Mature seeds had high water content (50%) and were able to germinate immediately after fruit abscission. The submicroscopic cell structure was similar to the structure of functionally active cells. Free and conjugated indole-3-acetic acid (IAA), abscisic acid (ABA), zeatin riboside and dihydrozeatin content and gibberellin-like substances (GLS) activity were determined during embryo maturation. Decrease in ABA, free IAA and cytokinin levels was observed at the end of maturation. Mature seeds contained considerable amounts of conjugated IAA and had high GLS activity. |

GG 2 (DT) and drought sensitive cv. JL 24 (DS). Soil moisture stress was imposed by withholding irrigation at pod development phase. The decrease in photosynthesis (PN) under stress was associated with a decrease in stomatal conductance (gs) and relative water content (RWC). The PN and RWC were significantly higher under stress in DT than DS. On relief of stress the gs and RWC recovered more quickly in DT than DS. The maintenance of higher RWC (>80 %), gs and PN under stress appears to be imparting drought tolerance in peanut. |

Organ-specific expression of the stress-related anionic peroxidases in cucumber flowersV. Repka, M. JungBiologia plantarum 37:523, 1995 | DOI: 10.1007/BF02908832 The study revealed a marked qualitative and quantitative differences in the pattern of expression of three stress-related cucumber (Cucumis sativus L., cv. Laura) isoperoxidases. Activity staining, as well as the protein gel blot (Western blotting) confirmed that the proteins studied are differentially expressed in both male and female sepals, in male but not in female pedicels and in, pistil. By using antibodies specific to three stress-related peroxidases, one serologically related pistil-specific anionic peroxidase was detected. This specific band had never been observed in other flower organs. Differential appearance of the stress-related peroxidase isoenzymes in both male and female flowers demonstrates that these proteins are developmentally regulated, showing an organ-specific expression. |

Levels of free, soluble ester- and glycoside-bound phenolic acids and cell wall-bound phenolics were determined in cortical parenchyma of tubers (periderm). The enhancement of phenolic acids in tubers from treated plants was caused primarily by the increase in the contents of free vanillic, caffeic andp-hydroxybenzoic acids and cell wall-bound ferulic, vanillic andp-coumaric acids. |

RichardsBiologia plantarum 36:37, 1994 | DOI: 10.1007/BF02921265 The cytological investigation of 12 taxa ofPanicum L. revealed that the vast majority of them have the basic number x=9 at different ploidy levels. The basic number x=8 was recorded only in the tetraploid speciesP. maximum with 2n=32. The diploid number 2n=18 was encountered inP. capillare, P. laevifolium, P. antidotale andP. coloratum (2) with 3B-chromosomes recorded in the latter species. The tetraploid chromosome number 2n=36 was found to exist inP. miliaceum, P. miliare, P. coloratum (1) andP. virgatum. The hexaploid number 2n=54 was recorded inP. bulbosum, P. dichotomiflorum andP. esculentum. The karyotypes of all accessions were mostly symmetrical and mainly comprised of meta- and submetacentric chromosomes with little variation in length among them within each karyotype. Investigation of chromosome association during metaphase I of meiosis revealed that the frequency of bivalents/cell was the highest among all investigated diploid, tetraploid and hexaploid accessions. Univalents were also frequently encountered in various accessions. These results may indicate that segmental alloploidy has been the major process by which polyploid species have originated. |

BolarinBiologia plantarum 36:247-254, 1994 Two tomato species (Lycopersicon esculentum andL. pennellii) were grown under unheated plastic greenhouse and irrigated with 0 or 140 mM NaCl. Salinity induces a more important reduction in predawn leaf water potential (ψpd) inL. esculentum than inL. pennellii. In both species the osmotic adjustment was achieved by active solute accumulation. The leaf water potential at turgor loss point (ψtlp) seemed to be controlled by leaf osmotic potential (ψos). The results revealed the existence of limits to the accumulation of osmotic solutes in leaf tissues and the existence of an ontogenetic effect on the solute accumulation. In both species, but essentially inL. pennellii the inorganic solutes contribution especially Na+ and Cl- accumulation to ψos was higher than the organic solutes. Therefore, wild species save energy more markedly. |
Effect of 2,4-dichlorophenoxyacetic acid on nuclear division in stem segments ofPisum sativum L. cultivatedin vitroL. Gantchev, W. BleissBiologia plantarum 36:335, 1994 | DOI: 10.1007/BF02920927 We have investigated the influence of 2,4-dichlorophenoxyacetic acid (2,4-D) on the appearance of nuclear fragments, caused by direct nuclear division, as well as on mitotic activity in cultivated internodial stem segments ofPisum sativum L., cv. Bördi, during 180 d of cultivation. Direct nuclear fragmentation (dNF) was indicated by the shape and structure of the nucleus as well as by the occurrence of 1C- and 3C-values of DNA, investigated cytophotometrically. The dNF occurred during the whole cultivation period in segments treated by 2,4-D in concentrations from 4 to 32 mg 1-1. In the presence of 2 mg 1-1 of 2,4-D the dNF existed in the explants only up to 90 d. Mitotic activity was not observed in the 2,4-D-free control but occurred during the whole cultivation period when 2,4-D was added in concentrations from 2 to 16 mg l-1. In the presence of 32 mg l-1 of 2,4-D the level of mitotic activity was very low at the beginning and ceased after 60 d in culture. |

