Cytosine Methylation Changes in Rice Centromeric and Telomeric Sequences Induced by Foreign DNA IntrogressionB. Liu, Z.L. Liu, Y.Z. Dong, J.H. Zhao, B.Q. Hunag, M.Y. HeBiologia plantarum 44:203-206, 2001 Cytosine methylation changes (hyper- or hypomethylation) in centromeric and telomeric sequences were observed in all three studied rice introgression lines containing DNA from wild rice, Zizania latifolia Griseb. The changed genomic Southern hybridization patterns were complex and non-concordant between a pair of isoschizomers (HpaII/MspI) digests, indicating methylation modifications at both the inner and outer cytosines of the CCGG sites. The changed patterns were inherited through generations. Possible mechanism for the methylation changes and their potential implications for the phenotypic variation and genome organization are discussed. |

Desirée callus, was studied. Like frost-tolerant species S. commersonii, cells of the frost-tolerant cell line contained starch grains in their plastids. The cells of this frost-tolerant line also possessed an increased number of microbodies containing protein crystals which suggests the involvement of proteins in frost tolerance but the mechanism may differ from that in frost-tolerant species. |

Salt Tolerance of In Vitro Established Salt-Tolerant Rice Plants during Further Growth in SoilY. Miki, M. Katoh, S. HisajimaBiologia plantarum 44:463-466, 2001 | DOI: 10.1023/A:1012452406918 In vitro salt tolerant rice plants established by step up treatment with 0.5, 1.0, 1.5 and 2.0 % NaCl at 3-week intervals were examined to determine whether they could grow in potted paddy soil containing 0, 0.55 or 0.75 % NaCl till harvesting. All the control plants were necrotic by the 4th week in the culture. At the 10th week of culture, 100 % of the salt-tolerant plants subjected to 0 or 0.55 % NaCl survived, and 78 % of the plants at 0.75 % NaCl. The Na+ and Cl- contents in the leaves of salt-tolerant plants grown at 0.55 and 0.75 % NaCl were about 4 times of those without NaCl. The ion contents in non-tolerant plants and seedling plants were 10 to 12 times of those in 0 % NaCl treatment. One of the hypotheses to explain the present data is that the in vitro step up salt selection induces the capability to maintain no lethal concentration of NaCl in the leaves. |

The Response of Nitrate Reductase Activity and Nitrate Assimilation in Maize Roots to Growth Regulators at Acidic pHN. Shankar, S.R. Khan, H.S. SrivastavaBiologia plantarum 44:599-601, 2001 | DOI: 10.1023/A:1013711224402 Nitrate and total nitrogen contents, and nitrate reductase (NR) activity of the excised maize roots in buffered or unbuffered nitrate solution (at pH 6.5 or 4.5) as affected by putrescine (PUT), abscisic acid (ABA) and salicylic acid (SA) were investigated. In unbufferred solution, the NR activity was lower at pH 4.5 as compared to that at pH 6.5, but in bufferred solution the activity was higher at lower pH. Supply of 100 µM PUT or 500 µM SA, promoted NR activity and 50 µM ABA inhibited the activity at pH 6.5. However, at pH 4.5, PUT and SA inhibited NR activity and ABA had no effect. In most cases, the increase in NR activity was positively correlated with total organic nitrogen and a negatively with nitrate content. A reverse situation was found when NR activity was inhibited by the growth regulators. |
Maternal Inheritance of Chloroplast DNA in Interspecific Crosses of BromusM. Pillay, K.C. ArmstrongBiologia plantarum 44:47-51, 2001 | DOI: 10.1023/A:1017914219789 Inheritance of chloroplast DNA (cpDNA) was examined in 41 F1 progeny obtained from the following interspecific Bromus crosses: Bromus arvensis (2n = 14) × B. inermis (2n = 4x = 28); B. arvensis × B. inermis (2n = 8x = 56); B. arvensis × B. erectus (2n = 6x = 42); B. arvensis × B. erectus (2n = 8x = 56); B. arvensis × B. erectus (2n = 10x = 70). Chloroplast DNA of the parental species was digested with BamHI, EcoRI and HindIII and species-specific restriction fragment length polymorphisms were identified by observation of ethidium bromide stained agarose gels as well as by filter hybridization experiments involving heterologous cloned barley cpDNA probes. The stability of these point mutations was verified by examining the cpDNA restriction patterns of at least 28 individual plants raised from seed of each of the parental species. No intraspecific cpDNA variability was detected. All the F1 progeny examined exhibited the cpDNA restriction fragment patterns of the female parent. There was no evidence of any paternal or biparental cpDNA inheritance. The results provided evidence for the uniparental-maternal inheritance of cpDNA in the Bromus crosses examined. |

