Somatic embryogenesis and plant regeneration in diploid and triploid Arachis pintoi

Plants of two cytotypes (2n=2x=20, and 2n=3x=30) of pinto peanut (Arachis pintoi Krapov. & W.C. Gregory) were regenerated through somatic embryogenesis. Embryogenic calli were induced from shoot tips or immature leaves dissected from in vitro growing plants. In the case of the diploid peanut the best somatic embryogenesis was achieved when shoot tips were cultured on Murashige and Skoog (MS) medium supplemented with 10 mg dm-3 Picloram (PIC) and 0.1 mg dm-3 6-benzylaminopurine (BAP) or when explants from immature leaves were cultured on MS + 10 mg dm-3 PIC + 0.01 mg dm-3 BAP. In the case of triploid peanut the highest number of somatic embryos was obtained when shoot tips were cultured on MS + 10 mg dm-3 PIC + 0.01 mg dm-3 BAP or when immature leaves were cultured on MS + 20 mg dm-3 PIC + 0.01 mg dm-3 BAP. Somatic embryos were converted into plants by culture on MS + 0.01 mg dm-3 naphthaleneacetic acid + 0.01 mg dm-3 BAP. Plants were successfully transferred to pots in greenhouse.

Cadmium accumulation in Medicago sativa seedlings treated with salicylic acid

Growth parameters and cadmium accumulation were investigated in alfalfa seedlings treated with 10 μM salicylic acid (SA) at the beginning of seed imbibition. Shoot and root growths were accelerated by SA treatment and suppressed by Cd both in presence and absence of SA. Cd accumulation was stimulated by SA in alfalfa seedlings in dependence of the treatment duration. K, Mg, Ca and Fe contents in roots are decreased in the presence of Cd alone, while SA induces a decrease of Mg, Ca and Fe. Shoot K, Mg and Ca concentrations are increased by Cd only in the absence of SA, while SA induces also an increase of these concentrations, but only in the absence of Cd. High negative correlation of Cd concentration with K and Ca concentrations in root indicates a competition for the same carrier not regulated by SA. Positive correlation between Cd and Mg concentrations in shoots, which is decreased by SA pre-treatment, together with the increase of positive correlation between Cd and Fe concentrations in shoots under the influence of SA, indicates a possible mechanism of SA action through maintenance of ionic homeostasis.

Peroxidase activity and lignification in soybean root growth-inhibition by juglone

The changes in activities of soluble and cell wall-bound peroxidases and lignin contents in juglone-stressed soybean (Glycine max) seedlings and their relationships with root growth were investigated. Soybean seedlings (3-d-old) were cultivated in nutrient solution supplemented with 0.5 to 25 μM juglone for 24 h. Length and dry mass of roots decreased after 5 to 25 μM juglone treatments. Low juglone concentrations (≤ 1 μM) increased soluble peroxidase activity, while high concentrations (≥ 10 μM) inhibited activities of soluble and cell wall-bound peroxidases. Juglone (≤ 1 μM) did not affect lignin content but highly increased lignification after 5 to 25 μM treatments. Results indicate that lignification may be an important step in root growth reduction of juglone-stressed soybean.

Antioxidant enzyme induction in pea plants under high irradiance

Exposure of pea plants to high irradiance (HI) for 60 min caused a reversible photoinhibition as shown by changes in variable to maximum fluorescence ratio (Fv/Fm). A significant decline in Fm was observed in leaves from both pea cultivars subjected to HI, the decrease being higher in JI281 than in JI399 plants. The values recovered during the post-stress period in both cultivars. In both cultivars, minimal fluorescence (F0) increased under HI, but in cultivar JI399 F0 recovered to initial value during the post-stress period. The expression of antioxidant enzyme genes was higher in JI399 than in JI281, both in control and stressed plants. In JI281, after 60 min of HI, an induction of the transcripts of CAT, chlMDHAR, cytAPX and cytCu,Zn-SOD was observed, whereas there was a slight increase in PHGPX, stAPX and chlCu,Zn-SOD mRNAs. After 24 h of the recovery period, the induction of some transcripts was not maintained (CAT, cytAPX and cytCu,Zn-SOD), whereas the induction of others was maintained (PHGPX and chlCu,ZnSOD) or even increased (cytGR, stAPX and chlMDHAR). In JI399, CAT and cytAPX were increased strongly after 60 min of HI, and slight increases were observed in cytGR, chlGR and chlMDHAR. In the post-stress period the expression of stAPX, cytGR and chlMDHAR was even slightly higher than after 60 min of HI, however, expression of CAT, cytAPX, cytCu,ZnSOD, chlCu,ZnSOD and chlGR decreased.

