Relationship between Calcium and Pyruvate KinaseJ.M. Ruiz, I. López-Cantarero, L. RomeroBiologia plantarum 43:359-362, 2000 | DOI: 10.1023/A:1026757320303 Tobacco plants (Nicotiana tabacum L. cv. Sevilla) were grown under controlled conditions. The leaf content of Ca2+, Mg2+ and K+, and the activity of the pyruvate kinase were analyzed. The increased application of Ca2+ diminished the content of K+ and Mg2+ in leaves, and decreased the activity of pyruvate kinase. Taking into account these results, we suggest the pyruvate kinase activity as an bioindicator of the contents of the Ca2+, Mg2+ and K+. |
Effect of Increasing Concentrations of Lead and Cadmium on Cucumber SeedlingsV.V. Talanova, A.F. Titov, N.P. BoevaBiologia plantarum 43:441-444, 2000 | DOI: 10.1023/A:1026735603890 The effect of lead and cadmium on biomass accumulation of cucumber seedlings (Cucumis sativus L.) as well as the contents of abscisic acid (ABA), free proline and soluble proteins in leaves were studied. Seedlings were subjected to lead nitrate or cadmium bromide in low concentrations (1 - 5 µM) for 1, 4 or 7 d, and then to the action of the same substances in high concentrations (500 - 1000 µM). The pretreatments of the seedlings with heavy metals in low concentrations enabled them to tolerate the subsequent high concentrations of cadmium and lead without injury. The plant responses to heavy metal treatment were accompanied by the accumulation of ABA, free proline and soluble proteins in leaf tissues. |
Leaf Structure of Tobacco In Vitro Grown Plantlets as Affected by Saccharose and IrradianceB. Radochová, A. Vičánková, J. Kutík, I. TicháBiologia plantarum 43:633-636, 2000 | DOI: 10.1023/A:1002820620688 Tobacco plantlets were cultured in vitro under high (200 µmol m-2 s-1) or low (60 µmol m-2 s-1) irradiance with or without saccharose in the medium. Light microscopy and image analysis were used to evaluate the effect of these culture conditions on leaf anatomy. Addition of saccharose resulted in thicker leaves (all leaf layers) and larger mesophyll cells under both growth irradiances. Various irradiance affected leaf anatomy differently when plantlets had been cultivated in presence or absence of saccharose in the medium. While under high irradiance in presence of saccharose leaf thickness and number of chloroplasts per cell section were increased, plantlets grown under high irradiance in absence of saccharose had thinner leaves and less chloroplasts per cell section. The changes were more pronounced in palisade parenchyma layer. |

Two soybean (Glycine max L. Merr.) genotypes were examined: BOSA (drought tolerant) and L 121 (drought susceptible). Seed samples were taken from plants at three stages of seed maturation (50 and 70 d after flowering, and at full maturity). The earlier synthesis of the β-subunit of the 7S protein occurred in the drought susceptible cultivar. We have not found such differences in the synthesis of the α- and α'-subunits of the 7S protein. Our results did not confirm significant genotypic differences in protein composition of the mature seeds between the cultivars studied, but have pointed out to the differences in the dynamics of protein biosynthesis during seed maturation and desiccation. |

The chromosomes of this species are small, morphologically similar, median and submedian. In the basic chromosome set the most distinguishable is chromosome number 1 which was subjected to detail analysis. It was found that there are two types of this chromosome which contribute differentially in diploid, triploid and tetraploid plants. |

