Successful micropropagation protocol of Piper methysticumZ. Zhang, L. Zhao, X. Chen, X. ZhengBiologia plantarum 52:110-112, 2008 | DOI: 10.1007/s10535-008-0020-9 An efficient in vitro propagation of kava (Piper methysticum) was established. Utilizing 15-d-old tender shoots from dormant auxiliary buds as explants, significant induction of vigorous aseptic cluster shoots was achieved in Murashige and Skoog (MS) medium containing 0.5 mg dm-3 6-benzyladenine (BA), 0.5 mg dm-3 indole-3-acetic acid (IAA), and antibiotics after 30 d. In vitro rooting was achieved at 100 % efficiency in MS medium containing 0.75 to 1.00 mg dm-3 IAA or indole-3-butyric acid and 3 % sucrose. The most robust and long roots were observed in medium with IBA. Moreover, the embryonic callus was induced from petioles in MS medium supplemented with 1.0 mg dm-3 BA and 0.1 mg dm-3 IAA, of which 70 % differentiated into shoots in the presence of 1.0 mg dm-3 BA and 0.5 mg dm-3 IAA. |

HassanBiologia plantarum 52:165-169, 2008 | DOI: 10.1007/s10535-008-0036-1 The effects of Al, Cd and pH on growth, photosynthesis, malondialdehyde (MDA) content, and some antioxidant enzyme activities of the two soybean cultivars with different Al tolerance were determined using a hydroponic culture. There were six treatments as follows: pH 6.5; pH 4.0; pH 6.5 + 1.0 µM Cd; pH 4.0 + 1.0 µM Cd; pH 4.0 + 150 µM Al; pH 4.0 + 1.0 µM Cd + 150 µM Al. The results showed that the low pH (4.0) and Al treatments caused marked reduction in the growth (root and shoot length and dry mass), chlorophyll content (SPAD value) and net photosynthetic rate. Higher malondialdehyde content, superoxide dismutase (SOD) and peroxidase (POD) activities were detected in the plants exposed to both Al and Cd than in those exposed to Al treatment alone. An expressive enhancement of SOD and POD was observed in the plants exposed to 150 µM Al in the comparison with the control plants, especially in Al-sensitive cv. Zhechun 2 which had also significantly higher Al and Cd content than Al tolerant cv. Liao-1. Cd addition increased Al content in the plants exposed to Al + Cd stress, and cv. Zhechun 2 had relatively lower Al content. The present research indicated that Al and Cd are synergistic in their effects on plant growth and some physiological traits. |

A total of 352 products were amplified by 34 10-mer arbitrary primers, among which 337 products (95.74 %) were found to be polymorphic. 5-14 polymorphic bands were amplified by each polymorphic primer, with an average of 9.91 bands. The data of 352 RAPD bands were used to generate Jaccard's similarity coefficients and to construct a dendrogram by means of UPGMA. Great genetic diversity in genus Leymus was observed, the genetic diversity among the different species more abundant than that of the different accessions, and the different accessions in a species or the species from the same areas were clustered together. |
Direct shoot regeneration from leaf explants of Spilanthes acmellaK. V. Saritha, C. V. NaiduBiologia plantarum 52:334-338, 2008 | DOI: 10.1007/s10535-008-0068-6 Multiple shoots of Spilanthes acmella Murr. were induced from nodal buds of in vivo and in vitro seedlings on Murashige and Skoog (MS) medium containing 1.0 mg dm-3 6-benzyladenine (BA) and 0.1 mg dm-3 α-naphthalene-acetic acid (NAA). Adventitious shoots were successfully regenerated from the leaf explants derived from the above mentioned multiple shoots. The efficiency of shoot regeneration was tested in the MS medium containing BA, kinetin, or 2-isopentenyl adenine in combination with NAA, indole-3-acetic acid (IAA), or indole-3-butyric acid (IBA) and gibberellic acid. Maximum number of shoots per explant (20 ± 0.47) was recorded with 3.0 mg dm-3 BA and 1.0 mg dm-3 IAA. An anatomical study confirmed shoot regeneration via direct organogenesis. About 95 % of the in vitro shoots developed roots after transfer to half strength MS medium containing 1.0 mg dm-3 IBA. 95 % of the plantlets were successfully acclimatized and established in soil. The transplanted plantlets showed normal flowering without any morphological variation. |

