Genotype- and Sex-Specific Protocols for in vitro Micropropagation and Medium-Term Conservation of JojobaR.K. Tyagi, S. PrakashBiologia plantarum 48:19-23, 2004 | DOI: 10.1023/B:BIOP.0000024270.02186.1f Nodal explant cultures from field-grown five jojoba genotypes (EC 99690, EC 99692, EC 99692, EC 267779 and EC 171284; male and female plants), could be established on Murashige and Skoog (MS) medium. The nodal explants of different genotypes as well as sex elicited differential requirements of N6-benzyladenine (BA) for optimum shoot regeneration and medium-term conservation. Female nodal explants of EC 99692 produced maximum shoots (10 shoots per explant) followed by male of EC 171284 (9.3 shoots per explant) on MS + 10 μM BA. The pulse treatment of 50 μM indole-3-butyric acid for 20 min caused in vitro rhizogenesis in 44 - 67 % cultures of various genotypes tested. A significant difference was observed for the conservation period of male and female cultures of all the genotypes. MS + 10 μM BA supported the shoot cultures of EC 99690, EC 99691 and EC 267779 for maximum conservation period, while MS + 5 μM BA proved to be optimum for conserving the shoots of EC 99692 and EC 171284. Generally, the female shoot cultures of genotypes survived for longer period than male ones. |

Leaves from middle part of the shoots and segments obtained from middle portion of leaf showed highest potential to regenerate shoot buds. Histological examination of developing shoot buds showed their de novo regeneration with clear vascular connection with the mother tissues. |

Benzyladenine Induced Somatic Embryogenesis and Plant Regeneration of Leptadenia reticulataK.P. MartinBiologia plantarum 48:285-288, 2004 | DOI: 10.1023/B:BIOP.0000033457.09115.f3 Plant regeneration through indirect somatic embryogenesis was attempted from leaf, internode, node and shoot-tip derived callus of Leptadenia reticulata. Somatic embryos at the highest frequency was induced on Murashige and Skoog (MS) medium supplemented with 8.87 μM benzyladenine (BA) and 2.46 μM indole-3-butyric acid (IBA). From different explants, only shoot-tip and node explant derived calli induced somatic embryos. Transfer of the embryogenic callus to suspension cultures of the same concentration of growth regulators facilitated the development of embryos. Suspension cultures with reduced concentration of BA (2.22 μM) either alone or in combination with 0.49 μM IBA fostered maturation of embryos. Half-strength MS solid medium with 1.44 μM GA3 and BA (0.22 or 0.44 μM) facilitated conversion of embryos into plantlets at higher rate compared to that on with BA alone. About 77 plantlets were recovered from 10 mg callus. Plantlets transferred to small cups and subsequently to field survived in 80 %. All the plantlets established in the field exhibited morphological characters similar to that of the mother plant. |

BackBiologia plantarum 48:361-366, 2004 | DOI: 10.1023/B:BIOP.0000041087.17353.d8 Fatty acid ω-3 desaturase (FAD) is the key enzyme catalyzing the formation of trienoic fatty acids. We utilized an Arabidopsis FAD7 gene and the seven independent transgenic rice plants harbouring 1 to 3 copies of this gene were generated. The expression of FAD7 mRNA was different among independent transgenic lines regardless of the copy number. The total linolenic acid (18:3) contents reduced by about 7 - 32 % in transgenic rice plants but the linoleic acid (18:2) content increased accordingly. With or without wounding treatments, the jasmonate content was higher in transgenic lines than in wild-type rice plant. The transgenic lines overproducing jasmonate also showed increased expression of PR1b mRNA and allene oxide synthase inresponse to wounding. |

