Development of the CELLOP optimisation model for plant cell cultivation

A mathematical computer-aided model CELLOP was constructed in which the desirability functions in a three-dimensional experimental design can be used to find the optimal growing conditions for plant cells. CELLOP is intended for the optimisation of 3 to 6 physical, chemical, or biological variables in the cultivation conditions of plant cell cultures. The model was used to optimise the culturing conditions (calcium, inorganic nitrogen, and sucrose concentrations) for coumarin-producing, spontaneously embryogenic cell lines of angelica Angelica archangelica L. subsp. archangelica and hogs fennel Peucedanum palustre (L.) Moench. For A. archangelica the overall optimum concentrations were 0.47 mM Ca2+, 5.06 mM NO3 -, 0.40 mM NH4 +, and 96.25 mM sucrose. The dry mass was 24.7 % higher and the coumarin content 40.5 % higher than those achieved in the standard 75 % Gamborg B5 medium. For A. archangelica the highest embryogenic activity was reached in the media containing 1.25 mM Ca2+. In the case of P. palustre the overall optimum concentrations were 1.60 mM Ca2+, 2.84 mM NO3 -, 0.23 mM NH4 +, and 85.10 mM sucrose. For P. palustre the dry mass production increased by 61.8 % and the coumarin content by 58.1 % compared to the values achieved in the Gamborg B5 medium. For P. palustre the highest embryogenic activity was reached in the presence of 50.0 mM NO3 - and 4.01 mM NH4 +.

Salinity induced changes in proline and betaine contents and synthesis in two aquatic macrophytes differing in salt tolerance

The present work investigated the effect of NaCl and seawater salinity (SWS) on accumulation of proline and glycinebetaine (betaine) and expression of two key enzymes of their biosynthetic pathway, betainealdehyde dehydrogenase (BADH) and Δ1-pyrroline-5-carboxylate synthetase (P5CS) in two aquatic macrophytes, salt-tolerant Najas gramenia and moderately-salt-tolerant Najas indica. N. gramenia did not show any accumulation of betaine or expression of BADH in response to NaCl or SWS treatments. The content of proline increased significantly in response to the salt treatments. High expression of P5CS was also observed, however, the transcript abundance did not correlate with the amount of proline. N. indica, exhibited significantly greater accumulation of proline in response to SWS, which contained Ca2+ in addition to NaCl, than in response to NaCl alone.

Influence of UV-B radiation on young triticale plants with different wax cover

Ultraviolet-B radiation (biologically effective dose 2.6 kJ m-2 d-1) had negative influence on morphology and physiology of the young triticale plants. Plants exposed to UV-B were of lower height than control plants, their leaves were narrow, and the rate of net photosynthesis was decreased. The line RAH 336, which wax cover is lesser than that of traditional cultivar Magnat, was more susceptible to UV-B radiation, considering primary photosynthesis reactions, recorded by chlorophyll a fluorescence. An activation of protective mechanisms was observed: plants responded to UV-B by an increase of the content of UV-B absorbing compounds, and changes of antioxidant enzyme activities.

Production of human papillomavirus type 16 E7 oncoprotein fused with β-glucuronidase in transgenic tomato and potato plants

The human papillomavirus type 16 (HPV 16) oncogene E7 fused with the gene for β-glucuronidase (gus) was used in plant transformation experiments. The E7 gene modified for lower cancerogenicity and fused with the 5' end of the gus in cassettes with cauliflower mosaic virus 35S promoter and transcription terminator produced high contents of fusion proteins in potato protoplasts. Expression vectors harbouring E7 fusion cassettes were used for Agrobacterium tumefaciens LBA4404 mediated transformation of either potato (Solanum tuberosum L. cv. Bintje) or tomato (Lycopersicon esculentum Mill. cv. Moneymaker). A fusion gene was found in all rooted regenerants using polymerase chain reaction with primers providing amplified fragments from E7 and gus genes. GUS activity was revealed in all regenerants obtained. Nevertheless, the level of GUS expression in different constructs varied much more than in transient expression experiments with potato protoplasts. Especially, expression level in plants carrying vectors with the whole E7 gene fused with gus was lowered by 2-3 orders of magnitude comparing with fusion of the first 41 codons of E7 and gus. Southern hybridisation of 18 tomato and 23 potato regenerants revealed mostly multiple tandem integration of T-DNA into the plant genome and Western blot proved the presence of the fusion protein in 9 tomato and 11 potato plants out of 41 tested individuals.

