Growth and nutation parameters of primary root of pedunculated oak (Quercus robur L.)

The elongation of primary roots of pedunculated oak (Quercus robur L.), which is associated with nutation oscillations, was registered by a time lapse cinematography. The following characteristics were expressed graphically: relative growth rate [mm h-1], frequency [rev. h-1] and amplitudes [mm] of nutation oscillations, their direction and location of nutation curvatures. The growth-oscillation mechanism is interpreted as a part of the geocontrol system of the root and was evaluated on the basis of its physiological efficiency. Comparison of nutation periods showed that the control system of pedunculated oak is three times more and two times less efficient than that of pea and maize, respectively. Comparison of the oscillation amplitudes of growing roots gives similar results.

Water stress alters characteristics of acid phosphatase in wheat

Effect of water stress at -1, -3, and -5xl05Pa osmotic potential on some properties of acid phosphatase has been studied in two wheat cultivars. Acid phosphatase from S-308 cultivar, which is less drought resistant than C-306, was less susceptible under varying conditions of pH and temperature treatments.

The growth of callus tissue cultures and the infectivity of twenty fiveSolanum laciniatum Ait. plants and of sixteenNicotiana tabacum L. cv. Samsun plants were investigated. The plants were obtained from callus tissue cultures derived from stem pieces of the respective plants infected with a mycoplasma-like organism (MLO) evoking potato witches' broom. The tissues were cultivated on synthetic nutrient medium with kinetin and IAA. Allde novo obtainedS. laciniatum plants were healthy. On the contrary twelve of the sixteen reconstituted tobacco plants showed MLO presence.
Summarizing these and previous results, the authors suppose that the most important factor influencing MLO persistence in callus tissues cultivated on the applied nutrient medium may be the callus growth rate and the organogenesis set. Both these conditions are determined by the metabolism of the investigated plant species.

The localization of starch in root tips

As an extension of earlier studies performed in our laboratory on enzyme localization on tissue level, the distribution of starch was examined in seedling root tips of peaPisum sativum L., broad beanFaba vulgarisMoench., maizeZea mays L., lupinLupinus albus L., and pumpkinCucurbita maximaDuch., and in tips of adventive roots of onionAllium cepa L. by means of the alcian blue /JJK procedure applied to paraffin sections. In pea, some genetic (various cultivars) and eco-physiological (different harvests, various localities, diverse way of germination, uneven seedling age, unequal stage of cell growth) factors were studied concerning the present problem. In all these cases the starch localization pattern remains constant though the amounts of starch vary in particular cell complexes. From the evaluation of the starch localization at the level of the beginning cell growth in particular species and from comparison of the findings in different species it follows that starch localization and histogenesis patterns are different. This supports the view that the two phenomena are caused or controlled by uneven factors.

The Influence of hydroxyurea on DNA synthesis and germination of seeds ofAgrostemma githago andVaccaria pyramidata

Application of hydroxyurea to intact seeds ofAgrostemma githago as well asVaccaria pyramidata causes a strong inhibition of radicular3H-thymidine incorporation into DNA, but germination rate is only insignificantly reduced. The seedling development, however, is strongly affected. Consequently, DNA synthesis is not a prerequisite for the radicle protrusion.

The sequence and duration of mitotic cycles in the apical root meristem ofVicia faba l

A study was made on the mitotic cycle times in meristematic cells of Vicia faba root tips and on the relationship between their duration and the position of a certain cell in the column of proliferating cells. For the demonstration of the sequence and duration of mitotic oycles a single-column model was used. The results of experiments show that the great variability in the duration of mitotic cycles (from 12 to l20 h)is the result of a different program of the apical meristem cells. The long duration of the cycle of initials corresponds to the sum of cycle times of their descendants.

Acid and alkaline inorganic pyrophosphatase activity after treatment with various herbicides

Various herbicides are applied on seventeen-day-old cucumber seedlings growing in a nutrient solution.
After 1 and 3 days, acid and alkaline pyrophosphatase activities are measured in the leaves and the roots of seedlings treated with lethale dosis of atrazine, chlortoluron, ioxynil, MCPA, dicamba, alachlor. The same enzymes are determined after 5 h in seedlings on which previously paraquat and DNOC were pulverised. In the leaves, acid pyrophosphatase activity is stimulated by almost all the herbicides, especially by ioxynil, paraquat and DNOC while alkaline pyrophosphatase remains similar to the controls. The alkaline and acid pyrophosphatase activity in the roots is either unchanged or stimulated or inhibited without any correlation with the various modes of action of the herbicides.

