The relationships between quantitative characteristics of stomata and epidermal cells of leaf epidermisJ. Most correlations were statistically significant at theP = 0.01 level. Positive linear correlations were established between the area and the thickness of epidermal cells. These correlations were near to theP = 0.05 level of statistical significance. A high variation coefficient "v%" of the total number of stomata per leaf was found in both plant species. |
Isolation of alcohol dehydrogenases from germinating seeds of pea, broad-bean, lentil and kidney-beanSylva Leblová, P. Mançal, Dana Sofrová, Jana BarthováBiologia plantarum 15:405-411, 1973 | DOI: 10.1007/BF02922760 The maximum of alcohol dehydrogenase (ADH) activity of germinating pea and broad-bean seeds sediments from 40 to 60% ammonium sulfate saturation, from lentil and kidney-bean seeds between 40 and 50%. This operation increased the specific activity of ADH preparations roughly tenfold. Chromatography on DEAE-eellulose and gel filtration increased the activity of the resulting preparation when compared with the initial preparation 178 times with pea, 334 times with broad-bean, 122 times with lentil and 77 times with kidney-bean. The ADHs resemble each other in coenzyme specificity: the reaction rate with NAD is one hundred times greater than with NADP. The substrate specificity is quite wide: besides ethanol, these enzymes oxidize 2-propene-l-ol (actually faster than ethanol), 2-butene-l-ol (at the rate of one half t h a t of ethanol) and butanol (even more slowly). In general, saturated alcohol analogues are oxidized more slowly than unsaturated ones. Methanol is a substrate for the enzym from pea only. The ADHs of the plants studied did not oxidize diols, sugar alcohols and cyclic alcohols. The enzyme from pea has the widest substrate specificity oxidizing isobutanol, phenylalcohol and mercaptoethanol. ADHs, which are widely encountered in plants, resemble each other to a certain degree - they have identical coenzymes, equal Km values and equal values of the pH optimum, they differ in the purification process and in substrate specificity. |
Ploidy chimeras in plants regenerated from the tissue cultures ofBrassica oleracea LJ. Autoradiographic data of3H-thymirdine incorporation indicated a higherpercentage of cells undergoing DNA synthesis in adventive embryos than in proembryonic masses. A definite pattern of DNA synthesis was observed during the transition from a proembryonic mass to the torpedo stage of embryogenesis. In particular the labeling pattern for both the heart and torpedo stages was found to be coincident with specific morphogenetic events. |

The device used includes a couple of thermometers in a differential connection and plate electrodes, through which a controlled heat input flows into a denned studied segment of the xylem. The present method was tested first with a laboratory simulating system, then with the stem of a living adult treePrunus avium L. The data are registered continually and automatically; continuous measurement of sap flow rate of long duration with a greater number of trees is thus made possible. |

This gradient of root water potential was found although the "mother" plants continued to be watered while watering all daughter plants had been stopped one week before the water potential was measured. Thus the whole polycormon consisting of a series of interconnected individua behaves as one hydrodynamic system where all individual root systems act as if being parts of one sole root system. |

This acropetal inhibition affects the tuber tip more strongly than its lateral buds, which develop into long stolon-like shoots after the tuber decapitation. The local supply of ABA shows no tuberizing effect, but enhances the dormancy of the tuber top. MH interrupts the correlation between the tuber laterals tuberizing without previous stolon formation. The uppermost leaf structures participate in the apical dominance, inhibiting their own axillaries on intact tubers. Mature scales disclose this correlative influence only on decapitated or dissected tubers on which IAA or BA release their inhibitory effect, but ABA increases it. Two scale pairs occurring regularly at the top of dormant tubers and seen later at the erect base of the stem are involved in the initiation of foliage leaves for the next-year growth period. BA applied to an axil at the top of the tuber provokes its sylleptic branching. |

The plants were labelled with32P three hours prior to sampling. The uptake of32P into the cotyledons was higher in light than in darkness in all cases, however, it was not in correlation with32P incorporation into the NA fractions. After one dark period lasting 8 or 16 h NA synthesis in light did not increase in comparison with darkness. After two or more photoperiodic cycles NA synthesis was higher in light than in darkness irrespective of whether the dark period lasted 8 or 16 h. NA synthesis was distinctly highest after two inductive cycles lasting 16 h. In buds NA synthesis was slightly shifted in favour of ribosomal RNA as compared with cotyledons. In the cotyledons the increase in light was mainly duo to a raise of rRNA synthesis whereas in the buds synthesis of sRNA and DNA increased, as well. |