Plant regeneration through direct somatic embryogenesis from leaf explants of Dendrobium

A protocol for induction of direct somatic embryogenesis, secondary embryogenesis and plant regeneration of Dendrobium cv. Chiengmai Pink was developed. Thidiazuron (TDZ) at 0.3, 1 and 3 mg dm-3 induced 5-25 % of leaf tip segments of in vitro grown plants to directly form embryos after 60 d of culture, and 1 mg dm-3 TDZ was the best treatment. Somatic embryos mostly formed from leaf surfaces near cut ends, and occasionally found on leaf tips. Higher frequency of embryogenesis was obtained in light than in darkness. During subculture, secondary embryos developed from outer cell layers of primary embryos. All combinations of NAA (0, 0.1, 1 mg dm-3) and TDZ (0, 0.3, 1, 3 mg dm-3) increased the multiplication rate of embryos. It takes about 8 months from embryo induction, plantlet formation to eventually acclimatization in greenhouse.

Changes in spectral reflectance of a foliar lichen Umbilicaria hirsuta during desiccation

Water potential (ψw) and water saturation deficit (WSD), and several reflectance (R) indexes were assessed in an aerophytic lichen Umbilicaria hirsuta (Sw. ex Westr.) Hoffm. The water index (WI, R900/R970) and normalized difference vegetation index, NDVI [(R900-R680)/(R900+R680)] were strongly correlated both with the ψw and the WSD of lichen thalli. No significant changes during desiccation were found in structural independent pigment index, SIPI [(R800-R445)/(R800-R680)]. Sensitivity of the spectral detection of water status was rather small at high hydration level (WSD < 25 %, or ψw > -1 MPa), but this is not much limiting its value and potential use, because physiological processes in lichens are usually inhibited at much lower values of ψw than in leaves of vascular plants.

Cadmium mitigates ultraviolet-B stress in Anabaena doliolum: Enzymatic and non-enzymatic antioxidants

Impact of ultraviolet-B (UV-B) and Cd, applied individually and in combination, measured in terms of oxygen-evolution, chlorophyll (Chl) and protein contents, lipid peroxidation, and enzymatic and non-enzymatic antioxidants of Anabaena doliolum, revealed a greater oxidative damage induced by UV-B than by Cd. While superoxide dismutase (SOD) showed a greater stimulation by UV-B than Cd, the activities of catalase (CAT) and glutathione reductase (GR) declined after UV-B treatment. Cd treatment, however, enhanced the activities of ascorbate peroxidase (APX) and GR. CAT activity increased at low but decreased at high dose of Cd. Increase in carotenoid (Car) content in UV-B treated cells suggested a shielding effect of Car against UV-B stress. A 15-and 10-fold rise in α-tocopherol (α-TOC) content at high dose of Cd and/or UV-B offered testimony to the antioxidant role of α-TOC.

Metal hyperaccumulation and bioremediation

The phytoremediation is an environment friendly, green technology that is cost effective and energetically inexpensive. Metal hyperaccumulator plants are used to remove metal from terrestrial as well as aquatic ecosystems. The technique makes use of the intrinsic capacity of plants to accumulate metal and transport them to shoots, ability to form phytochelatins in roots and sequester the metal ions. Harbouring the genes that are considered as signatures for the tolerance and hyperaccumulation from identified hyperaccumulator plant species into the transgenic plants provide a platform to develop the technology with the help of genetic engineering. This would result in transgenics that may have large biomass and fast growth a quality essential for removal of metal from soil quickly and in large quantities. Despite so much of a potential, the progress in the field of developing transgenic phytoremediator plant species is rather slow. This can be attributed to the lack of our understanding of complex interactions in the soil and indigenous mechanisms in the plants that allow metal translocation, accumulation and removal from a site. The review focuses on the work carried out in the field of metal phytoremediation from contaminated soil. The paper concludes with an assessment of the current status of technology development and its future prospects with emphasis on a combinatorial approach.

Daily variations in water relations of apricot trees under different irrigation regimes

Mature apricot (Prunus armeniaca L. cv. Búlida) trees, growing under field conditions, were submitted to two drip irrigation treatments: a control (T1), irrigated to 100 % of seasonal crop evapotranspiration (ETc), and a continuous deficit (T2), irrigated to 50 % of the control throughout the year. The behaviour of leaf water potential and its components, leaf conductance and net photosynthesis were studied at three different times during the growing season, when they revealed a diurnal and seasonal pattern in response to water stress, evaporative demand of the atmosphere and leaf age. The deficit-irrigated trees showed, among other effects, a pronounced decrease in leaf water potential (ψw), decreased in leaf conductance (gs) and no osmotic adjustment. For this reason, gl and ψw can be considered good indicators of mature apricot tree water status and can therefore be used for irrigation scheduling.

