Biointeraction between precarthamin and cell components in florets ofCarthamus tinctorius L.K. SaitoBiologia plantarum 36:397, 1994 | DOI: 10.1007/BF02920938 The relative rate of precarthamin extractability and floret protein inactivation by organic solvents were compared. Upon trituration of the floral tissues ofCarthamus tinctorius L., usually less extractable precarthamin is released at a markedly high level, while the releasability rate is changed conspicuously by macerating the florets in the test solvents of increased concentrations; the amount of releasable precarthamin also varies according to the solvents used. 30% (v/v) acetone promotes the pigment solubility about twice, whereas, its capacity decreases abruptly in reverse proportion to the increment of acetone content. Methanol accelerates the precarthamin release, but the rate is very low. Ethanol acts in far lesser extent. The data indicate that these varied aspects are more or less attributed to the inactivation of possible factors dissociating precarthamin from cellular components in freezed safflower florets. |

Electrolyte concentration inF. trinervia in control and salt treated plants is lower than inF. pringlei. Fluorescence data do not indicate a damage of PS 2 charge separation in both species. Whether the C4 photosynthetic pathway inF. trinervia is responsible for the improved salt tolerance compared toF. pringlei remains an open question. |

Five days after sowing, the plants were exposed to non-inductive (15 long days-LD) or inductive (6 short days-SD + 9 LD) photoperiodic conditions. The length of hypocotyl and cotyledon blade were measured and percentage of flowering was scored. Gibberellic acid (GA3) stimulated hypocotyl growth of green and photobleached plants under SD and inhibited under LD conditions. Indole-3-acetic acid (IAA) slightly stimulated hypocotyl growth of green plants only under LD conditions. Benzylaminopurine (BAP) inhibited hypocotyl growth regardless of photoperiodic regime. The optimal concentration of glucose or saccharose for flowering in green and SANDOZ-treated plants was 5%. In green SAN 9785-treated plants exogenous saccharides compensated lack of photosynthates to bring about full flowering, but SAN 9789-treated plants needed in addition GA3. |

The leaves of all the seven tree species showed significantly higher PN during summer than in winter and these rates differed from one species to the other. A positive correlation was found between PN of different tree species and their Chl content or biomass production. There was no significant correlation between ribulose-1,5-bisphosphate carboxylase activity and PN when these were expressed on leaf area basis. |

PlevkováBiologia plantarum 36:65, 1994 | DOI: 10.1007/BF02921271 Glyphosate tolerant cell lines were selected from highly embryogenic cell suspension culture ofMedicago sativa L. Resistant cell lines showed significant reduction of embryogenic ability and during long-term culture in the presence of glyphosate gradual loss of this ability was observed. After glyphosate treatment the increased activity of 5-enolpyruvylshikimate-3-phosphate synthase in tolerant cell lines overcame the block in aromatic amino acid synthesis which was observed in control cell lines. Glyphosate caused marked increase in the content of shikimic acid in both control and tolerant cell lines but the accumulation of shikimic acid was considerably lower in tolerant calli. Significant qualitative and quantitative differences were found in the content of individual phenolic acids. The considerable decrease in the amount of cinnamic acid derivates and broader spectrum of hydroxybenzoic acids suggest in tolerant cell lines the activation of alternative pathway not regulated by phenylalanine ammonia lyase. The possible role of altered pool of phenolic acids on the embryogenic ability is discussed. |

At low temperature treated cells, the superoxide dismutase, catalase and glutathione peroxidase activities were significantly higher and the protein content lower than in high temperature treated cells. The increase of hydroxyl free radical level and malonyldialdehyde formation, when algal cells were exposed to low temperature, were due to the stimulated production of superoxide radicals O2 - and hydrogen peroxide (H2O2). |
Changes in cell membrane permeability and lipid content of wheat root cortex cells induced by NaClM. M. F. Cell membrane permeability to nonelectrolytes and water was determined by the plasmometric method for individual intact cells. NaCl increased membrane permeability to urea, methylurea and ethylurea and decreased permeability to water. Membrane lipid partiality was decreased by NaCl. The effects of NaCl on cell permeability parallel changes in the lipid composition of the plasma membranes induced by NaCl stress suggesting that nonelectrolyte permeability is a useful tool to probe alterations in the lipid matrix of the membrane. |

