Benzylaminopurine induced changes in photosynthesis and photorespiration of barley plants

Benzylaminopurine (BA) caused an enhancement of chlorophyll and protein content and a reduced elongation of primary barley leaves. BA did not change the rhythmic pattern of14CO2 fixation and activities of RuBP carboxylase, RuBP oxygenase, glycolate oxidase and phosphoglycolate phosphatase, but the enzyme activities were enhanced and the level of14CO2 fixation was reduced. Light/dark14CO2 evolution ratio was affeoted by BA only in older leaves. BA acts sequentially on the activities of photosynthetic and photorespiratory enzymes.

Studies on the Transmission of Floral Effects of Photoperiod and Gibberellin from One Branch to the Other inImpatiens balsamina

In two branched plants ofImpatiens balsamina with intact apex and leaves floral buds are induced only in the branch which is either exposed to 8-h (inductive) photoperiods or receives GA3 treatment if maintained under 24-h (non-inductive) photoperiods. GA3 induces floral buds on the treated branch even if the leaves on that branch are removed, showing that while leaves are essential for photoperception, these are not neoessary for GA3 to cause induction. The effect of the inductive photoperiods or GA3 treatments to a branch is not transmitted to the other branch which is treated with water and is maintained under non-inductive photoperiods even when the latter is defoliated but is transmitted if the apioal or both the apical and axillary buds on the branch receiving inductive photoperiods or GA3 treatment are excised. It, therefore, appears that the existence of strong sinks in the form of axillary and apical buds on the treated branch prevents the transmission of photoperiodic as well as GA3 effects to the other branch in this plant.

Mitotic cycle kinetics of root meristems of isolated barley embryos and intact seedlings. Labelling of nuclei by tetraploidy

The minimum length of the mitotic cycle of root meristems of cultivated barley embryos and intact seedlings was longer than that measured by the construction of the labelled mitoses curve; it was 10-12 h for intact seedlings and 16 h for cultivated barley embryos. Action of colchicine on interphase was detected. Colchicine induces the increase of the frequency of prophases starting from the fourth hour. The most probable explanation is shortening of the S-phase. As the whole mitotic cycle duration is increased in comparison with that after3H-thymidine, it is most probable that G1 phase duration is increased by colchicine treatment. Different cytogenetic effects of colchicine were analysed in detail. A basic difference between the response of root meristems of isolated embryos and of intact seedlings was found. In isolated embryos, the effect of 0.1% and 0.4% colchicine (i.e. blockage of anaphase movement, metaphase arrest and contraction of chromosomes) disappears within 2-5 h after removing the colchicine. In intact seedlings, the effect of colchicine is maintained for a considerably longer time. It leads to gradual accumulation of metaphases over 9 h after pulse treatment and this accumulation of metaphases leads to a gradual increase of the incidence of tetraploid mitoses starting from 10th - 12th till 22nd hour after the pulse. This is the reason why maximum frequency of tetraploid cells in root meristems of cultivated isolated embryos was 16 h after the pulse (i.e. at the beginning of their incidence) and it reached the value 5.4% while in seedlings the maximum was 22 h after colchicine treatment and it reached the value 38%.

Effects of mutagens on androgenesis in some species of the genusNicotiana L.

Chosen species and cultivars(Nicotiana sanderae, N. glutinosa, N. otophora, N. paniculata, N. sylvestris, N. velutina, N. tabacum cv. Samsun, N. tabacum cv. Samsun - a dihaploid line, and an amphidiploid N.sylvestris x N tomentosiformis) were treated with adequate concentration of n-buthylmethane sulphonate. The anthers of M1 plants were cultured on the medium of NITSCH 1969 plus 0.1 mg IAA/1. Special attention was paid to quantitative characteristics of androgenesis. Androgenesis occurred more often in the treated variants. The conclusions have been confirmed of a heterosis effect of adequate chemomutagen concentration at the diploid level (an increase in the number of androgenic anthers in the treated variants, and a decrease in the mean number of haploids per androgenic anther) in the broad range of the material. We have recorded a high androgenic ability in the amphidiploidN. sylvestris x N. tomentosiformis, and androgenesis inN. velutina for the first time.

Function of the basal pore in the germination of sugar-beet seed balls

Anatomical analysis of the sugar-beet seed ball indicates two possible ways for the entrance of water and oxygen during germination: cells from the operculum periphery and cells from the basal pore characterized by absence of lignin and a thin layer of cellulose. Impermeabilisation of these zones by vaseline considerably reduces the percentage of germination and stresses the major role played by the basal pore. This can also be confirmed by variation in the germination percentage when sowing the seed ball with the basal pore up or down to the substrate.

