YeoBiologia plantarum 53:329-333, 2009 | DOI: 10.1007/s10535-009-0060-9 Long-term (30 d) effects of 100, 200, 300, and 400 mM NaCl on photosystem 2 (PS 2)-mediated electron transport activity and content of D1 protein in the thylakoid membranes of chrysanthemum (Dendranthema grandiflorum) cultured in vitro at low irradiance 20 µmol(photon) m-2 s-1 were investigated. 100 mM NaCl increased contents of chlorophylls (Chl) a and b, carotenoids (Car; xanthophylls + carotenes), and the ratio of Chl a/b, and Car/Chl a+b. However, further increase in NaCl concentration led to the significant reduction in the contents of Chl a, and Chl b, and increase in the ratio of Chl a/b and Car/Chl a+b. NaCl treatment decreased the PS 2-mediated electron transport activity and contents of various thylakoid membrane polypeptides including D1 protein. |

MendesBiologia plantarum 53:395-399, 2009 | DOI: 10.1007/s10535-009-0075-2 In vitro organogenesis of Citrus volkameriana and C. aurantium was studied considering three explant types: epicotyl segment, internodal segment, and hypocotyl segment with attached cotyledon fragment. The explants were cultured in medium according to Grosser and Gmitter (EME) supplemented with 0, 0.5, 1.0, 1.5, and 2.0 mg dm- 3 6-benzyl-aminopurine (BAP), incubated firstly in darkness for 4 weeks, and then transferred to 16-h photoperiod for 2 weeks. Comparing epicotyl and internodal segments, a higher percentage of responsive explants and a higher number of shoots per explant were obtained with epicotyl segments, regardless of the BAP concentration. For C. volkameriana the highest percentage of responsive epicotyl segments (42 %) was obtained in EME with 1.0 mg dm-3 BAP, while for C. aurantium (59 %) in EME with 0.5 mg dm-3 BAP. The organogenesis efficiency was the best with the use of the hypocotyl segment with attached cotyledon fragment (77 % for C. volkameriana and to 75 % for C. aurantium). With this explant the morphogenesis occurred only in the hypocotyl region. The in vitro organogenesis was characterized by histological analyses showing that the morphogenic process started in the cambium region near the explant cut end. |
Responses to drought stress in two poplar species originating from different altitudesF. Yang, X. Xu, X. Xiao, C. LiBiologia plantarum 53:511-516, 2009 | DOI: 10.1007/s10535-009-0092-1 Cuttings of Populus kangdingensis and Populus cathayana, originating from high and low altitudes in the eastern Himalaya, respectively, were examined during one growing season in a greenhouse to determine their responses to drought stress (soil moisture decreased from 100 to 55 or 25 % field capacity). Compared to control plants grown under 100 % field capacity, those poplars grown under 55 and 25 % field capacity possessed lower increases in height and stem diameter, and higher contents of soluble sugars, free proline, malondialdehyde (MDA) and hydrogen peroxide, and higher activities of catalase (CAT), superoxide dismutase (SOD), peroxidase (POD), ascorbate peroxidase (APX) and glutathione reductase (GR). Compared with P. cathayana with greater leaf area, P. kangdingensis with greater root/shoot ratio exhibited lower MDA and H2O2 contents, higher soluble sugar and free proline contents, and higher activities of CAT, SOD, POD, APX and GR. These results suggested that P. kangdingensis was more drought tolerant than P. cathayana. |

ChenBiologia plantarum 53:593-596, 2009 | DOI: 10.1007/s10535-009-0108-x The lengths of open reading frame (ORF)100 and ORF29-TrnCGCA, the intronic sequence of rps16 and the transcribed spacer of TrnTUGU-TrnLUAA in chloroplast from different lines of cytoplasmic male sterility (CMS) rice were studied using indica types, japonica types and common wild rice as controls. The results show that the lengths of ORF100 and ORF29-TrnCGCA in CMS lines are similar to those of typical indica. The sequences of the rps16 intron and the TrnTUGU-TrnLUAA spacer in sporophyte sterile types (wild-abortive type, Yinshui type and K type) are almost the same, and they also share a molecular marker of GTTGAG at nucleotide positions 220-225 in the rps16 intron. Therefore, it is speculated that the source of these three types is the same. In contrast, a gametophyte sterile type, Yuetai A does not contain such a GTTGAG sequence in the rps16 intron and has a unique G at position 595, which may works as a molecular marker distinguishing the sporophyte sterile type from the gametophyte sterile type. Based on the observation that CMS rice has much lower cytoplasmic polymorphism than indica, japonica and wild rice, it is concluded that CMS rice lack cytoplasm diversity. Therefore, it is important to introduce new sources of cytoplasm into hybrid rice. |

