These treatments enhanced the contents of ABA and expression of genes coding 9-cis-epoxycarotenoid dioxygenase 1 (NCED1) and basic leucine zipper (b-ZIP).expression of genes coding 9-cis-epoxycarotenoid dioxygenase 1 (NCED1) and basic leucine zipper (b-ZIP). Simultaneous treatment of potato plants with Cd and fluridone (Flu), an inhibitor of ABA biosynthesis, completely halted the Cd-induced transcription of StPCS1, NCED1 and StbZIP genes and limited the increases in PCS activity and ABA content. The data suggest that ABA participates in transduction of the Cd signal to the cells of potato roots. |
Reduced protein secretion and glycosylation induced by ammonium stress inhibits somatic embryo development in pumpkinA. Crnković, R. Garić, D. Leljak-Levanić, S. MihaljevićBiologia plantarum 58:209-217, 2014 | DOI: 10.1007/s10535-014-0400-2 Extracellular proteins and glycoproteins secreted by ammonium- or auxin-induced somatic embryogenic cultures of pumpkin were analyzed. Despite an overall similarity in developmental characteristics between these embryogenic cultures, distinct expression patterns of extracellular proteins and glycoproteins were observed. Ammonium, when supplied as the sole source of nitrogen, caused acidification of the culture medium and significantly reduced protein secretion. Buffering pH in the ammonium-containing medium restored extracellular protein secretion and glycosylation and an enhanced cell aggregation but not the development of later embryo stages. As revealed by Concavalin A (Con A) immunodetection, extracellular glycoproteins containing α-D-mannose and α-D-glucose were most abundant in proembryogenic cultures grown in a buffered ammonium-containing medium and in a medium supplemented with 2,4-dichlorophenoxyacetic acid (2,4-D). We assume that extracellular proteins (Mr 28, 31, and 44 kDa) and Con Abinding glycoproteins (Mr 26, 30, 40, 53, and 100 kDa) found in both proembryogenic cultures may have a role during somatic embryogenesis induction. The glycan components of proteins were further characterized by affinity blotting with different lectins. Binding patterns of mannose-specific lectin from Galanthus nivalis partially correlated with those detected with Con A, whereas no signal was observed with lectins from Datura stramonium and Arachis hypogea regardless of the treatment applied. Results indicate that complex N- or O-glycans are not typical for early phases of pumpkin embryo development. The accumulation of extracellular glycoproteins with high-mannose-type glycans from 30 to 34 kDa, observed after the transfer from the ammonium- or 2,4-D-containing media into a maturation medium, appeared to be associated with development of later embryo stages. This study also revealed the presence of EP-3-like endochitinases in pumpkin embryogenic cultures, particularly in cultures grown in the buffered ammonium-containing medium, however, these proteins should be examined further. |

Most cereals are poor in n-3 and n-6 PUFAs. Using biolistics, barley (Hordeum vulgare L. cv. Golden Promise) was transformed with an artificial gene encoding Δ6-desaturase (D6D) under an endosperm-specific promoter. This artificial gene was designed from the sequence of D6D of the filamentous fungus Thamnidium elegans, but codon usage was optimised for cereals. A signal sequence from the gene encoding for high molecular mass glutenin Dx5 was added to a destinate mature protein. Successful transformation was confirmed in T0 plants at the genomic level and in T1 seeds at the transcriptomic and metabolomic levels. Transformed plants produced up to 0.141 % of γ-linolenic acid (GLA) and 0.294 % of stearidonic acid (SDA) of the total amount of fatty acids in their grains. Although the content of these fatty acids was relatively low, the current study provides the first evidence that transgenic barley can be a source of GLA/SDA. |

