Comparison of seed proteins of some representatives of the genusPisum from the point of view of their relationship comparison by disc electrophoresis

Ten taxa of the genusPisum were examined by disc electrophoresis in gels according to Davis and to Reisfeldet al. For evaluation of band patterns the Jaccard Index was applied.
The results in both types of gels show thatPisum abyssinicum and especiallyP. fulvum have biochemically a relatively isolated position.Pisum elatius and its subspeciescaspicum andpalestinicum form a subgroup withP. cinereum;P. sativum var.zeylanicum and cv. Jupiter form another subgroup withP. syriacum.
Our results are in good agreement with the results of Przybylskaet al. (with the exception ofP. cinereum) and also with immunoelectrophoretic analyses performed by Turkováet al. (1980), with the same exception.

Adaptability ofMentha piperita l. to irradiance. Growth, specific leaf area and levels of chlorophyll, protein and mineral nutrients as affected by shading

Height growth ofMentha piperita L. is better at 44% than 100% daylight. At 14% daylight, growth is limited, probably by lack of photosynthate, and no flowering occurs. Specific leaf area and protein content increase significantly with the decrease of irradiance. Shade leaves contain more chlorophyll and potassium than sun leaves. Sun leaves show higher levels of calcium because of their more xeromorphic structure. Transfer experiments between light and shade indicate thatM. piperita responds very effectively to irradiance also in a late stage of the life cycle with changes of specific leaf area and chlorophyll content.

Study of xylem pressure potential daily dynamics by means of autocorrelation analysis

The xylem pressure potential (Ψxylem) of the leaves ofQuercus cerris, Acer campestre andCarpinus betulus was measured under anticyclonic weather types. The autocorrelation analysis revealed the daily course of the Ψxylem values approaching the stationary random process. A close statistical relation was found between the results obtained in three successive measurements of the Ψxylem (interval 2 h). A close statistical relation also between the value of the base potential (Ψb) measured at dawn and the actual values of the Ψxylem allowed the prediction of the Ψxylem values on the base of the known Ψb-values by means of a simple linear regression model.

Esterase ontogeny in cotton fibre

Growth parameters and esterase isoenzyme patterns were studied in developing cotton fibre. Fibre length and dry weight when plotted against boll age showed four distinct stages; (i) initiation, (ii) elongation, (iii) secondary thickening, and (iv) maturation. Stage specificity and changing intensities of esterase isoenzyme suggested a finely modulated control of gene expression during fibre development.

Effect of phosphorus and nitrogen on growth, chlorophyll, amino nitrogen, soluble sugar contents and algal heterocysts of water fernAzolla pinnata

The P uptake inAzolla pinnata in mineral medium increased linearly with increasing phosphate doses and optimum growth and chlorophyll content of fern were recorded at 1.2 mM of K2HPO4. The higher doses of phosphate reduced growth and chlorophyll, but an increase in soluble sugar and amino nitrogen was noticed. Addition of combined nitrogen sources decreased the fern growth, soluble sugar, chlorophyll and heterocyst frequency but amino nitrogen was comparatively higher. The growth and chlorophyll content were always high when the fern was grown in the absence of added nitrogen sources; this supports the thesis about the specific importance of symbiosis for the nutrition of fern with nitrogen which cannot be replaced by mineral compounds.

Antiviral activity of substituted 5-chlorodiphenylamine-2-carboxylic acid hydrazides and their N-benzylidene derivatives

The substituted 5-chlorodiphenylamine-2-carboxylic acid hydrazides and their N-benzylidene derivatives were tested for their antiviral activity against gomphrena mosaic virus in a hypersensitive hostin vitro as well asin vivo. Most of the compounds exhibited potential antiviral activity. These compounds may be useful in controlling viral infections in garden as well as in field crops.

Some pea cultivars as local lesion hosts for potato virus X

SixteenPisum sativum cultivars were mechanically inoculated with two isolates of PVX. Distinct necrotic local lesions on the inoculated leaves were found in six cultivars 5 to 6 days after inoculation. The best results were obtained with the Meteor and Raman cultivars, 17 to 23 days old.

A Prague isolate of wisteria vein mosaic virus

An isolate of wisteria vein mosaic virus (WVMV) obtained fromWisteria sinensis in Prague resembled in its properties WVMV isolates described in Italy and Holland.Nicotiana megalosiphon is reported as a new host of WVMV. Other known host plants showed reactions similar to those described formerly. The incubation period extended in some hosts up to two or four weeks. Pea plants showed symptoms within five to seven days. Species ofApium, Brassica,Datura and others were not susceptible. TIP of Prague isolate of WVMV was 61 °C, at a dilution 1: 5000 47% of plants were infected, and 48 h > LIV > 28 h. Modal particle length was 743 nm. Massive granular inclusions were seen in pea epidermal cells, usually adhering to nuclei that did not show alterations. No serological relation to turnip mosaic virus could be established.

