Transformed tissues showed elevated levels of cytokinins and grew as teratomas forming clumps of short, branched shoots with small modified leaves. Roots appeared rarely in later subcultivations of some teratomous clones. |

SheikhBiologia plantarum 38:309, 1996 | DOI: 10.1007/BF02873866 Seven plant species including three chenopods:Suaeda fruticosa, Kochia indica, Atriplex crassifolia and four grasses:Sporobolus arabicus, Cynodon dactylon, Polypogon monspeliensis, Desmostachya bipinnata, varied greatly in their seed germination and growth responses to soil moisture or salinity. The germination percentage of each species was significantly lower at soil moisture level of 25 % of water holding capacity than at the levels ranging from 50 to 125 %. Increase in salinity resulted in gradual decrease in seed germination of each species. Growth responses of species to salinity varied widely from significant decrease with slight salinity to stimulation up to salinity levels of 20 dS m-2. Higher K+Na+ratios in plant shoots of all species compared to that in the root medium indicated selective K+uptake. Higher tolerance in chenopod species seems to be attendant on their ability for internal ion regulation. |

ČanigováBiologia plantarum 38:585, 1996 | DOI: 10.1007/BF02890611 The striking diversity in the expression pattern of the stress-related anionic peroxidase was observed during development of female cucumber flower. While the isoenzyme Prx3 was accumulated constitutively in the course of flower development, the expression patterns of other two isoenzymes (Prx1 and Prx2) were restricted to the period after flower opening. The virus infection was simulated by careful opening of the intact female flower buds 3 d before anthesis followed by exposition to the glasshouse environment for 3 d. The results obtained in this experiment revealed a marked accumulation of the isoenzyme Prx1 and Prx2 at anthesis. Under normal flower development, the pistils did not accumulate these isoenzymes at this stage. In contrast, the pattern of expression of Prx3 as well as of the pistil-specific peroxidase isoenzyme remained unchanged, confirming a constitutive type of expression. Beside the pistil, a 3-d exposition of the stripped flowers resulted in a marked accumulation of Prx1 and Prx2 isoenzymes also in both adjacent flower organs - the ovary and the pedicel. At the same time of the normal development of female flower these organs did not accumulate these isoenzymes. |

One-year old seedlings of three trees, one bush and one annual plant were grown in controlled conditions. For all investigated plant species the dark transpiration rate (E), the free energy of respiratory substrate, the entropy production and the free energy balance (FEB) of the dark respiration were higher at low than at high RH. Effect ofCalotropis latex on laticifers differentiation in callus cultures ofCalotropis proceraS. S. Suri, K. G. RamawatBiologia plantarum 38:185-190, 1996 | DOI: 10.1007/BF02873844 Laticifers differentiation in callus cultures of Calotropis procera (Asclepiadaceae) as affected by own latex and its fractions incorporated in Murashige and Skoog (MS) medium is described. Callus cultures have been maintained on MS medium with 2.3 ΜM 6-furfurylaminopurine (FAP) and 3.0 ΜM 1-naphthylacetic acid (NAA). Marked increase in laticifers differentiation (from 10.1 to 28.4 %) was observed on this medium supplemented with 1 % (v/v) of latex. Latex fractions containing proteins + complex polysaccharides or inorganic salts also increased laticifers differentiation (by 21.8 % and 24.1 %, respectively). Other fractions (free amino acid + saccharides, phenols and terpenes + sterols) had no marked effect on laticifers differentiation while alkaloid fraction inhibited it. Effect of latex on laticifers differentiation was much more profound than the reported optimal concentration of plant growth regulators (4.6 ΜM FAP + 1 ΜM IAA). |

KastoriBiologia plantarum 38:281-287, 1996 | DOI: 10.1007/BF02873861 In young sugar beet plants cadmium suppressed the activity of nitrate reductase, glutamine synthetase and glutamate dehydrogenase, whereas sulphur exhibited a protective role towards activity of these enzymes, except of glutamine synthetase. Protein synthesis was suppressed in the absence of S in nutrient medium; the lowest level was at 10-3 M Cd2+. Chloroplast pigment contents were increased by S while Cd2+, even in the lowest concentration, (10-5 M) showed a repressive effect. The highest concentrations of Cd2+ (10-3 M) caused a decrease in dry mass, whereas S induced its increase. Nitrate content was increased in the presence of Cd2+ and decreased by increased concentration of S. |

Catalase activity is very low in endosperm and in embryo of germinating poppy seeds. During postgerminative growth and development the enzyme activity increases rapidly with maximum in endosperm on day 2 and in developing seedling on day 3. A rapid decline of enzyme activity parallells the extension growth of poppy seedlings. Three electrophoretic forms of catalase have been detected in isolated glyoxysomes and partially purified catalase preparation. Electron microscopic observation indicates the presence of catalase in glyoxysomes of parenchyma cells of poppy seedling cotyledons. Numerous lipid bodies and electron-dense deposits in vacuoles are the most characteristic feature of these cells. |

A simple modified Feulgen staining procedure was employed for SCE-analysis. Maleic hydrazide and paraquat were included for comparison. An evaluation of genotoxicity of the above test chemicals made on the basis of SCE-assay was found positive for all the test chemicals with exception of zinc sulphate which gave a weak positive result. |

The nitrogenase activity and leghemoglobin content of nodules markedly decreased under drought and nodules of potassium fed plants showed better recovery after rehydration. The proline content significantly increased under drought but declined upon reirrigation. Also, the C, N and K contents of nodules significantly declined under drought. |