Mensuali-SodiBiologia plantarum 58:601-610, 2014 | DOI: 10.1007/s10535-014-0434-5 Five in vitro culture systems with different ventilation rates were used to investigate the influence of vessel environment on photosynthesis, dark respiration, ethylene evolution, and rosmarinic acid (RA) production in sweet basil (Ocimum basilicum L.) micropropagated shoots. The systems under comparison were two bioreactors with either temporary (RITA™) or stationary (Growtek™) immersion, and three types of vessels (Magenta™, Microbox ECO 2 ™, and PCCV25™) that are largely used for plant micropropagation. Shoots of green-leaved cv. Genovese and purple-leaved cv. Dark Opal were cultured on a modified Murashige and Skoog medium containing 0.25 mg dm-3 6-benzylaminopurine. The instantaneous rates of photosynthesis, dark respiration, and ethylene production were determined by gas chromatography measuring CO2 and ethylene concentrations in vessel headspaces. The tissue RA content was determined by HPLC in HCl-methanol extracts. The explant growth and morphology were significantly affected by culture conditions and cultivars. The largest biomass production was observed under the photomixotrophic culture conditions provided by Growtek™, whereas the highest RA content in shoot tissues was found in the RITA™ photomixotrophic system, where ethylene accumulated to the greatest extent. |

Zn is an essential micronutrient for all organisms and it is reasonable to hypothesize that Zn status is essential for maintaining salt tolerance in plants. In this study, the physiological and molecular mechanisms of Zn-based alleviation of salt stress in wheat seedlings were investigated. Our results indicate that sufficient Zn nutrition maintained antioxidative enzyme activities and decreased a reactive oxygen species over-accumulation in wheat seedlings. Our data also reveal that sufficient Zn nutrition improved the expression of Na+/H+ antiporter genes, TaSOS1 and TaNHX1, thereby decreasing the Na+ accumulation and subsequently improving salt tolerance in wheat seedlings. |

No. HM235775) encoding a Pollen_Ole_e_I conserved domain was identified in roots of soybean drought resistant cv. Jindou 23. In the present study, GmPOI was cloned and functionally characterized. Real-time quantitative PCR indicated that the expression of GmPOI was induced by drought, cold, salt and abscisic acid in wild-type soybean. The soybean plants overexpressing GmPOI showed higher tolerance to drought stress than wild types. We concluded that GmPOI is probably a novel gene that is involved in the response to various stresses in soybean. |

Transcriptional analysis of T2 homozygous tobacco plants overexpressing Osmyb4 revealed that Myb4 activated the transcription of several genes of the phenylpropanoid pathway such as PAL, C4H, 4CL1, 4CL2 (encoding phenylalanine ammonia-lyase, cinnamic acid 4-hydroxylase, 4-coumarate: Co A ligase1, 4-coumarate: Co A ligase2). Moreover, the Myb4 increased expression of HQT encoding hydroxycinnamoyl-CoA: quinate transferase, which specifically triggers the accumulation of chlorogenic acid (CGA). In addition, increased acccumulation of rosmarinic acid (RA) was found in transgenic plants of both species. These results open the possibility of using the Osmyb4 gene to increase the production of specific bioactive hydroxycinnamates. |

BieBiologia plantarum 58:179-184, 2014 | DOI: 10.1007/s10535-013-0349-6 Self-grafted and pumpkin rootstock-grafted cucumber plants were subjected to the following four treatments: 1) aerated nutrient solution alone (control), 2) nutrient solution with 10 mM Ca(NO3)2 (Ca), 3) nutrient solution with 90 mM NaCl (NaCl), and 4) nutrient solution with 90 mM NaCl + 10 mM Ca(NO3)2 (NaCl+Ca). The NaCl treatment decreased the plant dry mass and content of Ca2+ and K+ but increased the Na+ content in roots and shoots. Smaller changes were observed in pumpkin rootstock-grafted plants. Supplementary Ca(NO3)2 ameliorated the negative effects of NaCl on plant dry mass, relative growth rate (RGR), as well as Ca2+, K+, and Na+ content especially for pumpkin rootstock-grafted plants. Supplementary Ca(NO3)2 distinctly stimulated the plasma membrane (PM) H+-ATPase activity which supplies the energy to remove excess Na+ from the cells. The expressions of gene encoding PM H+-ATPases (PMA) and gene encoding a PM Na+/H+ antiporter (SOS1) were up-regulated when Ca(NO3)2 was applied. The pumpkin rootstock-grafted plants had higher PM H+-ATPase activity as well as higher PMA and SOS1 expressions than the self-grafted plants under NaCl + Ca treatment. Therefore, the addition of Ca2+ in combination with pumpkin rootstock grafting is a powerful way to increase cucumber salt tolerance. |

