Effects of salt stress on nitrogenase activity and growth of four legumes

The effects of salt stress on nitrogenase (N2-ase) activity, growth and nitrogen content ofVicia faba (L.),Medicago sativa (L.) Merrill,Glycine max andVigna sinensis (L.) were investigated. Four levels of salinity were applied and salt treatments were imposed on inoculated and N-fertilized plants.M. sativa tolerated mild levels of salinity but higher salt concentrations depressed N2-ase activity of this species. The other three legumes were considerably affected by salt treatments, and N2-ase activity was significantly reduced by salinity.
Vicia faba, carrying elongate nodules, could restore a partial N2-ase activity upon recovery from salt stress whereasG. max andV. sinesis, both with spherical nodules, could not regain significant activity when salinity was removed.
Salt stress retarded growth of both inoculated and N-fertilized plants. The nitrogen content of both treatments was also affected by salinity and the effect was more severe for inoculated than N-fertilized plants.

The role of auxin stored in scots pine trunk during spring activation of cambial activity

In a 9-year-old pine girdled during the winter cambial activity was observed below the girdle in the next spring. This indicates that cambial activity was initiated without auxin produced in the spring by buds. The auxin produced in apical shoots successively flows down the stem, where as a result of periodic restriction in transport it remains over the winter till the next year. This auxin of apical origin but locally stored over the winter in the stem is responsible for the activation of cambium before the new flow of auxin produced in the apical meristems arrives. Calculations based on seasonal changes in auxin levels can explain both, earlier spring activation of cambium in the crown and the temporary cambial divisions below the girdle, without assumption of direct auxin synthesis in the lateral meristems.

Drought resistance in pearl millet

The influence of wilting on the levels of free proline, soluble proteins, reducing sugars, starch and on the activities of nitrate reductase, invertase, amylase and pyrophosphatases have been studied in the leaf tissue of five cultivars of pearl millet at their vegetative stage under pot culture conditions. The metabolic changes could not be correlated with the yield behaviour of the cultivars under a drought condition in the field.

The effect of temperature on the uptake and loss of anions by seedling roots ofZea mays L

Intact maize seedlings were examined for the uptake and leakage of labelled sulphate and phosphate anions affected by temperature. Control plants, grown at 25 °C were compared from the aspects of uptake capacity and leakage with plants incubated in nutrient solutions cooled to 15 °C and 5 °C, respectively. Short time intervals as well as 1-7 d exposure to cooling were used. Already after 1 h exposure at 5 °C and 5 h cooling at 15 °C and at 5 °C, considerable changes were manifested in anion uptake and leakage.
The uptake of32P declined more than that of35S. So, after a 30 min uptake interval the uptake of35S decreased at 15 °C to 49.84% and at 5 °C to 6.05% comparing with the uptake at 25 °C, while the uptake of32P declined to 28.64% at 15 °C and to 4.45% at 5 °C. The leakage of both anions was the highest at 25 °C in absolute rates, but relatively most of the uptaken35S and32P was released at 5 °C. Longer exposure to a chilling temperature of 5 °C (1-7 days) resulted in two patterns of sulphate and phosphate uptake.

Changes in the anatomical structure of the first two leaves of barley caused by the absence of nitrogen or phosphorus in the nutrient medium

The absence of nitrogen or phosphorus in the nutrient medium affected quantitative anatomical characteristics of the first two leaves of spring barley: nitrogen deficiency brought about changes in both the leaf size and characteristics of stomata, whereas the effect of phosphorus absence was insignificant. N deficiency had an opposite effect on certain characteristics connected with volume ratios of tissues than that of P. The most marked finding is the decrease in the leaf volume and specific volume, and in specific volumes of most tissues at N deficiency and their increase at P deficiency. On the other hand, the number of stomata per unit leaf or photosynthetic tissue volume increased as the result of N deficiency and decreased at P deficiency.

Identification of cytokinins present in root nodules ofPhaseolus mungo L

Root nodules ofPhaseolus mungo were extracted for their cytokinin content. Four cytokininsviz. zeatin, zeatin riboside, N6(‡2-isopentenyl) aminopurine and its riboside were identified on the basis of Rf values obtained by co-chromatography with authentic samples of cytokinins on thin layer of a mixture of polyvinylpyrrolidone/CaSO4.