The reduction of aldehydes by broad bean and maize alcohol dehydrogenases and a study of the substrate binding to the enzyme protein

Alcohol dehydrogenase was prepared from 2-day germinating maize and 3-day germinating broad-bean seeds by ammonium sulphate fractionation of sodium phosphate extracts, chromatography onDEAE cellulose and Sephadex G-200. The activity of the broad beanADH amounted to182 800 units per mg protein, that of maizeADH 79 000 units per mg protein. Besides oxidation of a series of alcohols at pH optimum in the alkaline region and with KM equalling 10-2M, alcohol dehydrogenases isolated from both plants catalyze the reduction of acetaldehyde, n-propanal, n-butanal, isobutanal and crotonal at pH optimum in the neutral region with KM equalling 10-3M. The inhibition studies using fatty acids and chloride ions revealed that the oxidation of alcohols is inhibited competitively by both types of inhibitors, with inhibition constants of 10-2M and 10-1M, respectively. The inhibition in the presence of acetaldehyde is non-competitive since the inhibitors do not compete with acetaldehyde and do not form an enzyme-NADH-inhibitor complex, yet they obviously react with the enzyme-NAD product only, thus giving rise to an enzyme-NAD-inhibitor complex. These differences in the behaviour of inhibitors may be interpreted in the sense that the binding sites of ethanol and acetaldehyde as substrates for broad bean and maize alcohol dehydrogenases are non equivalent. Mathematical description of nitrogen and phosphorus accumulation in developing barley kernels

The changes of kernel nitrogen (N), phosphorus (P) and dry weight (DW) were determined during the time period from 10 days after anthesis till maturity in three spring barley strains. The plants were cultivated under field conditions in Gatersleben (German D.R.) and Kroměříž (Czechoslovakia). The course of N, P and DW changes was described by Richard's comprehensive growth function dW/dt = aWm + bW, where W is the amount of N, P or dry matter per kernel, a, b, m are coefficients. The integral of this function was used and several parameters calculated. There was a remarkable synchrony between N or P and total dry matter accumulation in the kernels. Thus, the N or P concentration per unit kernel dry weight was relatively constant during the investigated period and with few exceptions corresponded to 80% to 100% of the final value. However, considerable differences between strains, years or places of cultivation were found.

The action of penicillin in mungbean (Phaseolus aureus L.) seedlings and wheat (Triticum vulgare L.) coleoptile sections

Penicillin caused elongation inhibition of mungbean (Phaseolus aureus L.) seedlings at concentrations above 100 mgl-1. Inhibition of hypocotyl could be reduced to different degrees by manganese, pyruvate, succinate, fumarate, malate, GA3, purines, pyrimidines and nucleosides, whereas the amino acids except cysteine were not effective. Penicillin inhibited neither elongation of wheat coleoptile sections nor the cell enlargement induced by IAA in this tissue. Thus penioillin inhibits intact seedling elongation probably through an Effect on cell division apparently without inhibiting cell enlargement.

Oxidation of glutathione by peroxidase isoenzymes in fenugreek

During the germination of fenugreek (Trigonella foenum graecum L.) sulfhydryl groups rapidly declined in cotyledon and seedling axis, while peroxidase activity increased. Studies on purified isoenzymes showed that GSH was oxidized by the isoenzymes and was accomplished in presence of cofactors, Mn2+ and DCP along with H2O2 (0.01 mM). This reaction was found to be peroxidatic in nature. The oxidation was inhibited by catechol but was enhanced by malic acid.

Changes in total contents of saccharides, proteins and chlorophyll in malformed mango inflorescence induced by fusarium moniliforme var. subglutinans

Fusarium moniliforme var.subglutinansWr. etRg. causes malformations of mango inflorescences that finally fail to fruit. The host-cultivar Himsagar showed higher contents of total saccharides, chlorophyll and protein than the cultivar Bombai. Considerable loss in saccharides and chlorophyll contents and increase in protein content as a result of malformation of inflorescence was recorded in both the host-cultivars which progressed with time.

Ultrastructure of differentiating epidermal cells of maize root under water deficit conditions

The ultrastructure of differentiating epidermal cells of maize root (within the distance of 1 and 2 mm from the root body apex) were studied under conditions of non-lethal and lethal osmotic stresses of nutrient medium containing polyethylene glycol 4000, as well as regeneration of their ultrastructure following rehydration. The structural response to water deficit of the cells investigated was dependent on both the stress duration and the stage of their ontogenic development. Following non-lethal stress, in younger cells investigated (1 mm), condensation of nuclear chromatin, decrease of polyribosomes and increased density of free ribosomes in cytoplasm, reduction of mitochondrial cristae and occurrence of intramitochondrial inclusions, less compact dictyosomes were observed. Pastid structure remained unchanged. Microtubules were lacking in treated cells. In the more differentiated cells (2 mm) protoplast retreat from cell walls was also observed as well as a general decrease of ribosomes and ER elements in parietal cytoplasm, an increased number of intramitochondrial inclusions and mitochondrial membrane fragmentation. In these cells, Golgi apparatus was also lacking. The ultrastructure regeneration of the more differentiated cells was less pronounced. Lethal osmotic stress would cause more severe structural damage in all the cell components investigated.