Old leaves of CKX plants maintained higher pigment content and photosystem 2 activity compared to WT leaves of the same age. Chloroplasts of old CKX plants showed better antioxidant capacity represented by higher superoxide dismutase, dehydroascorbate reductase and glutathione reductase activities. |

G. SilveiraBiologia plantarum 53:764-768, 2009 | DOI: 10.1007/s10535-009-0140-x Na+ accumulation was investigated in the roots of 11-d-old cowpea [Vigna unguiculata (L.) Walp.] plants. The relative contribution of different membrane transporters on Na+ uptake was estimated by applying Ca2+, K+, NH4 +, and pharmacological inhibitors. Na+ accumulation into the root symplast was decreased by half in the presence of 1 mM Ca2+ and it was almost abolished by 100 mM K+. The inhibitory effect of external NH4+ on Na+ accumulation was more pronounced in the roots of NH4 +-free growing plants. Na+ accumulation was reduced about 73 % by 0.1 mM flufenamate and it was almost blocked by 2 mM quinine. In addition, 20 mM tetraethylammonium and 1.0 mM Cs+ decreased Na+ accumulation by 28 and 30 %, respectively. These results evidenced that low-affinity Na+ uptake by cowpea roots depends on Ca2+-sensitive and Ca2+-insensitive pathways. The Ca2+-sensitive pathway is probably mediated by nonselective cation channels and the Ca2+-insensitive one may involve K+ channels and to a lesser extent NH4 +-sensitive K+ transporters. |

NarangajavanaBiologia plantarum 53:37-44, 2009 | DOI: 10.1007/s10535-009-0006-2 The import of ATP into plastids is facilitated by members of the plastidic ATP/ADP transporter (AATP) family. Our results indicate that the cassava (Manihot esculenta Crantz) genome possesses two genes encoding for putative ATP/ADP translocases, which we have designated as MeAATP1 and MeAATP2. Their deduced products are 92 % identical, and phylogenetic reconstructions of plant AATP sequences suggest that MeAATP1 and MeAATP2 are the result of a relatively recent duplication event. Both genes were found to be expressed in a wide range of plant organs via RT-PCR, including young and mature leaves, fibrous and tuberous roots, and green stems. Neither MeAATP1 nor MeAATP2 showed evidence of increased transcription in the presence of exogenous sucrose. Interestingly, the transcriptional activity of MeAATP1 in leaves appeared to be upregulated after wounding, potentially indicating its involvement in the wound response mechanism of cassava. |

LozoyaBiologia plantarum 53:125-128, 2009 | DOI: 10.1007/s10535-009-0017-z We used a random amplified polymorphic DNA (RAPD) amplification method to identify molecular markers associated with high quercetin accumulation in the leaves of Psidium guajava L. trees, selected from four different Mexican agronomic regions. We identified six polymorphic RAPD fragments of 620, 590, 370, 690, 480 and 460 bp among individuals of P. guajava. Genetic linkage disequilibrium analysis revealed that three RAPD profiles considered as DNA markers (620/590 bp, 370 bp and 480/460 bp) had a positive, direct association with quercetin content. These informative molecular markers can be used for selective identification of plants with the highest accumulation of flavonoids. |

Zhongdan 9409 seedlings grown under different soil moisture regimes (40-45 % and 70-75 % of soil saturated water content) were studied. Zn application did not enhance maize plant adaptation to drought stress. The relative water content and the water potential of leaves were not affected by Zn treatment. Moreover, The PN of drought-stressed plants was not improved by Zn supply. The increases of plant biomass, stomatal conductance and quantum yield of photosystem 2 due to Zn addition were notable in well-watered plants. |

Six different types of somatic embryos based on cotyledon development were observed. The growth hormones (2,4-dichlorophenoxyacetic acid and kinetin) used in induction and maintenance media did not affect embryo rooting and germination. The 95 % conversion of normal embryos (with two cotyledons) was achieved, while an overall conversion was only 38 %. Horn shaped embryos failed to exhibit shoot growth. Poorly developed apical meristems were responsible for lower conversion percentages in some of embryo classes. However, regenerated plants phenotypically resembled to seed grown control plants regardless of somatic embryo morphology. |

