Somatic Embryogenesis, Plantlet Regeneration and Micropropagation of Cultivars and F1 Hybrids of Manihot esculentaM.N. OgburiaBiologia plantarum 46:429-432, 2003 | DOI: 10.1023/B:BIOP.0000023888.82993.b1 Explants of four F1 hybrids (OMR 36-41/1, OMR 36-41/2, OMR 36-41/4 and OMR 36-41/5) and two cultivars (Rayong 1 and Rayong 60) of cassava (Manihot esculenta Crantz) were subjected to different combinations of 2,4-dichlorophenoxyacetic acid (2,4-D), 1-naphthaleneacetic acid (NAA), kinetin (KIN) and N6-benzylaminopurine (BAP) to induce somatic embryogenesis, organogenesis and micropropagation. Shoot apices of the F1 hybrids exhibited higher frequency (62 - 74 %) of proliferation of somatic embryos than the cultivars (21 - 43 %) in Murashige and Skoog basal medium supplemented with 8 mg dm-3 2,4-D and 0.5 mg dm-3 NAA. Nodal explants of regenerated plantlets were rapidly micropropagated with 90 % efficiency on a medium containing 0.1 mg dm-3 NAA and 0.05 mg dm-3 BAP irrespective of explant source. |

Selecting Bacterial Strains for Use in the Biocontrol of Diseases Caused by Phytophthora capsici and Alternaria alternata in Sweet Pepper PlantsA. Sid, M. Ezziyyani, C. Egea-Gilabert, M.E. CandelaBiologia plantarum 46:569-574, 2003 | DOI: 10.1023/B:BIOP.0000041063.38176.4a More than 500 isolates of bacteria were obtained from the aerial part and rhizosphere of sweet pepper (Capsicum annuum L.) plants harvested from different places in the Region of Murcia (Spain). The isolates were purified and assayed in vitro against Phytophthora capsici and Alternaria alternata. Sixty isolates (12 %) produced an inhibition zone against at least one of the pathogens, while ten had a strongly inhibitory effect on both pathogens assayed. Microscopic observation of interactions zone showed cell vacuolisation, hyphae lysis and spilling of cytoplasm content of the pathogens in the culture media. These ten isolates were then chosen for biocontrol of Phytophthora root rot and Alternaria leaf spots of pepper plants in vivo. Four of them denominated HS93, LS234, LS523 and LS674 reduced P. capsici root rot by 80, 51, 49 and 54 %, respectively, and A. alternata leaf spots by 54, 74, 62 and 53 %. HS93 belongs to the genus Bacillus and probably the species subtilis, while LS234, LS523 and LS674 belong to the genus Bacillus and probably the species licheniformis. Dry mass of plants treated with these bacteria was significantly higher than that of non-treated and inoculated plants. |

Total leaf area, dry mass of leaves, length and dry mass of shoots and roots increased with the age of the plant but the magnitude of increase declined significantly under the influence of Zn treatment. Net photosynthetic rate, intercellular carbon dioxide concentration and stomatal conductance were highest at flowering stage in control and treated plants and decreased at post flowering stage. Contents of chlorophyll a, chlorophyll b, carotenoids, proteins and nitrate reductase activity in leaves increased from pre-flowering to maximum level at flowering stage and decreased thereafter in both control and treated plants. Presence of Zn in the soil drastically decreased/inhibited all the parameters, and the magnitude of decline increased with increasing Zn concentration. |

