Histochemical demonstration of non-specific esterase in wheat root

Azo-coupling methods were used for demonstrating non-specific esterase in the wheat root in all parts of a transverse section, usually with the exception of the woody parts of the vascular bundle. The central cylinder gave a more intense reaction than the primary cortex and the rhizoderm. The reaction was not inhibited by dodecyl sulphate. A weakening of the reaction intensity was observed after application of AgNO3. The Tween method did not yield reliable results.

Proteins found during maturation and germination of seeds ofPhaseolus vulgaris L.

Seeds of the beanPhaseolus vulgaris L. (Veltruská Saxa cultivar) were gathered gradually at different stages of development, starting at fertilization up to full maturity. Seeds were freeze-dried and the dry solid used for preparing extracts which were analyzed by immunoelectrophoresis for the presence of proteins resembling those contained in the cotyledons of a mature seed. Proteins from cotyledons of the first stages of development of bean seedlings were analyzed similarly.
After a preparatory period, approximately from the second-third seed development stage, there is a period of intense protein synthesis that characterizes cotyledons of a mature seed. These proteins increase in quantity and are differentiated in quality up to maturity when a single antiserum detected a total of 12. After germination both the quantity and number of these proteins decreases.
It was found that some proteins are metabolically more active, both during synthesis and cleavage. This holds e.g. for phaseolin during maturation, as well as during germination. In addition, phaseolin changes its electrophoretic mobility, which is apparently due to proteolytic hydrolysis of phaseolin molecules.
During the last phase of maturation, viz. dehydration of seeds, some new proteins suddenly appear, apparently synthesized from pre-formed peptide chains.
In the discussion the possibility is taken up that the beginning on the synthesis of specific proteins characteristic for mature seeds is the cause underlying the disturbances in the embryonal development of distant hybrids.

Peroxidase isozymes in wheat infected withTilletia controversa Kühn and rye infected withPuccinia recondita Rob. exDesm

Peroxidase isozymes were studied in the case of two diseases:Tilletia controversa in wheat andPuccinia recondita in rye. The method employed was that of disc-electrophoresis on acrylamid gel. In the case of systemic disease (Tilletia controversa) no change was found in the isozyme pattern until a concentration of pathogen took place in the ears that began to form. Proofs are given that the new isozyme in unmatured ears comes from the pathogen. In the case of rye infected withPuccinia recondita a new isozyme was found; the latter does not originate in the pathogen, its origin is being discussed.

Vessel member length-diameter and perforation plate-vessel member length relations inDioscorea alata L.

The vessel member length-diameter and perforation plate-vessel member length relations inDioscorea alata L. have been investigated. The two types of vessel elements, one with scalariform perforation plate and the other with foraminate perforation plate, have been considered for correlation tests; the variables being vessel member length and diameter, and vessel member length and perforation plate length. Significant positive correlation was found only in the case of perforation plate length and vessel member length for types with scalariform perforation plates.

Der Einfluss von Pyrazon auf die Katalase-Aktivität in den Blättern der Senfpflanze (Sinapsis alba L.) und der Zuckerrübe

Senf- (Sinapis alba L.) und Zuckerrübenpflanzen wurden in der Phase des ersten echten Blattpaares mit dem Herbizid Pyrazon (=1-Phenyl-4-amino-5-chlorpyridazon-6) in Form eines Spritzpulvers-Pyramin-in der Konzentration 0,4% besprüht. 1, 4, 7 und 14 Tage nach der Besprühung wurde mit Hilfe der volumetrischen Methode nachJermakov und Mitarbeitern die Katalase-Aktivität im Blattsaft gemessen. Es wurde festgestellt, dass die Katalase-Aktivität in den Blättern der sensitiven Pflanze (Senf) durch die Einwirkung von Pyrazon im Vergleich mit dem Blattsaft aus unbesprühten Blättern herabgesetzt erschien, wogegen sie in den Blättern der resistenten Pflanze (Zuckerrübe) nicht abgesunken war.