Both species showed decreasing quantum efficiency of photosystem 2 (Fv/Fm) with decreasing water content. Extreme water loss observed after 25 d of dehydration resulted in considerable damage of leaf tissue ultrastructure. After rehydration, both species need several days to reconstitute their photosynthetic machinery. |

In prophase I, in triploid species B. holboellii and B. gunnisoniana the individual chromosomes condensed into long strands within the nucleus. Then, in metaphase I, each PMC formed a restitutional nucleus thereby bypassing the rest of the first meiotic division. This is interpreted as representing apomeiosis. Subsequently, the restitution nuclei underwent a single cytokinesis as evidenced by the production of dyads. The cells within each dyad were separated by a callose wall. Most of the PMC in B. holboellii and B. gunnisoniana produced dyads, but a small proportion generated conspicuous tetrads. In contrast, diploid apomict B. divaricarpa produced only tetrads by simultaneous cytokinesis. |

Seedling cotyledonary nodes on Murashige and Skoog medium (MS) containing 2 mg dm-3 N6-benzyladenine (BA) produced highest number of multiple shoots. The shoot numbers were increased further upon subculture on MS medium supplemented with 0.5 mg dm-3 BA. By repeated subculture of derived shoots, a high multiplication rate was established. The excised shoots were rooted on MS basal medium without growth regulators. The in vitro formed shoots were also rooted ex vitro by dipping them in 2 mg dm-3 of indole-3-butyric acid (IBA) solution for 2 min before transferring them onto the hardening medium. Successful hardening and further establishment (survival 90 %) of micropropagated plants under natural conditions was observed. |
QTLs mapping of physiological traits related to salt tolerance in young rice seedlingsH. Sabouri, A. M. Rezai, A. Moumeni, A. Kavousi, M. Katouzi, A. SabouriBiologia plantarum 53:657-662, 2009 | DOI: 10.1007/s10535-009-0119-7 Oryza sativa L. F2 population and F2:3 derived from a cross between salt tolerance cv. Tarommahali and salt sensitive cv. Khazar were used in this study. A linkage map based on F2 population was constructed (74 SSR markers on 192 individuals), which covered a total of 1231.50 cM with an average two locus interval of 19.83 cM. Two QTLs related to Na+/K+ ratio were found on chromosome 3 and 6. qDM-3 and qDM-8 (for dry mass of shoot) are major QTLs with very large effects explained 20.90 and 17.72 % of the total phenotypic variance, respectively. Major locus for DM (qDM-3) was bracketed by RM1022 - RM6283 spread over 13.6 cM on chromosome 3. Major part of the variability for standard tolerance ranking (STR) was explained by the qSTR-6 flanked by RM3727 - RM340 on chromosome 6, which exhibited phenotypic variance of 17.25 % and peak likelihood ratio (LR) of 17.51. The length of this QTL is 8.8 cM and identification of any tightly linked markers in this region will serve as a candidate gene for fine-mapping. qSTR-3 overlapped with qNA-3 and qNAK-3. The qSTR-3 may contain a new major gene for salt stress tolerance at seedling stage in rice. Major QTLs identified in this paper, after fine-mapping, could be used for marker assisted selection. |

ZhangBiologia plantarum 53:741-744, 2009 | DOI: 10.1007/s10535-009-0135-7 DNA variations of forty-eight Eucalyptus globulus plants, regenerated by successive culture from seven different explants were assessed by AFLP analysis using 18 primer combinations. At least one variation showed 66.7 % of the analyzed plants, and the numbers of polymorphic bands per plant ranged from 1 to 22. The proportion of polymorphic fragments did not correlate with the numbers of the regenerated plants. However, the more times of successive culture were done the more of polymorphic bands were found within the groups. On average, between 97.39 and 99.88 % of all fragments were shared within the same group. AMOVA analysis showed 39.33 % of the variation was found among the accessions that originated from different calli while 60.67 % was from same calli. |

With increasing Cu concentration total biomass was not affected, leaf area slightly decreased, while chlorophyll content decreased considerably. Cu content increased significantly both in roots and in leaves, but the contents of other ions were only slightly affected at the highest Cu concentration (Mn content decreased both in roots and in leaves, P content decreased only in leaves and Zn content increased in roots). Superoxide dismutase (SOD) activity increased up to day 7 after copper application. Peroxidase (GPOD) and polyphenol oxidase (PPO) activities also increased, while catalase (CAT) activity remained constant. |