Molecular characterization of Mauritanian date palm cultivars using plasmid-like DNAs markers

Mauritanian date palm cultivars and progenies of two controlled crosses were analyzed according to the identity of mitochondrial plasmid-like DNAs. Starting from total genomic DNA and appropriate primers, polymerase chain reaction was designed to amplify either a 373-bp or a 265-bp fragments corresponding to the S and the R-plasmid respectively. Data proved that 5 cultivars out of 10 studied have exhibited the R-plasmid suggesting their resistance to the fusariosis. The existence of intra-cultivar variability has also been revealed in the cv. Ahmar. In addition, analysis throughout progenies of two controlled crosses suggested the strict maternal transmission of the date palms' mitochondrial genome.

Agrobacterium-mediated transformation of Cry1C, Cry2A and Cry9C genes into Gossypium hirsutum and plant regeneration

Three constructs harbouring novel Bacillus thuringiensis genes (Cry1C, Cry2A, Cry9C) and bar gene were transformed into four upland cotton cultivars, Ekangmian10, Emian22, Coker201 and YZ1 via Agrobacterium-mediated transformation. With the bar gene as a selectable marker, about 84.8 % of resistant calli have been confirmed positive by polymerase chain reaction (PCR) tests, and totally 50 transgenic plants were regenerated. The insertions were verified by means of Southern blotting. Bioassay showed 80 % of the transgenic plantlets generated resistance to both herbicide and insect. We optimized conditions for improving the transformation efficiency. A modified in vitro shoot-tip grafting technique was introduced to help entire transplantation. This result showed that bar gene can replace antibiotic marker genes (ex. npt II gene) used in cotton transformation.

Low night temperature effects on photosynthetic performance on two grapevine genotypes

The functional activities of the photosynthetic apparatus of two grapevine genotypes (Vitis vinifera L. cvs. Müller-Thurgau and Lagrein) were investigated after low night temperature (LNT) treatment for 7 d. LNT caused important reductions of the net photosynthetic rate (PN) of Lagrein plants due to non-stomatal components. These non-stomatal effects were not evident in Müller-Thurgau. At LNT treatment, the contents of photosynthetic pigments decreased significantly in Lagrein, but in Müller-Thurgau the contents of chlorophyll (Chl) remained unchanged whereas the contents of carotenoids (Car) increased. An increase and decrease of Chl a/b was shown in Mü ller-Thurgau and Lagrein stressed plants, respectively. RuBPC activity and content of soluble proteins were also significantly reduced in Lagrein. Under LNT treatment, photosystem (PS) 2 was markedly more inhibited in Lagrein than in Müller-Thurgau. The decrease in PS 2 activity in Lagrein was mostly due to the marked loss of D1, 47, 43, 33, 28-25, 23 and 17 kDa proteins determined by immunological and SDS-PAGE studies.

Steady presence of cadmium and nickel affects root anatomy, accumulation and distribution of essential ions in maize seedlings

When growing in the field, plants are exposed to the effect of heavy metals as soon as the seed comes into contact with the soil solution. Therefore, we found important to study the effect of Cd and Ni on maize exposed to these heavy metals since sowing. The aim of this work was to examine which anatomical changes are induced by continuous intoxication of young maize root system with 0.1 mM Cd and Ni, thus modifying its growth and capacity for water and nutrient uptake. Concomitantly, the effect on concentration and distribution of Cd, Ni and some essential ions (Ca, Mg, Zn, Fe, Cu and Mn) was studied.

UV-B response of green and etiolated barley seedlings

7-d-old etiolated and green barley seedlings (Hordeum vulgare L. cv. Alfa) were irradiated with UV-B for 30 min and then kept for 24 h in light or darkness. Chlorophyll (Chl) synthesis was inhibited by about 30 % as a result of UV-B irradiation, but there were no significant changes in photochemical activity measured by variable to maximum fluorescence ratio (Fv/Fm), quantum yield (ΦPS2) and oxygen evolution rate. Electron transport of etiolated seedlings was similar to that of green ones, nevertheless, the Chl content was more then 2-fold lower. Ribulose-1,5-bisphosphate carboxylase/oxygenase large and small subunits were diminished as a result of UV-B irradiation in etiolated and green plants, especially in those kept in the darkness. Catalase activity decreased and total superoxide dismutase activity increased in green and etiolated plants following UV-B treatment. When benzidine was used as a substrate, an isoform located between guaiacol peroxidases 2 and 3 (guaiacol peroxidase X) appeared, which was specific for UV-B treatment. As a result of irradiation, the contents of UV-B absorbing and UV-B induced compounds increased in green seedlings but not in etiolated seedlings.