Sorption of some ions by algae related to their trophic conditions

The unicellular algaeScenedesmus obliquus (125),Chlorella pyrenoidosa (82) andCoccomyxa solorinae saccatae (111) were studied with respect to the form of uptake of potassium, phosphate, calcium and zinc ions and to the energy sources involved: light under autotrophic conditions, glucose under mixotrophic or heterotrophic conditions (in light and in darkness or together with yeast extract as an auxotrophic substrate). We respected the trophic conditions of algae when preparing the experimental material (precultivation). The following conditions were reached:
(1) The three algae grow faster in a glucose medium under mixotrophic conditions and are capable of growing on it also heterotrophically:Ch. pyrenoidosa andSc. obliquus grow substantially better thanC. solorinae saccatae. The first two algae grow more intensively in a glucose medium containing yeast extract whileCoccomyxa does not. After cultivation under mixotrophic conditions the first two diminish endogenous respiration, the third raises it. Glucose stimulates respiration in the first two when grown autotrophically, while after mixotrophic cultivation the effect of glucose is suppressed inCh. pyrenoidosa and in the other two only after growth on glucose with yeast extract.
(2) A relationship between sorption and metabolism was established for potassium in all the algae investigated. The effect of light is generally positive, heterotrophic components (glucose and possibly yeast extract) stimulate the uptake in the dark as long as they represent a suitable mixotrophic medium for the given alga.
(3) The uptake of K+ was proportional to concentration in solution and independent of Ca2+.
(4) The uptake of phosphate is clearly stimulated by light, while no stimulation under mixotrophix or heterotrophic conditions on glucose was observed-actually, a depression usually took place.
(5) At concentrations from 10-6M to 10-2M the uptake of phosphate is non-linearly but positively related to the concentration in the medium. The presence of Ca2+ is the cause of non-metabolic forms of phosphate sorption.
(6) Two forms of calcium uptake were found: the predominant one is adsorption which can be associated with the presence of phosphate ions; the metabolic uptake represents a considerably lower fraction which is characterized either by a light dependence or at least by a rise of calcium content between t=0.1-6 h.
(7) The absorbed Ca2+ can be washed out up to 75% with dilute hydrochloric acid. The total adsorption and remainder after washing depend on the pH of the solution from which the sorption took place (a considerable rise beyond pH 6.5). A substantial effect on the formation of the adsorption system was found to be due to organic nutrients. Algae after mixotrophic precultivation possess a decreased sorptive capacity.

The Mechanical transmission and some properties of a virus disease of cow-parsnip,Heracleum sphondylium L

The described virus of cow-parsnip,Heracleum sphondylium L., was found in three ruderal localities of Greater Praha. The symptoms are manifested by decolorations which consist of bright yellow areas spreading from the centre of the leaf blade along the main veins. These symptoms appear severely in May. Under higher temperatures and in a chronic stage of infection the symptoms are more or less masked. The disease is mechanically transmissible to parsley, coriander, parsnip, dill, sowbane,Chenopodium quinoa and C.giganteum. The author failed to transmit the disease to celery, carrot, caraway and to 27 species of differential host plants, he failed in the transmission of the virus by the dodder,Cuscuta campestris YUNCK., too. Thermal inactivation point of the virus lies between 51° and 55° C. Infectivity of extracted sap was lost after 2 days at room temperature.

Interaction of indoleacetic acid with synthetic and native growth regulators during transfer of32P into epicotyls of etiolated pea seedlings

If both cotyledons are amputated from non-decapitated pea seedlings, the intensity of transport of32P into intact epicotyls is raised more than twofold. This is apparently connected with the regulatory-inhibitory effect of the cotyledons that can be simulated by a 0.1% paste of indoleacetic acid (IAA). If both amputated cotyledons are replaced by this paste, the intensity of32P transport is again decreased to the level found in plants with intact cotyledons.
When etiolated pea seedlings grown in the dark in 100% relative humidity were decapitated, the32P transport to the top of the epicotyl stumps covered with 0.5% IAA paste was strikingly decreased if the stumps were covered below with a 10% paste containing chlorocholine chloride (CCC) or with a 0.5% paste containing triiodobenzoic acid (TIBA). On the other hand, the intensity of32P transport was significantly increased if the IAA paste was put above a 0.5% paste with kinetin which alone does not raise the transport intensity. No such synergism toward32P transport could be established between IAA and gibberellin.