Effects of Cadmium, Lead, Mercury and Zinc on °-Aminolevulinic Acid Dehydratase Activity from Radish LeavesV.M. Morsch, M.R.C. Schetinger, A.F. Martins, J.B.T. RochaBiologia plantarum 45:85-89, 2002 | DOI: 10.1023/A:1015196423320 The purpose of the present study was to investigate the in vitro and the in vivo effects of cadmium, zinc, mercury and lead on δ-aminolevulinic acid dehydratase (ALA-D) activity from radish leaves. The in vivo effect of these metals on growth, DNA and protein content was also evaluated. The results demonstrated that among the elements studied Cd2+ presented the highest toxicity for radish. 50% inhibition of ALA-D activity (IC50) in vitro was at 0.39, 2.39, 2.29, and 1.38 mM Cd2+, Zn2+, Hg2+ and Pb2+, respectively. After in vivo exposure Cd2+, Zn2+, Hg2+ and Pb2+ inhibited ALA-D by about 40, 26, 34 and 15%, respectively. Growth was inhibited by about 40, 10, 25, and 5% by Cd2+, Zn2+, Hg2+, and Pb2+, respectively. DNA content was reduced about 35, 30, 20, and 10% for Cd2+, Zn2+, Hg2+, and Pb2+, respectively. The metal concentration in radish leaves exposed to Cd2+, Zn2+, Hg2+, and Pb2+ was 18, 13, 6, and 7 μmol g-1, respectively. The marked ability of radish to accumulate Cd2+ and Zn2+ raises the possibility of using this vegetable as a biomonitor of environmental contamination by these metals. |

Effects of NaCl and polyamines (PAs) on plasma membrane (PM) lipids, phospholipids, fatty acids, and free sterols were determined. NaCl treatment caused a decrease in total phospholipids, increase in saturated fatty acids and altered distribution of sterols and phospholipids. NaCl also induced increase in sterol/phospholipid ratio. PAs treatments (particularly Put and Spd) counterbalanced the NaCl deleterious effects on PM lipids. |

Changes in Nitrogen Metabolism of Vigna Radiata in Response to Elevated CO2A.C. Srivastava, M. Pal, U.K. SenguptaBiologia plantarum 45:395-399, 2002 | DOI: 10.1023/A:1016269717817 With the aim to determine the effects of CO2 on nitrogen metabolism mungbean (Vigna radiata) plants were grown from seedling emergence to maturity inside open top chambers under ambient CO2 (CA, 350 ± 25 µmol mol-1) and elevated CO2 (CE, 600 ± 50 µmol mol-1) concentrations at the Indian Agricultural Research Institute, New Delhi. Leaflet blades of the same physiological age were sampled at 20, 35 and 50 d after germination. Total nitrogen concentration in dry mass was consistently lower under CE than in CA. Non-protein nitrogen and protein nitrogen were also decreased under CE Total soluble protein content also decreased up to 35 d after germination under CE. However, a 27 % increase in protein content at 50 d after germination due to CE was observed. A significant decrease in total free amino acid under CE at 20 d after germination was observed. CE also brought about a remarkable decrease in the activity of nitrate reductase in leaves at 20 d after germination but increase at 35 d and 50 d after germination. Nitrogenase activity increased at all growth stages due to CE. Although total harvested leaves of CE plants accumulated more nitrogen, the relative amount of nitrogen on a percentage basis was low, probably due to a comparatively greater accumulation of sugars in the leaves of CE plants. |

Relative water content (RWC) of leaves of all cultivars declined with water stress, but in the two drought tolerant cultivars (UPASI-3 and UPASI-26), higher RWC were maintained in comparison to the others. Phenol content and activities of phenylalanineammonialyase, polyphenoloxidase and peroxidase initially increased, but decreased during extended drought. Chlorophyll contents decreased, whereas proline contents increased during water stress. SDS-PAGE analysis of proteins revealed increased accumulation of proteins of intermediate molecular masses (42 - 44 kDa) and low molecular masses (14 - 26 kDa). After 12 d of water stress, most of these proteins disappeared in T-78 and HV-39, but in the other cultivars they were still detectable. |

