Isolation and functional analysis of cDNAs similar to Hyp-1 involved in hypericin biosynthesis from Hypericum erectumM. L. Jin, J. C. Ahn, B. Hwang, H. -S. Park, H. S. Lee, D. -W. ChoiBiologia plantarum 54:725-729, 2010 | DOI: 10.1007/s10535-010-0129-5 Hypericin, a naphthodianthrone, has been identified as the principal active compound found in St. John's wort (Hypericum perforatum L.). To generate a gene resource for hypericin and other valuable metabolites, we generated expressed sequence tags (ESTs) from H. erectum. Analyses of the ESTs enabled us to select three cDNAs, HeHyp1, HeHyp2, and HeHyp3, evidencing significant sequence homology to Hyp-1 that were involved in hypericin biosynthesis from H. erectum. The deduced amino acid sequence of HeHyp1 cDNA exhibits 95 % identity with Hyp-1. The HeHyp2 and HeHyp3 polypeptides also exhibit 81.1 % identity with Hyp-1. The transcripts of HeHyp1, HeHyp2, and HeHyp3 were detected in the root, stem, leaf, flower, and callus cells. Study using recombinant protein suggests that Hyp-1, HeHyp2, and HeHyp3 may be involved in the biosynthetic of hypericin or other emodin derivatives. |
Chromosomal location of 45S rDNA and dfr gene in Citrus sinensisX. Lu, W. Zhou, F. GaoBiologia plantarum 54:798-800, 2010 | DOI: 10.1007/s10535-010-0146-4 Dihydroflavonol 4-reductase (DFR) is a key enzyme in the anthocyanin biosynthesis. In this study, the localization of 45S rDNA and dfr gene (named as CsDFR-bo) of the blood orange (Citrus sinensis Osbeck cv. Ruby) on chromosomes was investigated by fluorescence in situ hybridization (FISH). A karyotype of C. sinensis was reconstructed based on the length of mitotic metaphase chromosomes. The 45S rDNAs were localized on chromosomes 2p and 7q. The detection ratios of 45S rDNA in the two chromosomes were 69.5 and 77.3 %, respectively. The CsDFR-bo was proved to be a single-copy gene and localized on chromosome 3p. The detection ratio of CsDFR-bo was 8.2 %. |

MegličBiologia plantarum 54:54-60, 2010 | DOI: 10.1007/s10535-010-0008-0 Three genetic similarity coefficients were estimated and compared for their usefulness: simple matching (S SM), Jaccard's (S J) and Dice's (S D), all based on dominant markers data from individuals representing predominantly self-pollinating species. AFLP markers were used to analyze 139 Phaseolus vulgaris L. (common bean) and 67 Lactuca saligna L. (least lettuce) accessions, and RAPD markers were used to analyze 110 Triticum dicoccoides Koern. (wild emmer wheat) accessions. Similar discriminating structure and power based on the three genetic similarity coefficients was found for each of the three species. This discriminating power was high for both P. vulgaris and L. saligna but moderate for T. dicoccoides. With closely related individuals, as in our study, the absence of a band in two individuals should be due to an identical cause inherited from the same ancestor. Accordingly we propose the use of S SM, which alone out of the three examined coefficients involved shared absence of DNA bands, as contributing to genetic similarity. When RAPDs are employed, inferences about population structure and nucleotide divergence should be made with prudence as the nature of genetic variation uncovered by RAPDs is often unclear. |
Effects of exogenous spermine on sweet sorghum during germination under salinityY. Y. Chai, C. D. Jiang, L. Shi, T. S. Shi, W. B. GuBiologia plantarum 54:145-148, 2010 | DOI: 10.1007/s10535-010-0023-1 Seedlings of Sorghum bicolor (L.) Moench were subjected to 180 mM NaCl with or without 0.25 mM spermine (SPM) for 7 d. NaCl treatment resulted in the inhibition of growth and increased the content of free proline, soluble protein and malondialdehyde (MDA). Additionally, it also enhanced the activity of catalase (CAT), peroxidase (POX) in both shoots and roots, while decreased that of glutathione reductase (GR). When exogenous spermine was added to the test solution, the growth of sweet sorghum seedlings was improved, and a smaller increase in the free proline and MDA contents was observed. The addition of spermine also partially increased the activities of POX and GR, but had no effects on soluble protein content or the activity of CAT. |