The increase in osmotic potential observed from spring to early autumn seems to result from the decrease in the non-osmotic water fraction, and, conversely, its decrease from early autumn to winter seems to be due to an increase in the non-osmotic water fraction. These observations support the hypothesis that the non-osmotic water fraction may be correlated to starch concentration in rose leaves. |

LercariBiologia plantarum 36:553, 1994 | DOI: 10.1007/BF02921177 The effect of radiation of different spectral composition on axillary proliferation of lavandin (Lavandula officinalis Chaix ×Lavandula latifolia Villars cv. Grosso) was studied in combination with application of exogenous benzyladenine (BA) and putrescine (Put) and endogenous ethylene production. The effect of BA was predominant over the radiation. Continuous far-red showed a fluence rate-dependent promotion of shoot proliferation in the presence of BA. On BA-free medium, shoot number was enhanced under blue, white, and red radiation, at low photon fluence rates. BA, however, could reduce the inhibiting effect of blue and ultraviolet radiation, at high photon fluence rates. Exogenous Put stimulated axillary bud proliferation under some radiation treatments in the presence of BA. Moreover, Put, analogously to BA, could overcome the detrimental effect of ultraviolet radiation. A positive correlation between biotic ethylene production and shoot formation was evidenced under far-red at high photon fluence rate in the presence of BA, and under white, red and blue radiation at low photon fluence rate in the BA-free medium. However, when abiotic ethylene (released from the agarized medium) was stimulated by UV, no improvement of shoot formation was observed. |

A greater increase in the POD activity was measured whenin vitro plants were transplanted into a weaning growth medium. The POD activity was further promoted by removal of the phenolic substances from the extracts by polyvinyl-polypyrrolidone (PVPP). |
Isolation and culture of suspension-derived protoplasts ofBeta vulgaris L.A. Majewska-Sawka, H. Nakashima, K. MoriBiologia plantarum 36:9-13, 1994 | DOI: 10.1007/BF02921260 Sugar beet protoplasts (Beta vulgaris L.) were isolated from hypocotyl-derived suspension cells and cultured on modified Murashige and Skoog medium supplemented with 5 μM naphthaleneacetic acid (NAA) and 2 μM 6-benzyl-aminopurine (BAP). Protoplasts were plated at a density 1.0-1.5×105 cm-3 and incubated in either liquid medium or in medium solidified by 1.2% agarose, at 25°C in the dark. Comparison of two methods of culture unequivocally showed the second to be superior. Immobilizing the protoplast in agarose proved to be essential for obtaining sustained protoplast division and reproducible colony formation. The plating efficiency after two weeks of culture, expressed as the percentage of protoplasts which developed to form colonies, reached 40%. Subsequent subcultures of protoplast-derived callus to regeneration media with different concentrations of BAP (5 μM, 10 μM, 20 μM, 30 μM) resulted in very good callus proliferation at the three lowest concentrations, although organogenesis was not achieved. |
Interrelationships between vesicular-arbuscular mycorrhiza and rhizosphere microflora in apple replant diseaseV. ČatskáBiologia plantarum 36:99-104, 1994 | DOI: 10.1007/BF02921276 Phytotoxic micromycetes appear to be responsible for the apple replant disease (ARD). This was suppressed by the inoculation of apple-tree seedlings with some species of vesicular-arbuscular mycorrhizal (VAM) fungi-Glomus fasciculatum andG. macrocarpum. After the inoculation, growth of apple-tree seedlings improved in dependence on the type of soil, on VAM fungus species and on the ARD appearance. After 12-month cultivation, plant biomass (height, shoot and root dry masses) was markedly increased by inoculation withG. fasciculatum. Similarly, the numbers of colony forming units per unit soil (CFU) of phytotoxic micromycetes and of diazotroph bacteria (associative dinitrogen-fixing bacteria) in the rhizosphere was affected; CFU of phytotoxic micromycetes decreased, whereas CFU of the genusAzospirillum was higher. These bacteria could also serve as antagonists against phytotoxic micromycetes. It is also suggested that the ratio of CFU of diazotroph bacteria to CFU of phytotoxic micromycetes can be used as an indicator of the degree of ARD. It may be assumed that the use of some VAM fungi can replace the chemcial treatment of the soil with ARD. |

This adaptation to γ-radiation possesses the following characteristics: (1) this phenomenon can be induced by an optimal range of low doses of γ-radiation applied acutely or fractionally; (2) the expression of this response seems to be transitory and depends on the time interval between adapting and challenging doses of γ-radiation. |