The highest frequency of adventitious shoot regeneration was achieved from hypocotyl and cotyledon explants on MS media supplemented with 0.75 mg dm-3 TDZ and either 0.25 or 0.50 mg dm-3 IBA. Efficient in vitro clonal propagation was also induced from cotyledonary nodes on a range of media supplemented with 0.75 mg dm-3 TDZ and 0.05 mg dm-3 NAA or 2 mg dm-3 KIN and 1 mg dm-3 NAA. Regenerated shoots were excised and rooted in MS medium supplemented with 1 mg dm-3 NAA. The rooted plantlets were finally transferred to pots. |

After five days of drought imposition the contents of chlorophylls, carotenoids, ascorbate and glutathione, and activities of guaiacol peroxidase and glutathione reductase decreased. Simultaneously, contents of proline, H2O2 and superoxide anion, lipid peroxidation and activities of catalase and superoxide dismutase increased. These parameters recovered to different degrees during subsequent rehydration. |

The first roots appeared approximately after 12 d on the rooting medium. By contrast to non-rooted stems, rooted stems showed a maximum of soluble peroxidase activity on the 9th day, of ionically bound peroxidase to cell wall on the 6th and 12th day and of catalase on the 6th and the 15th day. A time course study of changes of soluble peroxidases isoenzymes showed that there was a band visible only in the rooted stems and also a new band appeared three days before the emergence of roots. |
Thermotolerance of Pearl Millet and Maize at Early Growth Stages: Growth and Nutrient RelationsM. Ashraf, M. Golden) was assessed at germination and vegetative stage. Final percentage of germinated seeds and rate of germination (number of days to 50 % germination) decreased due to high temperature (45 °C) similarly in the both species. In contrast, at the vegetative stage, high temperature (38/27 °C) caused a significant reduction in shoot dry mass of maize, whereas this attribute remained almost unchanged in pearl millet. Relative growth rate and net assimilation rate (NAR) increased significantly in pearl millet due to high temperature, but in contrast, in maize NAR was slightly reduced. Concentrations of N, P, and K in the shoots of both species increased at high temperature, but N accumulation was more pronounced in pearl millet than in maize. High temperature caused a marked increase in both shoot and root Ca2+ concentration in maize, but it did not affect that of pearl millet. S concentration in the shoots of maize decreased significantly due to high temperature, whereas that in pearl millet remained unaffected. Shoot Na+ concentration of both species was not significantly affected by high temperature. High temperature caused a significant increase in uptake of N, P, and K+ in pearl millet, but the uptake of Ca2+, Mg2+, Na+ and S remained unaffected in this species. In contrast, in maize, a significant increase in uptake of K+ and Ca2+, and a decrease in uptake of N, S, Mg2+, and Na+ were found at high temperature. Overall, maize showed lower tolerance to high temperature compared with pearl millet. |

YordanovBiologia plantarum 48:153-156, 2004 | DOI: 10.1023/B:BIOP.0000024295.27419.89 Fatty acid content and composition of chloroplast membranes, ethylene production associated with thylakoid lipids degradation as well as photosynthetic electron transport involving photosystems 1 and 2 were used to determine the effects of increasing Cd concentrations in the growth medium [0, 14, 28, and 42 mg (Cd) kg-1(sand)] on the photosynthetic performance of barley plants (H. vulgare L., cv. CE9704). High concentrations of Cd triggered serious disturbances of the chloroplast membranes. Ethylene production increased whereas a drop of 18:3 fatty acid content occurred, indicating that Cd mediates lipid peroxidation in the thylakoids. The enhanced ethylene production could be used as an early indicator of Cd-induced membrane degradation, yet at very high concentration (42 mg kg-1) Cd decreased ethylene production. |
Protein Patterns, Characterized by Computer Image Analysis, of Lentil Embryo Axes Germinating Under Salt StressA. Dell'AquilaBiologia plantarum 48:237-242, 2004 | DOI: 10.1023/B:BIOP.0000033450.16985.07 Following 16, 40 and 64 h exposure to 0.33 M NaCl given after 8 h water imbibition, lentil seeds showed a gradual decrease of germination upon their transfer to water. These salt related changes were accompanied by modifications in the protein patterns of embryo axes as revealed by two-dimensional electrophoresis separation and by the computer image analysis of protein spots. In comparison with 8 h water imbibed seeds, prominent proteins comprised between the 5.1 - 7.6 pH isoelectric point in the first dimension and 75 - 50 kDa molecular mass in the second dimension showed a significant increase in their abundance as salt exposure increased. On transfer to water to complete germination, the content of many of these proteins decreased at 24h in 2 - 3 cm length embryo axes in comparison with the corresponding embryo axes of seeds continuously imbibed in water for 24 h. Some groups of proteins ranging between 15.5 - 17.3 kDa, already present after 8 h water imbibition, were not detectable after 24 h but were expressed in seeds exposed to NaCl and transferred to water for 24 h. Up- and down-regulated proteins in lentil embryo axes, imbibed under non-lethal salt stress conditions, have been tentatively identified by comparison with the protein map of germinating seeds of the model plant Arabidopsis. |