Stomatal and cuticular transpiration of beans (Phaseolus vulgaris M.) attacked by rust (Uromyces appendiculatus [Pers.] Link)

The changes of stomatal and cuticular transpiration of bean plants were investigated by graphical transpiration curves method (Slavík 1958). Bean leaves were infected by fungusUromyces appendiculatus (Pers.) Link. After the infection the intensity of stomatal transpiration had a decreasing tendency. Beginning with the sixth day after infection, the proportion of stomatal and cuticular transpirations becomes more expressive, i.e. the leaves transpire more by cuticles than by stomata. The higher share of cuticular transpiration brings extensive water relations to the diseased plants.

Studium autopathischer und allelopathischer Eigenschaften von Kulturpflanzen durch Folgekulturen-Methode

In der Arbeit wurde der Einfluss der Vorkulturen von Lein, Korn, Mohn und Ackersenf auf die folgenden Kulturen der gleichen oder einer anderen Art bei unmittelbar aufeinanderfolgenden Aussaaten sowie bei Aussaaten nach verschieden langen Intervallen unter Abstufung der Dauer der Lagerung des Bodensubstrates untersucht. Die Versuche wurden in Gefässen mit Komposterde durchgeführt, die im Versuchsgarten aufgestellt worden waren. Es wurde das Wachstum der Erst- und der Folgekultur mit Hilfe der Bestimmung der Trockensubstanz der oberirdischen Teile und der Wurzeln gewertet. Im Laufe der Wachstumsentwicklung der Folgepflanzen wurden Bodenproben entnommen und in denselben der Gehalt an physiologisch zugänglichen Stickstoff-, Phosphor- und Kali-Mengen bestimmt.
Alle vier in Betracht gezogenen Vorpflanzen bewirkten nachweisbare Veränderungen des Wachstums der Folgepflanzen. Lein und Mohn wirkten auf lein und Zuckerrübe als Folgepflanzen durch autopathische oder allelopathische Faktoren im Boden. Die Wirksamkeit von Korn als Vorpflanze auf Korn und Ackersenf als Vorpflanzen auf Gerste war nicht so deutlich. Von den Ergebnissen der beiden letzten Fälle lässt sich nicht direkt auf das Vorhandensein autopathischer oder allelopathischer Faktoren schliessen.
Bei unmittelbarer Kultivierung von Folgepflanzen im Boden der Vorpflanzen ohne Lagerung wurden nur Wachstumshemmungen festgestellt. Versuche mit abgestufter Lagerungsdauer bieten die Möglichkeit eine ganze Skala von Wachstumsänderungen an den Folgepflanzen von der Hemmung bis zur Stimulierung zu erfassen. Die Wirksamkeit der Vorpflanze auf die Folgepflanze ändert sich erheblich mit der Lagerungsdauer des Bodens nach der Vorpflanze.
Die Wachstumsänderungen der Folgepflanzen ergaben mit Ausnahme der Versuche mit Korn und teilweise mit Ackersenf weder Beziehungen zu den Veränderungen des Gehalts der untersuchten Nährstoffe, noch auch zur angewachsenen Pflanzenmenge der Vorkultur.

Adaptability in enzymes

In studying inhibitors and activators of enzymes it has often been found that inhibitors act as activators when in very low concentrations and that certain activators (in suitable concentrations) show inhibition. Similar results were obtained in the present work in studying urease from soya bean in the presence of lead acetate. Lead acetate is normally an inhibitor of urease. It was found, however, that in small concentrations of lead acetate increase in the activity of urease occurs. On prolonging the time of the reaction inhibition develops and still later increased enzymatic activity again occurs. The results were elaborated statistically and are discussed in relation to the use of "adaptation" of urease in unfavourable conditions in reaction mixtures by the action of lead acetate.

Bestimmung der Intensität des Gaswechsels mit85Kr

Während Assimilations- und Transpirationsmessungen die Durchlüftung den Pflanzenteile und die Reaktion der lebenden Zellen mit der sie umgebenden Luft nur gemeinsam widerspiegeln, lässt sich mit dem radioaktiven Edelgas85Kr der Gaswechsel zwischen Aussenluft und Interzellularluft unabhängig vom Stoffwechsel und ohne künstliche Druckdifferenzen erfassen. Der Gassaustausch zwischen der Interzellularluft und der Aussenluft wird dabei durch die Austrittsgeschwindigkeit des vorher kurzfristig durch Diffusion oder Unterdruck eingeführten85Kr aus dem Pflanzenteil charakterisiert.
Orientierende Versuche unter Freilandbedingungen an den benadelten Zweigen einiger Koniferen sowie Zweigstücken mit und ohne Rinde erbrachten Unterschiede in der Geschwindigkeit des Gaswechsels sowohl zwischen verschiedenen Pflanzen unter gleichen Bedingungen als auch bei der gleichen Pflanze unter verschiedenen Bedingungen.