Increase of nitrate availability accelerated nitrate reductase activity (NRA) in parent and offspring ramets of E. crassipes, and there was greatly different pattern in inter-connected ramets during clonal growth stage. Leaf NRA was lower in offspring than that in parent ramets in phase 1, while significantly higher leaf NRA in offspring ramets was detected during phase 2. The results indicated NRA in inter-connected ramets of E. crassipes was highly dependent on nitrate availability and growth stage. |

SonBiologia plantarum 53:271-277, 2009 | DOI: 10.1007/s10535-009-0050-y To obtain an insight into the comprehensive molecular characteristics of the salt tolerance mechanism, we performed a screening for salt inducible genes in a halophytic plant, Salicornia herbacea, using mRNA differential display. A comparative analysis of gene expression in Salicornia grown in control and salt-stressed conditions led to the detection of a gene that was induced by salt. Both sequence analysis and a subsequent database search revealed that this gene was highly homologous to tonoplast intrinsic proteins (TIPs) from a variety of plant species. This gene, designated as ShTIP, is 1014 bp in size and contains a coding region of 762 nucleotides, which encodes a protein of 254 amino acids. Northern blot analysis revealed that ShTIP was predominantly expressed in shoots under normal conditions. However, salt stress induced high expression of ShTIP in both the shoots and roots. The expression of ShTIP in a salt-sensitive calcineurin-deficient yeast mutant (cnbΔ) resulted in a resistance to the high salt conditions. In addition, we compared the expression of a TIP gene in Arabidopsis with that of ShTIP under different conditions and found that the Salicornia TIP has a different regulatory mechanism for adapting to salt stress conditions compared with the glycophyte Arabidopsis TIP. These results indicate that ShTIP plays an important role in salt tolerance. |
Expression of Rhizobium pydA-pydB fusion gene in Nicotiana tabacum confers resistance to the toxic aromatic compound 3-hydroxy-4-pyridoneS. Jube, J. Awaya, D. BorthakurBiologia plantarum 53:355-359, 2009 | DOI: 10.1007/s10535-009-0066-3 The toxic aromatic compound 3-hydroxy-4-pyridone (HP) is an intermediate in both synthesis and degradation of mimosine, which is produced by the tree legume Leucaena leucocephala. The L. leucocephala root-nodule symbiont Rhizobium TAL1145 contains a dioxygenase (pydA) and a hydrolase (pydB) gene that produce enzymes necessary for the degradation of HP. In order to coordinately express both genes in plant tissues under a single promoter, three different pydA-pydB fusion constructs (G0, G3, and G7) with varying glycine linkers between the two genes were developed. Prior to transferring the fusion constructs into L. leucocephala, which is highly recalcitrant to genetic transformation, we tested the expression and activity of the hybrid proteins in Nicotiana tabacum, a model plant system that can be easily transformed and analyzed. Seven independent transgenic tobacco lines were generated by Agrobacterium-mediated transformation, and stable integration and expression of pydA-pydB in these transgenic lines were confirmed by polymerase chain reaction (PCR), reverse transcriptase PCR (RT-PCR) and Western analysis. Only one of the fusion constructs, G3, containing a 9-nucleotide linker between pydA and pydB, provided significant levels of resistance to 3 mM HP, indicating that the hybrid protein produced by this fusion construct could degrade HP. |
Characterization of β-tubulin 4 regulated by gibberellins in rice leaf sheathG. Yang, A. Jan, S. The expression of OsTUB4, one of eight β-tubulin isotypes identified in the rice genome, was characterized. OsTUB4 was expressed in root primodia and the shoot apical meristem in basal parts of the leaf sheath in rice seedlings. OsTUB4 transcript abundance in leaf sheath increased by treatment with gibberellic acid (GA3) in a dose- and time-dependent manner. OsTUB4 transcript levels in gibberellin (GA)-deficient mutants were less than those of wild type rice. An OsTUB4 promoter::GUS assay also confirmed the responsiveness of OsTUB4 to exogenous GA3, suggesting that OsTUB4 expression was regulated by GA and may be involved in GA-regulated leaf sheath growth. In addition, OsTUB4 could interact with different specific proteins in vitro as assayed by a yeast two-hybrid system, indicating that OsTUB4 may have diverse functions through interaction with different proteins. |