Huds.) was initiated on Murashige and Skoog (MS) medium. In the presence of benzylaminopurine (BAP) numerous adventitious buds formed, which developed into shoots. Presence of indole-3-butyric acid (IBA) in media significantly decreased number of buds, but increased development of lateral roots. On hormone-free medium shoots successfully rooted and developed flowers and viable seeds that formed another generation. Shoot cultures of B. perfoliata inoculated with suspension of Agrobacterium rhizogenes strain A4M70GUS developed hairy roots at 3 weeks and they were cultured on hormone-free MS medium. Spontaneous shoot regeneration occurred in 3 clones. |
TMV-RNA Biosynthesis in the Light-Green and Dark-Green Regions of Tobacco LeavesM. İindeláĝová, L. İindeláĝBiologia plantarum 48:419-423, 2004 | DOI: 10.1023/B:BIOP.0000041096.21583.61 Changes in the proteins, chlorophyll, virus content and activity of key enzymes of viral RNA biosynthesis were investigated in the light- and dark-green regions of tobacco leaves systemically infected with tobacco mosaic virus. The protein content was increased to 118 % in the dark-green islands in contrast to 60 % in the light-green regions when compared with the control healthy leaves. The comparative analysis of soluble proteins from healthy and light- or dark-green regions of leaves by means of SDS-PAGE revealed that the main soluble proteins are equal in pattern but differ in quantity. The contents of chlorophylls did not differ from healthy tissues in the dark-green islands but were considerably lower in the light-green regions. The content of virus in light-green tissues was about 10 times higher than in the dark-green islands. The activities of key enzymes of oxidative pentosephosphate cycle -- glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase -- did not differ from healthy tissues in the dark-green islands but were considerably higher in the light-green tissues when compared with healthy control. Similar relationships were observed for ribonuclease, phosphomonoesterase and phosphodiesterase activities. The biosynthesis of viral RNA in the dark-green islands is probably restricted by the steady (or reduced) activities of key enzymes of these metabolic pathways. |