Alfa) seedlings treated with 150 mM NaCl was promoted in the light and suppressed in the dark. The light/dark changes of proline content was enhanced with each 12 h light/12 h dark cycle and the proline content increased steadily. Root and shoot concentrations of Na+ and Cl- in salt treated plants increased about 10 to 25 times as compared to the control. The content of these ions and the content of malondialdehyde were higher in the shoot of seedlings exposed to salt stress for 4 d in the light in comparison with the seedlings exposed to NaCl for 4 d in darkness. Light stimulated both ions and proline accumulation in the leaves and has no effect in the roots. Oxygen uptake was higher in the seedlings kept 4 d in the light which have higher endogenous free proline content. Chlorophyll fluorescence measurements showed that the photochemical activity of PS 2 slightly decreased as a result of salt stress and was not influenced by light regimes during plant growth. |

Laboratory experiments were conducted to determine the effect of salinity (0, 200, 400, 600, 800 and 1000 mM NaCl) and temperature (day/night: 5/15, 10/20, 15/25, 20/30, and 25/35°C) on seed germination. S. vermiculatus showed 100% germination in non-saline controls, at all thermoperiods. Percentage and rate of germination decreased with increases in salinity and few seeds germinated at even 1000 mM NaCl. High salinity exposure caused the loss of viability at higher temperature regimes, while some recovery was recorded in low salinity treatments. |

In this way a multicomponent system encompassing anthocyanins, proline, and peroxidase may act coordinately to overcome abiotic stress in cotton. |

The minimum range of variation (1.2-fold) is shown by Annonaceae and Sapindaceae while the maximum range (6.23-fold) is shown by Euphorbiaceae, and Fabaceae and Rutaceae show 1.7 and 1.9-fold differences, respectively. On an average, Euphorbiaceae and Rutaceae have been found to possess the largest and the smallest genome sizes respectively. |