Many monosomics were derived directly from irradiation, but a number of monosomics were also recovered in the progeny of plants with translocations and of desynaptic plants. Only 28 primary monosomics showed normal pairing at metaphase-1 of meiosis. The others formec rare trivalents or additional univalents. Partial desynapsis was detected in some monosomics. The pollen fertility levels of monosomics are presented. New morphological characters were detected among the monosome plants of cotton. |
Nodule Structure and Functioning in Chickpea (Cicer Arietinum) as Affected by Salt StressS. Babber, S. Sheokand, S. MalikBiologia plantarum 43:269-273, 2000 | DOI: 10.1023/A:1002764512963 Cicer arietinum L. plants raised in sand culture under natural light were subjected to salinity stress induced by mixture of NaCl, CaCl2, MgCl2 and MgSO4 (40, 60 or 80 meq dm-3). Acetylene reduction activity (ARA) of nodules, leghemoglobin content and nodule structure were followed 55, 75 and 85 d after sowing. ARA declined significantly under salt treatments and the lowest ARA was observed at day 85 after sowing. Decrease in ARA was consistent with decreased nodule leghemoglobin content. The leghemoglobin content of control plants decreased by 50 % at day 85 indicating senescence of nodules. This senescence was further accelerated by salt treatment after which the leghemoglobin content fell to negligible levels. The structural changes associated with salt stress were mainly reduction in size of the nodules, decreased meristematic zone, reduced number and degradation of symbiosomes, reduced intercellalar spaces and deposition of electron dense material in the intercellular spaces in the cortex of nodules. |
Linkage Mapping of DNA Markers Generated with Specific and Non-Specific Gene Primers in SoybeanK.M. Soliman, M. Paladugu, T.E. DevineBiologia plantarum 43:337-346, 2000 | DOI: 10.1023/A:1026703823512 Polymerase chain reaction (PCR) has been used extensively in the construction of linkage maps for many cultivated crops including soybean, [Glycine max (L.) Merr]. In this study, four sets of oligonucleotide primer pairs of known genes (pearl millet Adh 1, nodule specific proline-rich protein, Drosophila homeobox, heat shock protein), several different combinations from kits A, D, E, and J of arbitrary primers and five primer pairs of soybean simple sequence repeats of varying length (Satt 9, Satt 20, Satt 42, Satt 64, and Satt 30) were utilized in PCR to identify molecular markers which were then used to construct a genetic linkage map. DNA for the PCR reactions was isolated from 65 recombinant inbred soybean lines resulting from crossing PI 290,136 and BARC-2 (Rj 4 ), followed by self-pollination for seven generations without selection. Mapmaker 3.0, a computer package, was used for construction of the linkage map. A total of 43 polymorphic markers were identified; 30 markers were linked and distributed among 5 linkage groups while 13 markers were unlinked. Arbitrary primers revealed more polymorphisms than specific primers. A combination of arbitrary primers A5 and A18 revealed the maximum number of polymorphic bands. Five observed linkage groups can be expanded in future soybean research by using additional markers. |
Amelioration of NaCl Stress by Arginine in Rice Seedlings: Changes in Endogenous PolyaminesR.K. Kakkar, S. Bhaduri, V.K. Rai, S. KumarBiologia plantarum 43:419-422, 2000 | DOI: 10.1023/A:1026715032115 Effects of NaCl (0.1 - 0.2 M) alone or in combination with 1 mM arginine on growth and endogenous polyamine (PA) content have been observed in two cultivars of rice differing in NaCl stress tolerance. The germination, seedlings fresh mass and water content decreased with increase in salinity in both the cultivars. This inhibition was partially alleviated by application of arginine. Cv. CSR-27 exhibited relatively better germination than cv. Bas-370 at different salinities. Total PA content increased in both the cultivars under NaCl stress alone and in combination with arginine. Putrescine to spermidine and spermine ratio was higher in NaCl-treated seedlings being more in cv. Bas-370 as compared to cv. CSR-27 and the ratio reversed to almost control level when arginine was applied along with NaCl. |

Pigment Diverse Mutants of Pseudomonas sp.: Inhibition of Fungal Growth and Stimulation of Growth of Cicer arietinumA.K. Goel, S.S. Sindhu, K.R. DadarwalBiologia plantarum 43:563-569, 2000 | DOI: 10.1023/A:1002877917537 A Pseudomonas strain MRS16 inhibited growth of different pathogenic fungi (Aspergillus sp., Fusarium oxysporum, Pythium aphanidermatum and Rhizoctonia solani) in vitro. Larger inhibition zones were obtained on nutrient agar and King's B media compared to potato dextrose agar and pigment production media. Mutants altered in production of fluorescent pigment were derived by nitrosoguanidine mutagenesis. The pigment overproducer mutant MRS16M-1 was more inhibitory whereas nonproducer mutant MRS16M-5 was less inhibitory than parent strain on nutrient agar medium. Addition of iron (100 µM FeCl3) in the medium decreased inhibition of fungal growth, suggesting the involvement of siderophores and other antifungal secondary metabolites. Seed bacterization of two cultivars of chickpea (Cicer arietinum cvs. H8618 and C235) differing in susceptibility to wilt caused initial root and shoot stunting at 5 d of growth followed by proliferation of secondary root growth at 10 d. Coinoculation of chickpea with Pseudomonas strain MRS16 or mutants and Rhizobium sp. Cicer strain Ca181 enhanced nodulation, nitrogen fixation and plant dry mass as compared to single inoculation with Rhizobium strain under sterile conditions. |