Studies on the morphogenetic response of maize tissue cultures of different origin

The participation of the genotype and of organ specifity effect in the quality of morphogenetic response (callogenesis, bud and root formation) of primary maize explants has been investigated. The presence of synthetic auxins - especially 2,4-D at 1 to 5 mg 1-1 conc. - in cultivation medium was essential for both callus formation and continuous growth of tissue and suspension cultures. Anatomic structure of callus cultures is permanently heterogeneous, their growth is ensured by the action of meristems of the type found in root tips, and by repeated callogenesis from malformed roots. Adventive buds and plants could be regenerated only from cultures of embryonal origin (of one line). The presence or absence of the endosperm gene "opaque" did not influence callogenesis intensity in cultures of isolated embryos; however the morphogenetic response was clearly "line specific".

The regulation of glutamate dehydrogenase isoenzymes by nitrite in isolated pea roots

Unmetabolized nitrite occurs in 3 cm long apical segments of primary pea roots(Pisum sativum L., cv. Raman), isolated from 6 d old seedlings, grown under aseptic conditions for 10 h in nutrient solutions containing 0.1 to 2 mM nitrite concentration, its amount being dependent on the concentration in the nutrient solution. With increasing amount of nitrite in the roots, the ratio between NAD+-GDH1 and NAD+-GDH2 isoenzymes is changed without influencing significantly the total NAD+-GDH activity. After 48 h cultivation unmetabolized nitrite occurred in the roots only in small amounts and the ratio of activities of these isoenzymes was not affected significantly. NADP+-GDH activity, detected in the gels at the same site as NAD+-GDH1, was depressed by increasing nitrite concentrations.

Nutation rhythm of growing pine hypocotyl (Pinus silvestris L.) interferred with a phototropic stimulus

The interaction of nutation oscillation and phototropic curvature mechanisms of growing pine hypocotyl(Pinus silvestris L.) was investigated. Nutation parameters of growing hypocotyl are not affected by continuous lateral illumination. Frequency and nutation amplitudes undergo changes which are dependent on plant development and are nearly the same when plants are illuminated from one side or from above. Lateral illumination, however, induces a phase shift. This deviation from a normal nutation rhythm usually disappears after a period of time equal to three nutation revolutions,i.e. after 12 h.
The positive phototropio response of nutating hypocotyl is dependent on the phase in which the hypocotyl is exposed to the lateral illumination:(a) nutation perihelium,(b) ahelium and(c) nutation interphase. These response-variants are compared with the expected gradients of plant hormones at the opposite sites of the hypocotyle. Analysis of location of the individual movement reactions(i.e. oscillations as a component of the geocontrol system and phototropic curvature) together with the determination of the zone of maximum elongation indicates the existence of different receptory centres of these two movement systems. In the case of the phototropic response reaction it is possible to consider the presence of different photo- and biochemical mechanisms at least for a part of this system.

A macerating ammonium oxalate fixative and schedule for some root meristems

0.25 % ammonium oxalate in a mixture of ethyl alcohol, chloroform, formaldehyde, acetic acid and hydrochloric acid works as a very fast and efficient macerating fixative for the root meristems ofAllium cepa, Allium sativum, Ornithogalum thyrsoides, Pisum sativum andVicia faba. Slicing the tip region of excised meristems hastens spindle poison pretreatment, fixation and staining. Owing to the dissolution of the middle lamella and eliminating a physical barrier to permeability an extremely rapid fixation and staining schedule involving several stains has thus been developed.

Response of "storage" effects in barley seeds to the dose of ethyl methanesulphonate and X-ray treatment

1. Ethyl methanesulphonate (BMS)-treated barley seeds stored at 30 % water content for 1 - 4 weeks are capable of recovering from the mutagen induced injury. The level and speed of the recovery depends on the EMS dose and storage period. Seeds treated with extremely severe EMS doses reducing the% of germination to 5 - 10 % cannot recover from the induced injury. 2. Due to a 1 to 6 weeks storage of EMS-treated seeds with 20 % water content the amount of induced injury increases in dependence on the applied EMS dose and storage period. Doses of EMS, reducing the seedling height in the M1 generation to 90 - 95 % of the control reveal no storage after-effects. 3. Storage of X-ray exposed seeds at 20 % or 30 % water content did not result in any significant changes in the yield of induced genetic damage.