Water stress and photoinhibition, often accompanying the acclimatization of in vitro grown plantlets to ex vitro conditions, are probably the main factors promoting production of reactive oxygen species (ROS) and in consequence oxidative stress. The extent of the damaging effects of ROS depends on the effectiveness of the antioxidative systems which include low molecular mass antioxidants (ascorbate, glutathione, tocopherols, carotenoids, phenols) and antioxidative enzymes (superoxide dismutase, ascorbate peroxidase, catalase, glutathione reductase, monodehydroascorbate reductase, dehydroascorbate reductase). This review is focused on ROS production and development of antioxidative system during in vitro growth and their further changes during ex vitro transfer. |
In vitro clonal propagation of Nyctanthes arbor-tristisG. R. Rout, A. Mahato, S. K. SenapatiBiologia plantarum 52:521-524, 2008 | DOI: 10.1007/s10535-008-0101-9 Rapid shoot multiplication of Nyctanthes arbor-tristis L. was achieved from axillary meristems on Murashige and Skoog (MS) basal medium supplemented with 1.0-1.5 mg dm-3 6-benzylaminopurine (BA), 50 mg dm-3 adenine sulfate (Ads) and 3 % (m/v) sucrose. Inclusion of indole-3-acetic acid (IAA) in the culture medium along with BA + Ads promoted a higher rate of shoot multiplication. Maximum mean number of microshoots per explant (6.65) was achieved on the MS medium supplemented with 1.5 mg dm-3 BA, 50 mg dm-3 Ads and 0.1 mg dm-3 IAA after 4 weeks of culture. The elongated shoots rooted within 13 to 14 d on half-strength MS medium supplemented with either indole-3-butyric acid (IBA), IAA or 1-naphthaleneacetic acid (NAA) with 2 % sucrose. Maximum percentage of rooting was obtained on medium having 0.25 mg dm-3 IBA and 0.1 mg dm-3 IAA. About 70 % of the rooted plantlets survived in the greenhouse. The in vitro raised plants were grown normally in the field. |

The plants were subjected to 100 mM NaCl and sodium nitroprusside (SNP; the donor of NO) at different concentrations. The plants showed lower NaCl-induced oxidative stress and proline accumulation after application of low concentrations of SNP together with the NaCl treatment. The reduction in the proline content was related to increased activity of proline dehydrogenase. These results suggest that the NO could be capable of mitigating damage associated with salt stress. |
Involvement of exopeptidases in dehydration tolerance of spring wheat seedlingsA. Miazek, B. ZagdañskaBiologia plantarum 52:687-694, 2008 | DOI: 10.1007/s10535-008-0133-1 The observed inability of 6-d-old seedlings of spring wheat (Triticum aestivum L.) to tolerate the same water deficit as compared to the 4-d-old seedlings seems to be associated with the higher carboxypeptidase and lower aminopeptidase activities. Free amino acid pools differentiated also the 4-d-old seedlings from the older ones. Dehydration decreased the amino acid content in 4-d-old seedlings, increased it in 6-d-old seedlings and changed composition of amino acid pool. In tolerant phase of wheat seedling growth carboxypeptidase activity increased in response to water deficit and aminopeptidase activity increased in dehydrated seedlings, independently of their age. |
Flooding tolerance and genetic diversity in populations of Luehea divaricataM. C. C. G. De Carvalho, D. C. G. Da Silva, P. M. Ruas, M. E. Medri, E. A. Ruas, C. F. RuasBiologia plantarum 52:771-774, 2008 | DOI: 10.1007/s10535-008-0150-0 We investigated some aspects of flooding tolerance in two riparian populations (exposed and no exposed to flooding) of Luehea divaricata C. Martius. Plants derived from seeds collected in each population were submitted to flooding (30 and 60 d), submergence and re-aeration treatments. Plants exposed to flooding showed development of aerenchyma, hypertrophic lenticels and new adventitious roots. Interestingly, whereas the plants originated from population naturally exposed to flooding developed some of these alterations more markedly, they could not survive when totally submerged. The random amplified polymorphic DNA (RAPD) markers, showed a significant difference between populations, suggesting that seasonal flooding on riparian populations of L. divaricata has been selecting individuals who are more adapted to survive in these conditions. |

