Population differentiation inSpartina patens: Water potential components and bulk modulus of elasticity

Pressure-volume technique was utilized to evaluate salinity response among three populations ofSpartina patens (Ait.) Muhl. from Louisiana Gulf coast marshes. Plants were subjected to salinities of 85 and 425 mol m-3 for 77 d in a greenhouse. Ψw and Ψπ decreased in all populations in response to increases in salinity. There were 32% decrease in Ψsat, 42% decrease in Ψtlp in response to salinity changes from 85 to 425 mol m-3 in the Ferblanc population. Similarly, there were 35% and 41% decrease in Ψsat in the Clovelly and Lake Tambour populations, respectively. All populations showed the ability to adapt to the increased salinity as was evidenced by osmotic adjustment. However, the Lake Tambour population appeared to have superior ability to adapt to high salinity through having a significantly lower osmotic potential at saturation (Ψsat), osmotic potential at turgor loss point (Ψtlp), and maximum turgor potential (ΨP(max)) compared to other populations. Ferblanc and Clovelly populations revealed the ability to adapt to saline environments to a lesser extent as compared to the Lake Tambour population. Results indicate that there is a potential for selection of superior strains ofSpartina patens for use in marsh restoration projects aiming at prevention of wetland loss in certain coastal areas.

Transport of14C-IAA from leaves and shoots to different fruit parts

Transport of14C-IAA was studied in apple spurs of a 20-year-old McIntosh with one fruit and one shoot. Water solutions of IAA were applied to intact, pricked or scratched leaf blades, to decapitated shoots or to petioles (leaf-blade removed) at the end of June, July and August.14C-IAA (in an unknown form) was transported from intact leaves and shoots to pedicel, pericarp and seeds. Radioactivity of the pedicels increased every month while that of seeds reached maximum at the end of July and then markedly decreased in August. Total radioactivity of whole fruit doubled, at least, with every month due to enlargement of the pericarp. Pedicels deprived of fruits had their retention prolonged on spurs with leaves or shoots treated with 1% IAA in lanoline. It is assumed that auxin delivered from shoots or still growing leaves at the time of its deficiency in seeds, restrains fruits from premature dropping. At the same time seeds seem to be protected by a regulatory system in pedicel against too massive flow of auxin from outside.

Hormone-like product of vascular tissue stimulating starch accumulation in pith explants of kale and endogenous cytokinins

When stem explants of kale (Brassica oleracea L. var.medullosa), containing pith parenchyma and a strip of vascular tissue, were cultured on simple sucrose medium, a hormone-like factor was transported from the vascular tissue to the adjacent pith, where it stimulated accumulation of starch. Similarly, up to a sevenfold increase of starch content in explants could be induced by cytokinins added to the culture medium. The relative stimulatory effect of several cytokinins (5×10-6 M) and hormone-like product of vascular tissue (HPVT) in a typical experiment were: control (1.0), trans-zeatin (6.7), HPVT (6.2), N6-[2-isopentenyl]adenine (5.4), transzeatin riboside (5.2), N6-[2-isopentenyl]adenosine (5.4), kinetin (3.6), 6-benzylaminopurine (3.5), and adenine (2.1). Concentration of endogenous cytokinins was determined using ELISA (trans-zeatin, N6-[2-isopentenyl]adenine and their ribosides) andAmaranthus bioassay (total cytokinins). No effect of vascular tissue on the level of endogenous cytokinins in explants was found. The results support the conclusions of previous experiments that the HPVT stimulating starch accumulation is not a cytokinin.

Effects of metribuzin herbicide on nitrogen, pigments, protease and nitrate reductase activity of normal and NaCl-stressed castor bean and maize plants

Addition of 0.5 and 2.5 gm-3 of metribuzin into Hoagland nutrient media, either alone or in combination with NaCl, induced significant decreases in nitrate-, amino-, ammonia-, and total soluble-N contents, whereas significant increases in these nitrogen fractions were apparent in maize and castor bean seedlings and plants treated with high concentrations (5 and 10 g m-3) of the herbicide, again either alone or in combination with NaCl. Protein- and total-N contents increased and decreased at low and high concentrations of the herbicide, respectively. The contents of chlorophyllsa andb, as well as carotenoids of both castor bean and maize seedlings and plants treated with low concentration of herbicide, either alone or supplemented with NaCl, were unaffected, whereas at high concentrations of the herbicide a significant decrease in chloroplast pigments was found. Nitrate reductase activity (NRA) was increased significantly at low concentrations of the herbicide alone and decreased significantly at high levels. Inclusion of NaCl into the herbicide media induced significant decreases in NRA of both castor bean and maize seedlings and plants. Unlike NRA changes, protease activity was increased significantly with high concentrations (5 and 10 g m-3) of metribuzin and decreased significantly with its low (0.5 and 2.5 g m-3) concentrations.