The effect of 2,4-dinitrophenol on the absorption and translocation of calcium by pumpkin plants

The study deals with the absorption and translocation of Ca2+ by án intact plant of pumpkinCucurbita pepo L. and with the effect of various concentrations of 2,4-dinitrophenol (DNP) on these processes. The absorption of Ca2+ was reduced by the application of this inhibitor, but not completely stopped. The translocation of Ca2+ into shoots was affected more expressively: it was almost completely inhibited by higher DNP concentrations. The uptake of Rb+ is aubstantially more influeneed than that of Ca2+: higher inhibitor concentrations caused its complete cessation.

Proteinaceous crystals in cells of virus infected plants

Crystal-containing organelles in cells of virus infected plants lying at chloroplasts and mitochondria are identical with single membrane-bound microbodies containing crystals of catalase described in healthy plants. Massive complex inclusions caused by turnip mosaic virus very frequently contain the same microbodies with crystal inclusions; that phenomenon may be related to some pathophysiological changes of virus infected plants. Comparable proteinaceous crystals, but not lying within microbodies limited by a membrane, may also be found in cytoplasm of infected cells. These crystals are sometimes surrounded by a substance resembling the microbody matrix. Disintegrated cytoplasm of virus infected cells may also contain the same crystals lying free in "empty spaces". Cytopathological effects responsible for this phenomenon and possible artifacts as well are discussed.

Syndrome of apricot witches broom

The syndrome of a new apricot disease - "witches broom" is described. Diseased trees have an abnormally high number of lateral, thin shoots, which again have many abnormal secondary shoots at a very narrow angle. Witches broom shoots are without any blossom buds. We may assume therefore that the apricot witches broom - according to the syndrome - may be listed among the group of witches broom diseases which are caused by a mycoplasma like organism. Some of the witches broom shoots are characterized by very small leaves.

The effect of different combinations of soil salinity and CCC on dry matter accumulation and yield of cotton plants

CCC in either absence or presence of salinity slightly and irregularly affected the shoot dry weight, but consistently increased the seed cotton yield. The extent of such positive effect upon yield tended to be larger in the presence rather than in the absence of salinity when using CCC as a seed-soaking medium alone, but the reverse if applied as a spray after seed-soaking.

Anther cultures of maize

The presence of synthetic auxins (2,4-D at a concentration as low as of 0.5 to 5.0 mg I-1, NAA at least at 5 mg I-1) in the cultivation medium was essential for the induction of callogenesis in anther cultures ofZea mays L. The application of IAA was ineffective. Kinetin induced bursting, darkening and a rapid anther necrosis, but at an appropriate concentration ratio with 2,4-D it stimulated pollen maturation at the same time.

Jerusalem artiohoke (Helianthus tuberosus L. cv. D-19) dormancy is normally broken by a 16-week treatment with 4 °C. A thermoperiod of 2 °C-12 h, 8 °C-12 h allows one to reduce this time to 12 weeks. Anoxy applied at 24 °C is effective after 4 weeks. By coupling anoxy and low températures varying daily between 2 °C - 12 h and 8 °C - 12 h, the breaking of dormancy is obtained after a one-week treatment.

Various types of meiotic anomalies in Mi generation such as multivalents, fragments, bridges, mioronuclei, polyads and in particular the multispindle formation, were observed after seed X-ray irradiation inCapsicum annuum L. As the dose increased there was a gradual increase in the number of aberrations.

Distribution pattern of gas exchange in the area of maize leaf blades during the generative phase

Net photosynthetic CO2 uptake (P N ) and transpiration (E) rates through adaxial and abaxial leaf surfaces were measured gasometrically on the area of maize leaf blades of different insertion levels in the phase of male florescence and in the phase of spadices development. The distribution pattern of gas exchange on the area of all measured leaf blades found in the phase of male florescence (significantly lower value in the basal parts) was changed in the phase of spadices development: PN andE of abaxial surfaces of older leaves,i.e. 9th, 10th (under which the spadices were growing) and 11th, rapidly increased in the basal parts in comparison with the middle ones and positively influenced the gas exchange of the whole leaves. It appears that PN may be maintained or increased in the older leaves in the phase of spadices development to prevent shortage of photosynthates.