ZhuangBiologia plantarum 52:93-96, 2008 | DOI: 10.1007/s10535-008-0015-6 A new micropropagation system for Lycium barbarum (L.) was developed using root explants as starting material. Callus can be produced from root explants on Murashige and Skoog (MS) medium containing 0.2 mg dm-3 2,4-dichlorophenoxyacetic acid. After three subcultures on the same medium, callus was then transferred onto the MS medium supplemented with 500 mg dm-3 lactalbumin hydrolysate to induce somatic embryogenesis (SE). After 20 d, about 60 somatic embryos per 0.25 g(f.m.) of embryogenic callus were obtained but only about 10 % of embryos converted into plantlets. After acclimated in the greenhouse, all of the randomly selected plantlets had survived and were similar phenotypically to zygotic seedlings. In addition, the effects of irradiance, photoperiod, growth regulators, explant age and cold treatment on SE of root-derived callus were evaluated. |
Limited variability of CTG/CAG repeats in Lycopersicon nuclear DNAD. Dimitrova, O. Georgiev, C. Valkova, B. Atanassova, L. KaragyozovBiologia plantarum 52:149-152, 2008 | DOI: 10.1007/s10535-008-0031-6 Seven clones containing (CTG)n/(CAG)n repeats (n ≥ 4) were isolated by screening Lycopersicon esculentum genomic DNA. Four of the clones contained more than one simple sequence repeat (SSR). The SSRs were analyzed in several L. esculentum cultivars after polymerase chain reaction (PCR) amplification. No length variations were observed, suggesting considerable locus stability. Five clones are from transcribed regions, which might explain the lack of cultivar variations. However the conservation of CTG repeats was limited as differences in some transcribed loci were registered between L. pennellii and other Lycopersicon species. It is noted that in Lycopersicon trinucleotide repeat variation might be used for species identification. |

