Modification of cultivar-specific regeneration capacity of potato expiants by phytohormones and byAgrobacterium oncogenesJ. Ovesná, I. Burdová, L. Křížková, Z. A replacement of 6-BAP with zeatin in the inductive MS medium stimulated bud/shoot formation in only two of three tested cultivars with naturally low regeneration capacity. GA3 did not affect the bud initiation phase (i.e. regeneration capacity of cultivars), it only stimulated shoot development. The insertion of some bacterial oncogenes (in particular genes 4, rolB, C and 5) enhanced bud/shoot formation, especially in the case of low-regenerating cultivars. |

Dry mass of shoots and roots, 14 and 20 weeks after the commencement of salinity treatment, increased at low levels of salinity (0.1 - 0.2 % NaCl) but decreased with higher NaCl concentrations (0.4 - 1.2 %). Nodulation occurred at NaCl concentrations up to 0.8 %. Nodule mass decreased with increasing salinity levels. The nodule size remained unaffected at NaCl concentrations up to 0.4 % but was reduced at higher concentrations. |

Significant differences between the two seasons for pollen fall, pollen germination, tube growth and pod set were observed. In kharif season with an average temperature of 26 to 30 °C and 75 to 85 % humidity all the characters showed higher values than in the spring season with a higher temperature of 32 to 36 °C and low humidity of 40 to 50 %. Application of gibberellic acid (GA) alone and a combination of gibberellic acid, naphthaleneacetic acid (NAA) and kinetin (KIN) significantly increased the pollen fall by 7-13 %, pollen germination by 10-12 % and pollen tube growth by 27-30 %. More pollen tube abnormalities in interspecific crosses only indicates partly incompatible reaction of pollen tube in the stylar tisue. Hormonal treatments, especially GA, significantly increased the pod set and pod harvest by 20 % and 34 %, respectively. Higher pod harvest was seen in crosses withV. unguiculata as female parent. GA treatment enhanced pollen germination and pollen tube growth, and by partially overcoming embryo abortion for 10 to 12 d, immature embryos were successfully rescued forin vitro production of hybrid plantlets. |

Segments of seedlings germinated on media supplemented with different growth regulator combinations were used as explants. Cultivation was carried out in two phases, which alternate permanently: (I) without and (II) with growth regulators. Thus it was possible to establish meristematic tissue cultures from 5-10% of the seedlings tested. They have produced buds now for over 3 years with multiplication rates of about 1.5 within 5-6 weeks. |

A positive correlation was found between the activity of 1,3-β-glucan synthase and the degree of barley resistance. On the contrary, the 1,3-β-D-glucanase activity in whole leaves was negatively correlated to host plant resistance. This phenomen is evident only in the early phase of plant pathogen interaction. However, in epidermal cells the 1,3-glucanase activity was not significantly changed after attack and the 1,3-glucan synthase activity was practically zero. Chitinase activity in inoculated leaves and epidermis was higher than in healthy ones, but no unambigous correlation was found between the enzyme activity and host resistance. |

F1 hybrids were studied morphologically and flow cytometry was used to estimate 2C nuclear DNA content both in parentalBrassica species and their hybrids. It was found that in comparison with the A genome, the B and the C genomes ofBrassica contained 26.9 % and 43.9 % more DNA, respectively. This finding may be used to distinguish interspecific hybrids containing various genome combinations. It was concluded that flow cytometric analysis of nuclear DNA content might be useful tool inBrassica breeding. |

RaoBiologia plantarum 35:267, 1993 | DOI: 10.1007/BF02925951 Sodium chloride tolerant cell lines of finger millet were isolated from embryogenic cultures growing on MS medium supplemented with picloram (2 mg I-1), kinetin (0.1 mg l-1) and sodium chloride (1 %) at the end of 6 passages. The sodium chloride tolerant cell lines showed better growth in comparison with control at all concentrations of sodium chloride tested, with optimum growth at 0.25 % NaCl. When the tolerant lines were grown for 3 passages in absence of NaCl, the growth was lower than that of the tolerant lines tested immediately at the end of 6 passages of selection. NaCl tolerant calli had more Na1 in comparison with control and they regenerated plants in presence of 1 % NaCl, while the control lines failed to differentiate. When screened in a hydroponics system with 1 % NaCl, the tolerant plants grew to maturity while the control plants failed to grow. |

Protein content showed similar slight variations in explants during the course of budding and callusing, whereas an important increase of protein content was found during the first 48 h in explants cultured on root forming medium. |