Shoot apices, hypocotyls and nodes of 10-d-old seedlings were used as explants. High frequency (100 %) embryogenic calli was initiated from all tested explants on Murashige and Skoog (1962) (MS) media supplemented with 1 g dm-3 polyvinylpyrrolidone (PVP), 1 mg dm-3 6-benzylaminopurine (BAP), 0.5 mg dm-3 kinetin for Nazilli M-503 and 1 g dm-3 PVP, 2 mg dm-3 BAP, 0.5 mg dm-3 kinetin for Nazilli-143. Globular stage somatic embryos were produced 4 months after transfer to hormone-free MS medium supplemented with 1 g dm-3 PVP. Subsequent subculture of globular embryos every 3 weeks on hormone-free MS medium led to the development of torpedo and cotyledonary stage embryos with the frequency of 75 and 83.2 % from hypocotyl explants of Nazilli M-503 and Nazilli-143, respectively. Afterwards, mature somatic embryos were isolated and cultured on hormone-free MS medium for germination and development into plantlets. The highest germination frequency (42.9 %) for Nazilli M-503 somatic embryos were obtained on hormone-free MS medium after 5 months with hypocotyl explants, whereas germination frequencies of Nazilli-143 embryos from hypocotyl, node and apex explants varied between 22 - 30 %. |
Calcium Ameliorates Effects of Lead in Protonema of Funaria hygrometrica Hedw.M. Krzes³owska, A. Woĵny, J. Konieczna-KoperskaBiologia plantarum 48:569-574, 2004 | DOI: 10.1023/B:BIOP.0000047154.16119.32 Two-days-old in vitro grown protonemata of Funaria hygrometrica Hedw. were treated with a mixture PbCl2 (4 μM Pb2+) and CaCl2 (16 μM Ca2+) (Ca+Pb) for 48 h. The results were compared with the control: distilled water (H2O) and the solution of PbCl2 (4 μM Pb2+) (Pb). Protonemata treated with Ca+Pb were longer and contained more cells than those treated with Pb. Moreover, a lower number of cells showed apical cell deformations typical for lead toxicity: swollen tips and wall thickenings at the apex. If deformations were present they were not as extended as in Pb. In comparison with the control, however, protonemata treated with Ca+Pb were shorter, contained a lower number of cells and some apical cells in this material were altered. It can be concluded that the presence of calcium partially neutralised toxic effects of lead in Funaria hygrometrica protonemata cells. |

The results presented here suggest that during the early stages of fruit ripening, efficient antioxidant system protects the tomato fruits against the damaging effect of progressive oxidative stress. At later stages, however, oxidative damage occurs due to decreased activities of the ROS scavenging enzymes. |

Four treatments (pH 6.0, 5.0, 4.0, and 3.0) were compared for effects on biomass, leaf and root length, crude protein, amino acid content and key enzymes of sugar metabolism. Reduction in biomass, root and leaf length, amino acid contents, glucose-6-phosphate dehydrogenase (G6PDH) and pyruvate kinase (PK) content was observed only at pH 3.0, in association with increased leaf proline content. Kikuyu grass is able to grow normally under mild acidity (down to at least pH 4.0). |

Wilczek and subjected to RAPD analysis using 14 random decamer primers. These cultivars revealed polymorphism with respect to RAPD markers and were subjected to hierarchical cluster analysis. A dendrogram was prepared based on these data. Analysis of banding patterns confirmed that two strongly aromatic cultivars IC1, IC4, were closely linked. But another aromatic cultivar, B1, formed a separate cluster. The high yielding cultivars were closely related to B1. The phylogenetic tree constructed by the neighbour joining method showed that RAPD results were correlated with morphological characters like plant height, leaf and seed size, seed colour, etc. |

Glycine was shown to be the main amino acid in mature seeds, while the nonprotein amino acid canavanine exhibited a high concentration in 7-d-old seedlings. Canavanine was lower in the seeds when compared to other tissues analyzed. This does not support the nitrogen-storage function of canavanine, however, it suggests that it is involved in the translocation of amines during the early stages of the development. |

Hassan). The best results were obtained with mature embryo in J25-8 medium supplemented with 2.0 mg dm-3 2,4-dichlorophenoxyacetic acid where about 75 % developed friable calli. Some 80 - 85 % of these calli regenerated barley plants in the same J25-8 medium supplemented with 1.0 mg dm-3 indole-3-butyric acid and 0.1 mg dm-3 kinetin. |
Comparison of Two Methods for Preparing Mitochondria from Tomato Leaves to Study the Ascorbate-Glutathione Cycle ActivityE. Kuĵniak, M. A comparative study on the ascorbate-glutathione cycle activity revealed that there was no difference in the specific activity of the antioxidative enzymes and ascorbate and glutathione concentrations between mitochondria prepared by these two methods. However, the contents of dehydroascorbic acid and oxidised glutathione in Percoll-separated organelles were 260 and 210 %, respectively, of those found in mitochondria purified by the differential centrifugation method. |