Natural infections of Sisymbrium loeselii Jusl. with cabbage black ringspot and tobacco mosaic viruses

Investigating weeds for viruses in ruderal localities of Greater Prague two forms of mosaic diseases inSisymbrium loeselii Jusl. were demonstrated (green and yellowish-green mosaic). Transmission tests carried out on differential host plants showed that the green mosaic is caused by cabbage black ringspot virus (CBRV) and the yellowish green by mixed infection of CBRV and tobacco mosaic virus (TMV).
TMV-isolate is characterized as an unusual necrotic strain; its capability to reproduce in cruciferous plant in nature is unique.
It was ascertained that green mosaic was commonly spread overSisymbrium plants in ruderal ***DIRECT SUPPORT *** A01GP029 00004 associations on Prague territory; epidemiological significance of this discovery is discussed.

The effect of 5-azacytidine on the root meristem ofVicia faba

The primary roots ofVicia faba seedlings were placed in a solution of 5-azacytidine and their further growth was observed after being replaced in running tap water. No inhibition of elongation occurred during the action of the 10-5 M solution of 5-azacytidine for 24 hours, but during subsequent cultivation in water in the absence of inhibitor, further growth was blocked. This inhibition could be overcome by cytidine, uridine, sodium azide, 5-azidomethyluracil and simultaneously with the 5-azacytidine solution. Inhibition was accompanied by a high incidence of chromosome stickiness and to a less extent by an incidence of chromosome aberrations. The occurrence of stickiness and chromosome aberrations was prevented by adding excess cytidine to the 5-azacytidine solution.

Untersuchungen über die allelopathischen Eigenschaften der Getreidearten bei deren Nachkultivierung

In dieser Arbeit wurden allelopathische Eigenschaften von Roggen, Weizen, Gerste und Hafer studiert, die sich bei gegenseitigen Beziehungen dieser Getreidearten geltend machen können.
Als Beweis für die Wurzelbeeinflussung der angeführten Getreidearten wurde eine Methode verwendet, bei der die erwähnten Getreidearten in derselben Kultivierungserde wuchsen, und zwar teils unmittelbar nacheinander, teils in abgestuften Zeitintervallen nacheinander mit verschiedenlanger Lagerungszeit der Erde.
Die Versuchsergebnisse haben die Ansichten zahlreicher Verfasser bestätigt, die behaupten, dass die angegebenen Getreidearten allelopathish aktiv sind. Roggen, Weizen und Hafer beeinflussten signifikant das Wachstum der nachfolgenden Kulturen; dabei erwiesen die Ergebnisse nicht deutlich jene Beeinflussung, die durch das Erschöpfen der Nährstoffe durch die vorhergehende Pflanze bedingt worden wäre.
Die angewandte Methode einer nachfolgenden Kultivierung erwies sich geeignet als Beweis für die allelopathischen Eigenschaften der Pflanzen unter natürlichen Bodenbedingungen. Die erwähnte Methode lässt sich vorteilhaft insbesondere in jenen Fällen anwenden, bei denen der Unterschied im Habitus der oberirdischen Teile der Pflanzenarten das Studium des Charakters ihrer gegenseitigen Beziehungen bei gemeinsamem Wachstum unmöglich macht. Diese Methode könnte ebenfalls eine Anwendung in der landwirtschaftlichen Praxis finden, wenn eine geeignete Zeitfolge der Pflanzen bei der unmittelbare Reihenfolge auf demselben Standort ermittelt werden soll.

The effect of inhibitors on the mutagenic activity of N-methyl-N-nitrosourea and N-ethyl-N-Nitrosourea inArabidopsis thaliana

NaN3-a respiratory inhibitor and anaerobic conditions (nitrogen) decrease the mutagenic activity of N-ethyl-N-nitrosourea and N-methyl-N-nitrosourea inArabidopsis thaliana. DNP-an uncoupling agent increases the effect of both nitrosoureas, but this may not be caused by the specific action of DNP.