These promotory effects were recorded at 40 and 80 mg m-3 triacontanol, but they decreased with increasing growth regulator concentration. |
Effect of external calcium on the control of stamen movement inBerberis vulgaris L.Z. Lechowski, J. BiałczykBiologia plantarum 34:121, 1992 | DOI: 10.1007/BF02925805 The mechanical stimulation of the sensitive internal lower part of Berberis vulgaris stamen resulted in its rapid bending. In the present study we have examined the influence of external Ca2+ concentration on stamen movement. The external Ca2+ reduced the extent of the response and the effect was dependent on Ca2+ concentration and duration of the treatment. Addition of calcium ionophore A 23187 to the medium reduced the response and the effect was dependent on the external Ca2+ concetration. This result might suggest an increase in Ca2+ level in cytosol. The inhibitory effect of higher Ca2+ concentration on stamen bending was cancelled by Ca2+-chelating agents. Ca2+-channel blockers prevented the stamen response in higher external Ca2+ concentration with different effectiveness. In these conditions, during 4-h experiments, La3+, verapamil and nifedipine gave ability of stamen movement at about 85 %, 58 % and 10 %, respectively. An energy-dependent Ca2+ efflux was confirmed in experiments by using vanadate, a non-specific ATPase inhibitor. The lack of inhibition of stamen bending after application of calmodulin antagonists suggests that it might not be directly involved in regulation of the response. The inhibitory effect of higher Ca2+ concentration on stamen movement might result from: (a) the binding with cell wall materials, (b) changes of structure of cytoplasm and metabolic activity and (c) influence on transport processes. |

Supplemented 30 mM L-proline was shown to be the most effective; at higher concentration it reduced seedling growth and K+/Na+ ratio. Thus 30 mM L-proline can alleviate the salinity stress in rice seedlings. |

Role of calcium and calmodulin antagonist in photosynthesis and salinity tolerance inChlorella vulgarisR. Abdel-BassetBiologia plantarum 35:237-244, 1993 | DOI: 10.1007/BF02925946 To cast light upon the role of Ca1+ and calmodulin on photosynthetic rate (Pn), dark respiration (RD) and amino acid and protein contents in salinity stressed and non-stressedChlorella cultures, the Ca2+ chelator EGTA [ethylene glycol-bis-(2-aminoethyl ether)-N,N- tetraacetate] and the calmodulin antagonist TFP (trifluperazine) were used. TFP markedly inhibited PN while EGTA exerted a slight, if any, effect on PN. NaCl tolerance, on the other side, was markedly abolished by TFP that inhibited PN and lowered rate of proline accumulation. Calmodulin might be involved in osmoregulation and salt tolerance ofChlorella. RD, however, was markedly enhanced by EGTA and Ca2+-free medium and hence the Ca2+ deprivation increased stress severity exerted by NaCl. Combinations of Na+ and Ca2+ enhanced PN, decreased RD and proline content in comparison with an osmotically equivalent reference culture containing only NaCl. Addition of Ca2+ to TFP treated cultures failed to reactivate calmodulin for proline synthesis. However, when Ca2+ was added to EGTA-treated cultures, only relatively reduced proline contents were recorded. |

Flowering and male reproductive functions of chickpea (Cicer arietinum L.) genotypes as affected by salinityH. R. Dhingra, T. M. VargheseBiologia plantarum 35:447, 1993 | DOI: 10.1007/BF02928525 The influence of salinity, given at germination (stage I) or 75 d after sowing (stage II), on flower production and characteristic features of male reproductive structures was studied in three promising genotypes of chickpea (Cicer arietinum L. cv. ICCV 88102, H 82-2 and C-235). In ICCV 88102 and H 82-2 salinity 20 meq 1-1 increased the number of flowers when applied at both stages whereas in C-235 only when applied at the later stage. The salinity also delayed flowering; the higher salinity the greater was delay in flowering. In H 82-2 and C-235 salinity treatment given at stage II (when few flower buds appeared) hastened the flowering. The salinity curtailed pollen production; the reduction being minimum in C-235 and maximum in H 82-2. Germination was not significantly affected in C-235 pollen collected from plants grown under salinity conditions upto 60 meq 1-1 applied at stage I but the tube elongation was inhibited. The inhibition of tube elongation was greatest in C-235. Salinity treatment administered at stage II did not affect significantly pollen germination excepting C-235 in which a consistent decline with increasing salinity was evident. Salinity stimulated tube growth in ICCV 88102. Na2SO4 in the germination medium was more detrimental for both pollen germination and tube growth than NaCl. |

Utilization of metabolic energy under saline conditions: changes in properties of ATP dependent enzymes in plant cells grown under saline conditionsM. ReuveniBiologia plantarum 34:181, 1992 | DOI: 10.1007/BF02925865 The effect of growth in saline medium on the activity of two ATP utilizing enzymes was studied. Hexokinase in carrot (Daucus carota L.) cells grown in suspension culture either in the absence or presence of 150 ml NaCl, and tonoplast H+-ATPase in tobacco (Nicotiana tabacum L. cv. Wisconsin 38) cells grown in suspension culture either in the absence of presence of 428 mM NaCl. There was no difference in the pH profiles, NaCl sensitivity and kinetic parameters towards glucose of hexokinase activities from carrot cells grown in the presence or the absence of NaCl, but the activity from cells grown in the presence of NaCl was more resistant to inhibition by N-ethylmaleimide and to inactivation by heat. Two separate apparent Km values toward ATP were delineated in the extract from cells grown in presence of NaCl while extracts from cells grown in the absence of NaCl had only one apparent Km value. The tonoplast H+-ATPase from NaCl grown tobacco cells showed changed kinetic compared to this activity from cells grown in the absence of NaCl. These data may indicate that growth in NaCl results in the appearance of isozymic activity that enhances the ability of plant cells to utilize metabolic energy more efficiently. |