SadderBiologia plantarum 58:419-426, 2014 | DOI: 10.1007/s10535-014-0421-x VERNALIZATION INSENSITIVE 3 (VIN3) is a chromatin remodelling protein that is induced by low temperatures and is required for the vernalization response in Arabidopsis thaliana. VIN3 is one of the polycomb group (PcG) proteins, which mediates epigenetic repression of FLOWERING LOCUS C (FLC) in A. thaliana. Here, we present cloning, characterization, and expression of a putative SlVIN3 gene in tomato (Solanum lycopersicum L.) by isolating cDNA clones corresponding to SlVIN3 gene using primers designed based on conserved sequences between PcG genes in A. thaliana and tomato. The SlVIN3 cDNAs were cloned into a pBS plasmid and sequenced. Both 5' and 3' RACE were generated and sequenced. The flcDNA of 2 823 bp length for the SlVIN3 gene was composed of 5'UTR (336 bp), ORF (2 217 bp), and 3'UTR (270 bp). The translated ORF encoded a polypeptide of 739 amino acids. Alignment of deduced amino acids indicates that there are highly conserved regions between tomato SlVIN3 predicted protein and plant VIN3 gene family members. Both unrooted phylogenetic trees constructed using the maximum parsimony and maximum likelihood methods indicate that there is a close relationship between SlVIN3 predicted protein and VIN3 protein of Vitis vinifera. The expression of SlVIN3 gene remained high during floral organ differentiation and growth and decreased when the fruit started to develop. |

The activities of enzymes including catalase (CAT), guaiacol peroxidase (GPX), ascorbate peroxidase (APX), superoxide dismutase (SOD), glutathione reductase (GR), lipoxygenase (LOX), and phenylalanine ammonia lyase (PAL) were measured. In addition, the content of H2O2, ascorbate (ASC), dehydroascorbate (DHA), reduced glutathione (GSH), chlorophyll (Chl) a+b, carotenoids, proteins, malondialdehyde (MDA), membrane aldehydes, and electrolyte leakage (EL) were determined. Results suggest that the excess of Ni increased the content of H2O2, MDA, membrane aldehydes and proteins in roots as well as GPX, LOX, APX activities, and EL in leaves, whereas Ca and His ameliorated these effects. Moreover, decreasing leaf GSH and DHA content and GR activity were observed under the Ni stress, but these parameters were raised by Ca plus His treatment. However, no improvement in leaf protein, ASC, root GSH content, and activities of PAL and CAT were observed by using Ca or His under Ni stress. |

The growth was inhibited and chlorophyll content, net photosynthetic rate, stomatal conductance, intercellular CO2 concentration, probability that a trapped exciton moves an electron into the electron transport chain beyond QA - (ETO/TRO), performance indexes (PIABS), and amount of active reaction centres per excited cross section (RC/CSO) were decreased with the increasing Cd concentration. The leaf reflectance increased in visual range (500-670 nm) and decreased in near-infrared range (750-1 000 nm). The accumulation of Cd in the roots was much higher than that in the stems and leaves. |
Production of triploid plants from endosperm cultures of Phlox drummondiiA. Razdan Tiku, M. K. Razdan, S. N. RainaBiologia plantarum 58:153-158, 2014 | DOI: 10.1007/s10535-013-0372-7 Triploid plants of ornamental Phlox drummondii Hook. were raised from cultures of endosperm excised from immature fruits having zygotic embryo at early dicotyledonous stage. Endosperm tissue was firstly cultured with the embryo on the Murashige and Skoog's (MS) medium supplemented with 5 μM 6-benzylaminopurine (BAP) + 10 μM α-napthaleneacetic acid (NAA) for 7 d and recultured after the embryo was removed. A friable callus appeared two weeks after removal of the embryo and it became compact callus mass in another three weeks. Upon transfer of this 5-week-old callus to the MS medium with 10 μM BAP + 2.5 μM indole-3-acetic acid (IAA), maximum percentage of green nodular shoot buds appeared from which regenerated dwarf shoots. Elongation of the dwarf shoots, however, required transfer of the individual dwarf shoots excised from the callus on the fresh medium and best results achieved on medium with low concentration of IAA (0.5 μM) in presence of 10 μM BAP. The shoots were then rooted in vitro and plants subsequently established in pots containing soil. Over 70 % of plants were triploid with a chromosome number of 2n=3x=21. Size of stem, leaves, flowers, pollen, and stomata of these triploid plants were higher and the plants were more vigorous as compared to naturally occurring diploid plants. In particular, flowers showed bright colour with enlarged central eye adding to their ornamental value. |