Morphology and function of the Golgi apparatus

The polymorphism of the dictyosomes in the root meristeme ofFagopyrum is connected with their various functions in secretory processes and cell differentiation. The dictyosomes containing vesicular dilatations of the cisternae, which in this object occur more frequently than in others, probably participate in a similar way as the Golgi apparatus of the animal cell in the formation of lysozomes, in the formation of elements belonging to the group of dense bodies analogical lysozomes. These bodies are present in large numbers in the cytoplasm of cells, containing dictyosomes with vesicular dilatations. The other forms of the dictyosomes reveal indications of their participation in the production of the carbohydrate material of the cell walls, like most dictyosomes of other plant objects. However, no fusion of the Golgi vesicles with the plasmalemma was observed. According to their morphological appearance the typical forms of dictyosomes were classified on the basis of their relationship to secretory processes. Simultaneously the morphology and function of the Golgi apparatus was compared in the animal and plant cell. Several morphological varieties of the dictyosomes of plant cells, observed after the action of pathogenic factors and the effect of the fixation procedures, were also noticed in small quantities in the cells of the investigated objects.

Attempts to diminish the injurious consequences of low winter temperatures by means of vitamins and growth substances

Plants of two varieties of winter wheat-Besostoya 1 and Etoile de Choisie- were raised in the field. The same number of selected plants from each variety were frozen once in a refrigerator at a temperature of -15°C for 24 hrs. After freezing, the plants were gradually thawed and returned to the field. Early in the spring, test and control plants were treated with one of following diluted solutions: vitamin B1, vitamin B6, indolyl-3-acetic acid and adenine (the four solutions at a concentration of 0.01% and 5% urea solution in water) by immersion for 15 hrs. at a temperature of 18-20°C. The plants were measured in the spring and in the summer, the yield being determined after ripening.
It was found that all substances used helped to diminish the deleterious consequences of overwintering and to raise the yield of the plants. Vitamin B6 exerted the strongest influence. Vitamin B6 treated plants produced higher or slightly lower yields than those of the non-frozen control. Plants treated with vitamin B1 gave yields close to, but in most cases lower than those of the non-frozen control.
The yields of plants treated with solutions of the remaining substances were appreciably lower than those of the non-frozen controls.
Plants of the more winter-hardy variety Besostoya 1 recovered relatively more easily under the effect of the substances used than the plants of the less winter-hardy variety Etoile de Choise.

The classic Golgi apparatus and vacuoles

The dense vacuoles, considered to be the classic Golgi apparatus in the root meristem ofFagopyrum, were studied by the following methods: 1. Impregnation methods for the demonstration of the Golgi apparatus, 2. cytochemical methods, 3. electron microscopic methods in the light microscope and 4. the electron microscope. A comparison was made with the classic Golgi apparatus in animal cells in the light and electron microscope.
Dense vacuoles inFagopyrum and also evidently in other plants, were taken for the classic Golgi apparatus on account of their morphological similarity to the Golgi apparatus in animal cells on impregnation with silver and osmium and their staining preperties with lipoid methods. Dense vacuoles differ from the classic Golgi apparatus in other chemical properties, such as content of phenol substances, etc. No formations were found in animal cells which were similar to dense vacuoles on investigating by electron microscopy. In the electron microscope dense vacuoles have the appearance of derivatives of the normal light vacuoles known in plant cells. They therefore belong to vacuome of plant cell and cannot be analogous to the classic Golgi apparatus in animal cells. Thus the use of the term Golgi apparatus for dense vacuoles is not well founded.
A comparison was made of fixation and impregnation used in the light microscope with fixation in the electron microscope. After fixation with permanganate, dense vacuoles have the same shape as after impregnation. After fixation with permanganate, they stain an intense black in the same way as after impregnation with silver and osmium. The form of the vacuoles is dependent on the fixation used. The comparison was made in the light microscope.

Reactions and resistance ofChenopodium species to tobacco mosaic virus

Chenopodium species react on infection with tobacco mosaic virus by the formation of chlorotic or necrotic lesions and later by the abscission of infected leaves. A transition of local infection into the stem has been observed exceptionally inChenopodium quinoa, C. hybridum, andC. rubrum, but no systemic infection of the leaves followed. Systemic infection was demonstrated only inC. polyspermum andC. murale. The recovery of new sprouts was demonstrated in C.murale in the late chronic phase of infection.

