Inhibition of peroxidase by algal humic and fulvic acids

In contrast to their soil counterparts, algal fulvic acids were more inhibitory than the corresponding humic acids. Fulvic and humic acids fromFucus vesiculosus were more efficient than the correspondingLaminaria digitata acids in inactivating the enzyme.Laminaria humic acids, which have no phenolic hydroxyls, showed a concentration dependent inhibition hardly in accordance with the presumed role played by these groups in the activity of oxidases.

Time-lapse microcinecmatography showed that the negative prototactic response of the chloroplast lasted 11-19 min and was completed 40 min after the beginning of irradiation; the contraction reached 11% in 60 min.

A propagation method for someFreesia cultivars usingin vitro cultures is described. Bud and root formation was evoked in callus tissue culture on a modified Murashige and Skoog's medium. Reconstituted plants were transferred into soil and cultivated in the glasshouse.

The regeneration of the tips of the rhizomorphs ofsphaerostilbe repens

The tips of the rhizomorphs ofSphaerostilbe repens transferred on agar medium regenerate some aggregated organs without an intermediate mycelium stage. Each extremity of the drawn rhizomorphs can give rise to eoremia on its air face and to rhizomorphs on the contact with tho culture medium. These heterogeneous environmental factors impose polarized regeneration, easily inverted, upon the rhizomorphs. Nevertheless, a residual and stable polarity remains; it is attributed to the apex which is more capable of bearing new organs than the other extremity. Tho efficiency to regenerate depends on the ago of collulos and on tho size of tho rhizomorph tip; howover it is very difficult to provent the formation of new aggregated organs.

Growth correlations and rna synthesis in different parts of the shoot apical meristem ofChenopodium rubrunt L. induced to flowering

Uridine-3H incorporation and RNA concentration were investigated in different parts of the shoot apical meristem ofChenopodium rubrum using autoradiography and cytophotometry. A single inductive cycle was sufficient to bring about postinductive first events in the shoot apex but not for complete flower differentiation. The initial activation of RNA synthesis manifested itself in all zones of the apex. The first increase was more conspicuous in the peripheral than in the central zone. The indications of the first events in the apices after a single inductive cycle disappear prior to morphological reversal to the vegetative state. Induction by three short days led to rapid flower differentiation. The increase in RNA synthesis and concentration was most conspicuous in the central zone in this case. The ratio of RNA synthesis and content between bud and leaf primordia (B/L) also change in relation to photoperiodic induction. In vegetative plants the B/L ratio was low while after induction it increased.
The shifts in activity of RNA synthesis observed in the shoot apical meristem are related to the changes in growth activity of the different parts of the apex. The growth ratios in the apices bear the character of growth correlations. The change in the growth correlations following photoperiodic induction together with the total activation of RNA synthesis are considered to represent one of the first events of the transition to the reproductive state.

Effect of different sugars on stomatal behaviour inMerremia aegyptia (L.) Urban andM. dissecta Hallier F.

The effect of mannitol, glucose and sucrose on the stomatal behaviour of two desert species,Merremia aegyptia andM. dissecta has been studied. Stomatal opening did not uniformly depend on the decrease in turgor of the epidermal and subsidiary cells caused by the different osmotic potential of the sugars. Sucrose caused plasmolysis of the subsidiary cells only but this was not accompanied by the opening of the stomatal pore. InM. aegyptia, no plasmolysis was seen either in epidermal or subsidiary cells, even the stomata opened; inM. dissecta, on the other hand, plasmolysis occurred in these cells without any stomatal opening, after incubation in glucose or mannitol. Mannitol is least absorbed, glucose slightly more and sucrose is absorbed to a very large extent in the guard cells when the materials were inoubated in the respective sugar solutions. However, the absorption of these three sugars was almost always larger in isolated epidermal strips than in discs; in detached intact leaves it was still more reduced.