Cr and Co reduced germination percentage only at the highest concentration used but markedly decreased radicle growth which might be attributed to depressive effect of Cr and Co on the activity of amylases and subsequent transport of sugars to the embryo axes. Protease activity, on the other hand, increased with the metal treatment. The highest concentration (10-2 M) tested of both metals was harmful on plant growth, while the low and moderate concentrations (10-6 and 10-4 M) enhanced the contents of chlorophylls and sugars, and activity of Hill reaction. Fresh mass of the produced pods increased at low and moderate concentrations of Cr and at Cr+Co treatment, but decreased in plants treated with Co. |
Effect of Sucrose Concentration, Charcoal, and Indole-3-Butyric Acid on Germination of Abies Numidica Somatic EmbryosB. Vooková, A. Kormu»ákBiologia plantarum 44:181-184, 2001 The dependency of radicle elongation in Abies numidica somatic embryos on germination media has been studied. No significant differences were detected between the Murashige and Skoog (MS) and Schenk and Hildebrandt (SH) medium. The addition of 10 g dm-3 activated charcoal or 0.05 mg dm-3 indole-3-butyric acid (IBA) into both media had positive influence on embryo germination. Difference between activated charcoal and IBA effects were significant. The high rooting percentage (85 %) was recorded on half SH medium with 10 g dm-3 sucrose and activated charcoal. After IBA addition rooting percentage was increased to 95 %. During 7 months 73 % of plantlets survived transfer to soil and in 54 % of plantlets shoot growth was observed. |
Counteraction of Salinity Stress on Wheat Plants by Grain Soaking in Ascorbic Acid, Thiamin or Sodium SalicylateA.M.A. Al-Hakimi, A.M. HamadaBiologia plantarum 44:253-261, 2001 The interactive effects of salinity stress (40, 80, 120 and 160 mM NaCl) and ascorbic acid (0.6 mM), thiamin (0.3 mM) or sodium salicylate (0.6 mM) were studied in wheat (Triticum aestivum L.). The contents of cellulose, lignin of either shoots or roots, pectin of root and soluble sugars of shoots were lowered with the rise of NaCl concentration. On the other hand, the contents of hemicellulose and soluble sugars of roots, starch and soluble proteins of shoots, proline of either shoots or roots, and amino acids of roots were raised. Also, increasing NaCl concentration in the culture media increased Na+ and Ca2+ accumulation and gradually lowered K+ and Mg2+ concentration in different organs of wheat plant. Grain soaking in ascorbic acid, thiamin or sodium salicylate could counteract the adverse effects of NaCl salinity on the seedlings of wheat plant by suppression of salt stress induced accumulation of proline. |

Responses of Penultimate and Flag Leaves of Wheat to Different Nitrogen SupplyV. Jain, S.K. Guru, Y.P. AbrolBiologia plantarum 44:373-378, 2001 | DOI: 10.1023/A:1012494726966 Uniculm wheat (Triticum aestivum L.) was grown to maturity at four concentrations of nitrogen corresponding to 3 (N1), 6 (N2), 9 (N3) and 12 (N4) g m-2. Penultimate and flag leaves were examined throughout the ontogeny. Sub-optimal concentrations of N resulted in sharp decline in both area and dry mass of the leaves. Decline in leaf area was due to fewer mesophyll cells. Net photosynthetic rate (PN) increased up to full expansion, remained constant for about a week and then declined. PN, nitrogen, ribulose-1,5-bis-phosphate carboxylase/oxygenase (RuBPCO) amount and activity, chlorophyll and soluble protein contents were similar at all the N concentrations. Both amount and activity of RuBPCO in the flag leaf were about two fold higher as compared to penultimate leaf, but PN was similar. This indicates the presence of an excess amount of RuBPCO in the flag leaf. |