Effect of different disintegration techniques and media on yield and appearance of isolated nuclei

Cell nuclei have been released from various plant tissues (barley leaves, roots and embryos, tobacco leaves and tissue cultures,Vicia faba roots,Arabidopsis thaliana leaves) by several homogenization methods and the optimal method was established for each tissue. The effect of the composition of isolation medium on the yield and appearance of isolated nuclei was also studied. Longer incubation withn-octanol increases the yield considerably in most cases. Low concentrations of osmoticum increase the yield and their adverse effect on the integrity of nuclei is of little significance. Gum arabic has a favourable effect on nuclei isolation from roots only.

Ontogeny and histochemistry of axillary bud inMurraya koenigii L. spreng

Histochemical localization of total proteins, histories, nucleic acids, ascorbic acid and polysaccharides in the developing axillary bud ofMurraya koenigii and its vascular relationship with the main axis are investigated. All the above variable metabolites are richly distributed throughout the bud development. The shell zone is indicative of poor distribution of these metabolites. Histochemical tests prove that the axillary bud is metabolically very active. The initiation of the axillary bud is in the axil of the 2nd leaf where two cells of 2nd tunica layer become prominent and undergo periclinal divisions to give rise to the bud. The bud maintains tunica-corpus organization throughout its development. The cells of the shell zone at the base of the bud differentiate into pith meristem of the bud. The axillary bud has two bud traces which are associated with vascular strands that are not the traces of the subtending leaf.

Investigations on the causes of low susceptibility of scentless mayweed to 2,4-D

White mustard (Sinapis alba L.) plants, susceptible to 2,4-D, and scentless mayweed (Tripleurospermum maritimum L.) plants, resistant to 2,4-D, markedly differ in the distribution and metabolism of 2,4-D. When 2,4-D-14C was applied onto the leaf, radioactivity was found in mustard after 8 days in the entire plant, whereas in scentless mayweed radioactivity occurred mainly in the leaf onto which it was applied and in another one or two leaves, with the roots showing only traces of radioactivity. The two plant species differed both in the character and in the number and amount of metabolites detected in aqueous and ether fractions. The metabolite with Rf 0.63 -0.65, soluble in ethylether (39% of the total radioactivity applied), and that with Rf 0.35 -0.36 (23% of the total radioactivity) were present exclusively in the roots of scentless mayweed plants. The Rf values established indicate that the former metabolite may be a conjugate with the amino acids alanine and valine and the latter a hydroxylated derivate of 2,4-D. In the shoots of both plants, 2,4-D-14C was metabolized to identical metabolites with Rf 0.59 -0.61, which occurred in mustard plants only in trace amounts. Free 2,4-D-14C occurred in the shoots of both plants and in considerable amounts also in mustard roots; it could not be demonstrated in the roots of scentless mayweed. The two species did not differ in the uptake rate or in the amount of absorbed 2,4-D-14C.

Irradiation induced changes in flower formation inCannabis sativa L

Stimulation of flower formation was ohserved in the female plants ofCannabis sativa raised from seeds treated with gamma rays. These plants produced seeds, of smaller size. One of 195 plants formed sterile hermaphrodite flowers with reduced female structures and increased number of anthers.

The changes in metabolic utilization of galactose in tobacco mosaic virus infected tobacco plants treated with 2,4-dichlorophenoxyacetic acid

The study deals with the changes in metabolic processes of galactose breakdown in tobacco mosaic virus infected tobacco tissues treated with a 1.0 × 10-4 M solution of 2,4-D (2,4-dichlorophenoxyacetic acid). At the time of maximum virus reproduction the plants infected exhibited a considerable increase in galactokinase and galactose-1-phosphate uridyltransferase activities. The activity of both galactose metabolizing enzyme systems investigated was not affected markedly by 2,4-D, but its increase was induced preferentially by virus biosynthesis.

Atypical composition of seed proteins in cultivars ofPhaseolus vulgaris L.

In only one cultivar out of 1200 investigated cultivars ofPhaseolus vulgaris L. could we find an extreme change in the pattern of reserve proteins on the cathodic side: one of the proteins, called protein I, is completely absent in the cultivar 'Krupnaya sakharnaya' and is replaced on the same site by another protein,i.e. a protein completely different in its immunochemical specificity. The case is of interest from both the phylogenetic and systematic viewpoints and deserves further attention.