Foliar treatments with solutions, containing N6-benzyladenine (BA), indole-3-acetic acid (IAA) or (2-chloroethyl)-trimethylammoniumchloride (CCC) were conducted after chlorosis had been well manifested. Changes in growth, chlorophyll content, rate of photosynthesis, catalase and peroxidase activities in leaves, and the contents of Fe, Cu, Zn, Mn, and P in leaves were recorded. Growth regulators improved (CCC, IAA) or aggravated (BA) the physiological state of chlorotic plants. Their effect might be explained by changes in Fe transport towards the leaves, by increased efficiency of Fe utilization, and by effects on plant metabolism not involving Fe. |
Serologically-Related Anionic Peroxidases from Petunia and Cucumber can Substitute Flavonoid AntioxidantsV. Repka, I. FischerováBiologia plantarum 43:105-112, 2000 | DOI: 10.1023/A:1026515316171 In this study we have investigated whether naturally occurring flavonoid-deficient mutant Red Star of Petunia hybrida is capable of metabolizing H2O2 by invoking other antioxidant enzyme system. We demonstrated that reduced flower pigmentation due to a reduction in the chalcone synthase mRNA expression results in strong H2O2 accumulation accompanied by the induction of a specific set of anionic peroxidase (PRX), serologically-related to main cucumber srPRX. We found correlation between rate of H2O2 accumulation and qualitative, as well as quantitative changes in the srPRX expression which seems to be determined by flower phenotype. In detached flower buds cultured in vitro both abscisic acid and anther extirpation prevented anthocyanin pigmentation, and thus flavonoid biosynthesis, resulting in a marked accumulation of immunoprecipitable srPRX. In contrast, pigmented flowers cultivated under the same conditions did not accumulate corresponding srPRX. The results suggest that a specific set of anionic PRX can substitute for the absence of flavonoid antioxidants. |

Mediterranean species Lotus creticus creticus and Lotus creticus cytisoides. L. creticus creticus showed higher density of trichomes and stomatal density on the adaxial leaf surface than L. creticus cytisoides, whereas L. creticus cytisoides showed higher stomatal density in abaxial surface than L. creticus creticus. These morphological traits promoted clear differences in leaf surface water retention and leaf reflectance. Leaf water potential and photochemical efficiency were lower in L. creticus cytisoides than in L. creticus creticus. |

BarrenoBiologia plantarum 43:387-391, 2000 | DOI: 10.1023/A:1026738225755 The acclimation potential to high irradiance of two cultivars of watermelon, Reina de Corazones and Toro, calculated as the ratio of sun vs. shade activities of O2- and H2O2 scavenging enzymes and non-radiative energy dissipation, was similar. However, Reina de Corazones exhibited a higher capacity in absolute terms for photoprotection (harmless dissipation of absorbed light energy at PS 2 and ascorbate and O2- and H2O2 scavenging enzymes) suggesting a larger resistance of this cultivar to high irradiance. This could be seen as smaller decreases in fruit productivity and in lower oxidative injury as probed by malondialdehyde content in sun plants of Reina de Corazones than in Toro plants. Additionally, the results show that shading might be beneficial to both cultivars, presumably because it reduces the susceptibility of high irradiance-induced stress. |

The values of pH optimum for L-alanine aminopeptidase, L-proline iminopeptidase, and L-arginine aminopeptidase were 8.2, 7.4, and 7.9, respectively. The immobilized cells showed significantly lower aminopeptidase activity when compared to untreated cells. Alginate hydrogel was successfully used for immobilization of carrot cells retaining the activity of some aminopeptidases. |