XuBiologia plantarum 52:209-214, 2008 | DOI: 10.1007/s10535-008-0047-y Somatic embryogenesis and plant regeneration were successfully established on Nitsch and Nitsch (NN) medium from immature zygotic embryos of six genotypes of grapevine (Vitis vinifera). The optimum hormone combinations were 1.0 mg dm-3 2,4-dichlorophenoxyacetic acid (2,4-D) for callus induction and 1.0 mg dm-3 α-naphthalene acetic acid (NAA) + 0.5 mg dm-3 6-benzyladenine (BA) for embryos production and 0.03 mg dm-3 NAA + 0.5 mg dm-3 BA for embryos conversion and plant regeneration. The frequency of somatic embryogenesis varied from 10.5 to 37.5 % among six genotypes and 15.5-42.1 % of somatic embryos converted into normal plantlets. The analysis of DNA content determined by flow cytometry and chromosome counting of the regenerated plantlets clearly indicated that no ploidy changes were induced during somatic embryogenesis and plant regeneration, the nuclear DNA content and ploidy levels of the regenerated plants were stable and homogeneous to those of the donor plants. RAPD markers were also used to evaluate the genetic fidelity of plants regenerated from somatic embryos. All RAPD profiles from regenerated plants were monomorphic and similar to those of the field grown donor plants. We conclude that somaclonal variation is almost absent in our grapevine plant regeneration system. |
Stem CO2 efflux of a Populus nigra stand: effects of elevated CO2, fertilization, and shoot sizeM. Liberloo, P. De Angelis, R. CeulemansBiologia plantarum 52:299-306, 2008 | DOI: 10.1007/s10535-008-0063-y To determine whether long-term growth in elevated atmospheric CO2 concentration [CO2] and nitrogen fertilization affects woody tissue CO2 efflux, we measured stem CO2 efflux as a function of temperature in three different size classes of shoots of Populus nigra L. (clone Jean Pourtet) on two occasions in 2004. Trees were growing in a short rotation coppice in ambient (370 µmol mol-1) and elevated (550 µmol mol-1, realised by a Free Air Carbon dioxide Enrichment system) [CO2], and measurements were performed during the third growing season of the second rotation. Elevated CO2 did not affect Q10 or specific stem CO2 efflux (E10) of overall poplar shoots. The lack of any effect of N on stem CO2 efflux indicated that nutrients were sufficient. Specific stem CO2 efflux differed significantly between shoot sizes, emphasizing the importance of tree size when scaling-up respiration measurements to the stand level. Variation in stem CO2 efflux could not be satisfactorily explained by temperature as the only driving variable. We hypothesize that transport of CO2 with the sapflow might have confounded our results and could explain the high Q10 values reported here. Predicting the respiratory carbon loss in a future elevated [CO2] world must therefore move beyond the single-factor temperature dependent respiration model and involve multiple factors affecting stem CO2 efflux rate. |
Transient RNAi based gene silencing of glutathione synthetase reduces glutathione content in Camellia sinensis (L.) O. Kuntze somatic embryosP. Mohanpuria, N. K. Rana, S. K. Using degenerate primers with cDNA of Camellia sinensis, a 457 bp GSHS gene fragment was cloned through polymerase chain reaction. This fragment was used in making ihpRNA. For this it was cloned in sense at AscI and SwaI and in anti-sense at Bam HI and XbaI restriction sites of pFGC5941 that has chalcone synthase (Chs) intron between SwaI and BamHI restriction sites. Resultant RNAi construct was used for C. sinensis somatic embryos transformation through Agrobacterium. After 11, 13 and 15 d of transformation, embryo GSHS transcript levels and GSH content decreased to a great extent which documented the feasibility of RNAi based gene silencing in C. sinensis. |
Putative primary involvement of Arabidopsis phosphoinositide-specific phospholipase C1 within abscisic acid-induced stomatal closingA. CoussonBiologia plantarum 52:493-501, 2008 | DOI: 10.1007/s10535-008-0096-2 Stomatal closing to abscisic acid (ABA) was studied in leaf epidermal peels of a dexamethasone (Dex)-inducible transgenic line expressing the phospholipase C AtPLC1 antisense in the Columbia genetic background. In the absence of Dex, the Ca2+ buffer, ethylene glycol-bis(b-aminoethyl ether)-N,N,N',N'-tetraacetic acid (EGTA) and the phopholipase C inhibitor, 1-[6-{[17β-3-methoxyestra-1,3,5(10)-trien-17-yl]amino}hexyl]-1H-pyrrole-2,5-dione (U73122) specifically inhibited the response to 20 µM ABA, whereas the Ca2+ buffer, 1,2-bis(o-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid (BAPTA) inhibited the response to 20 or 30 µM ABA. Neither EGTA nor BAPTA increased the U73122 effect. Applying 30 µM Dex specifically affected 20 µM ABA-induced stomatal closing through reducing its magnitude as well as suppressing the EGTA, BAPTA and U73122 inhibitory effects. Neither Dex nor U73122 changed the specific inhibitory effects of both the antagonist of cyclic ADP-ribose synthesis, nicotinamide and the GTP-binding protein (G protein) modulators, pGlu-Gln-D-Trp-Phe-D-Trp-D-Trp-Met-NH2 (GP Ant-2) and mas17 on 30 µM ABA-induced stomatal closing. When tested in combination, substituting nicotinamide for mas17, but not for GP Ant-2, enhanced their inhibitory effect to an extent that BAPTA did not increase. These results supported that AtPLC1 primarily mediates the Ca2+-dependent stomatal closing response to 20 µM ABA as much as 30 µM Dex did not affect 20 µM ABA-induced stomatal closing when tested on the wild type Columbia-4 ecotype. Furthermore, the present study suggested that Ca2+ mobilization did not involve any dependency between AtPLC1 and a putative G protein-coupled ADP-ribosyl cyclase at the tested ABA concentrations. |

Zlota Saxa) grown hydroponically in nutrient solution and exposed to Pb(NO3)2 (0.1, 0.5, 1 mM) with or without equimolar concentrations of chelator ethylenediaminetetraacetic acid (EDTA). The roots treated only with Pb(NO3)2 accumulated up to 25 g(Pb) kg-1(d.m.), during 4-d exposure. However, in bean plants exposed to 0.5 mM Pb + 0.5 mM EDTA or 1 mM Pb + 1 mM EDTA 2.5 times less Pb was determined. In bean plants treated only with Pb, less than 6 % of total lead accumulated was transported to the aboveground parts, while in the case of plants grown with Pb + EDTA, around 50 % of total Pb was transported to the shoots. |