Phenolics, lignin content and peroxidase activity in Picea omorika lines

We analyzed low molecular mass phenolics, lignin content and both soluble and cell wall bound peroxidase activity in the needles of three Picea omorika (Pancic) Purkyne lines grown in the generative seed orchard. The highest values of the total phenol content as well as of catechine, caffeic acid, coniferyl alcohol, isoferulic acid and lignin concentration were detected in B5 line ("semidichotomy" line). The soluble guaiacol peroxidase activity was the highest in A3 line (line "borealis"). The highest activity of cell wall bound peroxidases was measured in B5 line, and it was in correlation with lignin content.

Effect of chilling on growth and nitrogen assimilation in Azolla caroliniana

Azolla caroliniana was exposed to 5 °C in darkness for 1, 2, 3, 5 or 7 d and then recovered for 7 d. Plants previously chilled for 2 or 3 d exhibited higher growth rates when transferred to normal temperature than either the control plants or those previously chilled for 5 or 7 d. Increased plant growth may be related to increased contents of chlorophyll, sucrose, and reducing sugars, due to increased photosynthetic capacity. In another experiment Azolla plants were chilled at 5 °C for 7 d and then transferred for 0, 4, 8, 12, or 16 d recovery to the N-free Hoagland solution or Hoagland solution containing 5 mM KNO3. In previously chilled plants, the growth rate was decreased. In the medium supplemented with nitrogen, the growth rate was greater than in the N-free medium in both chilled and nonchilled plants. In chilled plants the decrease in growth rate may be related to the disturbance of Anabaena azollae cells where the protecting envelope of the heterocysts was deorganized. During the recovery the rate of N2-fixation increased in both chilled and nonchilled plants up to 12 d after which both rates were similar. However, during the first 4 d the rate of the nonchilled plants was approximately 4-fold that of the previously chilled plants. Nitrate reductase and nitrite reductase activities in control plants were higher than in those previously chilled for 7 d. Both activities increased in nonchilled and previously chilled plants up to 12 d then decreased. The total protein content increased up to 12 d in chilled and nonchilled plants after which it decreased. Under all treatments, the values were higher in nonchilled plants than in those previously chilled ones and were also higher in presence of N than in its absence. Thus the presence of N-source in the medium counteracts the effect of chilling injury particularly during prolonged recovery.

Establishment of Camptotheca acuminata regeneration from leaf explants

Plantlet regeneration through shoot formation from young leaf explant-derived callus of Camptotheca acuminata is described. Calli were obtained by placing leaf explants on Woody plant medium (WPM) supplemented with various concentrations of 6-benzyladenine (BA) and naphthaleneacetic acid (NAA) or 2,4-dichlorophenoxyacetic acid (2,4-D). Callus induction was observed in all media evaluated. On the shoot induction medium, the callus induced on the WPM medium containing 19.8 μM BA and 5.8 μM NAA was the most effective, providing high shoot regeneration frequency (70.3 %) as well as the highest number of shoots (11.2 shoots explant-1). The good rooting percentage and root quality (98 %, 5.9 roots shoot-1) were achieved on WPM medium supplemented with 9.6 μM indole-3-butyric acid (IBA). 96 % of the in vitro rooted plantlets with well developed shoots and roots survived transfer to soil.

Comparison of induction frequency, maturation capacity and germination of Abies numidica during secondary somatic embryogenesis

Efficiency of the method for improving repetitive somatic embryogenesis and plant recovery of Algerian fir (Abies numidica De Lann.) was investigated by evaluating of induction frequency, maturation capacity and germination. Individual zygotic embryos differed only slightly in induction frequencies (6.8 %) from somatic embryos of the first (5.7 %) and second cycle (5.5-9.0 %). The yield of mature embryos differed significantly among the cell lines of the same cycle and among cell lines of the different cycles. Percentage of abnormalities was lowest in the first cycle of somatic embryos, whereas the second and the third cycles of somatic embryos were branded by a higher frequency of abnormalities. The differences in germination of well developed somatic embryos depended on cell lines rather than on cycle of somatic embryos.

Direct somatic embryogenesis and plant regeneration from ray florets of chrysanthemum

Direct somatic embryogenesis from ray floret explants of five chrysanthemum cultivars has been obtained within 12 - 15 d on Murashige and Skoog medium supplemented with 2,4-dichlorophenoxyacetic acid (2,4-D) and 6-benzyladenine (BA). Scanning electron microscopic observation also confirmed the direct origin of somatic embryos from explants. Somatic embryos developed asynchronously on the adaxial surface of explants. Among the five cultivars tested, Birbal Sahani was best responding (40 % explants responded on 4 mg dm-3 2,4-D and 2 mg dm-3 BA supplemented medium). Precocious germination of somatic embryos was noticed on the same medium. The best sucrose concentration in the medium was found to be 60 g dm-3 where 70 % explants responded while 55 % embryogenic response was obtained on medium supplemented with 400 mg dm-3 inositol. Plants developed from somatic embryos were transferred to soil and produced true-to-type flowers.