Complete absence of carbon source caused death of shoots within 20 d. Under elevated concentrations of CO2 (10.0 and 40.0 g m-3) the shoots grew photoautotrophically on sucrose-free medium. The growth of cultures was better at 40.0 g (CO2) m-3 than on 3.0 % sucrose under ambient air of growth room. However, the best response was obtained at 10.0 g (CO2) m-3 and 3.0 % sucrose where maximum number of shoots, shoot length, fresh and dry mass, total number of leaves and leaf area was observed. |

Full rooting (100 %) was observed in explants nourished with Fe-EDDHA, while less rooting was found in the absence of iron or in the presence of FeCl3. On the contrary, no root formation was observed in explants nourished with Fe-EDTA, which showed extremely lower chlorophyll and high iron contents at the end of the experiment. |
Abaxial and Adaxial Stomatal Density, Stomatal Conductances and Water Status of Bean Primary Leaves as Affected by PaclobutrazolI. TariBiologia plantarum 46:215-220, 2003 | DOI: 10.1023/B:BIOP.0000022254.63487.16 The plant growth retardant, paclobutrazol at 8.5 or 17.0 μM concentrations effectively inhibited the stem elongation and primary leaf expansion of bean seedlings. Although the retardant reduced the relative water content in well-watered plants, the water and pressure potentials remained high in the primary leaves. K+, Na+, Mg2+ and Ca2+ contents in the primary leaves of the paclobutrazol-treated plants were not significantly different from those in the control. The stomatal density increased on both surfaces but the length of guard cells was not reduced significantly on the adaxial epidermes of the paclobutrazol-treated primary leaves. The inhibitory effect of paclobutrazol on the abaxial stomatal conductances became more pronounced with time during the light period but the adaxial surfaces displayed similar or slightly higher conductances than those of the control. The transpiration rate on a unit area basis did not change significantly or increased in the treated leaves thus the reduced water loss of paclobutrazol-treated plants was due to the reduced leaf area. Stomatal conductances of the adaxial surfaces responded more intensively to exogenous abscisic acid and the total leaf conductance decreased faster with increasing ABA concentration in the control than in the paclobutrazol-treated leaves. Higher heavy metal (HM) concentrations strongly reduced the hyphal growth, however, the inhibitory effect was to a large extent eliminated by the addition of maize root exudates to the media. However, the capacity of exudates to ameliorate HM toxicity was limited and did not operate when a threshold HM concentration was reached. |

TommasiBiologia plantarum 46:387-393, 2003 | DOI: 10.1023/B:BIOP.0000023882.24490.51 The ascorbate system, one of the major antioxidant systems, has been studied in two bryophytes; a moss, Brachythecium velutinum (Hedw.) B., S. & G., and a liverwort, Marchantia polymorpha L. The moss and liverwort gametophytes contain ascorbate both in the reduced and oxidized form; utilize ascorbate in removing hydrogen peroxide by means of ascorbate peroxidase and reconvert to ascorbate its oxidation products by means of dehydroascorbate reductase and monodehydroascorbate reductase. Ascorbate oxidase activity was measured in the cytosolic fraction suggesting a localization of the enzyme different from more evolved organisms. The ascorbate content was maintained in the moss after drought stress while it declines in the liverwort, which seems more sensitive to water stress. Since ascorbate recycling is more efficient in the moss than in the liverwort, this seems to suggest a correlation between efficiency of ascorbate recycling and water stress tolerance. |

Meristematic activity in the superficial layers of cotyledons led to bud primordia formation. Bud primordia had characteristic zonal organization with needle primordia arising on the flanks of the meristem. Somatic embryo initial was formed on the cotyledon surface after an unequal transversal division of a cell. The smaller, apical cell gave rise to apical dome and later on secondary, filamentous suspensor while the larger vacuolated cell presented primary suspensor. |