Translocation of14C-abscisic acid from roots into the aboveground part of pea (pisum sativum L.) seedlings

The translocation of14C-ABA from roots into other parts of the plant was followed in intact and decapitated pea seedlings. In intact plants ABA from roots was translocated above all into the apical part of epicotyl. In decapitated plants the regulative ability of intact apex can be partly simulated by exogenous IAA. The growth of lateral buds occurring after decapitation was associated with an intensive flow of14C-ABA from roots into released lateral buds as late as 72 h after decapitation,i.e. in the stage of intensive elongation growth of buds.

The stomatal density, dimension and conductances to water vapour diffusion of sevenHevea species were studied. It was found that these parameters were significantly different between the species. However, no distinct relationship between stomatal density or dimension and stomatal conductances could be discerned.

Dimethyl sulfoxide: A solvent for cytokinins in theAmaranthus bioassay

Dimethyl sulfoxide present in the agar medium at concentration 0.2 % (v/v) and lower does not inhibit cytokinin-induced betacyanin synthesis in theAmaranthus caudatus seedlings. The activity of kinetin, N6-(Δ2-isopentenyl)adenine andtrans- zeatin is the same when these cytokinins are dissolved in either water or dimethyl sulfoxide and incorporated into the medium after autoclaving. A simple method is described which allows the cytokinin activity of slightly water-soluble and thermolabile compounds,e.g. aromatic urea and thiourea derivatives, to be determined in theAmaranthus bioassay.

Water potential and sap flow rate in adult trees with moist and dry soil as used for the assessment of root system depth

Sap flow rate (Qw) and leaf water potential (Ψw.leaf) in adult specimens of birch (Betula) and oak (Quercus) were measured under contrasting soil moisture conditions (Ψw.sofl). With sufficient soil moisture Qw reached about 250 cm3h-1 calculated per unit tree-trunk segment as given by 1 cm length of its circumference. In soil water-stress conditions (when Ψw.leaf = = -15 × 105Pa), birch stopped transpiration and wilted. Oak transpired even when Ψw.leaf fell below -20 × 105Pa. The relation between Qw and Ψw.leaf was always linear and with various Ψw.soil differed in the slopes of regression lines only. Hydraulic conductance (Kwcu) with nonlimiting moisture conditions reached about 6 × 10-9m3 10-5Pa-1s-1 and "conductivity" ("kwa") when calculated per leaf area unit reached about 23 m 10-5Pa-1s-1. Kwcu and "kwa" were of about one half to nine times greater in birch than in oak. On the basis of relations between Ψw.soil at various depths, Ψw.leaf and Qw (resp. Kw) it is possible to assess the maximal rooting depth and the effective depth where the maximum of absorption of roots occurs. It is to be seen that the root system macrostructure substantially participates in the drought avoidance of adult trees in a forest stand.

Cytokinin-like activity in sea water andFucus vesiculosus L

Cytokinin activity was demonstrated in the Baltic sea waters. This activity was always higher in the near-bottom water, taken from theFucus vesiculosuszone, than in the superficial zone. A cytokinin-like substance showing properties typical of 6-(3-methyl-2-butenylamino)purine (2iP) was also present in extracts from the thallus ofFucus. It was found that exogenously applied cytokinins, in some concentration ranges, markedly increased the number of adventitious branches formed on the cut surface of the thallus fragments. The growth responses of the investigated plant tissues to the different cytokinins varied according to season and kind of cytokinin applied. The possibility of exudation of some cytokinin like substances by the algal cells into the surrounding waters was considered.

Grain yield and ear development of spring barley as influenced by environmental conditions during early stages of plant development

Barley plants were grown until day 21 under conditions which were different in relation to photon flux density, carbon dioxide concentration and temperature. Dry weight and leaf area increase from day 7 until day 21, shoot apex development between day 15 and day 47, and yield of each treatment group were considered. Photon flux density was demonstrated to have a greater influence on net assimilation rate (NAR) of young plants than has carbon dioxide enrichment. High temperature treatment seems to influence NAR less than growth and developmental processes. Grain yield of high temperature treated plants was significantly lower than that of the other treatment groups. Significant correlations have been found between growth analysis values of young plants and some yield components of each treatment.

This paper describes a procedure for isolation and preservation of mesophyll cells, adapted toValerianella olitoria. We have tested the best conditions of temperature, pH, enzyme concentration, plasmolysis and the action of several adjuvants, dithioerythritol and mineral nutrients. The method described gives a high yield of viable cells. The ability of isolated cells and cellsin situ to take up methyl-glucose is compared.