Are the T-DNA Mutants Amenable to Standard Recombination Analysis?J. Relichová, J. ŘepkováBiologia plantarum 43:19-23, 2000 | DOI: 10.1023/A:1026586409811 Genetic analysis with T-DNA mutants often brings difficulties resulting from instability of the transgenic phenotype. In this work three different Arabidopsis thaliana T-DNA embryonic lethals and one T-DNA morphological mutant were analyzed in F2 progeny after 15 different crosses with marker lines for individual chromosomes. F2 analysis of 44 segregation ratios revealed segregation distortion of similar character consisting in abnormal excess of nontransgenic plants to the detriment of transgenic ones. We quantified this phenotypic drift (d) on the basis of phenotypic ratios given the respective formulas. The d values indicate the rate of F1 gametes which loose the T-DNA mutation or ability of its expression. The obtained d value were relatively high, 0.4 to 0.9 for individual crosses. It makes the standard recombination analysis with insertional mutants very problematic or even impossible. |
Quantitative Changes in Maize Membrane Proteins Induced by AluminiumI. Mistrík, L. Tamás, J. HuttováBiologia plantarum 43:85-91, 2000 | DOI: 10.1023/A:1026559131192 Sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) was used for monitoring Al-induced changes in polypeptide composition of membrane proteins isolated from 3-d-old maize seedlings subjected to aluminium stress. Analysis of peripheral membrane proteins isolated from maize root showed an Al-induced increase in accumulation of 14 polypeptides with apparent molecular mass from 10 to 135 kDa. Qualitative differences were found between peripheral membrane proteins isolated from root tip (increased accumulation of 4 polypeptides with Mr 42 000 - 135 000) and from root base (increased accumulation of 10 polypeptides with Mr 10 000 - 59 000). On the other hand, no Al-induced changes were observed in peripheral membrane proteins isolated from maize coleoptile and integral membrane proteins isolated either from root or coleoptile. These results indicate that peripheral membrane proteins undergo considerable changes during 24-h Al treatment while integral membrane proteins pattern is stable. |

The regenerated shoots elongated in the initial medium and most of them rooted in the presence of 2.41 µM indole-3-butyric acid, and flowered in vitro. Rooted plants could be transferred to the field after hardening. |

The study has been made to detect specific to Tripsacum isoesterases and isoperoxidases, zein subunits and SDS-protein protomers which could be used as markers for introgression of gene loci encoding these proteins from Tripsacum into hybrids of Tripsacum with Zea mays. Isoesterases and isoperoxidases as well protomers of SDS-protein complex specific to Tripsacum were detected in all hybrids analyzed. Zein subunits, specific to Tripsacum were detected in some of the analyzed hybrids which i that introgression frequency of the loci encoding proteins studied was different. Chromosome counts taken on the examined hybrids showed the addition of 9 - 13 Tripsacum chromosomes to maize chromosome complement. |
Triacontanol-Induced Changes in the Growth, Photosynthetic Pigments, Cell Metabolites, Flowering and Yield of Green GramG. Kumaravelu, V. David Livingstone, M.P. Wilezek] cultivar KM-2 were sprayed with different concentrations of triacontanol (TRIA) (0, 0.5, 1.0, and 2.0 mg dm-3) at 15 and 25 days after sowing. Foliar spray of 0.5 mg dm-3 TRIA significantly promoted the plant height, fresh mass, and contents of chlorophylls, saccharides, starch, soluble proteins, amino acid and phenols. Leaf nitrate content was reduced by 0.5 and 1.0 mg dm-3 TRIA with a corresponding increase in nitrate reductase activity. TRIA of 0.5 mg dm-3 stimulated the onset of flowering, pod production and retention, but less number of pods and seeds per plant were observed at 2.0 mg dm-3 treatment. |
Effect of Low Temperature on the Protein Metabolism of Wheat LeavesM.L. Pinedo, G.F. Hernández, R.D. Conde, J.A. TognettiBiologia plantarum 43:363-367, 2000 | DOI: 10.1023/A:1026765504374 The effect of low temperature on the protein metabolism of wheat primary leaves was examined. In seedlings transferred from 25 to 5 °C, total soluble protein accumulation, in vivo protein synthesis and breakdown, in vitro protein breakdown, and SDS-PAGE profiles of proteinases in gelatine-containing gels were analysed. Leaf protein content increased within a 7-d period (70 % over the initial value) in plants exposed to 5 °C. The fast protein accumulation observed on days 0 - 2 was mainly attributed to a decreased breakdown. In further days, parallelly to a slowdown in the rate of protein accumulation, the leaf proteolytic activity increased. The incubation temperature also had an influence on the proteolytic activity: Q 10 values for the 15 - 5 °C range were 80 - 200 % higher than those observed for the 25 - 15 °C range. On the other hand, the in vivo protein synthesis capacity, at either 25 or 55 °C, was not significantly modified in cold-treated plants. In addition to the enhanced activities of two serine-proteinases (previously found in control plants by SDS-PAGE analysis), cold-treated plants displayed a new proteinase, which had not been detected so far. |
Application of Esterase Isozymes for Garlic Ecotype IdentificationN. The isozyme patterns were obtained by means of vertical block electrophoresis in polyacrylamide gels. Quantitative and qualitative differences between loci of winter and summer garlic ecotype were found. This fact substantiates application of the isoesterases for identification of garlic clones. |