Veredelung der Walnuss und anderer Holzarten in der Zeit der echten Winterruhe

Im Verlaufe der Arbeit ist festgestellt worden, dass es nach einer Sprossverletzung von Holzarten in der echten Winterruhe unter günstigen Bedingungen zu einer Aufhebung der Teilungshemmung der Zellen der meristematischen Gewebe in der Nähe der Wunde kommt. Die Kambiumzellen in einer Entfernung einiger mm von der Stelle der Verletzung und die übrigen intakten Gewebe des Sprosses sowie seine Knospen verbleiben im Ruhezustand. Bei einem Veredlungsvorgang in der Ruhezeit verwachsen unter günstigen Bedingungen, insbesondere der Temperatur und Feuchtigkeit, die Veredlungskomponenten ohne dass Knospen an den Pfropfreisern oder den Unterlagen ausschlagen.
Die auf Grund dieser Unterlage ausgearbeitete Veredlungsart der Walnussbäume hat sich praktisch bewährt. Auf einem Quadratmeter Vermehrungshaus- oder Erwärmungskammerfläche erzielen bei 26°C und einer hohen relativen Luftfeuchte im Laufe von drei Wochen cca 500 Walnussveredlungen ein Verwachsen. Die Veredlungen werden im Keller oder in einem frostfreien Raume eingeschlagen und im Frühjahre ohne Schutz nicht ausgetrieben in der Baumschule aufgepflanzt. Der Prozentteil der verwachsenden veredelten Pflanzen ist hoch.

Action of volatile substances liberated from couch grass

A study was made of the action of volatile substances liberated from the rhizome of couch grass on wheat seedlings. The experiments were made in a closed atmosphere in glass vessels with continual removal of expired CO2 and the addition of O2.
Volatile substances liberated from couch grass produced numerous morphological and physiological changes in wheat seedlings. They caused retarding of the growth of overground parts and roots, the excessive formation of root hairs, curling of the roots and bending of the coleoptiles. The respiratory rate was higher in the leaves and roots of the experimental wheat seedlings than in the controls. The changes observed were similar to those produced by hydrocarbons of the ethylene series, which are well documented in the literature.

Research on substances inactivating the X virus present in sap from potato leaves

The work ofJermoljev andBrèák (1964) showed that the sap of potato leaves contains substances inactivating X, Y and S viruses and that there was an increasing trend in the content of these substances in varieties resistant to these viruses. In further research it was found that potato tubers of varieties which are resistant and susceptible to the viruses contained the same amounts of inactivating substances. Differences in the power of inactivating the viruses evidently appeared in the leaves. Inactivating substances could be removed by boiling, they did not pass through a dialysing membrane and were adsorbed by animal charcoal. On centrifuging sap from potato leaves in a Spinco L ultracentrifuge for 60 min. at 40,000 r.p.m., the inactivating substances remained in the supernatant. When sucrose gradient centrifugation for 60 min. at 24,000 r.p.m. was employed, the inactivating substances remained in the layer of sap and 10% sucrose. Inhibition of the activity of certain enzyme groups did not affect the power of sap to inactivate X virus. Inactivating substances could be isolated chemically. The best method of isolation, however, was fractionation of sap, after ultracentrifugation, on a Sephadex G 50 column, rinsing the column with McIlvaine buffer at pH 6.5. Inactivating substances can be isolated, concentrated and preserved by lyophilization by this method. It is difficult to say precisely to what chemical group the inactivating substances belong. Some reactions indicate that they may be low molecular proteins.

Application of growth analysis to potatoes in field culture and some specific features of potato growth

In field experiments, potato varieties with different lengths of the vegetation period were compared. The methods of growth analysis were applied to the evaluation of these experiments. Some important specific features of potato growth had to be taken into account to make the application of the growth analysis successful and to enable valid and reliable results to be obtained.

Electron microscopy of ribosomes isolated from young green fruit of the apple tree

Many studies have been made on ribosomes both in plant and animal material, on account of their importance in the proteosynthesis of protein. In plant material, studies have been made on the pea by Ts'o andBonner (1956), Ts'o,Bonner andVinograd (1958),Setterfield et al. (1960) andSisakyan et al. (1963). Ribosome from spinach were investigated byLyttleton (1962) andMurakami (1963) and fromClivia byMikulská et al. (1962). As part of a wider study of the mechanism of biosynthesis of nucleic acids in apple trees, we isolated ribosomes from the young green fruit and studied them by means of electron microscopy. Young apples were selected because cell division is very intense at this stage of growth of the apple.