BhattacharyaBiologia plantarum 15:412-418, 1973 | DOI: 10.1007/BF02922761 2.5 cm long etiolated stem segments ofPopulus nigra did not root at all in water or in IAA alone, but rooted in glucose + IAA. 5-Fluorodeoxyuridine (FUDR), 5-Fluorouracil (FU). actinomycin-D and cycloheximide all inhibited rooting, the inhibitory effect of each increasing with concentration and being more marked in glucose than in glucose + IAA. The inhibitory effect also decreased with the delay in the application of cycloheximide. The DNA, RNA and protein contents of segments grown in IAA + glucose where rooting occurred were higher than in water or in IAA + glucose + cycloheximide/or + aetinomycin-D where rooting did not occur. Auxin in the presence of adequate amount of carbon nutrition increases the fresh production of protein and that the magnitude of rooting is determined by the size of protein pool that is available in tho tissue at the time of root initiation. |
Comparison of effects of calcium deficiency and IAA on the pumpkin plant (Cucurbita pepo L.)M. Dvořák, Jana ČernohorskáBiologia plantarum 14:28-38, 1972 | DOI: 10.1007/BF02920899 In pumpkin plants suffering from calcium deficiency growth of the root system is characteristically suppressed. A biochemical symptom of calcium deficiency is a decrease or even complete disappearance of activity of one of the isoperoxidases (isoperoxidaseD). In the present study the two characteristics were used to compare the effects of calcium deficiency and of IAA in the supraoptimal concentration of 10-5 M. Peroxidases were separated by centrifugal fractionation: peroxidasesA andB were found in the sediment corresponding to the mitochondrial fraction, peroxidaseD in the supernatant. It was not possible, however, to obtain the sediment completely free of peroxidaseD. A negative correlation between the activity of isoperoxidaseD and the logarithm of concentration of IAA in nutritive solution was found and similarities in the effects of IAA in supraoptimal concentration and of calcium deficiency on the development of root system were demonstrated. Still, it would be difficult to imagine that the mechanism of the action of the two factors is the same. For this reason, a scheme of possible interactions was suggested with the aim to explain how similar changes in the content of isoperoxidases are brought about by the two factors. In this scheme, calcium is considered chiefly as a substance regulating membrane permeability. |

All the compounds inhibited protein synthesis and retarded protein breakdown. Cycloheximide,p-fluorophenylalanine, diazouracil and vanillin also inhibited the incorporation of uracil-14C into RNA of senescing discs. Abscisic acid and 2-chloroethylphosphonic acid accelerated yellowing both in darkness and in light and stimulated the protein breakdown in senescing discs. |

The influence of ABA on the decrease of two GAs levels was proved. The relation between the effect of ABA and the stage of stratification is closer in the case of GA4 than GA7. It is assumed ABA to inhibit the gibberellin biosynthesis. |

LimberkBiologia plantarum 14:112-115, 1972 | DOI: 10.1007/BF02920954 Tre potato cultivars Erika and Krasava are unable to reproduce two strains of TMV systemically except when in graft symbiosis under normal greenhouse conditions with tomato which has been infected with the virus. The escape resistance of potato to TMV infection due to slight hypersensitivity was not changed either by long-lasting non-infectious symbiosis with TMV-sensitive tomato or in a constant environment (32±2 °C, 9 700 lx) which was adequate to change the hypersensitive reaction of some other TMV-hosts. On the contrary, enhanced temperature provoked a still more severe local and necrotic response in the potato varieties. This led to systemic necrosis and the quick death of potato components systemically infected in graft symbiosis with tomato under normal greenhouse conditions when transferred into an environment of enhanced temperature. |

The carbohydrate management of leaves of prunus laurocerasus L. at low and Sub-Freezing temperaturesL. The leaves of field-grown bushes as well as those kept under controlled conditions were analyzed at one week intervals at the end of the winter and in early spring from March 11 to May 6, 1970, The effect of four different temperature regimes was compared (day/night) : A : - 2 to - 6 °C/ - 6°C. B: 6°C/ -6°C, C: 6 °C/6 °C and D: 12 °C/ - 6 °C. During the whole experimental period of 56 days a net increase in dry weight 246.84 mg dm-2, i.e. 21.7 % was found under field conditions. A negative balance in assimilate management was recorded only during the one week period. The total content of sugars decreased from 11.68 % to 6.42 % while the content of starch rose from zero to 3.12 %. The net increase in dry matter found under controlled conditions was 6.69 (A), 4.68 IB). 6.88 (C) and -0.43 (D) mg dm-2 d-1. |

The different behaviour of long and short shoots as regards apical dominance may be tested with this substance. Shoots growing out through ring fasciations first form leaves lacking the teeth and marginal primordia corresponding to bud scales. Are fasciations facilitate correlative studies on anisophylly. Sylleptic branching is cleared by additional fasciations provoked by the cytokinin supply. The effect of gibberellin which decreases these deformities corresponds to the correlative influence of roots. The TIBA induced modification of aerial roots on internodes treated with naphthalenacetic acid reveals the importance of polarity and periodicity in the shoot development. |
Senescence and IAA Induced Elongation of Oat Coleoptile SectionsMonique FantinetBiologia plantarum 14:286-292, 1972 | DOI: 10.1007/BF02935854 Five mm sections excised from 20, 30 and 40 mm long coleoptiles were transferred into a indole-3-acetic acid (IAA) solution after an incubation in a medium containing buffer and sucrose without IAA. The ability of sections to elongate in dependence on IAA treatment was rapidly decreased at a temperature of 23 °C within a period of time depending both on the duration of incubation and on the age of coleoptiles from which sections were obtained. Under the same conditions of incubation and age the same decrease of the ability of sections to elongate at 4 °C was, however, observed only after a period fourfold longer than at 23 °C. Incubation of sections in the presence of benzimidazole did not extend their reactivity to IAA. This phenomenon may be therefore considered as a senescence reaction which is more effective at 23 °C than at 4 °C. |