Rapid micropropagation of Ocimum basilicum using shoot tip explants pre-cultured in thidiazuron supplemented liquid medium

An efficient protocol has been developed for rapid micropropagation of Ocimum basilicum. Multiple shoots were induced by culturing shoot tip explants excised from mature plants on a liquid Murashige and Skoog (MS) medium supplemented with 5-100 µM of thidiazuron (TDZ) for different treatment duration (4, 8, 12 and 16 d). The optimal level of TDZ supplementation to the culture medium was 50 µM for 8 d induction period followed by subculturing in MS medium devoid of TDZ as it produced maximum regeneration frequency (78 %), mean number of shoots (11.6 ± 1.16) and shoot length (4.8 ± 0.43 cm) per explant. A culture period longer than 8 d with TDZ resulted in the formation of fasciated or distorted shoots. The regenerated shoots rooted best on MS medium containing 1.0 µM indole-3-butyric acid (IBA). The micropropagated shoots with well developed roots were successfully established in pots containing garden soil and grown in greenhouse with 95 % survival rate. The regenerated plants were morphologically uniform and exhibited similar growth characteristics and vegetative morphology to the donor plants.

Agrobacterium-mediated high frequency transformation in dwarf recalcitrant rice cultivars

The Agrobacterium-mediated transformation was done in rice (Oryza sativa L. var. indica) cv. HKR126 and elite cross-bred cv. Pusa Basmati1 (PB1), using strain LBA4404 containing pCAMBIA1300 cloned with gene cassettes; potato proteinase inhibitor and Bacillus thuringiensis endotoxin (plasmid JDW53) or mannitol-1-phosphate dehydrogenase (plasmid RKJ108). Co-cultivation with scutellar-calli derived from mature seeds showed stable and highly efficient transformation. In cvs. HKR126 and PB1, 35 % and 41 % of hygromycin resistant calli were obtained. The transformation efficiency in PB1 (22.0 %) was much higher than in HKR126 (12.5 %). Similarly, PB1 had higher plant regeneration efficiency than HKR126. The shoots regenerated per callus were, 3-4 in HKR126 and 5-6 in PB1. The transformation efficiency with pRKJ108 (18.6 %) was higher than pJDW53 (15.9 %). Polymerase chain reaction (PCR) analysis showed the presence of transgenes in regenerated transgenic plants of both cultivars.

Chitinase and peroxidase activities in sunflower hypocotyls: Effects of BTH and inoculation with Plasmopara halstedii

Systemic acquired resistance (SAR) can be induced in plants by incompatible pathogens, pathogen derived extracts, or certain chemicals as benzothiadiazole (BTH). The aim of this work was to compare changes in peroxidase and chitinase activities, enzymes considered as PR-proteins, caused by BTH and the pathogen Plasmopara halstedii. Hypocotyls from susceptible and resistant BTH-treated sunflower seedlings showed increased peroxidase and chitinase activities. Inoculation with P. halstedii increased chitinase and peroxidase activities in inoculated hypocotyls from susceptible but not from resistant sunflower seedlings.

High efficient transgenic plant regeneration from embryogenic calluses of Citrus sinensis

Transformation and high efficient regeneration of transgenic plants from embryogenic calluses of Bingtang sweet orange [Citrus sinensis (L.) Osbeck] was reported. Embryogenic calluses were inoculated with Agrobacterium tumefaciens strain EHA105, harboring the binary Ti plasmid pROK II and carrying a neomycin phosphotransferase II (NPTII) gene, an intron β-glucuronidase (GUS) gene and the Arabidopsis APETALA1 (AP1) gene. Transformation treatment was with inoculation time of 30 min, co-culture of 3 d at 23 °C and supplementation of the co-culture medium with 2 mg dm-3 acetosyringone (AS). Kanamycin (50 mg dm-3) was effective to inhibit the growth of non-transformed calluses while it did not affect the transformed ones. The total number of transformed callus lines was 7 with 100 % embryo induction. High efficient regeneration of the transgenic embryos (88 % with 4-5 shoots per embryoid) was realized within 3 months. Integration of the transgene into the citrus genome was confirmed by histochemical GUS staining, polymerase chain reaction (PCR) analysis with AP1-specific primer and Southern blot hybridization with a 712 bp PCR fragment of AP1 as the probe.