AmâncioBiologia plantarum 54:237-244, 2010 | DOI: 10.1007/s10535-010-0042-y Plantain (Musa ABB CEMSA 3/4) plantlets were micropropagated in temporary immersion bioreactors (TIB) or in gelled medium (GM). After ex vitro transfer ROS accumulation was determined by infiltrating leaves with nitroblue tetrazolium (NBT) and 3,3'-diaminobenzidine (DAB). Stomatal cells were more stained with NBT and DAB in GM plants than in TIB plants, but the difference disappeared at the end of acclimatization. At the end of the in vitro phase, GM plantlets showed higher activities of ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR), monodehydroascorbate reductase (MDHAR) and glutathione reductase (GR), while activities of catalase (CAT), superoxide dismutase (SOD) and glutathione transferase (GT) were higher in TIB grown plantlets. At the end of acclimatization GT, SOD, CAT and MDHAR stabilized at low values of activity in plantlets derived from both treatments. Concerning the correspondent genes, GM plantlets showed higher expression of all transcripts with the exception of CuZnSOD. The immunobloting of peroxiredoxins (PRXs) showed that chloroplast-located PRXs were expressed at higher levels in TIB plantlets, some showing polymerization. In conclusion, TIB grown plantlets had an improved anti-oxidative response when compared with GM. |

MaitiBiologia plantarum 54:334-338, 2010 | DOI: 10.1007/s10535-010-0058-3 Rapid micropropagation was achieved in Chlorophytum borivilianum Santapau and Fernandes using shoot base as explants. Multiple shoots were induced on Murashige and Skoog's (MS) medium supplemented with 3.0 mg dm-3 6-benzylaminopurine, 0.1 mg dm-3 1-naphthaleneacetic acid, 150 mg dm-3 adenine sulphates and 3 % saccharose. Rooting was readily achieved upon transferring the shoots onto half strength MS medium supplemented with 0.1 mg dm-3 indolebutyric acid and 2 % saccharose. Micropropagated plantlets were hardened in the greenhouse and successfully established in soil. Random amplified polymorphic DNA (RAPD) markers were used to evaluate the genetic stability of the micropropagated plants. Thirty one arbitrary decamers were used to amplify genomic DNA from in vitro and in vivo plant material to assess the genetic stability. All RAPD profile analysis from micropropagated plants was genetically similar to mother plants. |

ČellárováBiologia plantarum 54:515-519, 2010 | DOI: 10.1007/s10535-010-0090-3 The spatial and temporal expression of dbat and dbtnbt genes involved in the later steps of paclitaxel biosynthesis in relation to baccatin III and paclitaxel accumulation in Taxus baccata L. germinating embryos and seedlings was investigated. The steady-state of mRNA transcripts was measured by quantitative real time polymerase chain reaction (qRT-PCR), the content of taxanes was determined by HPLC. The spatial distribution of the metabolites was found to be in accordance with the transcript level of the respective genes. Higher content of mRNA transcripts in shoots of yew seedlings responded to higher content of taxanes in stems and needles. The highest increase in the transcript level of both genes was observed 8 d after placing the embryos on germination medium, before elongation of embryonic axis and emergence of the radicle. The pattern of temporal dbat expression was in line with the expression of the dbtnbt gene. The temporal distribution of the precursor baccatin III correlated well with paclitaxel, the final product of the pathway. |
Morpho-histological study of direct somatic embryogenesis in endangered species Frittilaria meleagrisA. Subotić, M. Trifunović, S. Jevremović, M. PetrićBiologia plantarum 54:592-596, 2010 | DOI: 10.1007/s10535-010-0107-y Direct somatic embryogenesis of Frittilaria meleagris L. was induced using leaf base explants excised from in vitro grown shoots. Somatic embryos occurred at the basal part of leaf explants 4 weeks after culture on a Murashige and Skoog (MS) medium supplemented with various concentrations of 2,4-dichlorophenoxyacetic acid (2,4-D) or kinetin (KIN). The highest number of somatic embryos (SEs) were formed (9.74) from leaf explant on MS medium supplemented with 0.1 mg dm-3 2,4-D after 4 weeks of culture initiation. An initial exposure to a low concentration of KIN in the medium also enhanced SEs induction. Our observations by light and scanning electron microscopy revealed that SEs originate directly from the epidermal and subepidermal layers of leaf explant. The developmental stages of somatic embryogenesis from the first unequal cell division through the meristematic clusters, multi-cellular globular somatic embryos to the fully formed cotyledonary embryos were determined. After 4 weeks on MS medium without plant growth regulators, SEs developed into bulblets. |