Studies on the physiology of hyacinth bulbs (Hyacinthus orientalis L.) the effect of plant growth regulators on metabolic activities in non-chilled hyacinth bulbs

The effect of gibberellic acid, benzyladenine and a mixture of these compounds on the activity of some hydrolytic enzymes was studied in hyacinth bulbs non-exposed to low temperatures. Plant growth regulators were applied on the heel of dormant bulbs in the middle of July. An intensive elongation of the inflorescence was observed only in the plants treated previously with GA3 and grown in a warm greenhouse for 63 and 84 days. The activities of amylase, invertase and acid phosphatase were higher in the flower buds of unrooted bulbs treated with growth regulators than in the control plants, although growth regulators did not affect the level of extractable proteins. The elongation of the inflorescence in the plants treated with gibberellic acid was correlated with a sharp increase of invertase activity in this organ. The effect of GA3 and BA on the activity of other enzymes in buds and scales varied with the period of plant growth.

Nitrate reductase, nitrite reductase and glutamate dehydrogenase levels in roots and leaves of maize seedlings

Total activities of nitrate and nitrite reductases were higher in 4 to 20 day old maize plants in the leaves than in the roots. The ratio of activities found in the leaves and in the roots respectively was much higher in the case of nitrate reductase than in the case of nitrite reductase. On the other hand higher glutamate dehydrogenase activity in the roots than in the leaves clearly indicates that the roots play a more important role in the assimilation of ammonium than in the assimilation of nitrate. When comparing the distribution of seminal and nodal adventitious roots of maize seedlings with the assimilation of inorganic nitrogen on the basis of enzyme levels, it could be deduced that during the first 20 days of seedling growth seminal roots were more involved in the assimilation of nitrate whereas nodal adventitious roots were more active in ammonium assimilation.

Effect of ammonium concentration on growth and nitrogen accumulation by soybean grown in nutrient solution

Soybean (Glycine max (L.) MERR. CV. 'Amsoy') plants were grown for 40 days in nutrient solution at various concentrations of ammonium. Maximum yield of dry matter was obtained at a concentration of 715 μM. Further increase in the concentration of ammonium resulted in a reduction in growth due to ammonium toxicity which affected both root and shoot development. The pattern of nitrogen accumulation in tops was consistent with the multiphasic uptake of ammonium and can be represented by 2 phases in the range 1.78 X 10-5-3.57 x X 10-3 M of ammonium.

Finding of lettuce big vein virus in Czechoslovakia

Big vein disease occurring on lettuce plants cv. 'Pražan' was identified and the susceptibility to this disease was tested on 9 lettuce cultivars of foreign origin. Lettuce big vein virus was confirmed to be the cause of the disease. The transmission of the LBVV by the fungusOlpidium brassicae (Wor.)Dang., the sporangia of which were found by microscopy in the cells of surface tissues of lettuce roots, was experimentally proved. Rod-like particles, the average length of which was estimated at 244 nm, were found in the roots of diseased plants. LBW transmission by mechanical inoculation and by aphids could not be proved.

Effect of a lack of manganese on the structure and functioning of the photosynthetic apparatus of Funaria hygrometrica spores.

Funaria spores must structure their photosynthetic apparatus before germination. A lack of manganese in the culture media affects neither the final structure of chloroplast nor the chlorophyll synthesis. Fixation of CO2, reducing power of chloroplasts (DPIP) and RPE photo-induced signal S II are modified by this lack of manganese. The study of Mn/Chl and CO2 fixed/Mn ratios shows that Mn is a limiting factor for CO2 fixation and that in the spores depleted of Mn the residual Mn is "structural" Mn.

Atypical composition of seed proteins in cultivars ofPhaseolus vulgaris L.

In only one cultivar out of 1200 investigated cultivars ofPhaseolus vulgaris L. could we find an extreme change in the pattern of reserve proteins on the cathodic side: one of the proteins, called protein I, is completely absent in the cultivar 'Krupnaya sakharnaya' and is replaced on the same site by another protein,i.e. a protein completely different in its immunochemical specificity. The case is of interest from both the phylogenetic and systematic viewpoints and deserves further attention.

Absence of biological activity in oxidation products of indoleacetic acid

The main enzymatic oxidation products of IAA have been tested for biological activity, using as bioassay the straight-growth test ofAvena coleoptile. After being purified by rechromatography, none of these products (including methyleneoxindole) exhibited biological activity within the wide range of concentrations employed; consequently, the results accord with the evidence that IAA is the true hormone of plant growth.