Significant positive correlations were found between the amounts of chlorophyll a (Chl a), carotenoids (Car), Chl a/b ratio, and the normal proceeding of meiosis. High probability of paternal type of inheritance of specifities of meiosis of pollen mother cells and variation in photosynthetic pigments during plant transition from the stalk-shooting to heading stage were demonstrated. |

RodevaBiologia plantarum 44:573-578, 2001 | DOI: 10.1023/A:1013746804837 Localization and changes in the activity of β-glucosidase were investigated in wheat caryopsis and glumes infected with Stagonospora nodorum as well as in lily ovaries and harvested tomato fruits both inoculated with Botrytis cinerea. It was established that the pathogen invasion caused splitting of wheat seed coat, xylem blocking in lily carpel and decay in tomato fruits. B. cinerea invasion evoked disorders of the embryogenesis accompanied by a decreased activity of β-glucosidase in all ovules. The activity of the enzyme was not changed considerably in wheat seeds as the infection occurred in the late embryonal stages and the embryonal processes were not affected. In the seeds of harvested tomatoes distant from the invaded area the enzyme activity was not changed as well. |

Dissa by PEG-mediated pGL2 plasmid DNA uptake containing hygromycin phosphotransferase (hph) gene under the control of 35S transcript of CaMV. Hygromycin resistant colonies were obtained, when protoplasts were incubated with hygromycin B (25 μg cm-3) at 3 - 4 cell stage. Heat shock treatment, prior to polyethylene glycol (PEG) treatment, markedly increased the protoplast transformation frequency. The presence of active hph gene product, and integration of hph gene was confirmed by hygromycin assay and Southern blotting, respectively. Analysis indicated that rearrangement had possibly occurred during the integration of plasmid DNA into genomic DNA of barley callus. These transformed calli showed no morphogenic response onto the regeneration medium. |
Salt Tolerance in Aquatic Macrophytes: Ionic Relation and InteractionN.P. Rout, B.P. ShawBiologia plantarum 44:95-99, 2001 | DOI: 10.1023/A:1017978506585 Effects of seawater salinity (SWS) and pure NaCl on the intracellular contents of Na+, K+, Mg2+, Ca2+, chlorophylls (Chl) and carotenoids (Car) were studied in three submerged aquatic macrophytes, Hydrilla verticillata, Najas indica and Najas gramenia, which differed in their tolerance to salinity. NaCl resulted in significant increase in Chl/Car ratio in the salt-sensitive H. verticillata and moderately salt-tolerant N. indica, but not in the salt-tolerant N. gramenia. SWS treatment did not result in any significant change in the ratio. The intracellular content of Na+ increased significantly in all the test plants upon exposure to both NaCl and SWS. The content of K+ decreased significantly in these plants upon salinity treatment, except in N. gramenia. The contents of Ca2+ and Mg2+ decreased significantly upon NaCl treatment and remained unchanged or increased upon SWS treatment. No relationship between salt tolerance and K+/Na+ ratio was observed. The maintenance of a minimal level of K+ was observed to be the most probable requirement of salt tolerance in aquatic macrophytes. |
Insertion Profiles in Stomatal Density and Sizes in Nicotiana Tabacum L. PlantletsM. Voleníková, I. TicháBiologia plantarum 44:161-165, 2001 Tobacco (Nicotiana tabacum L. cv. Samsun) plantlets were cultured in vitro on Murashige-Skoog medium photoautotrophically (without sucrose) or photomixotrophically (with 3 % sucrose) under two irradiances [70 or 230 µmol m-2 s-1]. Significant differences in stomatal density and sizes in leaves of different insertion levels (3rd, 5th and 7th leaves from bottom) in photomixotrophic plantlets but not in photoautotrophic ones were found after 35 d of culture. Stomatal density was higher in upper leaves and on abaxial leaf side. Higher irradiance enhanced stomatal density in photoautotrophic plantlets. Stomatal sizes decreased with leaf insertion level but no significant differences between leaf sides were found. Abaxial stomata were more circular than the adaxial ones. In photomixotrophic plantlets stomata tended to be more elongated in the 3rd and the 5th leaves, whereas stomatal elongation in photoautotrophic plantlets was similar in all leaves. |