Marechal] immersed in water or not were treated in microwave oven (2450 MHz, 800 W cm-2) for 1, 3, 5 and 7 s before culturing. Callusing and shoot emergence from these explants were enhanced up to microwave exposure lasted 5 s while longer treatment of water-immersed explants delayed callusing. One polypeptide (26.6 kD) was up regulated in the callus derived from microwave treatment in water-immersed explants. RAPD analysis detected alteration in DNA sequences due to microwave treatment in water-immersed explants for 7 s. The frequency of mutation was 1.6 % (4 bands out of 248) over all the cultures analyzed and the same was 13 % (4 bands out of 31), if amplicons generated at 7 s treatment alone were considered. |

We describe a simple protocol for the efficient biolistic transformation of protonemal tissue with minimum tissue handling steps. The transformation efficiency depends on the biolistic conditions. The bombardment of tissue with 1 μm gold particles yielded between 20 and 40 stable transformants per 1 μg of DNA. Transformation with circular plasmids generates higher frequencies of random transgene integration, whereas linear plasmids are more efficient in generating gene-targeted insertions. |

NairBiologia plantarum 54:21-32, 2010 | DOI: 10.1007/s10535-010-0004-4 Reproducible and high-frequency transgenic plant regeneration from callus and embryo axes of four different genotypes of chickpea (Cicer arietinum) was achieved after Agrobacterium-mediated transformation. Three different strains of Agrobacterium (EHA105, AGL1 and LBA4404) harboring the binary vector pCAMBIA1301 containing β-glucuronidase (GUS) and hygromycin phosphotransferase (hpt) genes under the control of a CaMV35S promoter were used. The highest number of transgenic plants was obtained from cotyledonary node-derived calli of genotype Pusa-256. A highly efficient rooting was achieved on Murashige and Skoog medium supplemented with indole-3-butyric acid. The stable integration of the gene was confirmed by molecular analyses of the transformed plants. Inheritance of GUS and hpt gene was followed through two generations and they showed the expected 3:1 inheritance. |

Somatic embryos were induced from immature cotyledons on Murashige and Skoog's (MS) medium supplemented with different concentrations of 2,4-dichlorophenoxyacetic acid (2,4-D), 2,4,5-trichlorophenoxyacetic acid (2,4,5-T), α-naphthaleneacetic acid (NAA) and picloram alone or in combination with 0.5 - 2.0 mg dm-3 N6-benzylaminopurine (BA) or kinetin (KIN). NAA was better for somatic embryo induction compared to other auxins. The well formed, cotyledonary shaped embryos germinated into plantlets with 36.6 % frequency on MS medium supplemented with 2.0 mg dm-3 BA + 0.5 mg dm-3 abscisic acid (ABA). The frequency of embryogenesis and plantlet regeneration was higher in cv. ICCV-10 as compared to cv. Annigeri. Regenerated plants were transferred to soil (40 % survival) and grown to maturity. Histological studies of explants at various developmental stages of somatic embryogenesis reveled that somatic embryos developed directly from the cotyledon cells and they were single cell origin. |
Concurrent occurrence of α-amylase inhibitor and stimulator in red kidney bean seed: physiological implicationsH. Alizadeh, D. W. M. Leung, A. L. J. ColeBiologia plantarum 54:195-197, 2010 | DOI: 10.1007/s10535-010-0034-y It is hypothesized that since protein α-amylase inhibitor (α-AI) and stimulator might be present together in red kidney bean (Phaseolus vulgaris L.) seeds, their in vitro interactions might influence their detection and quantification. Assay of α-AI using extracts from the embryonic axes revealed an unexpected finding in that the extracts stimulated rather than inhibited α-amylase activity. The cotyledon extracts exhibited inhibitory or enhancement effect on α-amylase activity depending on whether prior to the α-amylase assay they had been boiled for 10 min or not. Phytohemagglutinin (PHA-L in particular) is implicated in the present study as a stimulator of α-amylase activity co-extracted with α-AI from red kidney bean cotyledons. The importance of these findings is discussed in relation to the possible widespread occurrence of protein α-amylase stimulator in seeds and other plant parts. |