The evaluation of the root system of mustard by means of its dielectric characters with respect to the yield

When examining plants of white mustard (Sinapis alba L.), a significant relationship between the size of the roots and the yield was found, both in a pot experiment and in a field one. The yield was influenced by the size of the root system during and after flowering time. Especially the number of seeds has increased, and the average weight of seeds showed the same tendencies after the flowering time. The productivity of plants with an above-average yield was increasing more quickly than the size of their root system. From the beginning of the vegetation till the end of flowering these most efficient plants showed the lowest permitivity dispersion and the highest dispersion after the flowering time. The dispersion has increased towards the end of vegetation.

The effect of sugars on inverse relation between the growth and chlorophy11 synthesis in tobacco tissue

Cytokinin-dependent and cytokinin-autonomous strains of tobacco callus tissue (Nicotiana tabacum L. cv. 'Wisconsin 38') were grown on media containing sucrose, glucose and fructose, respectively. The tissues were kept 14 days in darkness and then transferred for 9 days to continuous light after which time the fresh weight and chlorophyll content were estimated. The highest chlorophyll concentration was recorded at sugar levels which were either suboptimal (sucrose in the case of cytokinin-dependent strain) or supraoptimal (all other sugars for both strains and sucrose for the cytokinin-autonomous strain) for tissue growth. The chlorophyll concentration was increased when the tissue was cultured on media containing glucose or fructose,i.e. sugars whioh did not support the growth as well as sucrose. Chlorophyll synthesis in the cytokinin-autonomous strain is significantly lower than in the cytokinin-dependent strain. This difference was independent of either sugar source or concentration. These results support the observed inverse relationship between tissue growth and plastid development and the limited metabolic activity of plastids in cytokinin-autonomous tissues.

Characteristics and inheritance of chlorophyll mutations inPhaseolus mungo

Viable mutations affecting chlorophyll production in black gram (Phaseolus mungo L.) were recovered following treatments with X-rays and ethyl methanesulphonate singly or in combination. Of these theviridis mutant was characterized by viridine green colour of all the leaves and reduced plant size. inchlorotica the emerging leaves were initially yellowish green which gradually changed to dark green at flowering. The terminal 2-3 leaves always remained yellowish green inchlorina-terminalis whereas inchlorina-virescence the emerging leaves were yellowish green that changed to dark green at flowering. Inalbo-virescence the first 3-5 leaves at emergence were white in which small irregular green dots or patches often developed. The subsequent leaves were normal green. The emerging trifoliate leaves ofaureo-virescence were light green which turned to turtle green within a week. It was established thatviridis, chlorotica, chlorina-terminalis, chlorina-virescence, andalbo-virescence mutants were conditioned by a single receissive gene, in each case. Theaureo-virescence character was found to be inherited through the cytoplasm.

Effect of calcium and sucrose concentration on pollen germination invitro of sixRosa species

The germination of the pollen from11 individuals of six wildRosa species was studied. The presence of calcium resulted in increased pollen germination, longer pollen tubes, and in a decrease of the requirement of sucrose concentration in cultivation media. Pollen germination in the medium with Ca reached the values of pollen viability estimated by tetrazolium test in all cases except with roses with balanced heterogamy, of the sectionCaninae. The stimulating effect of calcium was generally most pronounced in the pollen from roses of hybrid nature, such asR. jundzillii, R. canina, and especially in the case of the calciphilous speciesR. eglanteria.

Aphid transmission of Beet Yellows Virus affected by homologous antiserum

A thin layer of homologous antiserum (against the beet yellows virus - BYV) between the leaf surface and a Parafilm membrane totally inhibited the acquisition of BYV by aphidsMyzus persicae (Sulz.), but it did not affect the inoculation of BYV by infective aphids. BYV transmission decreased with aphids picking up the virus from leaves coated with a normal rabbit serum. Aphids sucking on purified BYV suspension through the Parafilm membrane as well as aphids allowed to probe into leaves of healthy plants spread with an infectious purified BYV suspension failed to transmit BYV. No BYV particles could be detected in eluates from stylets and labia cut off from aphids which had probed on BYV infected plants by electron microscopic examination. The acquisition seems to be the most important phase for the aphid transmission of BYV which is apparently carried on the stylet surface.