Utilization of exogenous sugars by excised maize embryos in culture

Sucrose was markedly superior to fructose and glucose in promoting growth of plantlets from immature maize embryos. The elongation of roots is shown to be more sucrose dependent than that of shoots. On the other hand, the exogenous sucrose was less effective than fructose as substrate for carbohydrate catabolism and for the synthesis of alcohol-insoluble compounds at the beginning of embryo cultivation. The absorbed fructose was found to be rapidly converted to sucrose and the level of endogenous sucrose derived from sugar supplied to the medium was higher in fructosethan in sucrose-fed embryos. The preferential utilization of fructose over sucrose, however, declined with the progress of germination which may be related to the decrease in proportion of scutellum in total mass and physiological activity of the embryo.

Nucleases in chloroplast of barley

Two barley chloroplast nuclease fractions were separated by the affinity chromatography and gel electrophoresis. Both were about 2 times more active to RNA than to native DNA and about half as active to denaturated DNA as to native DNA. Both fractions were as active to UV-irradiated (270 J m-2) native DNA as to intact DNA but their action was inhibited by apurinic sites. The enzyme activities were inhibited by high concentrations of EDTA, NaCl, Mn2+, Ca2+, Zn2+ ions and by N-ethylmaleimide. They do not require Mg2+ ions but are stimulated or at higher concentration inhibited by their presence. Both RNase and DNase were active over a wide pH range (5.5-9), the optimum for DNase action in the presence of Mg2+ being 6.5, for RNA decomposing activity at pH 8.0. As no mononucleotides were detected in acid soluble form, it seems likely that DNase acts in the endonucleolytic way.

Mycoplasma-like organisms inLupinus hybrids showing witches' broom symptoms

A previously undescribed disease onLupinus hybrids manifesting typical witches broom symptoms occurred spontaneously in a locality in North Bohemia. Broomed shoots appeared only in late summer and resulted from the abnormal proliferation of dormant axillary buds in leaf and fruit axils. The shoots were bushy and their leaves were dark green and not reduced in size. Electron micrographs of ultrathin sections showed the presence of numerous mycoplasma-like organisms (MLO) in diseased but not healthy phloem tissues.

Fluctuations of nitrogenase and cytosol glutamine synthetase activity in pea (Pisum sativum L.) nodules in the course of vegetation

Changes in glutamine synthetase activity located in the cytosol of root nodules were followed in pea (Pisum sativum L.) plants in relation to their nitrogenase activity. The highest glutamine synthetase activity was found in young nodules (15 days after inoculation) and its changes in 17-to 45-day-old plants showed a positive correlation with nitrogenase activity. In contrast to nitrogenase activity, changes in glutamine synthetase activity during the day and night period could not be unequivocally interpreted in terms of diurnal fluctuation.

Mitodepressive effects ofChelidonium alkaloids on root tip cells ofAllium cepa L

A comparison of the effects of a water extract of the fresh vegetation apex ofChelidonium majus L. var.majus, with those of berberine sulphate and of chelidonine hydrochloride is reported. The water extract of fresh stems and leaves ofChelidonium majus and the berberine sulphate solution had marked mitodepressive effects on onion root tip cells. Chelidonine hydrochloride had similar but less marked effects. On the basis of the results obtained it can be assumed that the most active group of substances with cytostatic effects, hindering the cells from entering mitosis, are protoberberine bases contained in the latex ofChelidonium majus. Within the range of the investigated concentrations the extract of fresh stems and leaves was less toxic for the cells than berberine sulphate. The data ascertained provide evidence that the mechanism of the cytostatic action of chelidonine, of berberine, as well as of the extract of fresh stems and leaves ofChelidonium, majus differs from the mechanism of the action of colchicine. Of the testedChelidonium alkaloids only chelidonine produced a partial inactivation of the mitotic spindle.

Using immunochemical methods a comparison was made of a complex of water soluble (albumins) and salt soluble (globulins) seed proteins, especially vicilin and legumin, in selected species of the genusPisum, to determine the degree of their taxonomic relationship.
Within the genusPisum the interspecific differences betweenP. abyssinicum, P. cinereum,P. elatius, P. fulvum, P. sativum, andP. syriacum are much smaller, and thus the taxonomic distances are shorter than is the casee.g. in the genusPhaseolus. In spite of this fact one may state thatPisum sativum, P. elatius andP. syriacum constitute a definite group, whereasP. abys-sinicum, P. cinereum and P.fulvum have longer taxonomic distances.

Histoenzymological compartmentation of β-glucuronidase in the germinating pollen grains ofPortulaca grandiflora

The contribution deals with the distribution of β-glucuronidase in the germinating pollen grains ofPortulaca grandiflora. In non-germinating pollen grains the enzyme is localized in the pollen wall; the cytoplasmic activity is subdued. With the initiation of germination, the activity of enzyme increases and the positive granules are richly packed in the pollen grains and pollen tubes. The stigma hairs also have such an activity. The functions of the enzyme in the metabolism of germinating pollen grains are discussed.

