots after 30 d. The changes in protein profiles in zygotic embryo axes at different stages of development correlated with their potential to form somatic embryos. Immature zygotic embryo axes exhibited high frequency somatic embryogenesis in the stage preceding abundant accumulation of 22 and 65 kDa proteins. The content of 22 and 65 kDa proteins decreased immediately after culture on medium fortified with 18.1 μM 2,4-D and increased again after 12 d of culture coinciding with the development of somatic embryos on the explants. The content of 22 and 65 kDa proteins was low at 15 d of culture on medium supplemented with 16.6 μM picloram possibly due to slow development of the somatic embryos on the explant. On maturation medium containing 21.5 μM NAA, a marked increase in the content of 22 and 65 kDa proteins in 15 d-old cultures was observed.

Effect of γ-radiation on development, yield and quality of microtubers in vitro in Solanum tuberosum L.

Explants obtained from in vitro-propagated plantlets of two potato cultivars, Shepody and Atlantic, were treated with five doses of γ-radiation (0, 2, 4, 6 and 8 Gy) to investigate the stimulating effects of low irradiation on the production and quality of microtubers in vitro. Microtubers of both cultivars treated with γ-radiation initiated 5 d earlier than in the non-irradiated control. The whole period of microtuberization was prolonged by 10 - 15 d with 4, 6 and 8 Gy irradiation treatment for cv. Atlantic. Irradiation of the plantlets (4 Gy) led to a significant increase not only in the microtuber number (116.7 and 34.5 % over the control) but also in the fresh mass (77.6 and 23.2 % in Shepody and Atlantic, respectively). Low dose irradiation (2 - 4 Gy) increased the starch content of microtubers. High doses (6 - 8 Gy) enhanced ascorbic acid and reducing sugar contents. 4 - 6 Gy doses also effectively increased the protein contents of microtubers.
