Mulberry Leaf Metabolism under High Temperature Stress

Effects of high temperature on the activity of photosynthetic enzymes and leaf proteins were studied in mulberry (Morus alba L. cv. BC2-59). A series of experiments were conducted at regular intervals (120, 240 and 360 min) to characterize changes in activities of ribulose-1,5-bisphosphate carboxylase (RuBPC) and sucrose phosphate synthase (SPS), photosystem 2 (PS 2) activity, chlorophyll (Chl), carotenoid (Car), starch, sucrose (Suc), amino acid, free proline, protein and nucleic acid contents in leaves under high temperature (40 °C) treatments. High temperature markedly reduced the activities of RuBPC and SPS in leaf extracts. Chl content and PS 2 activity in isolated chloroplasts were also affected by high temperature, particularly over 360 min treatment. Increased leaf temperature affected sugar metabolism through reductions in leaf starch content and sucrose-starch balance. While total soluble protein content decreased under heat, total amino acid content increased. Proline accumulation (1.5-fold) was noticed in high temperature-stressed leaves. A reduction in the contents of foliar nitrogen and nucleic acids (DNA and RNA) was also noticed. SDS-PAGE protein profile showed few additional proteins (68 and 85 kDa) in mulberry plants under heat stress compared to control plants. Our results clearly suggest that mulberry plants are very sensitive to high temperature with particular reference to the photosynthetic carbon metabolism.

Activities of Ribulose-1,5-Bisphosphate Carboxylase/Oxygenase and Phosphoenolpyruvate Carboxylase, and Oxygen Evolution in Transgenic Tobacco Plants

Three clones of tobacco transformed with the T-DNA of Agrobacterium rhizogenes Ri plasmid A4b cultivated in vitro on a solid agar medium neither showed pronounced morphological diversity nor significantly differed in chlorophyll (Chl) contents from control plants. However, the transformation caused a 27 to 83 % decay in leaf oxygen evolution and in both ribulose-1,5-bisphosphate carboxylase (RuBPC) and phosphoenolpyruvate carboxylase (PEPC) activities. Therefore, the transformation brought about a reduction of active PEPC as well as activable RuBPC amounts in plant tissues. Individual clones did not mutually differ. In tobacco transformed with A. rhizogenes strain TR101 and grown in soil only the mean leaf area tended to reduce. Chl contents, Chl a/b ratio, oxygen evolution, and activities of both RuBPC and PEPC were insignificantly affected by the transformation.

Factors Affecting Somatic Embryogenesis from Cotyledonary Explants of Safflower

Frequency of safflower (Carthamus tinctorius L.) somatic embryogenesis, number of somatic embryos per responding explant and somatic embryo maturation and germination were affected by genotype, explant age, carbon source, and ethylene. Among 8 cultivars tested, 7 were embryogenic with varying frequencies. The best response was obtained with cv. Girna. Whole cotyledonary explant from 10-d-old plants was best responding compared to 5- or 15-d-old ones. Among different carbon sources, sucrose at 87.6 mM concentration was most suitable for embryo induction, maturation and germination. Of the different ethylene inhibitors, silver nitrate at 50 [micro ]M concentration significantly increased the embryogenic frequency and also the number of embryos per responding explant. Silver nitrate has pronounced effect on embryo maturation but had no effect on germination.

Hop Latent Viroid (HLVd)-Caused Pathogenesis: Effects of HLVd Infection on Lupulin Composition of Meristem Culture-Derived Humulus lupulus

Season-dependent fluctuation of hop latent viroid in meristem tips enabled selection of viroid-free cultures from Osvald's clone 31, 72, 114, and cv. Premiant. These mericlones were used to evaluate effect of viroid infection on the composition of hop resins and essential oils in the first production year. Healthy plants were compared with naturally re-infected individuals under field conditions. On average, viroid infection decreased content of α-bitter acids by 40 %. The content of β-bitter acids, as well as the ratios of humulone/cohumulone and lupulone/colupulone was not influenced by viroid infection. The content of all monoterpenes was for 29, 37.4 and 41.6 % higher for myrcene, α- and β-pinene, respectively, in infected plants compared with the healthy controls. The contents of sesquiterpenes like β-caryophyllen α-humulene, α-copaene, γ-muurolene, β-bisabolene, γ-cadinene, and δ-cadinene decreased by 13.7, 13, 14, 18.5, 29, 21.7 and 18.5 %, respectively, due to viroid infection. The possible influence of some oxidative-reduction processes activated by viroid-caused pathogenesis was assumed to be involved in the accumulation of terpenes alcohols like geraniol and methylgeranate, and in the reduction of the contents of the majority of ketones detected in the spectra of essential oils.