Nevskii) tubers with 24-epibrassinolide (EB) resulted in prolonged deep dormancy, increased production of ethylene and higher contents of free and bound abscisic acid (ABA) in buds. EB at the most efficient concentration 0.021 mg dm-3, applied immediately after tuber harvest, inhibited sprouting by 36 - 38 d, increased ethylene formation after 1 and 7 d of storage by almost 300 and 150%, respectively, and increased the content of both free and bound ABA during the whole period of storage (on average by about 80%). Electron microscopic and morphometric studies showed that EB brings about a decrease in cell volume in tunica and all types of meristems and an increase in the number of vacuoles, accompanied by a decrease in their volume. |
Enhancement of Green Gram Nodulation and Growth by Bacillus SpeciesS.S. Sindhu, S.K. Gupta, S. Suneja, K.R. Seed inoculation with the rhizobacteria showed stunting effect on root growth whereas four Bacillus strains caused stimulation of shoot growth at both 4 and 7 d of observations. Coinoculation of some Bacillus strains with effective Bradyrhizobium strain S24 resulted in enhanced nodulation and plant growth of green gram. The shoot dry mass (ratio to uninoculated control) varied from 1.32 to 6.33 at day 30 and from 1.28 to 3.55 at day 40 of plant growth. Nodule promoting effect after 40 d of plant growth was observed with majority of Bacillus strains except for MRS9 and MRS26. Maximum gains in nodulation, nitrogenase activity and plant growth were observed with Bacillus strains MRS12, MRS18, MRS22 and MRS27 after 40 d of plant growth, suggesting the usefulness of introduced rhizobacteria in improving crop productivity. |
Anticytokinin Effect on Apical Dominance Release in in vitro Cultured Rosa Hybrida L.V. Kapchina-Toteva, M. Somleva, H.J. The compounds were applied at three different concentrations separately or in combination with benzyladenine. Cytokinin antagonists reduced the number of the sprouted buds in both cultivars at different extent. Their effect was strongly dependent on both concentration applied and culture period duration. The replacement of anticytokinins with benzyladenine in the medium resulted in overcoming of the bud break suppression. Both compounds significantly inhibited bud outgrowth as well. |
BTH-Induced Accumulation of Extracellular Proteins and Blackspot Disease in RoseY. Suo, D.W.M. LeungBiologia plantarum 45:273-279, 2002 | DOI: 10.1023/A:1015161110058 Treatment of rose shoots with 50 µM acibenzolar-S-methyl (BTH) resulted in increased protection against Diplocarpon rosae. This was accompanied by the induction and accumulation of a set of extracellular proteins as shown by SDS-PAGE and 2D-PAGE. Some of these proteins have been identified as PR-1, PR-2, PR-3 and PR-5 proteins by immunoblot analysis probed with tobacco antisera against PR-1c, PR-N, PR-Q and PR-S protein. Most of the extracellular proteins activated by BTH were also induced and found to accumulate in leaves upon infection with Diplocarpon rosae. However, their accumulation was much more pronounced in BTH-pretreated leaves than in water-pretreated leaves upon a challenge inoculation with D. rosae, particularly, the 15 kD PR-1, 36 and 37 kD PR-2 proteins. They may be more important in the expression of disease resistance. |
Germplasm Preservation of Wild Arachis Species through Culture of Shoot Apices and Axillary Buds from In Vitro PlantsR.F. Gagliardi, G.P. Pacheco, J.F.M. Valls, E. Nodal segments excised from in vitro-grown plants of A. retusa, A. macedoi and A. burchellii were used to examine the effects of explant position and age of the donor plant. Explants were excised from plants maintained in culture for 30, 60, 90 or 180 d, numbered I - V from top to bottom and cultured on MS medium supplemented with 2.7 µM NAA or different BAP concentrations (0, 4.4, 13.2 and 22 µM). The age of the donor plant has not influenced the responses of the four genotypes studied. In contrast, shoot regeneration ability was significantly affected by the original explant position, decreasing from top to bottom. In media supplemented with different BAP concentrations, multishoot formation was induced from apical segments at low frequencies (10 - 20%) and segments of all positions originated calluses at the explant basis after 30 d of culture. The culture of nodal segments in the presence of 2.7 µM NAA as the sole growth regulator is recommended for the multiplication of in vitro collections of wild groundnut species in order to avoid callusing and adventitious shoot formation. |
Variations in the Phenolic Contents of Cocoyam Clones in Correlation to Resistance to Pythium MyriotylumT.J.C. Temgo, O. All clones, infected or not, have phenolic compounds showing fungitoxic activity. The phenolic content of the tolerant and susceptible clones is less than that observed in the resistant one meanwhile in the resistant clone RO1075, a large increase in phenolic content is observed particularly in the roots during attack by pathogen. |

Number of plants regenerated per unit culture, formation of easily dissociating cell clusters and frequency of type I and type II calli were highest on NBKNB medium. Thus NBKNB medium is suitable for in vitro culture of even the hitherto recalcitrant indica genotypes. |

An optimized 2-D PAGE protocol was used and gave reproducible 2-D gels from leaf crude protein extracts with a high number of detected polypeptides. When applied for 24 h SA affected the expression of a number of soluble proteins. Most of them appeared to be down-regulated. Although no abundant expression of specific proteins was observed, we detected three polypeptides that were present only in SA-treated leaves. |

Millsp.] has been achieved using cotyledon segments of mature seeds as explants. A large number of globular somatic embryos were induced directly from cotyledons of genotypes T-15-15, GAUT-82-90 and GAUT-82-99 when cultured on EC6 basal medium supplemented with 2.22, 4.44, 13.32 or 22.2 μM N6-benzylaminopurine (BAP) and 0.45, 1.36, 2.27, 4.54 and 13.62 μM thidiazuron. Somatic embryos developed into cotyledonary stage when the globular embryos were transferred to Murashige and Skoog's (MS) basal medium containing 2.89 - 14.43 μM gibberellic acid. Maturation of somatic embryos was achieved on half strength MS medium with 0.38 μM abscisic acid.
