FelippeBiologia plantarum 30:48-52, 1988 | DOI: 10.1007/BF02876423 In negatively photoblastic seeds of Cucumis anguria L. the scarification of micropyle overcame the inhibitory effect of "white light ", although far-red radiation inhibited the germination in both intact and scarified seeds. The moistened testa powder transmitted more far-red than red radiation. Thus the testa could act as a filter of radiation and maintain a high far-red/red ratio at the photosensitive site of the embryo, which must be localized near the micropyle. |

SlovákováBiologia plantarum 36:121-132, 1994 | DOI: 10.1007/BF02921279 Three anionic peroxidases (EC 1.11.1.7), named Prx1, 2, and 3, which are rapidly accumulated in cucumber (Cucumis sativus L., cv. Laura) reacting hypersensitively to tobacco necrosis virus, were purified to homogeneity. The three enzymes had an isoelectric point about 4.3, and the relative molecular masses of Prx1, 2, and 3 estimated by SDS-PAGE were 40 700, 38 000, and 37 100, respectively. These peroxidases had a similar pH stability, but differed in their specific activity, pH optimum, and thermal stability By Ouchterlony double diffusion tests with antisera raised against the three purified enzymes, close serological relationships have been demonstrated between the three peroxidases. |

Effect of amino acids on rooting of apple dwarf rootstocksin vitroT. OrlikowskaBiologia plantarum 34:39, 1992 | DOI: 10.1007/BF02925788 The effect of twelve amino acids and lactalbumin hydrolysate in concentration of 200 mg 1-1 on rooting of the dwarf apple rootstocks P 2 and P 60 was testedin vitro. Arginine, omithine, glutamic acid and glycine enhanced root number of the P 60 rootstock; proline and lactalbumin hydrolysate were neutral; and asparagine, tyrosine, methionine, cysteine and glutamine lowered the root number. Tyrosine, methionine, cysteine and glutamine reduced almost completely rooting of P 60. In the recalcitrant P 2 rootstock aspartic acid, glutamic acid and omithine significantly enhanced the number of roots and rooted shoots, arginine and tryptophan increased the root number only slightly, asparagine was neutral, and proline reduced the root number. |

Attainment of the final plastochron index was considered as a point of completion of a vegetative phase. The population under study completed a vegetative phase within 50 d and exhibited flower initiation in 55 d. The gap between completion of the vegetative phase and flower initiation indicated the prevalence of juvenility in cowpea. The maturity period was found to be a sum of the periods for plastochron completion, lag I, pod development and lag II. Lag I and Lag-II periods existed due to variation in the relative sensitivity of successively formed leaves to the normal inductive conditions. A high genotypic variation for the two lag periods indicated a scope for designing an efficient plant architecture of cowpea either for grain or for vegetable purpose. |

The tuberization rate increased and tuber mass decreased to one half or one quarter in potato plantlets in a modified MS-medium with 3 % saccharose and decreased nitrogen content in comparison with normal MS-medium. Decreasing nitrogen content prolonged the time necessary for tuberization. The increase of sucrose concentration to 4 % decreased the inhibitory effect of higher contents of nitrogen on tuberization rate. |

The thermal denaturations of DNA, the number of DNA components reassociating with second order kinetics, the proportion of repeated DNA sequences, the frequency of the repeated DNA classes are reported and compared to previous data onVicia faba DNA. Feulgen absorptions at different thresholds of optical density+ of interphase nuclei in cytological preparations of the root meristems ofV. bithynica andV. narbonensis are determined and compared withV. faba analogous determinations. The results, confirming that plant genome is highly flexible, are discussed in relation to other data on the interspecific variations of the nuclear DNA content. |
Apple tree water relations studied by means of the relative rate of water flow in the trunk xylemN. E. NadezhdinaBiologia plantarum 34:431-437, 1992 | DOI: 10.1007/BF02923593 The results of long-term investigations of variations of rates of water transport through the trunk xylem, its diameter, the leaf water potential and the transpiration rate of the apple tree showed that the daily rhythm of the relative rate of moisture flow in the trunk xylem is an obvious index of the state of the apple tree water exchange. This enables us to determine the period of its unbalance at intensive transpiration as well as the level of the forming water deficit with high accuracy and operativeness. Moreover, by the daily curve of the relative rate of xylem flow one can judge the role of contribution of the trunk reservoir to transpiration. |