Merr.] cultivars: Aldana (more resistant) and Essor (less resistant to low temperature) were subjected to chilling at 5°C for 24 h, and then the ultrastructure of the root meristem cells was investigated. The ultrastructure of control root cells of the tested cultivars differed in the number of condensed mitochondria, plastids with phytoferritin, deformed vacuoles, as well as multivesicular bodies (MB) in cytoplasm and vacuoles. Chilling induced concentric endoplasmic reticulum (ER) arrangement in both soybean cultivars, while the circular Golgi apparatus (GA) occurred only in cv. Essor and MB in the cytoplasm of cv. Aldana cells. Additionally, in cv. Aldana chilling increased the number of condensed mitochondria, MB in vacuoles and multilamellar structures (MS) in cytoplasm whereas in cv. Essor it enlarged the population of plastids with phytoferritin and the number of MB in cytoplasm. After chilling treatment the population of deformed vacuoles with phenolic compounds in the form of electron dense granules increased but the number of multilamellar structures (MS) in the vacuoles of both cultivars decreased. The ultrastructural changes induced by the chilling stress were not lethal but rather adaptive, especially in more resistant cv. Aldana. |

A new CBF (CRT/DRE binding factor) gene was cloned from trifoliate orange [Poncirus trifoliata (L.) Raf.] by RT-PCR with degenerate primers and rapid amplification of cDNA ends (RACE) techniques. The full-length cDNA of CBF gene from trifoliate orange (designated as Ptcbfb) was 847 bp containing a 732 bp open reading frame (ORF), encoding a 243 amino acid protein. The predicted protein (designated as PtCBFb) had over 60 % identity to CBFs from some other plant species. Bioinformatical analysis showed that PtCBFb contained N-terminal bipartite nuclear targeting sequence, potential C-terminal acid domain and high conserved AP2 domain. Some other loci such as phosphorylation sites of several protein kinases, N-myristoylation site, tyrosine sulfation site and amidation site were also conserved in PtCBFb. Predicted three-dimentional structure of PtCBFb was similar to CBF from Arabidopsis thaliana. Expression pattern analysis revealed Ptcbfb expression in every tested organ, and Ptcbfb was cold induced. |

