Differences between fruit-bearing and non-bearing apple spurs in activity of an enzyme system decomposing phloridzinMaria J. Grochowska, Wiesława CiurzyńskaBiologia plantarum 21:201-205, 1979 | DOI: 10.1007/BF02884569 The activity of an enzyme system decomposing phloridzin was investigated in fruitbearing and nonbearing spurs of apple trees, Landsberger Reinette cv., throughout vegetation. Acetone powder obtained from xylem sap of apple spurs was incubated with phloridzinsubstratum in citric buffer at pH 5.5 for 12, 18 and 24 h at 30 °C. A paper and thin-layer chromatography as well aa a spectrophotometric assay were employed for tentative identification of enzymic degradation products. Phloretic acid (PA), co-factor of IAA-oxidase, as well as phloretin (Pin), and phloroglucinol (PI) were found after the digestion of phloridzin. The chromatographed enzyme reaction products were measured densitometrically. The activity of the enzyme system was estimated by its efficiency in PA production and phloridzin disappearance. Obtained values, expressed in percentages, showed that the enzyme activity in fruitbearing spurs was much higher than in nonbearing ones; 30 and 10% of released PA in July, respectively. Because fruitbearing spurs of the apple tree are possibly additionally supplied with auxin translocated from developing seeds, an adaptive character of the enzyme system producing PA, a known auxin repressor, is suggested. |

The seeds should contain at least 20% of moisture for a good performance at low temperatures. This hydration level can be reached by pre-sowing exposure of seeds in water saturated atmosphere (WSA). The exposure decreases both visible symptoms of chilling injury and leaching electrolytes during imbibition, and increases both germination and growth of seedlings at 10 to 12 °C. It is concluded that the cell membrane integrity is re-established in a course of the WSA exposure. |

The growth of isolated barley embryos cultivated under different conditionsBěla Landová, M. The growth of the primary root of each embryo was measured during four days of cultivation. Embryos were cultivated on three different consistencies of media: on agar plates, on cellulose tissue moistened with the medium and in liquid shaken cultures. The last way of cultivation yielded the highest degree of the growth of roots. Optimum combination of conditions in shaken liquid medium was selected on the basis of systematic study. The growth of roots of isolated embryos under appropriate conditions approaches that of roots of seedlings cultived on moistened blotting paper. |
Short term cultivation of isolated barley roots and their mitotic activityBěla Landová, M. The excised roots continued their growth but a progressive decrease in the growth rate was observed. There was a considerable short-term drop of the mitotic activity immediately after excision, which was followed by a compensatory increase and then equilibrium was reached 12 h after excision. During the next at least three days, the mitotic index of isolated barley roots varied between 5-6.5%, which is slightly lower than the mitotic index of the root meristems of isolated barley embryos under identical conditions. The mitotic cycle index of isolated barley roots and the size of the root meristem later decreased gradually. |

ShaddadBiologia plantarum 21:259-265, 1979 | DOI: 10.1007/BF02902207 When cultivated in culture solutions salinized with different concentrations (0 to 100 meq I-1), of either NaCl or Na2SO4, the oil producing castor bean, flax and sunflower plants exhibited significant changes in their water relationship parameters (stomatal frequency, rate of transpiration and water content). Although the trend of these changes was nearly the same, their magnitudes depended on the plant treated as well as on the type and level of the salinizing agent. In this respect the values of the stomatal frequency and the rates of transpiration increased significantly with the rise of salinization up to the level 40 meq I-1. Above this the values of these parameters were generally reduced with the rise of salinization up to the highest level used. In addition a considerable number of stomata in either the lower or upper epidermis was found to be closed at the levels ranging from 60-100 meq l-1. The water content of leaves of either castor bean or sunflower subjected to low or moderate salinization levels, remained more or less constant, but with a general tendency to decrease at high levels. However, in the case of flax the leaf water content was generally elevated at nearly all salinization levels. |

Comparison of the induced mutation frequencies at the haploid and diploid levels demonstrated that selection existed against the haploid embryoids with induced chlorophyll deficient mutations. The diploid vegetative stage with phenotypic expression of the chlorophyll mutation was more vital than the haploid one. The suitability of anther cultures for studying induced mutagenesis is discussed. |
Morphogenetic effects of various growth substances on the cotyledonary stomata of brinjal and tomatoG. S. R. Murthy, J. A. Anomocytic, paracytic, anisocytic and stoma with a single subsidiary cell are observed. Abnormal developments like persistent stomatal cells, degeneration of guard cells, unusual thickening, unequal guard cells, single guard cells and size and shape of the pore are noticed in various growth substances. The growth substances also affect the stomatal frequency, stomatal index, epidermal frequency and size of guard and epidermal cells in both the plants. The highest meristematic activity is found in MOR 100 ppm in brinjal and in GA 25 ppm in tomato. The largest size of stomata is found in COL 25 ppm in brinjal and in MH 50 ppm in tomato. The same growth substance responds differently in the two plants. |