Expiants cultured on media with cytokinins or in combination with auxins produced shoot buds. Maximum number of shoot buds (20-25 per culture) were differentiated from cotyledonary leaf segments on medium with 2 mg 1-1 each of KN and IBA. Rooting of regenerated shoot buds was acheived on medium with NAA. The obtained plantlets were successfully transferred to soil. |
Autotoxic impact of essential oil extracted fromLantana camara L.R. K. Arora, R. K. The contents of water and chlorophyll, of leaves apart from seed germination, seed vigour and length of seedlings of the parent plant were adversely affected with increasing concentration of theLantana oils; this indicated autotoxic potential of the oil. The correlations between oil concentration and the studied parameters were rather strong and obeyed the concentration response relationship, typical of chemical bases. The cell respiration, however, increased with increasing concentration of the oil vapours. Compared to control, less than 13 % of the seeds of the parent plant exposed to 30 cm3 m-3 of oil showed incipient germination with no growth of radicle beyond immeasurable protuberance, and no plumule emergence. |

Rates of apparent photosynthesis, respiration and dry matter accumulation in maize canopiesDong Shuting, Hu Changhao, Gao RongqiBiologia plantarum 35:273-277, 1993 | DOI: 10.1007/BF02925953 The rates of canopy apparent photosynthesis (PC) and canopy respiration (Rc) were studied during vegetation season in two erectophile and two planophile hybrids of maize (Zea mays L.) grown at two canopy densities [7.5 plants m-2 (HD) and 4.5 plants m-2 (LD)]. Large differences in PC, Rc, RC/PC, leaf area index (LAI), dry matter accumulation and grain yield were found among hybrids and plant densities. Variations in PC and RC were associated mainly with changes in LAI. There was also found change in PC per unit LAI with time. The average RC/PC was 28.9 % for all treatments throughout the vegetation season. PC and RC per unit dry matter were higher in LD than in HD and decreased throughout the measurement period. The HD stand had higher PC and yield in hybrids with erectophile foliage, whereas LD stand had higher PC after male tetrad and got higher yield in hybrids with planophile foliage. Only RC in hybrids of the two foliage types was higher under HD than under LD throughout the vegetation period. |
Absorption of sucrose by tissues of adult tubers of potato(Solatium tuberosum L.). modalities and influence of phytohormonesM. Penot, A. Hourmant, A. ReddahiBiologia plantarum 35:365, 1993 | DOI: 10.1007/BF02928512 Sugar uptake by potato tuber discs was studied. Discs were used "fresh" or after a 24-h ageing period. It was shown that ageing increases (by 3 to 4 times) the rate of glucose and sucrose uptake. Sucrose uptake by fresh tissues was insensitive to the presence of glucose or fructose while a competitive effect was observed after ageing. This indicates the development of an invertase activity, which was inhibited by tris-buffer. Sucrose and glucose uptake by aged discs was dependent on cellular metabolism as shown by the sensitivity to low temperature and metabolic inhibitors (NaCN, DNP, CCCP). Involvement of thiol groups was demonstrated by the inhibition with NEM and PCMBS. Orthovanadate, which decreases phosphate uptake by 85 % (Poderet al. 1986) did not produce any effect on glucose and sucrose uptake by aged tissues. Fusicoccin produced only a slight stimulation (15 %). These results argue in favour of the involvement of specific ATPases in ion and sugar uptake. No involvement of a redox system was observed. ABA and BAP inhibited the uptake induced by ageing but had no effect on the endogenous sugar content. BAP would act by its effect on the amount of ATP while ABA would act at the membrane level. The results are discussed in relation to the mechanisms of transfer of glucosyl groups and to the transport of sucrose by the symplasmic pathway. Reçule |

SinghBiologia plantarum 35:583, 1993 | DOI: 10.1007/BF02928036 Inclusion of cadmium as cadmium chloride and nickel as nickel chloride in media differentially induced dry matter mobilisation from cotyledons of germinating pigeon pea seeds. Cadmium depressed the activities of total amylase, protease, acid phosphatase and peroxidase in germinating seeds. The activities of these enzymes were stimulated at the lower concentrations of nickel and suppressed at the higher ones. This dual response of hydrolytic enzymes to cadmium and nickel is postulated to account for the promotory and inhibitory effects of these heavy metals on dry matter mobilisation. |