MohamedBiologia plantarum 58:265-273, 2014 | DOI: 10.1007/s10535-014-0391-z Drought is one of the main environmental stresses and many investigators identified beneficial effects of both silicon and selenium on plant growth and development. To examine the effects of Si and Se on rice (Oryza sativa L.) responses to drought, two cultivars Giza 177 and IET 1444 pretreated with 1.5 mM Si or 0.03 mM Se were then exposed to a water stress until leaf rolling was observed. The enhanced growth of Se or Si pre-treated plants was associated with a significant increase in the content of proline and glycine betaine in both shoots and roots. Furthermore, the transcription factors (TFs), dehydration responsive element-binding protein DREB2A, and NAC5 [no apical meristem (NAM), Arabidopsis thaliana activating factor (ATAF), and cup-shaped cotyledon (CUC)] were over-expressed in the drought stressed rice shoots. Notably, a pretreatment with either Se or Si significantly enhanced the expression of both TFs, DREB2A, NAC5, as well as the expression of the ring domain containing OsRDCP1 gene and some drought specific genes, such as OsCMO coding rice choline monooxygenase and dehydrin OsRAB16b. Expression of TFs and the studied genes was markedly enhanced in the Si-stressed shoots of cv. IET 1444 which favors its drought tolerance. |
Identification of a C2H2-type zinc finger gene family from Eucalyptus grandis and its response to various abiotic stressesS. Wang, X. L. Wei, L. J. Cheng, Z. K. TongBiologia plantarum 58:385-390, 2014 | DOI: 10.1007/s10535-014-0399-4 C2H2-type zinc finger proteins belong to a major family of transcription factors that play a crucial role in plant stress responses. In this study, seven C2H2-type zinc finger genes (EgrZFP1-7) in Eucalyptus grandis were cloned using the RACE PCR method. EgrZFP1-7 proteins included a QALGGH motif, two zinc finger domains, and an EAR motif. They belong to a Q-type C2H2 zinc finger protein family and are classified into the subset C1. EgrZFP4 and EgrZFP6 had a higher transcription in roots than in leaves and stems, whereas the expression of the other genes did not differ in various plant tissues. The expression of EgrZFP genes in seedlings was induced by low temperatures. Time course experiments at temperatures lower than 4 °C revealed oscillating expression of EgrZFP1-6 during the cold treatment. However, EgrZFP7 showed a phasic expression pattern at the same conditions. The expression of EgrZFP1-6 was found to be enhanced by 200 mM NaCl, whereas the expression of EgrZFP7 was inhibited. |

The responses of cucumber (Cucumis sativus L. cvs. Negin and Dominus) plants supplied with urea as sole N source to four Ni concentrations (0, 50, 100 and 200 μM) were investigated. Nickel at a 50 μM concentration stimulated growth and reduced urea accumulation and lipid peroxidation in the leaves. However, the application of 100 and 200 μM Ni reduced a shoot dry mass and increased a malondialdehyde (MDA) content. An activity of catalase (CAT) was not affected by 50 μM Ni, whereas it was significantly increased by 200 μM Ni. The application of Ni resulted in an enhancement of a guaiacol peroxidase (GPX) activity in the leaves. An ascorbate peroxidase (APX) activity was reduced by 200 μM Ni in cv. Negin and by 100 μM Ni in cv. Dominus. |