The role of gibberellic acid (GA3) in the removal of dormancy inFraxinus excelsior L. seeds

The seeds ofFraxinus excelsior L. were stratified at 17-20 °C (warm stratification), at 4-6 °C (cold stratification) and at alternating temperature (warm - cold stratification). The seeds subjected to warm stratification only, remained dormant. The seeds stratified only at 4-6 °C germinated gradually during a long period of time. The seeds subjected to warm - cold stratification, however, germinated with great intensity within a relatively short period of time.
GA3 was shown to stimulate the growth of embryos markedly, and its effect on the germination of seeds depended on the temperature of stratification. GA3 applied during the cold stratification accelerated the removal of dormancy by shortening the period of stratification and by influencing the germination of seeds. The results obtained indicate a similarity between the effect of temperature 17-20 °C during the warm stratification and that of gibberellic acid. Both those factors applied separately affect favourably after-ripening of the embryos and accelerate the germination of seeds.

The volumes of anatomical components in leaves oftypha angustifolia L. andtypha latifolia L.

The volumes of tissues in leaves of two species ofTypha with different habitus were measured. It was found that air cavities occupy more than 50%, parenchyma about 20% and photosynthetic tissue about 10% of the total volume of the leaf. If the cavities are omitted, parenehyma amounts to approximately 50% and photosynthetic tissue to little more than 20% of the total volume of tissues. The relative volumes of tissues vary continuously from the base to the tip of the leaf. The differences between the two species studied were not large. Besides the relative volumes, the real ones of leaves and tissues were calculated.

Proteinase inhibitors in meal ofAnacardium occidentale seeds

Trypsin and chymotrypsin inhibiting activities were detected in a "crude inhibitor" fraction of a lipid free cashew nut (Anacardium occidentale L.) meal. Both activities were shown to be heat resistant and not affected by pepsin at pH 3.0. Molecular weight distribution of tho trypsin inhibiting activity is similar to the known range of molecular weights for trypsin inhibitors of plant origin.
A "crude extract" of the cashew nut meal was shown to contain strong L-leucine-p-nitro-anilide and moderately strong α-N-benzoyl-DL-arginine-p-nitroanilide hydrolyzing activities. This last activity was strongly inhibited by the "crude inhibitor" fraction shown to contain trypsin and chymotrypsin inhibiting activities. The presence of this endogenous inhibitor-endo-peptidase system seems to indicate that the protein proteinase inhibitors of plant origin could be involved in the control of protein mobilization in the seed.

The fluctuations in the level of endogenous growth regulators in seeds ofAcer pseudoplatanus L. in the course of stratification

In the course of cold stratification ofAcer pseudoplatanus L. fruits a statistically significant alternation occurs in their seeds of a rise and fall in the level of endogenous growth regulators. In the initial weeks the inhibitory effect slightly declines, or, on the contrary, the stimulatory effect slightly increases; in the middle phase of stratification a marked increase in inhibitions and reduction of stimulations appears, and towards the end of stratification the stimulatory effect of isolated substances in the individual biotests rises again, or their inhibition effect is decreased. No direct dependence was found between the decrease of the degree of dormancy and the drop of inhibitor, or increase of promotor levels. However, a certain analogy was observed between the time course of fluctuations in the level of growth regulators and the germination energy of the seeds investigated. An enhancement of the growth activity of the substances isolated (e.g. those of a gibberellin-like nature) in the last weeks of stratification can already be considered as the result of the release of fruits from dormancy.

Detection of vitamin B12 and pantothenic acid in cell exudates of blue-green algae

The blue-green algaeChroococcus, Oscillatoria and Nostoc, excrete vitamin B12 and pantothenic acid in the culture filtrates during the phase of active growth. The excretion has an ecological significance.

Nitrate reductase, nitrite reductase and glutamate dehydrogenase levels in roots and leaves of maize seedlings

Total activities of nitrate and nitrite reductases were higher in 4 to 20 day old maize plants in the leaves than in the roots. The ratio of activities found in the leaves and in the roots respectively was much higher in the case of nitrate reductase than in the case of nitrite reductase. On the other hand higher glutamate dehydrogenase activity in the roots than in the leaves clearly indicates that the roots play a more important role in the assimilation of ammonium than in the assimilation of nitrate. When comparing the distribution of seminal and nodal adventitious roots of maize seedlings with the assimilation of inorganic nitrogen on the basis of enzyme levels, it could be deduced that during the first 20 days of seedling growth seminal roots were more involved in the assimilation of nitrate whereas nodal adventitious roots were more active in ammonium assimilation.

Physiological mechanisms of frost tolerance: subcellular localization and some physical-chemical properties of protein fractions accumulated under cold treatment

Two soluble protein fractions were found to increase under cold treatment in winter rape plants. They were the proteins of low molecular weight, impoverished in proline and methionine residues and localized in the citosol subcellular fractions. They seem to be linked in "situ" with the hydrophobic compounds of cytoplasm. Owing to those properties, they seem to be more resistant against salt-induced aggregation which may occur at freezing or at A01GP096.