D-Isomers of free amino acids were not revealed by the treatment with L- and D-amino acid oxidases. The noncationic fraction of the extract contained N-malonyl-D-tryptophan and no other N-acylated amino acids. A special search for endogenous N-malonyl-D-phenylalanine gave negative results. Exogenous14C-malonate was only incorporated in one Chromatographic zone corresponding to N-malonyl-D-tryptophan. It is concluded that drought stress does not induce the appearance of D-amino acids except for D-tryptophan which is accumulated in the malonylated form. |

SrivastavaBiologia plantarum 34:153, 1992 | DOI: 10.1007/BF02925812 Glutamate dehydrogenase (GDH) activity, protein and total nitrogen contents in the secondary leaves of maize(Zea mays L. cv. Ganga Safed-2) seedlings increased during early seedling growth and then declined after reaching a peak level at either 10 d (GDH) or 12 d (metabolites). While the effect of kinetin on enzyme activity was statistically insignificant, benzyladenine supplied with nutrient solution increased GDH activity in secondary leaves of both 10-d as well as 14-d seedlings. However, both growth regulators increased the contents of total soluble proteins, total nitrogen, chlorophyll(a+b) and carotenoids in both 10 and 14-d old leaves. |

GendraudBiologia plantarum 34:373, 1992 | DOI: 10.1007/BF02923582 Plasmalemma-enriched fractions were isolated from Jerusalem artichoke tubers along the time course of dormancy break produced by cold treatment. A decrease of membrane fluidity was noted from the 3rd to the 8th week of this treatment, as well as a decrease of plasmalemma NADH dehydrogenase activity from the 5th to the 8th week. The plasmalemma lipid extracts studied revealed two major phospholipidic components: phosphatidylcholine (PC) and phosphatidylethanolamine (PE). Their respective quantities decreased until the 12th week, where the phosphatidylcholine level is lower than the phosphatidylethanolamine one. The observed changes are discused in relation to dormant and non-dormant states of tubers and the breaking of dormancy. |
Effects of mineral composition and acidity of media, saccharose level, brand and quantity of agar on rooting of fruit rootstocksin vitroT. OrlikowskaBiologia plantarum 34:45-52, 1992 | DOI: 10.1007/BF02925789 The influence of the mineral composition and acidity of media, saccharose level, brand and quantity of agar onin vitro shoot rooting of P 60 and P 2 apple and quince S 1 root-stocks were compared. Among the tested salt compositions (full WMP and 1/2 WMP, full MS and 1/2 MS) the most suitable for rooting was composition of WMP. Further modifications in quantity of nitrogen, H3BO3 CaCl2 and MgSO4 of WMP medium did not have a positive effect on rooting. 30 g l-1 saccharose gave better results than 20 g l-1. Acidity of WMP medium tested in a pH range of 4.0 to 6.5 did not affect rooting of P 60 rootstock but media adjusted to pH 4.5 and 5.0 were better for rooting of recalcitrant P 2 rootstock. Among 4 agar brands (Difco Bacto, Oxoid No.1, Japanese commercial fibre and Japanese commercial powder) tested at 6 g l-1, the most effective was the last one, but its quantity had no effect in the range of 3 to 10 g l-1 for rooting of P 60 rootstock. |
Karyological and cytophotometric study of expiant derived clones of non-polysomatic and polysomatic species ofKniphofiaS. Nayak, S. SenBiologia plantarum 34:135-141, 1992 | DOI: 10.1007/BF02925807 Cytological and cytophotometric analysis of root tips of regenerated plants, derived from rhizome expiant ofKniphofia nelsonii andKniphofia uvaria, revealed marked difference in behaviour of chromosomes and level of 4C nuclear DNA content. Karyotypic stability could be retained in all 52 regenerants of K. nelsonii whereas inK. uvaria out of 75 regenerants analysed, 12 plants were exclusively diploid and the rest 63 plants were predominantly diploid comprising variable amount of aneuploid and tetraploid cells. Cytological data was further confirmed by nuclear DNA content estimation. Alteration in the structure of chromosomes could also be noted in 57 regenerants ofK. uvaria giving rise to two new karyotypes. The use of polysomatic tissue for securing variantsin vitro inK. uvaria has been suggested. |