One of the characteristics of monocot species is that their embryos show only one cotyledon. In this work, the occurrence of embryos with two cotyledons and fused cotyledons in A. tequilana is reported for the first time. The occurrence of dicotyledonar embryos in a species that by definition should have only one cotyledon could bring an opportunity to elucidate the mechanisms that have given the origin to the only one cotyledon present in monocots. Syncotyly is considered in this work as the possible mechanism that gave rise to the only cotyledon mostly present in this species. |

ZhengBiologia plantarum 57:201-209, 2013 | DOI: 10.1007/s10535-012-0282-0 It is widely accepted that fruit abscission is a highly regulated developmental process that is both influenced and activated in response to changing environment and plays crucial roles in the health and reproductive success of plants. Recent evidences showed that numerous genes related to metabolic and signalling pathways were coordinately implicated in regulating fruit abscission. Cross talks within hormones, between saccharides and hormones, as well as between polyamines and ethylene result in synergetic or antagonistic interactions which together play an important role in adjusting fruit abscission. Although hormones are the most studied internal factors related to abscission, the role of saccharides and polyamines during fruit abscission is emerging now. The characterizations of the molecular mechanisms of regulating fruit abscission are essential to develop effective strategies for controlling this process in plants. |

GichnerBiologia plantarum 57:783-787, 2013 | DOI: 10.1007/s10535-013-0345-x We applied the alkaline version of the single-cell gel electrophoresis (comet assay) to seedlings of heterozygous tobacco (Nicotiana tabacum L. var. xanthi) treated with zinc acetate dihydrate (20 to 80 mM Zn2+ for 2 h or 2 to 12 mM Zn2+ for 24 h). A dose dependent increase in DNA damage expressed by the tail moment values were observed in nuclei isolated from the roots after 2 and 24 h Zn2+ treatments. In contrast, Zn2+ did not induce significant DNA damage to leaf nuclei, with the exception of 10 or 12 mM Zn2+ for 24 h. Somatic mutations, identified as dark green, yellow, and dark green/yellow double sectors on the pale green tobacco leaves were not detected after any Zn2+ treatments. The accumulation of Zn in roots and shoots was determined by inductively coupled plasma optical emission spectrometry and the Zn content in roots was about three times higher than in shoots. |

PinaBiologia plantarum 58:114-120, 2014 | DOI: 10.1007/s10535-013-0370-9 Phenylalanine ammonia lyase (PAL) is a key enzyme in the phenylpropanoid pathway responsible for biosynthesis of many secondary metabolites, such as anthocyanins, flavanols, and lignins. The objective of this work was to determine the effect of different rootstock/scion combinations on Prunus tree growth, PAL gene expression, and PAL activity in order to identify compatibility of different graft combinations. The study was performed with peach (P. persica cv. Chimarrita) grafted on two peach rootstocks (Capdeboscq and Tsukuba1) and one Japanese apricot (P. mume cv. Umezeiro). Two or three years after grafting, the growth of peach scion on Umezeiro was weak and finally the death of some trees occurred. The peach rootstocks induced vigorous growth without any tree loss through the three years of evaluation. However, PAL activity and expression of PAL encoding genes were higher in Umezeiro as compared to the other rootstocks. These results show that the differential gene expression together with the PAL activity is a promising strategy to predict graft incompatibility. |

Changes in the electric potential (EP) inside plants have been observed during the propagation of electric signals. Increasing radiofrequency electromagnetic radiation (EMR) in the environment raise the question about possible effects of EMR on the EP of plants. In the present experiment, we investigated the effect of 2, 2.5, 3.5, and 5.5 GHz EMR with a maximum field intensity of 23-25 V m-1 on the EP in emergent Myriophyllum aquaticum plants. The 2 and 5.5 GHz exposures caused significant (16 and 13 %) decreases in the standard deviation of rapid fluctuations observed in the EP. The greatest change was caused by 2.5 GHz EMR (23 % increment), although it was not statistically significant. A recovery of the EP was only after 2.5 GHz EMR exposure. The temperature of the plants was not changed by the EMR exposure. These findings confirm the frequency-dependent non-thermal effects of EMR on the EP of plants. |