Host range and symptom differences between isolates of turnip mosaic virus obtained fromSisymbrium loeselii

Twenty-four isolates of turnip mosaic virus (TuMV) from spontaneously infectedSisymbriutn loeselii plants were collected in Bohemian localities. Single lesions on leaves ofNicotiana tabacum cv. Samsun served for inoculating petunia plants used as infection sources for twelve host species. In no case were two identical isolates obtained. 15 isolates could be transmitted toVicia faba, a new TuMV host. Almost all isolates infectedPhaseolus vulgaris cv. Prince locally andTrifolium incarnatum systemically. Seven isolates were transmissible toPisum sativum. No substantial differences between isolates were observed with infectedAn aranthus caudatus, Chenopodium quinoa, Datura innoxia andSinapis alba plants. Several isolates could not infectNicotiana glutinosa at all, other isolates caused in it latent systemic infection and some isolates only local infection contrary to normal local and systemic infections ofN. glutinosa. Attempts to transmit one isolate to cereals failed.

Proteolytic activity and nitrogen transfer in maize seeds during imbibition

Ethanol-soluble and insoluble nitrogen and protease activity in maize seeds during imbibition period of 6 to 60 h at 30 ± 2 °C were determined both in light and in the dark. In light, soluble and insoluble nitrogen in the embryo were similar to that in the dark. But the increase in soluble nitrogen in the endosperm up to 38 h was higher in light than in the dark. Decrease in insoluble nitrogen was correlated with increase in soluble nitrogen, the level always being higher in the dark. Light increased protease activity also in the endosperm. Among various light qualities, red light was most effective in inducing proteolysis, and loss of nitrogen from the endosperm. Further, the growth and organic nitrogen of primary leaves from seedlings raised from light pretreated seeds were better than those from dark pretreated ones.

Transport of benzyl-8-14C-adenine in pea seedlings in relation to stem apical dominance

In intact, decapitated and decapitated indole-3-acetic acid (IAA) treated pea seedlings the translocation of benzyl-8-l4C-adenin (14C-BA) from the roots was studied with regard to the release of lateral buds from apex-induced inhibition. In intact plants (controls) a substantial part of the activity was found in the apical part of the epicotyl. Decapitation resulted in the initiation of growth of lateral buds. As early as 24 h after decapitation and application of14C-BA a significantly higher activity was found in growing lateral buds (cotylars) of decapitated plants than in inhibited ones of intact or IAA-treated decapitated plants. The accumulation of14C-activity in stump tops of decapitated plants treated with IAA was associated with the thickening growth.

The reduction of aldehydes by broad bean and maize alcohol dehydrogenases and a study of the substrate binding to the enzyme protein

Alcohol dehydrogenase was prepared from 2-day germinating maize and 3-day germinating broad-bean seeds by ammonium sulphate fractionation of sodium phosphate extracts, chromatography onDEAE cellulose and Sephadex G-200. The activity of the broad beanADH amounted to182 800 units per mg protein, that of maizeADH 79 000 units per mg protein. Besides oxidation of a series of alcohols at pH optimum in the alkaline region and with KM equalling 10-2M, alcohol dehydrogenases isolated from both plants catalyze the reduction of acetaldehyde, n-propanal, n-butanal, isobutanal and crotonal at pH optimum in the neutral region with KM equalling 10-3M. The inhibition studies using fatty acids and chloride ions revealed that the oxidation of alcohols is inhibited competitively by both types of inhibitors, with inhibition constants of 10-2M and 10-1M, respectively. The inhibition in the presence of acetaldehyde is non-competitive since the inhibitors do not compete with acetaldehyde and do not form an enzyme-NADH-inhibitor complex, yet they obviously react with the enzyme-NAD product only, thus giving rise to an enzyme-NAD-inhibitor complex. These differences in the behaviour of inhibitors may be interpreted in the sense that the binding sites of ethanol and acetaldehyde as substrates for broad bean and maize alcohol dehydrogenases are non equivalent. The nonequivalency discussed in the text.

Oxidation of glutathione by peroxidase isoenzymes in fenugreek

During the germination of fenugreek (Trigonella foenum graecum L.) sulfhydryl groups rapidly declined in cotyledon and seedling axis, while peroxidase activity increased. Studies on purified isoenzymes showed that GSH was oxidized by the isoenzymes and was accomplished in presence of cofactors, Mn2+ and DCP along with H2O2 (0.01 mM). This reaction was found to be peroxidatic in nature. The oxidation was inhibited by catechol but was enhanced by malic acid.

The effect of cultivation temperature on nitrate reductase, glutamine synthetase and glutamate dehydrogenase levels in isolatedPisum sativum roots

Nitrate reductase (NR), glutamine synthetase (GS), and glutamate dehydrogenase (GDH) levels are differently influenced by cultivation temperature, NR level being influenced most and GDH level least. The differences caused by cultivation temperature are more pronounced in roots cultivated without sucrose in which a slower decrease in NR and GS levels and a lower increase in GDH level occur at 14 and 18 °C than at 24 °C in comparison to roots cultivated with sucrose. Sugar consumption also tends to be slower at 14 °C than at 24 °C in roots cultivated without sucrose.
