Growth correlations and rna synthesis in different parts of the shoot apical meristem ofChenopodium rubrunt L. induced to flowering

Uridine-3H incorporation and RNA concentration were investigated in different parts of the shoot apical meristem ofChenopodium rubrum using autoradiography and cytophotometry. A single inductive cycle was sufficient to bring about postinductive first events in the shoot apex but not for complete flower differentiation. The initial activation of RNA synthesis manifested itself in all zones of the apex. The first increase was more conspicuous in the peripheral than in the central zone. The indications of the first events in the apices after a single inductive cycle disappear prior to morphological reversal to the vegetative state. Induction by three short days led to rapid flower differentiation. The increase in RNA synthesis and concentration was most conspicuous in the central zone in this case. The ratio of RNA synthesis and content between bud and leaf primordia (B/L) also change in relation to photoperiodic induction. In vegetative plants the B/L ratio was low while after induction it increased.
The shifts in activity of RNA synthesis observed in the shoot apical meristem are related to the changes in growth activity of the different parts of the apex. The growth ratios in the apices bear the character of growth correlations. The change in the growth correlations following photoperiodic induction together with the total activation of RNA synthesis are considered to represent one of the first events of the transition to the reproductive state.

Effect of different sugars on stomatal behaviour inMerremia aegyptia (L.) Urban andM. dissecta Hallier F.

The effect of mannitol, glucose and sucrose on the stomatal behaviour of two desert species,Merremia aegyptia andM. dissecta has been studied. Stomatal opening did not uniformly depend on the decrease in turgor of the epidermal and subsidiary cells caused by the different osmotic potential of the sugars. Sucrose caused plasmolysis of the subsidiary cells only but this was not accompanied by the opening of the stomatal pore. InM. aegyptia, no plasmolysis was seen either in epidermal or subsidiary cells, even the stomata opened; inM. dissecta, on the other hand, plasmolysis occurred in these cells without any stomatal opening, after incubation in glucose or mannitol. Mannitol is least absorbed, glucose slightly more and sucrose is absorbed to a very large extent in the guard cells when the materials were inoubated in the respective sugar solutions. However, the absorption of these three sugars was almost always larger in isolated epidermal strips than in discs; in detached intact leaves it was still more reduced.

Studies on the physiology of hyacinth bulbs (Hyacinthus orientalis L.) the effect of plant growth regulators on metabolic activities in non-chilled hyacinth bulbs

The effect of gibberellic acid, benzyladenine and a mixture of these compounds on the activity of some hydrolytic enzymes was studied in hyacinth bulbs non-exposed to low temperatures. Plant growth regulators were applied on the heel of dormant bulbs in the middle of July. An intensive elongation of the inflorescence was observed only in the plants treated previously with GA3 and grown in a warm greenhouse for 63 and 84 days. The activities of amylase, invertase and acid phosphatase were higher in the flower buds of unrooted bulbs treated with growth regulators than in the control plants, although growth regulators did not affect the level of extractable proteins. The elongation of the inflorescence in the plants treated with gibberellic acid was correlated with a sharp increase of invertase activity in this organ. The effect of GA3 and BA on the activity of other enzymes in buds and scales varied with the period of plant growth.

Nitrate reductase, nitrite reductase and glutamate dehydrogenase levels in roots and leaves of maize seedlings

Total activities of nitrate and nitrite reductases were higher in 4 to 20 day old maize plants in the leaves than in the roots. The ratio of activities found in the leaves and in the roots respectively was much higher in the case of nitrate reductase than in the case of nitrite reductase. On the other hand higher glutamate dehydrogenase activity in the roots than in the leaves clearly indicates that the roots play a more important role in the assimilation of ammonium than in the assimilation of nitrate. When comparing the distribution of seminal and nodal adventitious roots of maize seedlings with the assimilation of inorganic nitrogen on the basis of enzyme levels, it could be deduced that during the first 20 days of seedling growth seminal roots were more involved in the assimilation of nitrate whereas nodal adventitious roots were more active in ammonium assimilation.

Effect of ammonium concentration on growth and nitrogen accumulation by soybean grown in nutrient solution

Soybean (Glycine max (L.) MERR. CV. 'Amsoy') plants were grown for 40 days in nutrient solution at various concentrations of ammonium. Maximum yield of dry matter was obtained at a concentration of 715 μM. Further increase in the concentration of ammonium resulted in a reduction in growth due to ammonium toxicity which affected both root and shoot development. The pattern of nitrogen accumulation in tops was consistent with the multiphasic uptake of ammonium and can be represented by 2 phases in the range 1.78 X 10-5-3.57 x X 10-3 M of ammonium.