Aspartate aminotransferase isozymes in plants: Comparison of two staining methods in polyacrylamide gels

Two staining methods for aspartate aminotransferase were compared after electrophoretic resolution of its isozymes in polyacrylamide gels. The first one uses L-aspartic acid and Fast Blue BB salt (classical method), the second uses L-cysteine sulfinic acid and a redox system with phenazine methosulfate and 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide. The seeds of pea, horse bean and soybean were used as a model plant source of the enzyme. The staining method with L-cysteine sulfinic acid is very reliable and more sensitive than the Fast Blue BB method and allows detection at very low isozyme activities in the gel.

Growth, carbohydrates and associated invertase and amylase activities in castor bean and maize as affected by metribuzin and NaCl

Growth parameters (leaf area, length of shoot and root, water content and dry matter accumulation), contents of reducing sugars and saccharose as well as activities of α- and β-amylases of castor bean and maize seedlings and adult plants supplemented with 0.5 μg g-1 and 2.5 μg g-1 of metribuzin either alone or in combination with 50 μg g-1 NaCl, were increased significantly whereas at high concentrations (5 and 10 μg g-1) of herbicide, an opposite response was apparent. On the other hand, polysaccharide content and invertase activity of castor bean and maize seedlings and adult plants were significantly decreased in response to low concentrations (0.5 and 2.5 μg g-1) of metribuzin and increased significantly at high concentrations (5 and 10 μg g-1) of the herbicide either alone or in combination with 50 μg g-1 NaCl. Total carbohydrate contents of castor bean and maize seedlings and whole plants treated with herbicide either alone or in combination with NaCl did not change significantly. Growth parameters, carbohydrate fractions contents and activities of enzymes in both castor bean and maize seedlings and whole plants treated with herbicide alone were consistently higher than those values detected in plants treated with herbicide in combination with NaCl.

The effect of nutrient medium and age ofErwinia amylovora andErwinia herbicola cultures on their reactivity in agglutination test

The influence of the type of nutrient medium (meat-peptone nutrient agar, KING B medium, YDC medium) and the age of bacterial cultures ofErwinia amylovora andE. herbicola on reactivity of their antigens with homologous antisera were examined. In the case ofE. amylovora, the best agglutination reactions were observed with isolates cultivated on the YDC nutrient medium. After 192 h of cultivation, seven outof ten isolates reacted positively. The reactivity ofE. herbicola antigen decreased in dependence on culture age more rapidly than withE. amylovora. The highest rates of positive agglutination reactions were observed withE. herbicola cultivated on the YDC and KING B nutrient media. After 144 h of cultivation on both these nutrient media eight out of ten isolates reacted positively.

Cytological relationships of selected species ofPanicum L.

The cytological investigation of 12 taxa ofPanicum L. revealed that the vast majority of them have the basic number x=9 at different ploidy levels. The basic number x=8 was recorded only in the tetraploid speciesP. maximum with 2n=32. The diploid number 2n=18 was encountered inP. capillare, P. laevifolium, P. antidotale andP. coloratum (2) with 3B-chromosomes recorded in the latter species. The tetraploid chromosome number 2n=36 was found to exist inP. miliaceum, P. miliare, P. coloratum (1) andP. virgatum. The hexaploid number 2n=54 was recorded inP. bulbosum, P. dichotomiflorum andP. esculentum. The karyotypes of all accessions were mostly symmetrical and mainly comprised of meta- and submetacentric chromosomes with little variation in length among them within each karyotype. Investigation of chromosome association during metaphase I of meiosis revealed that the frequency of bivalents/cell was the highest among all investigated diploid, tetraploid and hexaploid accessions. Univalents were also frequently encountered in various accessions. These results may indicate that segmental alloploidy has been the major process by which polyploid species have originated.

Net assimilation rate, relative growth rate and yield of pea genotypes under different NaHCO3 concentrations

The net assimilation rate (NAR), relative growth rate (RGR) and yield were negatively correlated with different exchangeable sodium percentage (ESP) levels; maximum reduction in NAR, RGR and yield was observed at 30 ESP. However, the cultivar NDP-2 showed maximum percent reduction in all the characters at 30 ESP as compared to cultivars NDP-7 and Rachna.