Our results show that increased salinity significantly decreased the survival rate, fresh mass, content of proteins, chlorophyll a (Chl a), and chlorophyll b (Chl b), and activities of antioxidant enzymes, whereas enhanced the content of malondialdehyde. However, the treatment with sucrose significantly enhanced salt stress tolerance in the Arabidopsis seedlings by decreasing lipid peroxidation and increasing the activities of superoxide dismutase, peroxidase, and catalase, the content of proteins, Chl a, Chl b, anthocyanins, and the transcription of genes involved in anthocyanin biosynthesis. Thus, sucrose might reduce ROS-induced oxidative damage by enhancing activities of antioxidant enzymes and the content of anthocyanins, thereby preventing membrane peroxidation and denaturation of biomolecules. |

Therefore, in the present study, the nucleotide and deduced amino acid sequences of STS genes from cultivars Campbell Early and Kyoho were compared. The deduced amino acid sequences of VlKSTS12, VlKSTS11, VICESTS13, and VlKSTS1 showed 100 % homology to VlCESTS12, VlCESTS11, VICESTS24, and VlKSTS13, respectively, in Campbell Early and Kyoho. In addition, the maximum transcription was observed 6 h after chemical treatments. In Campbell Early, the transcription of PAL, CHS, and CHI was higher in leaves treated with ethephon than in those treated with hydrogen peroxide, methyl jasmonate, and salicylic acid. The PAL, CHS, and CHI genes were more induced in Campbell Early than in Kyoho. The mRNA content of STS genes started to increase at 6 h and peaked at 48 h after the treatments. In Kyoho leaves, the expression of STSs was highly up-regulated at 1 h and peaked at 6 h after the treatments. The expression of the STS genes was induced in both the cultivars in leaves inoculated with B. cinerea. STS11 and STS12 showed differential expression patterns from STS1, STS24, and STS13 in Campbell Early leaves inoculated with B. cinerea. |

In this study, the gene expression of wheat trHb (TatrHb) was analyzed under various biotic and abiotic stresses. TatrHb transcript levels increased in NaCl-treated leaves and gibberellic acid (GA3)-treated roots. In addition, sodium nitroprusside (SNP), a nitric oxide donor, induced an increase in TatrHb transcript levels in roots and leaves. A yeast two-hybrid assay (YIIH) was used to screen a hypoxia-treated wheat seedling library with the goal of determining the putative function of TatrHb. In this YIIH assay, photosynthesis-related genes that showed high homology to the Hordeum vulgare chloroplast photosystem I PSK-I subunit and Zea mays photosystem II subunit PsbS1 were detected and their interactions with TatrHb were confirmed. Subcellular localization of a TatrHb-green fluorescent protein (GFP) fusion protein and bimolecular fluorescence complementation (BiFC) assay suggested that TatrHb is involved in photosynthesis. The TatrHb-GFP fusion protein was localized in the plastids and the yellow fluorescent protein signal indicated that the TatrHb protein interacted with PSK-I and PsbS1 in the chloroplast. |