Ribulose-1,5-Bisphosphate Carboxylase/Oxygenase Content and Activity in Wheat, Rye and Triticale

Photosynthetic parameters were measured in triticale and its parents wheat and rye. Soluble protein content in leaves, ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) content per fresh mass, total chlorophyll content, biomass yield, leaf area, leaf mass and specific leaf mass were higher but Rubisco content expressed as percentage of soluble protein, carboxylase activity, photosynthetic rate and stomatal conductance were significantly lower in rye than in wheat. Native-PAGE of Rubisco revealed that rye carboxylase was different from that of wheat. The difference was not related to either the small or large subunit of Rubisco but, may be, to the ionic and/or other properties of the Rubisco protein moiety. Triticale Rubisco was similar to wheat. For most of the studied physiological parameters, triticale showed much more similarity with wheat than with rye.

Response of Zea mays to the Inoculation with Azospirillum on Nitrogen Metabolism under Greenhouse Conditions

The maize (Zea mays L.) plants inoculated by N2-fixing bacterium Azospirillum showed increased activity of glutamate dehydrogenase (GDH) and glutamine synthetase (GS) in root cells free extracts over uninoculated control plants. Maximum differences in NADH-GDH activity were observed during the second and third weeks after sowing. The specific activity of GS showed a greater increase at the end of the assay. The percentage of nitrogen in leaves, root and foliage length, total fresh mass and nitrogenase activity were higher in inoculated plants than in the control ones.

Effect of Benzyladenine and Indolebutyric Acid on Ultrastructure, Glands Formation, and Essential Oil Accumulation in Lavandula Dentata Plantlets

Lavandin (Lavandula dentata) axillary buds were grown in Linsmaier-Skoog (LS) medium solidified with 10 % bactoagar (control) and supplemented with 0.1 mg dm-3 benzyladenine (BA), 0.1 mg dm-3 indolebutyric acid (IBA) or both plant growth regulators. In the studied conditions the axillary buds developed into plantlets. The addition of BA inhibited the formation of glands by 44 % as compared with the control plantlets and also inhibited their development: these plantlets had the highest number of unbroken glands (in pre-secretory state) when compared with plantlets grown in the other conditions. The presence of BA stimulated chloroplast formation, and increased the content of essential oils by 150 % with respect to the control plantlets. It also increased their secretion, and the number of lipid droplets in the chloroplasts, cytosol and plasmalemma. On the contrary, the presence of IBA decreased the essential oil concentration in plantlets by 31 % when compared with the control ones and inhibited their secretion capacity.

Determination of Frost Tolerance in Winter Wheat and Barley at the Seedling Stage

Detailed studies were made on changes in the quantity of 1.4 MDa rRNA precursor in barley and wheat cultivars with different degrees of frost tolerance. When analysing genotypes with different LT50 values a close negative correlation was found between the quantity of 1.4 MDa molecular mass rRNA precursor and the forst tolerance of the given barley or wheat cultivar. The results suggest that a technique based on low temperature phosphorus incubation at the seedling stage could be suitable for the selection of genotypes on the basis of this character in applied research.

Shoot Regeneration from Immature Cotyledons of Cicer Arietinum

Shoot regeneration was achieved from immature cotyledons of five chickpea (Cicer arietinum L.) genotypes: C235, ICC4971, ICC11531, ICC12257 and ICC12873. The cotyledons cultured on Murashige and Skoog (MS) medium supplemented with 3 or 5 mg dm-3 zeatin with or without 0.04 mg dm-3 indole acetic acid (IAA) showed formation of cotyledon like structures (CLS) at their proximal ends. Subsequently, shoot regeneration took place in some of the CLS forming explants. CLS were also formed in cotyledons cultured on MS + 0.2 - 1 mg dm-3 thidiazuron (TDZ); direct shoot regeneration was observed in cotyledons cultured on 1 mg dm-3 TDZ. The shoot buds elongated on media containing indole butyric acid (IBA), benzylaminopurine (BAP) and gibberellic acid (GA3). Complete plantlets were obtained by rooting of shoots following pulse treatment with 200 mg dm-3 IBA for 5 min and culture on growth regulator free half-strength MS medium.

Phytotoxic Effects of Cd, Zn, Pb, Cu and Fe on Sinapis Alba L. Seedlings and their Accumulation in Roots and Shoots

The inhibitory effects of Cd, Cu, Zn, Pb, and Fe on root elongation, contents of photosynthetic pigments, and metal accumulation in the roots and shoots of Sinapis alba were assessed. On the basis of growth inhibition metals can be arranged in a order Cu > Cd > Fe = Zn > Pb. All the metals, except Fe, were accumulated in significantly higher amount in the roots than in the shoots. Cd, Zn, Cu and Pb reduced chlorophyll a, and especially chlorophyll b content, and Zn and Pb reduced the carotenoid content, but less than that of chlorophyll a+b. The plants contained the highest concentration of Cd, and the lowest concentration of Zn.