JoshiBiologia plantarum 37:225-232, 1995 | DOI: 10.1007/BF02913218 The ontogenetic changes in growth, and the diurnal changes in net photosynthetic rate (PN) and stomatal conductance were studied in two peanut cultivars of different habit groups. Significant cultivar differences were noticed: the prostrate cv. M 13 was found superior to the erect cv. J 11 in all the parameters studied. Specific leaf mass and the rates of gross photosynthesis and respiration were higher in cv. M 13 than in cv. J 11. In vegetative phase, the maximum PN was in cv. J 11, but in pod filling phase, it was in cv. M 13. The differences in growth and PN of the cultivars were significant after the onset of reproductive sink. Therefore, the screening for higher PN has to be made at the pod-filling phase, and between 09.00 and 10.00 of the day (at optimum temperature). |
Detection of DNA polymorphism among pea cultivars using RAPD techniqueP. Samec, V. NašinecBiologia plantarum 37:321-327, 1995 | DOI: 10.1007/BF02913234 An influence of some Random Amplified Polymorphic DNA (RAPD) reaction factors on resulting banding pattern and the ability of RAPD technique to detect DNA polymorphism among six economically important pea cultivars was tested. Relatively high level of DNA polymorphism among peas was observed, using polyacrylamide/urea gels and silver staining. Altogether 13 arbitrarily designed primers produced 313 amplification products. In addition 59 polymorphisms were found. These polymorphisms can serve as potential genetic markers. RAPD data were processed using cluster analysis and plotted as dendrogram. Each tested cultivar was clearly distinguished from the others. Moreover,Pisum sativum andP. sativum subsp.arvense cultivars were separated into 2 different clusters, according to their systematic relationships. |

Belinka) were studied by electrophoresis in polyacrylamide gel for estimating ontogenetic variability of gene expression. Formation of individual isoesterases depended on the type of tissue and the stage of its development. Isoesterases characteristic of exclusively one or some tissues of the same developmental stage were revealed simultaneously with basic esterase isoforms active in all analysed parts of seeds, seedlings and field-grown plants. The revealed changes of esterase isoenzyme spectrum during germination show tissue and time specificity of the endogenous regulation of genes controlling their formation. |

Wisconsin No. 38 cells grown in suspension culture. The amount of the binding site detected in both these fractions changed during the culture cycle according to a strict pattern. The molecular mass of the binding site was estimated by gel filtration to be approximately 175 000 and it appears to be a protein. When partially purified by affinity chromatography and allowed to pre-incubate with IAA, the site had a significant stimulatory effect on total RNA synthesis, as measured by a cell-free assay system. Unpurified extracts had no such effects. The system behaves rather like the steroid hormone-receptor system in animals. |

Phase setting of the rhythm and length of its period were not affected, its amplitude decreased or changed insignificantly depending on the time and the site of IAA application. The intervals of significant inhibitory effect of IAA applied to apical buds terminated later than after IAA application to photoreceptive organs. The inhibitory effect did not correlate with the level of IAA uptake. Results obtained support the hypothesis that both photoreceptive organs and apical buds are the sites of inhibitory effect of applied IAA. |