Fairy Dance), the effects of shoot physiological status, medium ingredients, and culture thermoperiod on in vitro flowering were evaluated. Shoot height, subculture media for shoot multiplication, sucrose concentration, plant growth regulators (PGRs), mineral substances in media, and thermoperiod had a significant effect on the percentage of in vitro flowering. Shoots 3 ± 0.2 or 2 ± 0.2 cm in height cultured on Murashige and Skoog (MS) medium containing 2.0 mg dm-3 6-benzyladenine (BA), 0.2 mg dm-3 α-naphthaleneacetic acid (NAA), and 20 g dm-3 sucrose were more suitable for in vitro flowering than shoots 4 ± 0.2, or 5 ± 0.2 cm in height. The most suitable sucrose concentration for in vitro flowering was 50 g dm-3 and the most suitable PGRs were a combination of 3.0 mg dm-3 BA and 0.1 mg dm-3 NAA. Increasing the potassium nitrate to ammonium nitrate ratio or increasing the phosphate concentration in MS medium had a positive effect on in vitro flowering. The percentage of in vitro flowering was significantly higher at day/night temperature of 28/20 °C than at other constant temperatures. The percentage of in vitro flowering shoots reached 68.33 % despite the occurrence of abnormal flowers and some unique developmental patterns. It makes miniature rose a potentially new in vitro experimental platform for research on the molecular mechanisms of flowering ornamental plants. |
Impact of phloem girdling on leaf gas exchange and hydraulic conductance in hybrid aspenA. Sellin, A. Niglas, E. Õunapuu, A. KarusionBiologia plantarum 57:531-539, 2013 | DOI: 10.1007/s10535-013-0316-2 We investigated phloem-xylem interactions in relation to leaf hydraulic capacity in hybrid aspen (Populus tremula L. × P. tremuloides Michx.) by using phloem girdling method. Removal of bark tissues (phloem girdling) at the branch base resulted in a substantial decline in stomatal conductance (gS), net photosynthetic rate (PN), and leaf hydraulic efficiency, and in increase of leaf water potential (ΨL). Although gS declined more than PN (83 versus 78 %), the ratio of intercellular to ambient CO2 concentrations (ci/ca) increased from 0.67 to 0.87 in three days after girdling. Girdling induced a decrease in leaf hydraulic conductance (KL) on average by 43 % (P = 0.006). The changes in gS and leaf conductance to water vapour were co-ordinated with KL only in girdled branches whereas intrinsic water-use efficiency was invariant to KL. The declines in KL with girdling were not accompanied by changes in potassium ion concentration ([K+]), electrical conductivity, or pH of xylem sap. The results suggest that phloem girdling at the branch base does not influence the recirculation of ions between the phloem and xylem in hybrid aspen and the decrease of KL in response to the manipulation is not related to changes in [K+] and total ionic content of xylem sap. |

BarakatBiologia plantarum 58:64-70, 2014 | DOI: 10.1007/s10535-013-0351-z In order to identify target region amplification polymorphism (TRAP) markers linked to three physiological traits in wheat (Triticum aestivum L.), the segregating F4 population from the cross between drought-sensitive (Yecora Rojo) and drought-tolerant (Pavon 76) genotypes was made. The parents and 150 F4 families were evaluated phenotypically for drought tolerance using two irrigation treatments [2.5 and 7.5 m3(H2O) m-2(soil)]. Using 40 different TRAP primer combinations tested for polymorphism in parental and F4 family genotypes, the results revealed that quantitative trait locus (QTL) for chlorophyll content was associated with TRAP 5, TRAP 14, and TRAP 20 and explained 18, 16, and 23 % phenotypic variation, respectively. The genetic distance between chlorophyll content QTL and TRAP 5, TRAP 14, and TRAP 20 were 12.3, 19.8, and 13.6 cM, respectively. QTL for flag leaf senescence was associated with TRAP 2, TRAP 3, TRAP 15, and TRAP 16 and explained 33, 27, 28, and 23 % phenotypic variations, respectively. The genetic distance between flag leaf senescence QTL and TRAP 2, TRAP 3, TRAP 15, and TRAP 16 were 9.4, 14.7, 18.1, and 17.3 cM, respectively. QTL for cell membrane stability was associated with TRAP 8, TRAP 9, and TRAP 37 and explained 27, 30, and 24 % phenotypic variation, respectively. The markers TRAP 8, TRAP 9, and TRAP 37 had genetic distances of 17.0, 10.0, and 9.0 cM, respectively. Therefore, these TRAP markers can be used in breeding for drought tolerance in wheat. |
Protection of photosynthesis and antioxidative system by 24-epibrassinolide in Solanum melongena under cold stressX. X. Wu, J. He, Z. W. Zhu, S. J. Yang, D. S. The eggplant (Solanum melongena L.) seedlings were pretreated with five concentrations of EBR (0, 0.05, 0.1, 0.2 and 0.4 °M) and then exposed to day/night temperatures of 10/5 °C for 8 d. The results show that EBR, especially 0.1 °M EBR, dramatically alleviated growth suppression and a decrease in chlorophyll content and photosynthetic rate caused by the cold stress. In addition, EBR also decreased malondialdehyde content and O2 .- production rate induced by the cold stress, and increased the activities of superoxide dismutase, guaiacol peroxidase, catalase, and ascorbate peroxidase, and proline content. The results of the present study suggest that exogenous EBR could improve cold tolerance of eggplant by regulating photosynthesis and antioxidative systems. |