Spermine as compared to putrescine or spermidine retarded the loss of chlorophyll and protein contents to a greater extent in wheat primary leaves during dark incubation. Activities of whole chain electron transport, photosystem (PS) 1 and PS2, and absorbed excitation energy distribution in favour of PS 1 were protected by these amines in valency dependent manner during 72-h dark incubation.

Stimulation of Shoot Regeneration on Linum Hypocotyl Segments by Thidiazuron and Its Response to Light and Calcium

The basal segment from hypocotyl of Linum usitatissimum L. seedling readily regenerates, to produce a large number of shoots, in a short period of 5 [ndash ] 7 d. This response was stimulated by a low concentration (0.1 [micro ]M) of thidiazuron (TDZ). TDZ was also effective in inducing regeneration in dark. A drastic reduction in regeneration response on hormone-free as well as TDZ-supplemented medium was found after inclusion of an inhibitor of calcium-uptake, lanthanum (La3+). An essentiality of calcium in the regeneration was also evidenced by an increased response with increasing concentration of calcium. At Ca2+ concentration insufficient for regeneration, inclusion of TDZ resulted in shoot formation.

The final biological effect of auxin depends both on free auxin levels and on auxin perception capacity.RolB transformedBeta vulgaris L. hairy roots provide a system for studying both factors. Highly purified plasma membrane fractions were prepared with aqueous two-phase partitioning. Individual hairy root clones were assessed for the binding activities of plasma membrane-bound auxin binding proteins and for their free intracellular indole-3-acetic acid levels. The presence of a high affinity auxin binding protein with a dissociation constant of 9.07 x 10-7 M was detected in the plasma membrane fractions isolated from non-transformed seedling roots and the six clones ofrolB transformed hairy roots. However, the levels of specific IAA binding considerably varied among different hairy root clones and between transformed and non-transformed roots. The levels of the detectable polypeptide in immunoblotting with an antibody against maize 22-kD auxin binding protein subunit were in good agreement to the levels that were detected in auxin binding assays. Differences in the indole-3-acetic acid levels were found between transformed and non-transformed roots and also between different transformed hairy root clones. A negative correlation was observed between free intracellular IAA levels and its specific binding to the plasma membrane-bound auxin binding proteins. A latency study indicated that the binding site for auxin may be located on the exterior face of the plasma membrane

Effects of ferulic acid on L-malate oxidation in isolated soybean mitochondria

The effects of ferulic acid on L-malate oxidation in mitochondria isolated from soybean (Glycine max L.) seedlings were investigated. Oxygen uptake and the products of L-malate oxidation were measured under two conditions: pH 6.8 and 7.8. At acidic pH, the activity of the NAD+-linked malic enzyme (L-malate:NAD+oxidoreductase [decarboxylating] EC 1.1.1.39) was favoured, whereas at alkaline pH a predominance of the L-malate dehydrogenase activity (L-malate:NAD+oxidoreductase EC 1.1.1.37) was apparent. Ferulic acid inhibited basal and coupled respiration during L-malate oxidation either at acidic or alkaline pH, reducing also the amounts of pyruvate or oxaloacetate produced. The results suggest that the site of ferulic acid action is situated at some step that precedes the respiratory chain. An interference with the L-malate entry into the mitochondria could be an explanation for the effects of ferulic acid, but the possibility of a direct inhibition of both enzymes involved in L-malate oxidation cannot be ruled out.

The detrimental effect of NaCl was found at the oxidation site of photosystem 2 (PS2). An impairment of PS2 was caused by damage of the oxygen evolving polypeptides (33, 23 and 20 kDa) of thylakoid membranes as well as by changes in the unsaturated and saturated fatty acids. Application of triacontanol, TRIA (1 mg kg-1) ameliorated the effect of NaCl and promoted the ratio of unsaturated to saturated fatty acids, the rate of14CO2 fixation and the activity of ribulose 1,5-bis-phosphate carboxylase.

The Ecological Significance of Allelopathy in the Community Organization of Alhagi Graecorum Boiss

Field study revealed that species diversity index of the community was 0.597. The data of interspecific association index of Alhagi graecorum and each of its associates, Chenopodium murale, Glinus lotoides and Malva parviflora, were 0.35, 0.41 and 0.33, respectively. Therefore, a net negative association and low diversity among the species were the main characters of the community. Laboratory experiments verified the role played by competition and allelopathy in this community organization. The bioassay results indicated that A. graecorum exhibited phytotoxic activity on the tested species. Accordingly, water-borne allelopathic compounds may be released from A. graecorum into the environment and suppress the growth of its associates where they were not able to compete with A. graecorum in the field, so the latter species become dominant.