GrilliBiologia plantarum 52:660-667, 2008 | DOI: 10.1007/s10535-008-0128-y Acidic and neutral ribonucleases (RNases) were studied in embryos of Triticum durum cv. Cappelli and the effects of abscisic acid (ABA) and gibberellic acid (GA3) were analysed. RNases activities increased during germination and were comparable in dormant and non-dormant embryos imbibed for 24 h. ABA generally inhibited ribonucleolytic activities, while GA3 only affected dormant embryos. To assess whether changes in RNase activities during germination or following hormonal treatment required new transcriptional or translational action, cycloheximide or cordycepin were used. The action of inhibitors of acidic RNase activity was found only in non-dormant-embryos. Findings obtained in the present work concur with a change of the ribonucleolytic pattern in the shift from dormant to non dormant metabolism. |
Starch synthase activity and heat shock protein in relation to thermal tolerance of developing wheat grainsK. V. Sumesh, P. Sharma-Natu, M. C. GhildiyalBiologia plantarum 52:749-753, 2008 | DOI: 10.1007/s10535-008-0145-x Wheat (Triticum aestivum L.) cvs. HD 2285 (relatively tolerant) and WH 542 (susceptible) were exposed to ambient and elevated temperature (3-4 °C higher) in open top chambers during post anthesis period. The grain yield components were determined at the time of maturity. In order to elucidate the basis of differential tolerance of these cultivars, the excised developing grains (20 d after anthesis) of ambient grown plants were incubated at 15, 25, 35 and 45 °C for 2 h and then analysed for the activities of soluble starch synthase (SSS), granule bound starch synthase (GBSS), kinetic parameters of SSS and content of heat shock protein (HSP 100). The elevated temperature during grain development significantly decreased grain growth in WH 542 whereas no such decrease was observed in HD 2285. High temperature tolerance of HD 2285 was found to be associated with higher catalytic efficiency (Vmax/Km) of SSS at elevated temperature and higher content of HSP 100. |
Origin and development of secondary somatic embryos in transformed embryogenic cultures of Medicago sativaB. Uzelac, S. Ninkoviæ, A. Smigocki, S. BudimirBiologia plantarum 51:1-6, 2007 | DOI: 10.1007/s10535-007-0001-4 Non-transformed and transformed embryogenic cultures of alfalfa (Medicago sativa L. cv. Zajeèarska 83), long-term maintained on growth regulator-free medium, were histologically analyzed. In all examined cultures, somatic embryos at various stages of development were observed and secondary embryos were formed in the cotyledonary, hypocotylary and radicular region of the primary embryos. Detailed histological analysis of the torpedo shape somatic embryo revealed that secondary somatic embryos arose directly from single epidermal cells of hypocotylary axis after an unequal periclinal division. Bipolar proembryos were composed of one smaller cytoplasm rich cell and one larger more vacuolated cell. Further cell division pattern was similar for both non-transformed and transformed embryos. However, multicellular origin of secondary embryos in a direct process and even from callus can not be excluded. |

CarvajalBiologia plantarum 52:72-79, 2008 | DOI: 10.1007/s10535-008-0010-y Water relations, mineral composition, growth and root morphology were studied in pepper plants (Capsicum annuum L. cv California Wonder). Two NaCl concentrations (30 and 60 mM) and two nutrient solutions in which the concentrations of macronutrients were increased were used to assess the ionic and osmotic effects of NaCl in these plants. The hydraulic conductivity (Lo), stomatal conductance (gs), percentage of open stomata and pressure potential (Ψp) decreased with all treatments, in a similar way for 30 mM NaCl and for its iso-osmotic solution of macronutrients, however, the decrease was higher for 60 mM NaCl than for its iso-osmotic solution. Ion analyses also revealed that nutrient concentrations were altered greatly at 60 mM NaCl. Also, changes in morphology, such as increases in cortex cell size and in intercellular spaces, were detected. Therefore, at low salinity, the effect of NaCl was mainly osmotic, however, under higher salinity also the toxicity of Na+ and Cl- participate. |