This study aimed to isolate BBM orthologue genes from Rosa canina and analyse their functions. Two full-length cDNAs, designated RcBBM1 and RcBBM2, were isolated from R. canina by the rapid amplification of cDNA ends (RACE). The predicted amino acid sequences of the two RcBBMs contained the bbm-1 motif and the motifs typically conserved in the eudicotANT (euANT) lineage. Phylogenetic tree analysis showed that the RcBBMs were most closely related to the BBM orthologue genes identified in Glycine max and Medicago truncatula. The transcripts of the RcBBMs were detected in young roots, calluses, and protocorm-like bodies (PLBs), whereas they were undetectable in stems, leaves, and flowers. RcBBM1-GFP and RcBBM2-GFP fusion proteins were both localized in the nucleus. 35S::RcBBM1 and 35S::RcBBM2 transgenic Arabidopsis thaliana lines exhibited enhanced shoot regeneration capacity in tissue culture, but did not undergo spontaneous somatic embryogenesis. The results suggest that RcBBMs may be candidate genes for improving the shoot regeneration efficiency of R. canina. |
Aluminum alleviates boron-deficiency induced growth impairment in tea plantsR. Hajiboland, S. Bahrami-Rad, S. In this work, tea [Camellia sinensis (L.) O. Kuntze] plants were cultivated hydroponically and treated with adequate (control) or low B supply (-B) without or with 300 μM Al (-B+Al) for 14 weeks. Growth of B-deficient plants was completely resumed by Al supplementation or even surpassed control plants regarding shoot biomass. Net photosynthetic rate was negatively influenced by the low B supply, and the Al treatment increased it up to the level of the control plants that was reflected in the higher content of saccharides. The activity of ascorbate peroxidase (APX) in the younger leaves decreased at the low B supply accompanied with an increased H2O2 content. The Al treatment increased the APX activity up to the level of the control plants simultaneously with the reduction of H2O2. Activities of superoxide dismutase (SOD) and peroxidase (POD) increased in the low B plants and the Al treatment augmented this effect. The content of malondialdehyde (MDA) in the leaves increased by low B but declined upon the Al treatment. In the Al-treated plants, the activity of nitrate reductase (NR) and the content of free α-amino acids exceeded those of the control plants, and nitrite concentration diminished. The shoot and root B content of the B-deficient plants supplemented with Al was similar with the B-sufficient ones. The results demonstrate that the up-regulation of C and N metabolism, the activation of antioxidative defense, and the enhancement of B uptake and transport were mechanisms for growth amelioration of the B-deficient plants by Al supplementation in tea. |
Molecular cloning and expression analysis of a new stress-related AREB gene from Arachis hypogaeaL. Hong, B. Hu, X. Liu, C. Y. He, Y. Yao, X. L. Li, L. The gene contains a 1 338-bp open reading frame that encodes a putative protein of 445 amino acids. The corresponding genomic DNA containing four exons and three introns was isolated and analyzed. An upstream 1 060-bp DNA promoter fragment of the AhAREB1 gene was also amplified from peanut genomic DNA. Multiple sequence alignment of the deduced amino acids of AREB showed that the AhAREB1 protein shares high sequence homology with GmAREB1, S1AREB and ABF2. Quantitative real-time PCR analysis showed that AhAREB1 was induced by polyethylene glycol, NaCl, gibberellic acid, abscisic acid and salicylic acid. The cloning and characterization of the AhAREB1 gene will be useful for further studies establishing the biological role of AhAREB1 in plants. |