Finding of lettuce big vein virus in Czechoslovakia

Big vein disease occurring on lettuce plants cv. 'Pražan' was identified and the susceptibility to this disease was tested on 9 lettuce cultivars of foreign origin. Lettuce big vein virus was confirmed to be the cause of the disease. The transmission of the LBVV by the fungusOlpidium brassicae (Wor.)Dang., the sporangia of which were found by microscopy in the cells of surface tissues of lettuce roots, was experimentally proved. Rod-like particles, the average length of which was estimated at 244 nm, were found in the roots of diseased plants. LBW transmission by mechanical inoculation and by aphids could not be proved.

Effect of a lack of manganese on the structure and functioning of the photosynthetic apparatus of Funaria hygrometrica spores.

Funaria spores must structure their photosynthetic apparatus before germination. A lack of manganese in the culture media affects neither the final structure of chloroplast nor the chlorophyll synthesis. Fixation of CO2, reducing power of chloroplasts (DPIP) and RPE photo-induced signal S II are modified by this lack of manganese. The study of Mn/Chl and CO2 fixed/Mn ratios shows that Mn is a limiting factor for CO2 fixation and that in the spores depleted of Mn the residual Mn is "structural" Mn.

Atypical composition of seed proteins in cultivars ofPhaseolus vulgaris L.

In only one cultivar out of 1200 investigated cultivars ofPhaseolus vulgaris L. could we find an extreme change in the pattern of reserve proteins on the cathodic side: one of the proteins, called protein I, is completely absent in the cultivar 'Krupnaya sakharnaya' and is replaced on the same site by another protein,i.e. a protein completely different in its immunochemical specificity. The case is of interest from both the phylogenetic and systematic viewpoints and deserves further attention.

Absence of biological activity in oxidation products of indoleacetic acid

The main enzymatic oxidation products of IAA have been tested for biological activity, using as bioassay the straight-growth test ofAvena coleoptile. After being purified by rechromatography, none of these products (including methyleneoxindole) exhibited biological activity within the wide range of concentrations employed; consequently, the results accord with the evidence that IAA is the true hormone of plant growth.

Dynamics and some characteristics of natural free cytokinin content in growing apices of apple tree shoots during the vegetative season

The changes of content and some chromatographic characteristics and biological activities of natural free cytokinins were studied during the vegetation season in the growing apices of apple tree shoots. Biological cytokinin activity was discovered in two groups of compounds differing in the time of their biological actions. Rf values and in the time of their maximal content in the growing apices. The time of maximal content was in accordance with their biological activity.

'Dobrovická A.' A weed plantRaphanus raphanistrum L. was identified as a new natural host plant of BMYV. A modifiedAmaranthus betacyanin test for cytokinin bioassay

A modified procedure for the extraction of betacyanin fromAmaranthus seedlings is described. Application of this modification increased the absorbance of cytokinin-treatedAmaranthus explants in most cases. The modifiedAmaranthus test is compared with the soybean callus test in the bioassay of kinetin, 6-benzylaminopurine, and 6(δ,δ-dimethylallyl) aminopurine.

Transpiration in seven plant species colonizing a fishpond shore

In 7 species (Eleocharis palustrisR. Br.,Juncus bufonius L.,Gypsophila muralis L.,Trifolium repens L.,Agrostis stolonifera L.,Potentilla anserina L. andAchillea millefolium L.) growing in a gradient of habitats from aquatic to terrestrial, on a sandy fishpond shore in Southern Bohemia, Czechoslovakia, the daily course of transpiration rate and water content was assessed gravimetrically in their cut-off transpiring parts on two typical summer days. Transpiration rate was largely controlled by microclimate but depressions occurred inJuncus and in some species growing in the drier habitats. The highest instantaneous transpiration rate and total daily transpiration per unit dry weight, about 100 mg g-1 min-1 and 50 g g-1 d-1, respectively, were recorded inAchillea andTrifolium. The water turnover rate ranged from 16.1 g g-1 (H2O) d-1 inJuncus to 10.7 inGypsophila. The estimated daily maximum water saturation deficit was highest inEleocharis (45.7%) and lowest inAchillea (15.0%). The adaptational significance of the observed phenomena is discussed.

Streptomycin induced changes in growth and metabolism of etiolated seedlings

Effects of various concentrations of streptomycin sulphate either alone or in combination with different cations and hormones on mungbean (Phaseolus aureus L.) seedling growth were studied. The relative inhibition of root growth was stronger than that of hypocotyl growth. Root growth inhibition was completely overcome by calcium, while other cations were ineffective. Inhibition of hypocotyl elongation could not be prevented by cations. IAA and GA3 were capable of relieving streptomycin inhibition but kinetin was ineffective.
In the coleoptiles of streptomycin-treated rice (Oryza sativa L.) seedlings, there were accumulation of nucleic acids and decline in protein content resulting in increased RNA/protein and DNA/protein ratios. High nucleic acid content induced by streptomycin could be correlated with reduced activity of the nucleases.