MasarovičováBiologia plantarum 54:308-314, 2010 | DOI: 10.1007/s10535-010-0053-8 Effects of zinc (12-180 μM) alone and in mixtures with 12 μM Cd on metal accumulation, dry masses of roots and shoots, root respiration rate, variable to maximum fluorescence ratio (FV/FM), and content of photosynthetic pigments were studied in hydroponically cultivated chamomile (Matricaria recutita) plants. The content of Zn in roots and shoots increased with the increasing external Zn concentration and its accumulation in the roots was higher than that in the shoots. While at lower Zn concentrations (12 and 60 μM) the presence of 12 μM Cd decreased Zn accumulation in the roots, treatment with 120 and 180 μM Zn together with 12 μM Cd caused enhancement of Zn content in the root. Presence of Zn (12-120 μM) decreased Cd accumulation in roots. On the other hand, Cd content in the shoots of plants treated with Zn + Cd exceeded that in the plants treated only with 12 μM Cd. Only higher Zn concentrations (120 and 180 μM) and Zn + Cd mixtures negatively influenced dry mass, chlorophyll (Chl) and carotenoid content, FV/FM and root respiration rate. Chl b was reduced to a higher extent than Chl a. |
In vitro responses of grape rootstocks to NaClM. Alizadeh, S. K. Singh, V. B. Patel, R. C. Bhattacharya, B. P. YadavBiologia plantarum 54:381-385, 2010 | DOI: 10.1007/s10535-010-0069-0 An investigation was undertaken to adjudge the in vitro salt tolerance and biochemical changes due to NaCl on four grape rootstocks (Dogridge, SO4, H-144 and 3309C). The in vitro two-node micro-cuttings from the established cultures were sub-cultured on rooting medium comprising Murashige and Skoog (MS) basal medium supplemented with 0.5 μM indolebutyric acid (IBA), 200 mg dm-3 activated charcoal and different NaCl concentrations ranging from 0 to 125 mM. The surviving and proliferated cultures were further sub-cultured four times on respective media. Dogridge and H-144 tolerated 125 and 100 mM NaCl, respectively, while SO4 and 3309C survived only up to 75 mM NaCl. Contents of proteins, proline, K+ and Na+ in tissue increased in all genotypes due to NaCl supplementation, while contents of chlorophyll and total soluble sugars declined. Higher K+/Na+ ratio was registered in Dogridge and H-144 than in SO4 and 3309C. The relative NaCl tolerance for different grape rootstocks under study could be ranked as Dogridge > H-144 > SO4 and 3309C. |

KrekuleBiologia plantarum 54:483-487, 2010 | DOI: 10.1007/s10535-010-0085-0 The contents of endogenous free and conjugated polyamines, putrescine (Put) and spermidine (Spd), were determined during 9 week of vernalization (at 5 °C) in winter wheat seedlings cultivated on Murashige and Skoog media without (MS0) and with 2 mg dm-3 zearalenone (MSZEN). At the 4th week of chilling treatment, which is sufficient to induce generative development in 30 % of plants, the marked increase in free and conjugated forms of Put and free Spd were observed. The presence of ZEN in medium significantly accelerated the vernalization. About 20 % of plants treated with ZEN flowered already after 2 weeks and 40 % after 3 weeks of chilling. Significantly higher content of free Put was determined in roots grown on MSZEN compared with MS0 during the first 5 weeks of vernalization with maximum at the 4th week. After germination, a marked decrease in free Spd content was observed both in plants grown on MS0 and MSZEN. Application of ZEN significantly slowed down the Spd decline in leaves and roots during the first and second week of vernalization. The content of Spd and its conjugates decreased in vernalized plants after 1 week of cultivation at 20 °C. |

Carbon dioxide assimilation data support that Sasanishiki was less sensitive to UV-B than Norin 1 and Surjamkhi. UV-B radiation sharply decreased the content of Rubisco protein in Surjamkhi and has no effect in Sasanishiki. The photochemical activities of photosystem (PS) 1 and PS 2 was slightly affected by UV-B treatment. The content of H2O2 and the activities of antioxidant enzymes, catalase (CAT), peroxides (POX) and superoxide dismutase (SOD) were enhanced after UV-B treatment. The activities of CAT and POX isoenzymes in Sasanishiki were more enhanced by UV-B radiation than those in Norin 1 and Surjamkhi. |