SawhneyBiologia plantarum 21:206-213, 1979 | DOI: 10.1007/BF02884570 The three plant types ofAmaranthus namely,A. caudatus f.albiflorus, A. caudatus f.caudatus andA. tricolor var.tristis are qualitative short day plants with critical photoperiods 16.0, 15.5 and 15.0 h, respectively. Gibberellins A3, A4+7 and A13 affect extension growth, leaf differentiation and floral induction differently. Thus, in all the three plant types ofAmaranthus, whereas, GA3 and G4+7 enhanced extension growth, GA13 was completely ineffective under both, 24- and 8-h photoperiods. None of the three gibberellins could affect the leaf differentiation. In all the three plant types, flowering was promoted by GA13 and not by other gibberellins tried. GA13 caused promotion was manifested in two manners, firstly by lowering the critical dark period requirement in each inductive cycle, and secondly by shortening the total period taken for the initiation of inflorescence primordia under inductive photoperiods. The floral induction by gibberellins inAmaranthus is contrary to the gibberellin-anthesin concept of Chailakhyan. It is suggested that gibberellins other than GA3 may be playing an important role in floral morphogenesis of short day plants. |

In ungerminated embryos the phosphatase activity comprised three isozymes; the pattern of iaozymes changes during germination, but there was no influence of P1 in comparison with the control. |
Stomatal activity within the crowns of tall deciduous trees under forest conditionsP. EliášBiologia plantarum 21:266-274, 1979 | DOI: 10.1007/BF02902208 The variation in stomatal activity within the crowns ofAcer campestre, Carpinus betulus andQuercus cerris was measured by vapour exchange porometer on several summer days in an oak-hornbeam forest, in SW Slovakia, Czechoslovakia. Variation resulted from crown position in the forest stand and from leaf position within the canopy. The highest stomatal conductance was in sunlit sun leaves in the upper part of the canopy. Stomatal conductance decreased with increasing depth in the canopy. The steepest decrease was in the upper canopy, in the intermediate zone between fully sunlit and fully shaded leaves, and was caused by the decline in leaf irradiance and in stomatal density. In codominant trees, the conductance in shade leaves at the base of the crown was significantly lower than in the sun leaves at the top of the crown. In a dominant tree,Q. cerris, the differences in stomatal conductance were small and most frequently insignificant. Variation in incident light also determined the diurnal variation of stomatal conductance with respect to crown aspect. Differences between sun leaves on the east and west facing aspects of the overstory crown ofQ. cerris were demonstrated for several days. |
Poly(A)+ RNA synthesis in germinating pollen ofNicotiana tabacum LJ. Süss, J. TupýBiologia plantarum 21:365-371, 1979 | DOI: 10.1007/BF02878235 Poly(A)+RNA is synthesized during the first hours of pollen germination and is rapidly incorporated into polysomal structures. After a 2-h pulse with uracil-14C, 42% of the transcribed fraction of polysomal RNA is polyadenylated. Following 4 h of germination the amount of the newly-made poly(A)+RNA decreases steadily at the rate of about 14% per h, whereas that of rapidly-labelled poly(A)-RNA continues to grow. Beginning 1 h of cultivation the ratio of poly(A)-/poly(A)+RNA increases exponentially. Similarly as in non-polyadenylated mRNA the main portion of the synthesized polysomal poly(A)+RNA sediments at a rate of 4 to 14 S and its mean size decreases slightly with the time of labelling. RNA isolated from nuclei and cell wall containing pollen tube fraction differed from the polysomal one in higher apeoific radioactivity and the polyadenylated RNA exhibited higher size distribution. The comparison of the results with earlier observations suggests the involvement of poly(A)in mRNA translation in pollen tubes. |

Leaf diffusion resistance pattern in an oak-hornbeam forestP. EliášBiologia plantarum 21:1-8, 1979 | DOI: 10.1007/BF02888711 Four tree, five shrub, and ten herbaceous species growing naturally in an oak-hornbeam forest were used for simultaneous study of the leaf diffusive resistances in the course of several summer days. Absolute minima of the stomatal resistance in the sun tree, the shrub, and the herbaceous species leaves were 1.7 to 6.2 s cm-1, 6.1 to 10.8 s cm-1, and 4.8 to 9.7 (17.3 inConvallaria majalis leaves) s cm-1, respectively. Minimum daily leaf resistances in the course of a day were noted earlier in the morning in sun leaves of large trees than in shade leaves of other species. Stomata were fully opened later in the morning and they began to close sooner in the afternoon in usual shade leaves of the plants in the interior of the forest canopy than those in sun leaves in active surfaces of the canopy (tops of tree crowns). The relatively large differences in leaf resistances found among investigated species may be explained by differences in leaf anatomy (stomata frequency and size) and in ambient leaf or plant environment caused by leaf (plant) position in different vertical layers. |