Estimation of substances with the cytokinin activity in studies on the correlation between the cotyledon and its axillary bud in pea (Pisum sativum L.)

The content of substances showing the cytokinin activity was estimated on decapitated and one-cotyledon-deprived pea plants during that period when the promoting effect of the cotyledon excision had not yet been manifested. Results of the bioassay showed that after the excision an increase in the level of substances with cytokinin activity occurred only in cotylars growing in axillas of these excised cotyledons. These results coincide with earlier data about the content of gibberellins and auxins in the same object.

The possibility of obtaining seeds following placental pollination in vitro

The paper aims at verifying the possibility of obtaining seeds following placental pollination in more than 20 species of dicotyledons and monocotyledons. Excised placentae with ovules were cultivated in solid nutrient media in Petri dishes. Pollen was transferred from anthers onto placentae, its germinating capacity being enhanced by a drop of a 5 per cent sucrose in boric acid in certain cases. Fertilization of ovules resulting in the formation of viable seeds was obtained withNicotiana tabacum, Melandrium album andAgrostemma githago. Although withPaeonia officinalis, Viola tricolor, Antirrhinum majus, Galanthus nivalis, Narcissus pseudonarcissus, Clivia miniata andTulipa gesneriana normal seed set occurred, the seeds did not germinate even after being stimulated with GA3. The results indicate that the findings may be used for further embryological studies. The species are listed in which no seeds were obtained.

Comparative study of the regeneration potential of the carpophores of Coprinus congregatus developed under various conditions of temperature and light

The manner of regeneration phenomena of carpophores ofCoprinus congregatus planted on the fresh nutritional substrate was studied. In the cultures taken from the carpophores that had completed their development the first viable regenerated primordiums appear mainly on the mycelium developed from the carpophore-implant (indirect regeneration). On the contrary, in the cultures taken from the carpophores in which the final morphogenesis was checked by unfavourable conditions, temperature or light, the first regenerated structure developed essentially on carpophore-implants (direct regeneration). It appears that a relation exists between the regeneration potential an development of the phenomena of maturation in carpophore-implants.

A closed system for measurement of photosynthesis, photorespiration and transpiration rates

Manometric estimation of respiration of isolated protoplasts ofPetunia

Respiration of protoplasts isolated from the leaf mesophyll ofPetunia hybrida was estimated by the Warburg method. The time course of respiration rate shows an initial decrease followed by a prominent increase which is caused by shaking of the protoplast suspension. The absolute respiration rates are low and show an unusual spread, but the temporal form of the curve is always similar to that one shown in Fig. 1.
The RQ-values shift from below 1 during the initial phase to far above 1 during the increase of the respiration. For isolated protoplasts from petals the same is true. Feeding the protoplasts with DNA does not influence the respiration rate significantly. The physiological quality of protoplasts seems to suffer in a high degree from the isolation process.

Manifestations of non-specific antiviral factor originating in tobacco leaves during virus infection

A non-specific effect of antiviral factor (AVF) was proved by interference of five different viruses (CMV, PVX, PVY, TMVe, TMV vulgare) with CBRV.
The non-specific AVF originates in non-infected tissues around TMV lesions. Transmitted mechanically to other plants this AVF exhibits the same suppressive effect on multiplication of TMV and CBRV.
The AVF is formed in the non-infected apical part of the leaf which was infected in its basal region. The susceptibility of leaves is decreased even when AVF is added to virus inoculum and applied mechanically on the leaf. The apical part of the leaf the base of which is met with necrotic reaction to TMV, is considerably less susceptible and TMV multiplication in this region is decreased. The necrosis on the leaf base induced by mechanical injury or by cysteine hydrochloride may decrease the susceptibility of the leaf to some extent, but has no effect on multiplication of the virus.
The activity of AVF in healthy apical region of the leaf increases till the fifth to the eighth day after its basal part was infected. The apical region then exhibits lower susceptibility (decrease in number of lesions) and the growth of lesions is decreased. This indicates the virus multiplication being suppressed.

In comparison with controls, the respiratory activity in locally infected leaves was lower until the 8th day after inoculation, whereas it was higher with the leaves in which systemic infection was expected. In the course of the remaining period of investigation, the respiratory activity was almost identical in both cases, showing a rising tendency. Ascorbic acid oxidase whose activity curves correspond to those of respiration, represents the main terminal oxidase. Its activity is probably coupled to the respiratory chain in which glutathione, reduced by NADPH, is capable of reducing reversely dehydroascorbic acid originating from the action of ascorbic acid oxidase. Virus Y-infected plants containing excess NADPH, as found out in our previous paper, the respiratory chain mentioned is likely to be utilized preferentially.