KruzmaneBiologia plantarum 44:233-237, 2001 Stimulation of ethylene biosynthesis in pine needles by hydrogen peroxide and sodium bisulfite coincided with the activation of ACC oxidase at the level of protein synthesis. Decrease in ethylene production at high concentrations of sodium bisulfite (above 7 mM) was apparently due to inhibition of ACC oxidase activity. Treatment of pine needles with aminotriazole caused an inhibition of both ethylene production and ACC oxidase activity. Both methylviologen and methyl jasmonate stimulated ACC oxidase activity in a concentration-dependent manner with no parallel changes in ethylene production. The presented results suggest that ACC oxidase plays an important role in regulation of ethylene formation in pine needles in response to different stimuli. |

Protein and RNA contents increased gradually in the initial stages and rapidly in the final stages of storage period. SDS-PAGE analysis of storage proteins revealed synthesis of 56, 52 and 47 kDa proteins during the later stages of storage with concomitant disintegration of 23 and 18 kDa proteins. |
Timing of Transposition of Ac Mobile Element in PotatoD. Pavingerová, J. Bříza, H. NiedermeierováBiologia plantarum 44:347-353, 2001 | DOI: 10.1023/A:1012478323331 The timing of excision of maize transposable element Ac was studied using visual histochemical assay based on Ac excision restoring activity of β-glucuronidase (GUS). The Solanum tuberosum L. cv. Bintje was used for Agrobacterium-mediated transformation with pTT230 plasmid harbouring Ac-interrupted gus A gene and npt II gene as a selectable marker gene. Twenty-eight out of 72 kanamycin resistant calli did not express any GUS activity, 31 calli showed partial GUS expression and 13 out of assayed calli revealed strong expression of gus A gene. Plants were regenerated from calli without and/or with partial expression of gus A gene. The regenerated transformants which did not express GUS during the callus phase often contained many small GUS expressing spots on leaves. A phenotypic selection assay for excision of Ac has been also used. This non-detectable excision of Ac in callus tissue could be followed by a "late" timing excision during leaf development. After transformation with pTT224 plasmid harbouring Ac-interrupted hpt II gene and npt II gene transgenic calli containing Ac within the hygromycin resistance gene were derived and hygromycin sensitive plants were regenerated from them. Protoplasts isolated from leaves of transgenic regenerated plants were selected on hygromycin. Hygromycin resistant minicalli showed to harbour multiple copies of Ac and mark out low uniqueness of integration sites. |