Non regenerative friable callus from hypocotyls and cotyledon explants from in vitro raised seedling induced on solid Murashige and Skoog (MS) medium supplemented with 0.05 mg dm-3 2,4-dichlorophenoxyacetic acid (2,4-D) was used for initiation of cell suspension cultures on same MS medium but without agar. Single cells were isolated after 3 d and the optimum cell density was 1-3 × 104 cells per cm3 of the liquid MS medium. Plating efficiency was 29.6 % and callus formed within 4 weeks was subcultured and transferred to solid MS medium supplemented with 0.6 mg dm-3 benzyladenine (BA) along with 0.05 mg dm-3 α-naphthalene-1-acetic acid (NAA) for the induction of adventitious bud primordia. The shoots developed were isolated and re-cultured on MS medium containing 0.6 mg dm-3 BA. These microshoots after dipping in 1-2 cm3 of 10 mg dm-3 indole-3-butyric acid (IBA) for 24 h in dark were cultured on half strength solid MS medium supplemented with 0.05 % charcoal and showed 80-82 % rooting within 4 weeks. |

Jinan 177), we developed a specific PCR primer set to decide its locus with nullisomic-tetrasomic lines of Chinese spring wheat. The result showed that it was assigned to Glu-D3. The DNA fragment of 177-21 was then subcloned into the pGEX-4T-1 expression vector and expressed in E. coli with isopropyl-1-thio-β-D-galactoside induction. The result indicated that this gene encodes about 30 kD polypeptide and deduced amino acid sequence consists of eight cysteine residues. Of the eight, six may be related with the formation of intra-molecular disulfide bonds, the last two with the formation of inter-molecular disulfide bonds, which could be a potential extender in "glutenin polymer" to have positive influence on quality of wheat flour. |

Cloning of differential expression fragments in cauliflower after Xanthomonas campestris inoculationY. Gu, Y. W. Mao, C. G. Wang, Q. C. Zhao, D. L. Sun, W. Q. SongBiologia plantarum 52:462-468, 2008 | DOI: 10.1007/s10535-008-0091-7 A near isogenic line (NIL) of Brassica oleracea var. botrytis with resistant and susceptible lines C712 and C731, was used in this study. More than 100 differentially expressed cDNA fragments were obtained from black rot resistant cauliflower plants obtained using cDNA-amplified fragment length polymorphism (AFLP) after infection with the pathogen. Thirteen of these fragments were cloned and subjected to reverse Northern blot analysis using both infected and control cDNA pools. Two positive clones, M2 and M6, were isolated. Northern dot blot and Northern blot analyses showed that M2 was constitutively expressed, whereas M6 contained a gene that was differentially expressed during pathogen infection. Moreover, M6 cDNA fragment was also highly expressed 16-24 h after H2O2 treatment. Southern blots showed that M6 is a single copy gene in the cauliflower genome, and encodes a protein with 84 % homology to gene on Arabidopsis chromosome 1. The deduced M6 protein has 91 % positive homology with the Arabidopsis 2A6 protein, which regulates ethylene synthesis; 76 % homology with a 1-aminocyclopropane-1-carboxylate oxidase (ACO), the last enzyme in ethylene synthesis; and 70 % homology with an ethylene induced DNA binding factor. These results suggest that M6 gene fragment is a new H2O2 downstream defense related gene fragment and can be induced by Xanthomonas campestris pv. campestris and H2O2. |