High chlorophyll content and glandular trichomes were observed in the plants grown on Murashige and Skoog basal medium supplemented with 131.85 mM sucrose. The solasodine was quantified using reverse phase high performance liquid chromatography. The plantlets cultivated on this medium yielded 35.97 mg g-1 (d.m.) solasodine whereas the field plants used as control yielded only 2.32 mg g-1 (d.m.) of solasodine. |
Development of novel chloroplast microsatellite markers for Cucumis from sequence databaseJ. -B. Hu, X. -Y. Zhou, J. -W. LiBiologia plantarum 53:793-796, 2009 | DOI: 10.1007/s10535-009-0146-4 The development of chloroplast microsatellite (cpSSR) markers in Cucumis species and analysis of their polymorphism and transferability were reported. Fifteen microsatellite markers, represented by mononucleotide repeats, were developed from the complete sequence of Cucumis sativus chloroplast genome. Intraspecific variation was successfully detected in C. sativus and C. melo and revealed mean 1.6 and 1.9 alleles per cpSSR locus, respectively. With the exception of two exon region-located cpSSR markers being monomorphic, each of the others amplified polymorphic fragments in C. sativus or C. melo. A total of 34 polymorphic loci were detected with these cpSSR markers in the two species. Transferability of the newly developed cpSSR markers was checked on an additional set of 41 Cucurbitaceae accessions (belonging to 12 different species), and except for two markers with no amplification in Cucurbita maxima, the others could be transferable to all the accessions tested. Of the 15 cpSSR markers, 14 markers generated fragments with expected band sizes and 13 markers detected interspecific polymorphism among the accessions. Intraspecific polymorphism was also observed within four Cucurbitaceae species excluding C. sativus and C. melo. |
Waterlogging induced oxidative stress and antioxidant activity in pigeonpea genotypesD. Kumutha, K. Ezhilmathi, R. K. Sairam, G. C. Srivastava, P. S. Deshmukh, R. C. MeenaBiologia plantarum 53:75-84, 2009 | DOI: 10.1007/s10535-009-0011-5 The objective of this study was to examine the role of antioxidant enzymes in waterlogging tolerance of pigeonpea (Cajanus cajan L. Halls) genotypes ICP 301 (tolerant) and Pusa 207 (susceptible). Waterlogging resulted in visible yellowing and senescence of leaves, decrease in leaf area, dry matter, relative water content and chlorophyll content in leaves, and membrane stability index in roots and leaves. The decline in all parameters was greater in Pusa 207 than ICP 301. Oxidative stress in the form of superoxide radical, hydrogen peroxide and thiobarbituric acid reactive substances (TBARS) contents initially decreased, however at 4 and 6 d of waterlogging it increased over control plants, probably due to activation of DPI-sensitive NADPH-oxidase. Antioxidant enzymes such as superoxide dismutase, ascorbate peroxidase, glutathione reductase and catalase also increased under waterlogging. The comparatively greater antioxidant enzyme activities resulting in less oxidative stress in ICP 301 could be one of the factor determining its higher tolerance to flooding as compared to Pusa 207. This study is the first to conclusively prove that waterlogging induced increase in ROS is via NADPH oxidase. |
Cadmium-induced oxidative damage and antioxidant responses in Brassica juncea plantsY. K. Markovska, N. I. Gorinova, M. P. Nedkovska, K. M. Vitasso) plants exposed to 10, 30, 50 and 100 µM of Cd for 5 d in hydroponic culture were analysed with reference to the distribution of Cd2+, the accumulation of biomass and antioxidants and antioxidative enzymes in leaves. Cd induced a decrease in plant biomass. The maximum accumulation of Cd occurred in roots followed by stems and leaves. Cd induced a decrease in catalase (CAT) and guiacol peroxidase (GPX) activities but an increase in ascorbate peroxidase (APX) and monodehydroascorbate reductase (MDHAR) activities. Enhancement in dehydroascorbate reductase (DHAR) activity was also at 10 µM Cd. Glutathione reductase (GR) activity showed pronounced stimulation after all treatments, but glutathione S-transferase (GST) and glutathione peroxidase (GPOX) activities decreased. The effectiveness of ascorbate-glutathione cycle (AGC) was determined by the ratio of ascorbate to H2O2. This ratio decreased in the Cd-treated leaves which indicated that the cycle was disordered. |
Growth, lipid peroxidation and photosynthesis in two tall fescue cultivars differing in heat toleranceJ. Z. Wang, L. J. Cui, Y. Wang, J. L. LiBiologia plantarum 53:237-242, 2009 | DOI: 10.1007/s10535-009-0045-8 Seedlings (70-d-old) of two tall fescue (Festuca arundinacea Schreb.) genotypes, heat-tolerant Jaguar 3 and heat-sensitive TF 66, were exposed to a high temperature stress of 35/30 °C (day/night) for 20 d and both light-saturated and CO2-saturated leaf stomatal conductance decreased, especially in TF 66. Higher reductions of quantum efficiency, carboxylation efficiency and maximum photochemical efficiency of photosystem 2 in dark adapted leaves (measured as Fv/Fm) occurred in TF 66 than in Jaguar 3. High temperature stress increased photorespiration in the two plants, but more in TF 66. Moreover, high temperature stress also reduced the growth, chlorophyll content and caused cell membrane injuries in the two cultivars, the changes were again more pronounced in TF 66 than in Jaguar 3. |
Effects of salicylic acid and salinity on apoplastic antioxidant enzymes in two wheat cultivars differing in salt toleranceS. Mutlu, Ö. Atici, B. NalbantogluBiologia plantarum 53:334-338, 2009 | DOI: 10.1007/s10535-009-0061-8 The effects of salicylic acid (SA) and salinity on the activity of apoplastic antioxidant enzymes were studied in the leaves of two wheat (Triticum aestivam L.) cultivars: salt-tolerant (Gerek-79) and salt-sensitive (Bezostaya). The leaves of 10-d-old seedlings grown at nutrient solution with 0 (control), 250 or 500 mM NaCl were sprayed with 0.01 or 0.1 mM SA. Then, the activities of catalase (CAT), peroxidase (POX) and superoxide dismutase (SOD) were determined in the fresh leaves obtained from 15-d-old seedlings. The NaCl applications increased CAT and SOD activities in both cultivars, compared to those of untreated control plants. In addition, the NaCl increased POX activity in the salt-tolerant while decreased in the salt-sensitive cultivar. In control plants of the both cultivars, 0.1 mM SA increased CAT activity, while 0.01 mM SA slightly decreased it. SA treatments also stimulated SOD and POX activity in the salt-tolerant cultivar but significantly decreased POX activity and had no effect on SOD activity in the saltsensitive cultivar. Under salinity, the SA treatments significantly inhibited CAT activity, whereas increased POX activity. The increases in POX activity caused by SA were more pronounced in the salt-tolerant than in the salt-sensitive cultivar. SOD activity was increased by 0.01 mM SA in the salt-tolerant while increased by 0.1 mM SA treatment in the salt-sensitive cultivar. |