The decrease in PS 2 activity was biphasic in cells exposed at the surface (0 cm) and at 2 cm depth in the water column, while gradual in those exposed at 10 and 30 cm depth. Addition of supplemental "white light" reduced the extent of UV-B damage. Decrease in photosynthetic activity was primarily due to the loss of energy transfer from phycobilisome to chlorophyll as the former in cyanobacteria acts as the primary light-harvesting complex. This was supported by the absorption and fluorescence excitation and emission spectral studies. All these changes were proportionally reduced by the thickness of the water column that reduced UV-B irradiance. |

NagarBiologia plantarum 38:153, 1996 | DOI: 10.1007/BF02879652 In leaf discs of maintenance foliage of tea (Camellia sinensis) polyamines (PAs) and kinetin retarded chlorophyll (Chl) loss, whereas inhibitors of PA biosynthesis [difluoromethyl arginine, difluoromethyl ornithine, methylglyoxal-bis(guanylhydrazone)] and abscisic acid (ABA) promoted senescence. The contents of RNA and protein were significantly higher in PA and kinetin treated leaf discs as compared to those treated with inhibitors and ABA. The contents of total and reducing saccharides declined with the progressive loss of Chl, and the concentration of starch increased in all the PA treated leaf discs. Free amino acid content also increased under all the treatments, but the increase was comparatively larger in case of inhibitors application. |

The lead ions could enter the symplast probably by endocytosis but we can not exclude diffusion as the way of entering. Lead deposits inside a protoplast were observed after 2 h of lead application. After 12 h, Pb deposits were commonly present in all cell structures surrounded by a membrane. Some disturbances in ultrastructure of organelles were found after 8 h. The most frequent were: change of endoplasmatic reticulum configuration from linear to concentric one in many cases surrounding other organelles (e.g. mitochondria) and large and numerous plastoglobuli in chloroplasts. Longer treatment of protonemata with lead (24 h or 48 h) caused degeneration of organelles and even death of the cell. |

Pollen grains from tobacco plans infected with tobacco mosaic virus (TMV) were smaller, with rounded shape and conspicuous deformation of aperture unlike oval and smooth pollen grains from healthy plants. No morphological alterations were observed inC. quinoa andC. album plants infected with TMV and cucumber mosaic virus (CMV). Polyacrylamide gel electrophoresis of pollen proteins revealed substantial quantitative and qualitative differences in protein components of pollen grains collected from healthy and virus infected plants |
Effect of sodium and calcium chlorides, abscisic acid and proline on callus cultures ofArachis hypogaea L.A. Gupta, G. S. SinghalBiologia plantarum 38:525, 1996 | DOI: 10.1007/BF02890601 Callus cultures ofArachis hypogaea L. cv. JL-24 adapted to 200 mM NaCl (otherwise lethal to cells) were used for the study. Calli grew slowly when transferred to 250 mM NaCl, but the growth was enhanced when ABA was included in the medium. ABA induced increase in growth of callus was not accompanied by corresponding increase in internal free proline levels. 0.5 mM of CaCl2 ameliorated the negative effect of NaCl indicating that cells require a specific Ca2+/Na+ ratio for their growth. Proline content also increased at this ratio thereby suggesting that increase in growth at 0.5 mM Ca2+ may be due to an increase in proline content. However, exogenous proline did not increase the growth of callus (adapted to 200 mM), and higher concentrations even inhibited the growth. This shows that proline is not required for growth or adaptation of cells to salt stress, but is produced as a consequence of stress. |

MangalBiologia plantarum 38:611-615, 1996 | DOI: 10.1007/BF02890618 The effect of salinity on contents of water, soluble saccharides, phenols, minerals and on respiration rate in bulbs of five garlic (Allium sativum L.) cultivars differing in salinity tolerance was determined. Cultivar HG-6 was found to be the most tolerant followed by cvs. G-l and G-42, and cv. Aru the least tolerant to salinity. The cultivars which were tolerant showed lesser reduction in water content of the bulbs by salinity. Initial contents of phenolic compounds and sulphur were comparatively low in salinity tolerant cultivars but they increased under high salinity levels whereas reverse was found in salt sensitive cultivars. The fatty acids profile did not show significant changes under saline conditions. Contents of K and Ca were reduced, content of Na was increased and there were no changes in the contents of N, Mn, Cu, Zn and Fe. The changes in soluble saccharides content and respiration rate were not found to be associated with the salinity tolerance. |

TerekBiologia plantarum 38:33, 1996 | DOI: 10.1007/BF02879629 The thickening that appeared on maize roots under the influence of 6-benzylaminopurine and α-naphthylacetic acid (concentration 10-5, 10-6, 10-7 and 10-8 M) were analysed. The changes in length and width of maize roots at the edge of elongation zone after 24,48 and 72 h of treatment were studied. The growth in length of cells at the edge of elongation zone stopped abruptly but the growth in width slowly continued. So, the growth of cells in length and width under the influence of growth regulators was not simultaneous. They had distinct time limits. |

In contrast to pigment and protein contents, starch and saccharide contents increased in salt stressed seedlings. When the seedlings were subsequently sprayed with triacontanol (1 mg kg-1) the salinity effect was partially ameliorated and growth, biomass, chlorophyll and carotenoid contents increased. |

Gigas) grown in liquid medium for 11d absorbed more solution when the saccharose concentration was 2 % than when it was 10 %. When the ears were grown in 6 % saccharose solution, the rate of uptake from the solution was between that from the 2 and 10 % saccharose medium. Dry mass per grain increased with the saccharose concentration in the medium and the reduced uptake of solution did not decrease the moisture percentage of the grain. The culture of ears decreased pH of the solution with 2 % saccharose more than with 10 %. Addition of 0.5% chloramphenicol to the culture solution had no adverse effect on grain mass; it prevented contamination of the solution and maintained a higher pH |