Osmotic adjustment in leaves ofLycopersicon esculentum andL. pennellii in response to saline water irrigation

Two tomato species (Lycopersicon esculentum andL. pennellii) were grown under unheated plastic greenhouse and irrigated with 0 or 140 mM NaCl. Salinity induces a more important reduction in predawn leaf water potential (ψpd) inL. esculentum than inL. pennellii. In both species the osmotic adjustment was achieved by active solute accumulation. The leaf water potential at turgor loss point (ψtlp) seemed to be controlled by leaf osmotic potential (ψos). The results revealed the existence of limits to the accumulation of osmotic solutes in leaf tissues and the existence of an ontogenetic effect on the solute accumulation. In both species, but essentially inL. pennellii the inorganic solutes contribution especially Na+ and Cl- accumulation to ψos was higher than the organic solutes. Therefore, wild species save energy more markedly.

Effect of 2,4-dichlorophenoxyacetic acid on nuclear division in stem segments ofPisum sativum L. cultivatedin vitro

We have investigated the influence of 2,4-dichlorophenoxyacetic acid (2,4-D) on the appearance of nuclear fragments, caused by direct nuclear division, as well as on mitotic activity in cultivated internodial stem segments ofPisum sativum L., cv. Bördi, during 180 d of cultivation. Direct nuclear fragmentation (dNF) was indicated by the shape and structure of the nucleus as well as by the occurrence of 1C- and 3C-values of DNA, investigated cytophotometrically. The dNF occurred during the whole cultivation period in segments treated by 2,4-D in concentrations from 4 to 32 mg 1-1. In the presence of 2 mg 1-1 of 2,4-D the dNF existed in the explants only up to 90 d. Mitotic activity was not observed in the 2,4-D-free control but occurred during the whole cultivation period when 2,4-D was added in concentrations from 2 to 16 mg l-1. In the presence of 32 mg l-1 of 2,4-D the level of mitotic activity was very low at the beginning and ceased after 60 d in culture.

Axillary bud proliferation and ethylene production as controlled by radiation of different spectral composition and exogenous phytohormones

The effect of radiation of different spectral composition on axillary proliferation of lavandin (Lavandula officinalis Chaix ×Lavandula latifolia Villars cv. Grosso) was studied in combination with application of exogenous benzyladenine (BA) and putrescine (Put) and endogenous ethylene production. The effect of BA was predominant over the radiation. Continuous far-red showed a fluence rate-dependent promotion of shoot proliferation in the presence of BA. On BA-free medium, shoot number was enhanced under blue, white, and red radiation, at low photon fluence rates. BA, however, could reduce the inhibiting effect of blue and ultraviolet radiation, at high photon fluence rates. Exogenous Put stimulated axillary bud proliferation under some radiation treatments in the presence of BA. Moreover, Put, analogously to BA, could overcome the detrimental effect of ultraviolet radiation. A positive correlation between biotic ethylene production and shoot formation was evidenced under far-red at high photon fluence rate in the presence of BA, and under white, red and blue radiation at low photon fluence rate in the BA-free medium. However, when abiotic ethylene (released from the agarized medium) was stimulated by UV, no improvement of shoot formation was observed.

Peroxidase activity in leaves ofSyngonium podophyllum following transition fromin vitro toex vitro conditions

A low activity of peroxidase (POD, EC 1.11.1.7) was found in crude extracts fromin vitro plants ofSyngonium podophyllum cv. Butterfly sealed in the multiplication vessels. Removal of the lids from the vessels increased the POD activity. A greater increase in the POD activity was measured whenin vitro plants were transplanted into a weaning growth medium. The POD activity was further promoted by removal of the phenolic substances from the extracts by polyvinyl-polypyrrolidone (PVPP).

Purification and immunohistochemical localization of the auxin binding site i protein from maize coleoptiles

An auxin binding protein fraction prepared by means of affinity chromatography on 2-OH-3,5-diiodobenzoic acid-Sepharose and gel filtration was used as antigen. The obtained rabbit antisera contained antibodies against the auxin, binding protein (ABP) and several contaminating proteins (nonABP). The nonABP could be separated on an appropriate affinity matrix omitting the TIBA analogue. After their immobilization on Sepharose antibodies directed towards contaminating, the proteins were isolated and immobilized, too. This IgGanti nonABP-Sepharose retains almost all contaminating proteins present in the specific eluates of the auxin affinity matrix. In a final affinity chromatography step on IgG-Sepharose a highly purified ABP could be eluted. This ABP was immobilized on Sepharose for the separation of monospecific antibodies against ABP (IgGanti abp). Using these antibodies the ABP could be localized within the outer epidermal cells of the coleoptile by immunofluorescence microscopy. From the inhibition of auxin induced elongation of coleoptile tissue by IgGanti abp it is concluded that the ABP is localized at the plasmalemma of the epidermal cells and that the ABP is involved in auxin action as a true hormone receptor.