PuBiologia plantarum 54:664-670, 2010 | DOI: 10.1007/s10535-010-0118-8 The aim of this work was to study the short-term effects of salt stress on the antioxidant system and ascorbate peroxidase (APX) expression in two salt sensitive sweet potato cultivars Tainung 57 (TN57) and Tainung 66 (TN66), and one salt-tolerant cultivar Hsusu 18 (HS18). Plants were grown in plastic pots in a greenhouse for 30 d followed by NaCl treatments (0, 150, 300, and 450 mM) for 0, 24 and 48 h in a growth chamber. Young, fully expanded leaves of each treatment and period of time were clipped for enzyme activity measurements. In addition, different tissues (leaves, stems, and roots) were also harvested to analyze the tissue-specific APX gene expression using semiquantitative reverse-transcription polymerase chain reaction (RT-PCR). Three degenerated primers of APX isoforms from cytosol, peroxisomes and chloroplasts were used to amplify the APX complementary DNA of these cultivars. Our results show higher increase in APX activity at 24 and 48 h of salinity (450 mM of NaCl) in salt-stress tolerant genotype than in saltsensitive ones. The expression of APX isoforms in response to salinity was tissue specific and also dependent on stress duration. |
The effect of 5-azacytidine on wheat seedlings responses to NaCl stressL. Zhong, Y. H. Xu, J. B. WangBiologia plantarum 54:753-756, 2010 | DOI: 10.1007/s10535-010-0135-7 The effect of 5-azacytidine (5-azaC) on the alleviation of damaging effects of NaCl treatment was studied in two wheat (Triticum aestivum L.) cultivars differing in salt tolerance (salt-tolerant Dekang-961 and sensitive Lumai-15). The plants were pre-treated or not with 50 μM 5-azaC and then subjected to salt stress induced by 100 or 150 mM NaCl. Salinity caused reduction in biomass accumulation and increase in malondialdehyde content in root tissues in both cultivars, but less in pre-treated seedlings. The activities of superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD) in the roots of both cultivars increased during salt stress, but the rate of increase was higher in Dekang-961. Plants treated with 5-azaC had higher root SOD, CAT and POD activities under salt stress than untreated plants. Content of 5-methylcytosine (5mC) decreased in both cultivars under salt stress, and the level of demethylation was higher in Dekang-961 than that in Lumai-15. Moreover, the degree of methylation was lower in both cultivars under salt stress after 5-azaC application compared to only salt-treated groups. These findings suggested that 5-azaC could protect plants from salt stress. |

LiBiologia plantarum 54:89-96, 2010 | DOI: 10.1007/s10535-010-0013-3 The photosynthetic performance and related leaf traits of Incarvillea delavayi Bur. et Franch were studied at different water regimes to assess its capacity for photosynthetic acclimation to water stress. The initial response of I. delavayi to water stress was the closure of stomata, which resulted in down-regulation of photosynthesis. The stomatal limitation (SL) represented the main component to photosynthetic limitations but non-stomatal limitation (NSL) increased quickly with the increasing water stress, and had similar magnitude to SL under severe water stress (soil moisture 25-30 % of field capacity). Chlorophyll (Chl) a fluorescence parameters characterizing photosystem (PS) 2 photochemical efficiency (ΦPS2), electron transport rate (J) and photochemical quenching (qP) decreased with the increasing water stress, indicating impaired photosynthetic apparatus. However, the water-stressed plants had a increased mesophyll CO2 diffusional conductance, Chl a/b ratio, leaf nitrogen partitioning in RuBPCO and bioenergetics in later grown parts, indicating that I. delavay had a substantial physiological plasticity and showed a good tolerance to water stress. |

Vyskot, J. ®lůvováBiologia plantarum 54:173-176, 2010 | DOI: 10.1007/s10535-010-0029-8 Heteroduplex formation is known to occur during mixed-template polymerase chain reaction (PCR) using universal primers, and may represent a serious problem in several PCR-based analyses. A common way to eliminate heteroduplex formation is to use reconditioning PCR. Because we detected that reconditioning PCR was not always sufficient to prevent heteroduplex formation, we focused on developing methods for the elimination of heteroduplexes during PCR. We detected that the heteroduplex to homoduplex ratio can be decreased by the addition of Taq polymerase and by a decrease in the number of PCR cycles. An appropriate combination of both of these approaches can be a method generally applicable to decrease the formation of heteroduplexes. |