Biointeraction between precarthamin and cell components in florets ofCarthamus tinctorius L.K. SaitoBiologia plantarum 36:397, 1994 | DOI: 10.1007/BF02920938 The relative rate of precarthamin extractability and floret protein inactivation by organic solvents were compared. Upon trituration of the floral tissues ofCarthamus tinctorius L., usually less extractable precarthamin is released at a markedly high level, while the releasability rate is changed conspicuously by macerating the florets in the test solvents of increased concentrations; the amount of releasable precarthamin also varies according to the solvents used. 30% (v/v) acetone promotes the pigment solubility about twice, whereas, its capacity decreases abruptly in reverse proportion to the increment of acetone content. Methanol accelerates the precarthamin release, but the rate is very low. Ethanol acts in far lesser extent. The data indicate that these varied aspects are more or less attributed to the inactivation of possible factors dissociating precarthamin from cellular components in freezed safflower florets. |
In vitro propagation ofWrightia tinctoriaS. D. Purohit, G. KukdaBiologia plantarum 36:519-526, 1994 | DOI: 10.1007/BF02921172 In vitro method has been developed for propagation ofWrightia tinctoria R.Br. using cotyledonary node segments. Murashige and Skoog's (MS) medium supplemented with 5.0 mg dm-3 of 6-benzylaminopurine (BAP) and 0.01 mg dm-3 of naphthaleneacetic acid (NAA) induced up to eight shoots per explant with an average shoot length of 1.4 cm in 21 d. Three fold multiplication rate was achieved during every subculture of regenerated shoots on the same medium producing an average of 230 shoots per node within 84 d. Reduction in BAP concentration from 5.0 to 1.0 mg dm-3 during subculture promoted shoot length without affecting the rate of multiplication. The differentiated shoots could be rooted by a dip treatment into preautoclaved indole-3-butyric acid (IBA-500 mg dm-3 for 5 min) followed by their implantation onto MS medium containing 1/4 salts. Rooting was observed within 8-10 d in approximately 80% of shoots inoculated after IBA treatment. 15 d after rooting, the plantlets were transferred to culture bottles containing soil-SoilriteTM (1∶1) and liquid nutrient solution comprising 1/4 MS salts. After their partial hardening in these bottles for 10 d they were transferred to pots containing soil-Soilrite (1∶1) mixture with 60% transplantation success. Methods are being standardized to improve the rate of survival and large scale field transfer. |

PlevkováBiologia plantarum 36:65, 1994 | DOI: 10.1007/BF02921271 Glyphosate tolerant cell lines were selected from highly embryogenic cell suspension culture ofMedicago sativa L. Resistant cell lines showed significant reduction of embryogenic ability and during long-term culture in the presence of glyphosate gradual loss of this ability was observed. After glyphosate treatment the increased activity of 5-enolpyruvylshikimate-3-phosphate synthase in tolerant cell lines overcame the block in aromatic amino acid synthesis which was observed in control cell lines. Glyphosate caused marked increase in the content of shikimic acid in both control and tolerant cell lines but the accumulation of shikimic acid was considerably lower in tolerant calli. Significant qualitative and quantitative differences were found in the content of individual phenolic acids. The considerable decrease in the amount of cinnamic acid derivates and broader spectrum of hydroxybenzoic acids suggest in tolerant cell lines the activation of alternative pathway not regulated by phenylalanine ammonia lyase. The possible role of altered pool of phenolic acids on the embryogenic ability is discussed. |
Peroxidase activities and contents of phenolic acids in embryogenic and nonembryogenic alfalfa cell suspension culturesM. Hrubcová, M. Cvikrová, J. EderBiologia plantarum 36:175, 1994 Contents of phenolic acids, peroxidase activities and growth curves showed significant differences in embryogenic (EC) and nonembryogenic (NEC) suspension cultures ofMedicago sativa L. NEC gave a typical growth curve while in EC the distinct phases were absent. The total content of phenolic acids was higher in NEC (related to EC), changed during the growth cycle and most of the acids occurred in ester-bound methanol soluble form. The level of phenolic acids in EC was significantly lower and did not change during 12-d cultivation. The major fraction was formed by phenolic acids ester-bound to the cell wall. The cytoplasmic peroxidase activity in NEC increased continuously during the growth and reached the maximum value at the end of exponential phase. In EC the extremely low cytoplasmic peroxidase activity did not change during cultivation. Ionically bound peroxidases in NEC represented 14 to 30% of the total extracted activity in dependence on the growth phase while in EC formed about 50% of the total activity and did not change during studied period. A possible participation of ionic peroxidase in the incorporation of phenolics into the cell wall is discussed. |

Amelioration of the effects of ageing in onion seeds by osmotic priming and associated changes in oxidative metabolismA. S. Basra, B. Singh, C. P. MalikBiologia plantarum 36:365, 1994 | DOI: 10.1007/BF02920933 Osmotic priming of aged onion seeds with 25% polyethylene glycol-8000 for 5 d resulted in a marked increase in the rate of germination and early seedling growth. Priming reduced electrolyte leakage as well as lipid peroxidation in seeds implying the activation of membrane repair processes. Priming was also associated with increased levels of antioxidants,i.e. ascorbic acid and tocopherols particularly the latter and the activities of catalase and peroxidase involved in the mitigation of oxidative damage. In comparison with the priming of unaged seeds, the aged seeds experienced a diminution of response in terms of changes in the levels of antioxidants and scavenging enzymes. |