The relationship between respiration rate and peroxidase activities in maize root mitochondria

In the present study we provide the evidence of different respiration rates and peroxidase activities in maize (Zea mays L.) mitochondria isolated from germinated seeds and roots of 2-week-old seedlings. The negative relationships between mitochondrial respiration rate measured with NADH as substrate and activities of peroxidases that oxidized NADH in both oxidative and peroxidative cycles were found. The possible role of peroxidase in the regulation of reactive oxygen species metabolism in expense of NADH oxidation was hypothesized.

Localization of cadmium in the root cells of Allium cepa by energy dispersive x-ray analysis

Allium cepa L. roots were exposed to 0.1 and 1.0 mM Cd for 6, 24 and 48 h and the localization of Cd in the root tissue was investigated. Scanning electron microscopy (SEM) and energy dispersive X-ray analysis (EDXA) were performed on frozen-dried tissues of roots. No Cd was detected in the roots treated with only 0.1 mM Cd, while after exposure to higher Cd concentration (1.0 mM) Cd was observed in cell wall and in cytoplasm in the epidermis, cortex and vascular tissues in the roots.

Effects of mutagens on somatic embryogenesis and plant regeneration in groundnut

The embryogenic calli (EC) were obtained from hypocotyl explants of groundnut (Arachis hypogaea L.) cultured on Murashige and Skoog (MS) medium supplemented with different concentrations of 2,4-dichlorophenoxyacetic acid (2,4-D) in combination with 0.5 mg dm-3 6-benzylaminopurine (BAP). The EC were exposed to γ-radiation (10-50 Gy) or treated with 1-5 mM of ethyl methane sulphonate (EMS) or sodium azide (SA). The mutated EC were subcultured on embryo induction medium containing 20 mg dm-3 2,4-D. Somatic embryos (SE) developed from these calli were transferred to MS medium supplemented with BAP (2.0 mg dm-3) and 0.5 mg dm-3 2,4-D for maturation. The well-developed embryos were cultured on germination medium consisting of MS salts with 2.0 mg dm-3 BAP and 0.25 mg dm-3 naphthaleneacetic acid (NAA). Well-developed plantlets were transferred for hardening and hardened plants produced normal flowers and set viable seeds. The fresh mass of the EC, mean number of SE per explant and regeneration percentage were higher at lower concentrations of mutagens (up to 30 Gy/3 mM). Some abnormalities in regenerated plants were observed, especially variations in leaf shape.

Changes in growth, physiological and biochemical characteristics under salt stress with or without La3+ treatment in Saussurea involucrata Kar. et Kir. were investigated. The results showed that La3+ relieved the plant growth inhibition, improved the leaf water potential and water content, increased the soluble protein and the proline contents and decreased malondialdehyde content under salt stress. Further, addition of La3+ significantly increased the activities of superoxide dismutase, ascorbate peroxidase, catalase, and glutathione reductase, decreased the photosynthetic pigment decomposition and increased the ratio of total chlorophyll to carotenoids under salt stress.

Phylogenetic relationships among the bamboo species as revealed by morphological characters and polymorphism analyses

Phylogenetic relationships among 15 bamboo species were evaluated using 32 key morphological descriptors and 120 polymorphic loci of the genomic DNA generated using randomly amplified polymorphic DNA (RAPD) technique. The dendrogram generated for these two parameters based on similarity matrix computed from allelic polymorphism data using unweighted pair group method of analysis. Phylogenetic relationships as revealed from the dendrogram and principal component analysis was in concurrence with the reliable, widely referred system of bamboo classification, while, the cluster pattern generated from the similarity matrix derived from key morphological character analysis was discriminatory. The findings suggest that the molecular evidences need to be supplemented by morphological data to validate the phylogenetic relationships among taxa.

Analysis of genetic diversity among Swertia chirayita genotypes

Inter simple sequence repeat (ISSR) markers were used to analyse genetic diversity of Swertia chirayita genotypes collected from the temperate Himalayas of India. Allied species of Swertia chirayita were used in the study as outliers. Nineteen UBC primers generated a total of 315 ISSR bands, revealing 98.7 % polymorphism among the genotypes assayed. This was reduced to 42.5 % when the outliers were excluded. The results revealed a high genetic diversity within the genotypes.