Influence of soil heterogeneity on water regime and yields of lucerne, sugar beet and wheat

Lucerne plants in the first crop year as well as plants of spring wheat from different sites of the respective experimental plots showed differences in sap exudation from detopped roots or from stumps of the main shoots, reflecting differences in properties of the soil profile. The differences in sap exudation were observed at a time when the plants did not show any visible differences in water availability.
Differences in the water potential deficit of the leaf blades of lucerne plants in the second crop year and of sugar beet plants, related to differences in soil profile properties, were observed in another series of experiments. Sugar beet plants showed a higher physiological lability than lucerne plants.
The sites characterized by unfavourable plant-water-relations usually gave lower yields. The coefficients of variability for the yields of lucerne fresh matter from irrigated plots were three times lower than those for yields from plots without irrigation, influenced by soil heterogeneity.

Contribution to the study of heterogeneity in the leaves of a spring wheat plant

The effect of the individual leaf blades of spring wheat on the dry matter of stalks, chaff and grain (caryopses), of spikes and total overground part, was studied. In the experimental plants the individual leaf blades were detached according to the scheme given, at the beginning of shooting, (A), at the beginning of earing (B), and at the beginning of flowering (C). The dry matter (fresh weight) of the stalk was least decreased if either the lowest or the uppermost leaf blade was severed during the developmental phase of shooting. The dry as well as the fresh weights of chaff were least affected in those plants where the leaf blade was removed during the developmental phase of flowering. Both the dry and fresh weights of caryopses were least decreased if either the lowest or the uppermost leaf blade was removed during the developmental phase of flowering. The dry weight as well as the relative water content of chaff and ear grains were most decreased following removal of leaf blades during the developmental phase of shooting. The relative water content of chaff, grains and ears was most decreased following removal of developed leaf blades during the developmental phase of earing. It was confirmed that in addition to the photosynthetic activity of leaves the photosynthesis of other parts of the stem system (stalk internodes, ear awns etc.) participated in the production of total dry matter of experimental plants. The photosynthetic activity of leaf blades was particularly high up to the earing phase, while subsequently the photosynthesis of extrafoliar area (stalk internodes and ears) predominated. In spite of this, participation of the total leaf area is very high in the formation of grain dry matter (over 50%), as well as of the total dry matter of plant (over 80%).

Zur Problematik der Verlässlichkeit der refraktometrischen Methode für die Indikation des Wasserbedarfes der Kartoffeln

Bei Erforschung des Einflusses der herabgesetzten Bodenfeuchtigkeit auf die osmotischen Verhältnisse im Zellsafte der Kartoffelpflanzen wurde die refraktometrische Methode für die Bestimmung deren osmotischen Zustandes bewertet. Es zeigte sich die Notwendigkeit eine gewisse Vorsicht bei ihrer Anwendung zu bewahren. Die Elektrolyte, die von der refraktometrischen Methode verhältnismässig weniger erfasst werden, haben ein wessentliches Übergewicht in der Bildung der osmotischen Gesamtwerte des Zellsaftes der Kartoffeln über den anderen Hauptteil, nämlich die Fehlingslösung reduzierende Substanzen. Sie haben auch oft einen entgegengesetzten Vegetationstrend gegen den molekular aufgelösten organischen Substanzen die die Fehlingslösung reduzieren. Aus diesem Grunde ändert sich die Korrelation zwischen den osmotischen Gesamtwerten und dem Brechungsindex des Saftes in einzelnen Fällen sowohl als auch während der Vegetation. Diese Verhältnisse sind auch in einzelnen Organen der Kartoffelpflanzen verschieden.
Eine bedeutende Herabsetzung der Bodenfeuchtigkeit, dauernd oder zeitlich, verursacht nur eine relativ geringe Erhöhung der osmotischen Gesamtwerte im Vergleiche mit dem Einflusse der Wetteränderungen. Von diesen Ergebnissen folgt, dass die refrektometrische Methode nicht empfindlich genug ist, um den Einfluss der Bodenfeuchtigkeit auf die osmotischen Verhältnisse in der Kartoffelpflanze bestimmen zu können.