The main fungal inhabitants on the leaves or shoot surface of the test plants wereAspergillus niger, Penicillium chrysogenum andCladosporium herbarum. The qualitative analysis of the fungal species associating the parasite, the hosts and the non-infected plants showed similar pattern. However, the total population exhibited quantitative differences coupled with the amount and the chemical composition of the exudates on plant surface and the quantity of transpired water. The fungal catch from the aerial shoot (inflorescence) of the parasite was higher than that collected from either the leaves or aerial shoots of non-infected or host plants. The fungal density on the leaves ofL. delicatulum was higher than those isolated from the aerial shoots ofA. glaucum. Infection byC. coccineum caused a marked drop in the total fungal population on leaves or shoot surfaces of the hosts as compared to the corresponding non-infected individuals. The stimulative effect of washings on spore germination of some isolated fungal species was matched with the density of fungi on the target plants. |

Chl stability to heat significantly increased with increased soil acidity. The Chla/b ratio was increased significantly at pH 5 and 4 and decreased at pH 3 and 2. SprayingSorghum shoots with kinetin solutions counteracted the above adverse effects on Chl content and dry matter accumulation. Kinetin-treated plants showed a lower transpiration coefficient than the untreated plants. |
Organic substances responsible for salt tolerance inEruca sativaM. AshrafBiologia plantarum 36:255-259, 1994 Responses of a salt tolerant and a normal population of an oilseed crop,Eruca sativa Mill. were assessed after four weeks growth in sand culture salinized with 0 (control), 100, 200, or 300 mol m-3 NaCl. The salt tolerant plants produced significantly greater dry biomass than the normal population. The populations did not differ significantly in leaf osmotic potential, relative water content and leaf soluble proteins. However, the tolerant population accumulated significantly greater amounts of soluble sugars, proline and free amino acids in the leaves compared with the non-tolerant population. It is established that leaf soluble sugars, proline, and free amino acids are important components of salt tolerance inEruca sativa. |
Somatic embryogenesis inCicer arietinum L: Influence of genotype and auxinsS. Eapen, L. GeorgeBiologia plantarum 36:343, 1994 | DOI: 10.1007/BF02920928 Plant regeneration through somatic embryogenesis was obtained in chickpea (Cicer arietinum L.) using immature cotyledons and immature embryonal axes as explants. 2,4-dichlorophenoxyacetic acid (2,4-D), 4-amino-3.5-6-trichloropicolinic acid (picloram) and 3.6-dichloro-0-anisc, acid (dicamba) in concentrations 1, 2, 5, 10 mg dm-3 were found better than naphthaleneacetic acid (NAA) (10-20 mg dm-3) for the induction of globular and heart-shaped somatic embryos. The embryos developed upto the dicotyledonary stage on medium supplemented with saccharose, mannitol and silver nitrate (AgNO3) and developed further into plantlets on medium containing gibberellic acid (GA3) and abscisic acid (ABA). The frequency of somatic embryogenesis was dependent on the genotype and auxins used. |
Changes in growth, osmotic potential and cell permeability of wheat cultivars under salt stressM. M. F. Salt stress decreased growth of shoots and roots of both cultivars; fresh mass of sensitive cultivar being more affected. NaCl increased membrane permeability to urea, methylurea and ethylurea and decreased membrane partiality in root cortex cells of sensitive cultivar. Neither parameter changed by NaCl in resistant cultivar. NaCl treatment decreased water permeability and osmotic potential in both cultivars; sensitive cultivar was more affected. The results extends our previous data that cell membrane properties are different in salt sensitive and resistant genotypes and so cell permeability could be a potential trait indicating salt tolerance. |

However, under combination of NaCl with Ca2+ ions, these parameters generally raised. Contents of soluble saccharides, proline and quaternary ammonium compounds increased with increasing NaCl concentration, but under addition of CaCl2 or CaSO4, contents of these compounds were decreased. Low concentrations of NaCl stimulated soluble proteins, production, but higher concentrations decreased the content of soluble proteins. Addition of Ca2+ in the media did not improve the soluble protein production. Insoluble proteins content was increased with the rise of salinity level, but these effects were more pronounced with NaCl and CaCl2 or CaSO4 than with NaCl only. |