Thidiazuron (TDZ) was the key plant growth regulator for inducing somatic embryogenesis and kinetin (KIN) and 6-benzylaminopurine (BAP) were the key cytokinins for inducing shoot organogenesis from leaf explants. TDZ combined with BAP or KIN in the induction Murashige and Skoog medium induced both somatic embryos and adventitious shoots. Leaf explants with abaxial site in contact with the medium induced less somatic embryos or adventitious shoots compared to inversely placed leaf explants and the optimum pH was 6.5-7.0. Secondary somatic embryos or adventitious shoot could be induced from primary somatic embryos using TDZ and BAP. Shoots developed adventitious roots on rooting medium containing 0.5 μM indole-3-butyric acid and 0.2 % activated carbon. Over 90 % of plantlets survived following acclimatization and transfer to potting mixture (sand:Vermiculite:limestone; 1:2:1). |
DIANTHIN, a negative selection marker in tobacco, is non-toxic in transgenic rice and confers sheath blight resistanceJ. M. Shah, K. VeluthambiBiologia plantarum 54:443-450, 2010 | DOI: 10.1007/s10535-010-0080-5 The DIANTHIN gene encoding a ribosome-inactivating protein (RIP) from Dianthus caryophyllus L. was tested for negative selection in tobacco and rice. Tobacco leaf discs and scutellum-derived callus of rice were transformed with Agrobacterium tumefaciens strain LBA4404 (pSB1, pJAS1). pJAS1 harbors the DIANTHIN gene under the control of the CaMV 35S promoter. Tobacco transformation efficiency, in comparison to pCAMBIA1301, was reduced by 87 % in pJAS1-transformed leaf discs. The DIANTHIN gene proved to be completely toxic to tobacco as all the recovered hygromycin-resistant transgenic plants harbored truncated T-DNAs with deletions of the DIANTHIN gene. Transformation of the DIANTHIN gene under a Mungbean yellow mosaic virus (MYMV)-inducible promoter did not cause any toxicity in tobacco as reflected by the recovery of transgenic tobacco plants with the complete DIANTHIN gene. Transformation efficiency of pJAS1 did not decline in rice. Interestingly, all transgenic rice plants harbored the complete DIANTHIN gene and expressed the gene. The T1 transgenic lines showed reduction of sheath blight symptom in the range of 29 to 42 %. The difference in the sensitivity to DIANTHIN between tobacco and rice provides a new direction to study the mechanisms underlying RIP toxicity in plants. |
Study of the origin of the rarely cultivated edible Solanum species: morphological and molecular dataP. Poczai, K. Mátyás, J. Taller, I. SzabóBiologia plantarum 54:543-546, 2010 | DOI: 10.1007/s10535-010-0096-x The present study applies RAPD technique and morphometric analysis to study the diversity of some accessions belonging to section Solanum. A total of 252 products were amplified with 23 12-mer arbitrary primer pairs, among which 210 were found to be polymorphic. Sixteen morphological characters were measured and used to compile a dendrogram. Both the morphological and RAPD marker analysis clearly separated the different accessions into similar groups. The results indicate that the analyzed cultivars with unknown origin could be derived from S. retroflexum. We found morphological differences among the S. scabrum subsp. scabrum accession which were not reflected in the molecular data. Presumably these accessions represent cultivated forms selected for their habit, fruit quantity and/or quality and leaf size, respectively. |
Functional characterization of the apple MdSAMDC2 gene by ectopic promoter analysis and over-expression in tobaccoL. -L. Zhao, L. -Q. Song, C. -X. You, T. Moriguchi, Y. -J. HaoBiologia plantarum 54:631-638, 2010 | DOI: 10.1007/s10535-010-0113-0 The expression of MdSAMDC2 gene, which encodes S-adenosylmethionine decarboxylase (SAMDC) in apple, was up-regulated by low temperature, salt and drought stresses. To identify its in vivo biological functions in the responses to stresses, the promoter region of MdSAMDC2 was isolated and characterized by analyzing the cis-acting regulatory elements and GUS reporter gene by an Agrobacterium-mediated transformation. GUS activity was enhanced upon salt and cold stresses, indicating that MdSAMDC2 promoter region controls gene transcription under stresses. In parallel, several lines of the transgenic tobacco plants over-expressing MdSAMDC2 were obtained. The contents of three polyamines greatly increased in the transgenic lines compared with the non-transgenic WT control. Upon exposed to low temperature (4 °C), salt (150 and 250 mM NaCl) and osmotic (20 % polyethylene glycol) stresses, transgenic plants produced more free polyamines and more active antioxidative enzymes such as superoxide dismutase and catalase than the WT control. Meanwhile, malondialdehyde content, an indicator for membrane lipid peroxidation, decreased in transgenic plants relative to the WT control. Thus, over-expression of MdSAMDC2 in tobacco conferred tolerance to stresses. |
Response of grape rootstocks to salinity: changes in root growth, polyamines and abscisic acidK. K. Upreti, G. S. R. MurtiBiologia plantarum 54:730-734, 2010 | DOI: 10.1007/s10535-010-0130-z Effects of salinity (0, 50, 100 and 250 mM NaCl) on growth, root:shoot dry mass ratio, osmotic potential (ψx), electrolyte leakage and contents of Na+ and K+, polyamines and abscisic acid (ABA) were studied in the grape rootstocks Dogridge, 1613, St. George and Salt Creek. In control rootstocks, the root length was highest in Dogridge and contents of K+ and ABA in Salt Creek. Salinity treatments increased root Na+ and decreased K+ content and St. George exhibited highest Na+ content and Na+:K+ ratio. The root:shoot dry mass ratio in all rootstocks increased upto 100 mM NaCl. With increasing NaCl concentration, putrescine, spermine and spermidine contents showed consistent increase and putrescine increase was highest in St. George and spermidine and spermine in the Dogridge and Salt Creek. Under salinity, the ABA content increased in all the rootstocks but more in Salt Creek and Dogridge than in St. George. |