The effect of glucosinolates (mustard oil glycosides) and products of their enzymic degradation on the infectivity of turnip mosaic virus

The effect of glucosinolates sinigrin, progoitrin, epiprogroitrin, gluconapin, gluconapoleiferin, glucobrassicanapin, glucotropaeolin and gluconasturtiin without and with the glucosinolate-degrading enzyme (myrosinase, EC:3.2.3.1.), on the infectivity of turnip mosaic virus was studied. Little or no effect was observed when the intact glucosinolate (2.5 μmol cm-3) was added to the suspension of turnip mosaic virus (TuMV, isolate Ruzyně; 0.2 mg cm-3 in 0.01 M potassium phosphate buffer) at both pH 7 and pH 6. A significant decrease of virus infectivity was, however, observed when 0.25, 1.25 and 2.5 μmol cm-3 of the glucosinolate together with 0.31, 1.56 and 3.13 mg cm-3 of the myrosinase, respectively, was added to the virus suspension of both pH 7 and pH 6. The effect, which was greater at pH 6, was most intense with sinalbin and glucobrassicin substrates.

Effect of ATP concentration and temperature on firefly luciferase activity

The dependence of luciferase activity in the homogenate of leaves of transgenic tobacco plants with chimeric firefly luciferase gene on ATP concentration and temperature was studied. The optimum ATP concentration was between 0.625 mM and 2.5 mM. The activity rapidly decreased if the homogenate was kept in 25°C and is completely lost during 30 min.

Metabolic conversion of exogenous14C-aspartate and14C-gIutamate in the dark byFucus serratus L

The rates of uptake of exogenous L[U-14C] aspartate and glutamate into tissues of vegetative growing tips ofFucus serratus and their metabolism were studied in the dark. In these non-photosynthetic conditions, aspartate was fixed and metabolically converted more rapidly than glutamate. Radioactivity from14C-aspartate was principally transferred into glutamate. On the other hand, metabolism of absorbed14C-glutamate was very slow and its rate did not increase during incubation time, but produced more diversified soluble radioactive compounds. Thus inF. serratus, glutamate principally seems to be in the dark more a temporary14CO2 storage product coming from β-carboxylation than a rapidly turned over intermediate.

Effect of Zn2+ on water and K+ fluxes in detopped maize plants

Water and K+ fluxes were examined in detopped plants ofZea mays L. (cv. White Horse Tooth), which were grown and exuded on half-strength Long Ashton nutrient solution containing the appropriate concentration of Zn2+ at 20 °C. In light-grown plants, 100 and 500 μM Zn2+ increased both water and K+ fluxes in detopped maize plants whereas 1 000 μM Zn2+ inhibited both fluxes. In the dark-pretreated plants, 1 000 μM Zn2+ in the medium stimulated K+ flux. The fluxes of K+, Zn2+, Ca2+ and Mg2+ were usually higher in detopped plants than in intact ones. At 1 000 μM Zn2+ in the exudation medium, Zn2+ concentration was higher in the xylem exudate of dark-pretreated plants than in roots of plants maintained in light. The results are discussed in relation to the influence of Zn2+ on the membrane permeability and transport in plants.

Resistance to ACNU induced toxicity in transgenic tobacco suspension cultures withada gene transferred fromEscherichia coli

The protein coding region of theE. coli DNA repair geneada combined with the CaMV 35S promoter has been transferred to tobacco by means ofAgrobacterium tumefaciens Ti plasmid. In transgenic plants having theada gene in a sense orientation, detectable amounts of O6-alkylguanine-DNA-alkyltransferase has been found whereas in non-transformed plants this activity is absent. Cell suspension cultures derived from the former plants showed lower sensitivity to the toxic (growth inhibiting) effects of the bifunctional alkylating agent 1-(2-chloroethyl)-1-nitroso-3-(aminomethyl-1,3-diazinylo)-methylurea compared with cell cultures derived from a control non-transformed plant or from transgenic plants harbouring theada gene in an opposite, non-sense orientation.

Effects of salinity on growth and metabolism ofPhaseolus vulgaris

Increasing salinity induced a marked reduction in the plant growth, thoughPhaseolus seedlings tolerated salinity up to 120 mM NaCI. A great reduction in sugar and protein contents occurred with increasing salinity, whereas soluble nitrogen compounds and the relative contents of the photosynthetic pigments were increased in the treated plants. Increasing Ca concentration in the salinized medium appeared to improve the plant growth and to increase the contents of saccharides and proteins in the NaCl-treated plants. This suggests that Ca could be added to salinized media to overcome the deleterious effects of salinity on the growth and productivity of leguminous crop plants.