Activity of mitochondria isolated and purified from dwarf and tall, light-grown pea seedlings

Mitochondria from dwarf and tall light-grown peas have been separated from contaminating chloroplasts and other organelles on discontinuous sucrose gradients. A comparison between activities of the mitochondria from the two cultivars of light-grown pea seedlings was made. It was observed that both respiratory control (R.C.) indices and phosphate to oxygen (P : O) ratios were superior in mitochondria from the tall cultivar than the dwarf cultivar. The results are discussed in relation to energy status and possible gibberellic acid content in the two cultivars.

Fluctuations of Uridine Incorporation in the Shoot Apex ofChenopodium rubrum L. during Photoperiodic Induction

Uridine incorporation into the shoot apex of the short-day plantChenopodium rubrum was investigated during a 16 h period of darkness and the following transfer to light. Uridine incorporation during this single inductive cycle was compared to incorporation under non-inductive conditions of continuous light. After transfer of the plants from light to darkness RNA synthesis was reduced to about half after the first two hours. This occurred not only when the plants were precultivated in continuous light but also after an interruption of the dark period by light for 31/2 h. The low level of uridine incorporation was maintained for the whole duration of the dark period. Incorporation regained its initial level after exposure of the plants to light irrespective of the duration of the preceding dark period. After this immediate rise of uridine incorporation in plants transferred from darkness to light a slight temporary decrease was observed in light. In darkness the decrease of incorporation into the nucleoli was still more marked than the reduction of overall incorporation. After the termination of the dark period incorporation into the nucleolus rose slowly and extranucleolar incorporation was relatively enhanced during the first 10 h of light in induced plants. The fluctuations of RNA synthesis observed in the shoot apex during photoperiodic treatment may be regarded as a necessary condition for the transition from the vegetative to the reproductive state.

The regulation of lactate dehydrogenase activity in soy-bean seedlings

An electrophoretically homogeneous lactate dehydrogenase was isolated from soybean seedlings, the specific activity of which was approximately 1800 times higher than the crude extract. From the dependence of the rate of reaction catalyzed by lactate dehydrogenase on substrate concentration, Michaelis constants and Hill coefficients were determined for four natural substrates,i.e. lactate, pyruvate, NAD and NADH. The enzyme from soy-bean plants is non-competitively inhibited by oxalate and mesoxalate,i.e. by the compounds analogous to the substrate. At pyruvate concentrations above 0.8 mM, the rate of reaction catalyzed by lactate dehydrogenase from soy-bean plants decreases, fructose diphosphate and ATP function as inhibitors as well. The inhibition by ATP is pH dependent, which seems to be of importance for the regulation of enzyme activityin vivo.

Purification and properties of arabis mosaic and tomato bushy stunt viruses isolated from lilac (Syringa vulgaris L.)

The paper gives more detailed characteristics of Arabis mosaic virus (AMV) and tomato bushy stunt virus (TBSV) isolated from lilac, the latter being identified in lilac (from plants suffering from yellow ring disease) for the first time. The isolate of TBSV from lilac, from which an antiserum with a titre of 1024 was prepared, is closely related to the artichoke strain. Information is given about two types of ringspot disease and about chlorotic ringspot of lilac. Whereas in the leaves of lilac suffering from ringspot disease (of ring mosaic type) the presence of AMV was demonstrated, the sap transmission from the leaves diseased with ringspot of linepattern (and wave-like mosaic) type failed; from the leaves affected by chlorotic ringspot a mixture of AMV and cherry leaf roll virus was identified. In addition, the polyetiological nature of "spring" mosaic and necrotic mosaic of lilac, in which bacteriumPseudomonas syringaevan Hall, was found is dealt with. Regulation of seed germination and polarity in seedling development inOrobanche aegyptiaca by growth substances

InOrobanche aegyptiaca PEES. (Orobanchaceae) the mature seed is tiny and contains a subglobose embryo which is not differentiated into radicle, hypocotyl, plumule, and cotyledons. In aseptic seed cultures on medium TB supplemented with yeast extract or coconut milk, both roots and shoot originated from the morphological radicular pole of the embryo (monopolar pattern). The bipolar mode of seedling formation, that is a shoot originating from the plumular pole and roots from the radicular pole, ensued on the basal medium THS and on TB supplemented with certain concentrations of IAA, kinetin, GA3, or strigol.

Water relations in plants dominating phryganic ecosystems

The water potential and the osmotic potential in plants which dominate Greek phryganic ecosystems (Phlomis fruticosa, Sarcopoterium spinosum, Gistus sp.) were measured from April to Nowember. Water potential decreased considerably reaching a minimum in September. Higher values of osmotic potential than that of water potential were found during dry period (i.e. negative values of pressure potential). This interesting fact was confirmed by artificial desiccation.