KimBiologia plantarum 54:61-68, 2010 | DOI: 10.1007/s10535-010-0009-z Among the 400 soybean (Glycine max) landraces, we selected 3 tolerant (KAS150-9, KAS160-15, and KAS170-9) and 3 susceptible lines (KAS160-14, KAS160-20, and KAS201-6-1) by the survival percentage and injury scores. Susceptible lines showed decrease in chlorophyll content and increase in glucose and malondialdehyde (MDA) contents under waterlogging stress, while tolerant lines did not change significantly. For AFLP analysis, 8 EcoRI (+3) and 8 MseI (+3) primers used in 32 primer combinations generated a total of 2 566 bands with a mean of 80 bands per primer combination, of which 1 117 (43.5 %) were clearly polymorphic between the tolerant and susceptible lines. A genetic similarity coefficient, based on cluster analysis using an unweighted pair grouping method of average (UPGMA), was 0.79 for the tolerant group, while the susceptible landraces were genetically less related, with a genetic similarity coefficient of 0.17. The 10 reproducible polymorphic PCR products present in the 3 tolerant or susceptible lines were sequenced and converted into sequence tagged site (STS) markers. These STS primer sets were designated GmWT01-GmWT06 and GmWS01-GmWS04. Two STS primer sets, GmWT06 and GmWS02, generated a single monomorphic PCR product identical in size to the original AFLP fragments. For the broad application of these STS markers in marker-assisted selection (MAS) for soybean genotypes tolerant to waterlogging stress, two developed STS markers are being evaluated with putative waterlogging tolerant mutant lines induced by γ-radiation in soybean mutation breeding programs. |
Allelochemical effects on net nitrate uptake and plasma membrane H+-ATPase activity in maize seedlingsM. R. Abenavoli, A. Lupini, S. Oliva, A. SorgonàBiologia plantarum 54:149-153, 2010 | DOI: 10.1007/s10535-010-0024-0 Seven-day-old maize seedlings grown in a nitrogen-free hydroponic culture were exposed for 48 h to 0, 100 and 300 μM trans-cinnamic, p-coumaric, ferulic, caffeic acids, umbelliferone and 200 μM KNO3. Net nitrate uptake was affected by trans-cinnamic, ferulic and p-coumaric acids in a concentration-dependent manner, and trans-cinnamic acid appeared to be the strongest inhibitor. Conversely, at low concentrations, caffeic acid stimulated net nitrate uptake while umbelliferone did not influence it. After 24 h of treatment, plasma membrane H+-ATPase activity significantly decreased in a concentration-dependent manner in response to trans-cinnamic, ferulic and p-coumaric acids, while umbelliferone and caffeic acid had no effect on H+-ATPase activity. |