This suggests a prevailing shade aspect, regardless of collection sites and cultivated or wild origin. |
The Complexity of Factors Driving Volatile Organic Compound Emissions by PlantsJ. Peñuelas, J. LlusiàBiologia plantarum 44:481-487, 2001 | DOI: 10.1023/A:1013797129428 The emissions of volatile organic compounds, VOC, from plants have strong relevance for plant physiology, plant ecology, and atmospheric chemistry. We report here on the current knowledge of the many internal (genetic and biochemical) and external (abiotic - temperature, light, water availability, wind, ozone, and biotic - animal, plant and microorganisms interactions) factors that control emission rates of different VOC by altering their synthesis, vapour pressure or diffusion to the atmosphere. The complex net of these factors, their interactions and the different responses of the different VOC produces the large qualitative and quantitative, spatial and temporal variability of emissions and the frequent deviations from current standard emission models. The need for a co-operative multidisciplinary multiscale research to disentangle this complex and important issue of plant VOC emissions is reminded. |
Alleviation of Changes in Protein Metabolism in NaCl-Stressed Wheat Seedlings by ThiamineF. El-Shintinawy, M.N. El-ShourbagyBiologia plantarum 44:541-545, 2001 | DOI: 10.1023/A:1013738603020 NaCl-stress induced a pronounced suppression in growth of wheat seedlings. The most abundant amino acids (cysteine, arginine, methionine) constituting about 55 % of total free amino acid content in control wheat were reduced in 100 mM NaCl-treated plants. However, valine, isoleucine, aspartic acid and proline accumulated in response to NaCl stress and NaCl-treated wheat seedlings showed 1.6 fold increase in total free amino acids compared to the control. Addition of 2 [micro ]M thiamine alleviated the effects of NaCl on the amino acid composition and the amount of total free amino acids decreased to that in the control. Content of 26 kDa protein increased in NaCl-treated plants, stimulation was more pronounced in roots than in shoots. In contrast, the contents of 13 and 20 kDa proteins decreased. After addition of thiamine, the 24 kDa protein, which disappeared with NaCl treatment, has been initiated again. Moreover, thiamine treatment stimulated the accumulation of the 20 kDa protein. |

Effect of Benzylaminopurine on Rehydration of Bean Plants after Water StressJ. Rulcová, J. PospíąilováBiologia plantarum 44:75-81, 2001 | DOI: 10.1023/A:1017922421606 The possibility of improving the recovery of plant photosynthesis after water stress by cytokinin-induced stimulation of stomatal opening or delay of leaf senescence was tested. The 6-benzylaminopurine (BAP) in concentrations 1 and 10 μM was applied to the substrate (sand + nutrient solution) or sprayed on primary leaves of 14-d-old Phaseolus vulgaris L. plants sufficiently supplied with water or water-stressed for 4 d. The later ones having relative water content decreased to 69 % were fully rehydrated during the following three days. Parameters of photosynthesis and water relations were measured in primary leaves of 7-, 10-, 14-, and 17-d-old plants. Application of 1 μM BAP slightly delayed leaf senescence: in 17-d-old control plants, net photosynthetic rate (PN) and chlorophyll (Chl) content, and when sprayed on leaves also some of Chl a fluorescence kinetic parameters of BAP-treated leaves were slightly higher than those of untreated leaves. Both types of application of 1 μM BAP slightly improved recovery of plants during rehydration after water stress in terms of increased gad, gab and PN, i.e., parameters which were markedly decreased by mild water stress. However, contents of Chl a, Chl b and carotenoids and parameters of Chl a fluorescence kinetic were not markedly affected by mild water stress and after rehydration were not stimulated by 1 μM BAP. 10 μM BAP had mostly negative effects on the parameters measured. |

AA treatment resulted in a higher accumulation of proline at all the stages of growth. Both BA and AA enhanced PN, Chl content and NR activity, and ameliorate the negative effect of water stress. |

The plants (F2) were selected for the analysis at seedling stage and were selfed to obtain F3 generation. The analysis covered about 15 enzymes controlled by about 30 loci. The seeds of F3 family planted in the greenhouse for measuring some quantitative traits, recorded tasselling and silking during vegetation. At the end of vegetation grain yield, and some other quantitative traits of grain in F3 family were assessed. The relationship between marker loci and the loci for quantitative traits (QTLs) were estimated by computerized statistical method. |
Stimulation of Growth and Nutrient Uptake by VAM Fungi in Brassica Oleracea Var. CapitataR. Nelson, P.N. AcharBiologia plantarum 44:277-281, 2001 Cabbage (Brassica oleracea, var. capitata, cv. Hercules) seedlings were inoculated with vesicular-arbuscular mycorrhizal (VAM) fungi Glomus fasciculatum, G. aggregatum, and G. mosseae. Differential efficiency in mycorrhizal colonization and the specificity of fungal symbiont to stimulate the growth and nutrient uptake of the host were observed. In addition, there was an increase in phenol, protein, reducing sugar contents, and peroxidase activity in the VAM inoculated seedlings.