PicardiBiologia plantarum 52:548-552, 2008 | DOI: 10.1007/s10535-008-0107-3 Pericarp polypeptide profiles were analyzed at three ripening stages in the F1 hybrid and the F2 population from the cross between the accessions: LA1385 (Lycopersicon esculentum var. cerasiforme) and 804627 (L. esculentum, a homozygous genotype for the nor mutant). Six polymorphic polypeptides were observed in LA1385, while no polymorphic polypeptides among ripening stages was observed in 804627. On the other hand, some polypeptides in the F1 hybrid were not observed in the parents whereas others were present in both parental genotypes and were unnoticeable in the hybrid genotype. From a cluster analysis on the protein profiles of the F2 population, the differential expression of proteins allowed to distinguish mature green (MG) stage from the others two stages, while for breaker stage (BR) and red ripe stage, the genetic background was more important in forming groups. The differential expression of proteins could be associated with fruit morphology traits such as a 72 kDa polypeptide present in MG stage with fruit diameter, height and mass and a 47 kDa polypeptide found in BR with fruit shelf life. |
Genetic diversity in in vitro-conserved germplasm of Curcuma L. as revealed by RAPD markersZ. Hussain, R. K. Tyagi, R. Sharma, A. AgrawalBiologia plantarum 52:627-633, 2008 | DOI: 10.1007/s10535-008-0123-3 A set of 30 accessions of five Curcuma species-C. latifolia, C. malabarica, C. manga and C. raktakanta and 13 morphotypes (identified on the basis of morphological markers) of C. longa conserved in the In Vitro Genebank at National Bureau of Plant Genetic Resources, New Delhi, were subjected to RAPD analysis. Of the 200 RAPD primers screened, 21 polymorphic primers were selected for further study. Mean genetic similarities based on Jaccard's similarity coefficient ranged from 0.18 to 0.86 in accessions of cultivated species, i.e., C. longa and from 0.25 to 0.86 in wild species. The dendrogram derived from the RAPD data corroborated the morphological classification of the morphotypes. The efficiency of individual RAPD primers was also compared; primers OPC-20, OPO-06, OPC-01 and OPL-03 were adjudged highly informative in discriminating the germplasm of Curcuma. |
Induction of in vitro flowering in the orchid Dendrobium Sonia 17C. S. Tee, M. Maziah, C. S. TanBiologia plantarum 52:723-726, 2008 | DOI: 10.1007/s10535-008-0139-8 In this study, Dendrobium Sonia 17 plantlets were used to induce in vitro flowering. Inflorescences were induced and rooting was inhibited in the half-strength Murashige and Skoog medium containing 20 µM N 6-benzyladenine (BA). The medium with high P and low N contents was effective to induce inflorescences while the medium with low P and high N contents was only effective to promote forming of shoots. In addition, the induced in vitro inflorescences were able to multiply and maintain without exhibiting a distinctive vegetative phase. Different morphologies of in vitro flowers such as incomplete flower structures, abnormal and unresupinated in vitro flowers were observed. |
Contribution of putrescine degradation to proline accumulation in soybean leaves under salinityG. X. Su, X. BaiBiologia plantarum 52:796-799, 2008 | DOI: 10.1007/s10535-008-0156-7 Proline accumulation was studied in the leaves of Glycine max (L.) Merr. subjected to salt stress in the presence of aminoguanidine (AG, a specific inhibitor of diamine oxidase, DAO) and exogenous putrescine (Put). Both DAO activity and proline content were increased while endogenous Put content was decreased in soybean leaves under 50 to 150 mM NaCl. There was a negative correlation between proline accumulation and endogenous Put content. The addition of AG during NaCl stress inhibited DAO activity, caused Put accumulation and a 15 to 20 % decrease in proline content. Application of 1 mM Put to NaCl solution markedly increased proline content. The promotive effect of Put application could be alleviated by the treatment with Put plus AG. Moreover an application of AG had no effect on proline accumulation in soybean seedlings grown under normal condition. These results indicate that the quantitative contribution of Put degradation to proline formation is 15 to 20 %. |

TMV 6) through cotyledon derived callus cultures. The impact of different concentrations of auxins, cytokinins, additives, amino acids and sugars were evaluated for callus induction and shoot proliferation. Green compact nodular organogenic callus was obtained on the medium fortified with Murashige and Skoog (MS) salts, B5 vitamins, 2.0 mg dm-3 6-benzyladenine and 0.8 mg dm-3 α-naphthalene acetic acid (NAA). Multiple shoot proliferation from the callus cultures was achieved on the medium with MS salts, B5 vitamins, 2.5 mg dm-3 thidiazuron (TDZ), 0.4 mg dm-3 NAA and 15 mg dm-3 glutamine. During multiple shoot induction the phenolic secretion was controlled by the addition of 15 mg dm-3 polyvinylpyrolidone. The proliferated shoots were elongated on the medium comprising MS salts, B5 vitamins, 1.5 mg dm-3 TDZ and 0.3 mg dm-3 gibberellic acid. The elongated shoots were rooted on the medium containing MS salts, B5 vitamins, 0.3 mg dm-3 indole-3-butyric acid and 0.6 mg dm-3 silver nitrate. After root induction, the plants were hardened in earthen pots containing sand, soil and vermiculite. |