Extent of ipt gene expression and resulting amount of cytokinins affect activities of carboxylation enzymes in transgenic plants

Three types of transgenic plants of Solanum tuberosum cvs. Kamyk and Oreb, and Nicotiana tabacum cvs. Maryland Mammoth and Trapezond were selected according to intensity of introduced ipt gene expression and resulting amount of synthesised cytokinins (CKs). In comparison with controls, original transgenic regenerants grown in vitro showed a massive increase of CK contents, in tobacco by 379 % and in potato by 159 % (MAS). Potato grown in soil from tubers of transgenic plants demonstrated a moderate increase (44 %) of CK contents (MOD). Transgenic tobacco grown from seeds in vitro did not show any significant change in CK contents (NOT). Initial (RuBPCi and RuBPOi) and total (RuBPCt) activities of ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO), and the activity of phosphoenolpyruvate carboxylase (PEPC) were not significantly affected by the transformation in the NOT plants. In the MOD plants, the RuBPCO activities were stimulated by up to 34 % whereas the PEPC activity was decreased by 17 %. On the other hand, all the measured enzyme activities were 32 - 91 % lower in the MAS. Leaf area, fresh and dry masses, and chlorophyll and soluble protein contents also went down with increasing CK amounts in the transformants. Dependence of RuBPCi/RuBPOi and RuBPCt/PEPC ratios on the relative CK amounts in transgenic plants revealed that the individual enzyme activities were not affected uniformly. Endogenous CK contents in the MAS thus apparently exceeded an optimum needed for positive effects on many physiological traits and became a stress factor for such plants.

Changes in protein profile of pigeonpea genotypes in response to NaCl and boron stress

Two pigeonpea [Cajanus cajan (L.) Millsp.] genotypes, a salt tolerant Manak and a salt sensitive ICPL 88039 were subjected to stress treatment of 3 mM boron, 60 mM NaCl and boron + NaCl at the seedling stage. Radicle and plumule proteins were analyzed by SDS-PAGE. Boron treatment increased 28.3 kDa proteins in plumule and 38.3 and 51.9 kDa proteins in radicle of Manak, however, there was no specific protein in ICPL 88039 either in plumule or in radicle. In NaCl treatment 95.6 kDa proteins appeared in plumule and 67.5 kDa proteins in radicle of Manak. Conversely content of some proteins decreased by boron treatment alone or in combination with NaCl although they were present in the controls. Thus, 54.3 kDa protein disappeared in ICPL 88039 plumule, 68.4 kDa in Manak radicle and 28.1 kDa in ICPL 88039 radicle.

Intraspecific interactions in Arabidopsis thaliana and the stomatal mutants tmm1-1 and sdd1-2

Competition is a major density-dependent factor structuring plant populations and communities in both natural and agricultural systems. Seedlings of the model plant species Arabidopsis thaliana cv. Columbia, and the Columbia-derived stomatal mutants sdd1 and tmm1, were grown under controlled conditions at increasing densities of 1, 10, 20, and 50 plants per pot. We demonstrate significant effects of time (days after planting), density, genotype, density and genotype, and the three-way interaction with time upon several fitness components (plant height, silique number, leaf biomass and flowering stalk biomass) in Columbia and these mutants.

Optimization of primer screening for evaluation of genetic relationship in rose cultivars

Optimization of primer screening for evaluation of genetic relationship in 34 cultivars of rose through random amplified polymorphic DNA (RAPD) markers was investigated. Four series of decamer primers were used for screening and optimization of RAPD analysis between which A and N series performed good amplification of fragments as compared with other series. The primers OPN-07 and OPN-15 produced maximum number of DNA fragments in Rosa hybrida cv. Anuraag. Some primer either did not produce amplification or produced very poor amplification. Further, ten selected primers were used for genetic analysis of 34 rose cultivars. The primer OPN-15 amplified 21 fragments in all cultivars tested. A total of 162 distinct DNA fragments (bands) ranging from 100 to 3400 base pairs were amplified by using 10 selected random primers. The cluster analysis indicated that these rose cultivars formed nine clusters.

Molecular cloning and expression analyses of a new gene encoding 3-hydroxy-3-methylglutaryl-CoA synthase from Taxus × media

A new full-length cDNA encoding 3-hydroxy-3-methylglutaryl-CoA synthase (designated as TmHMGS, GenBank Accession No. AY644708), which catalyses the condensation of acetyl CoA and acetoacetyl CoA to form 3-hydroxy-3-methylglutaryl-CoA as an early step in the taxol biosynthetic pathway, was isolated from young leaves of Taxus × media by rapid amplification of cDNA ends (RACE) for the first time. The full-length cDNA of TmHMGS contained a 1431 bp open reading frame (ORF) encoding a deduced protein of 476 amino acid residues. The deduced protein had an isoelectric point of 5.23 and a calculated molecular mass of about 53 kDa. Amino acid sequence comparison analysis showed that TmHMGS had high similarity with a number of HMGSs ranging from Schizosaccharomyces pombe to humans, with much higher identity with other HMGSs from plants than those from yeast and humans. Phylogenic analysis showed that TmHMGS had closest relationship with HMGS from Pinus sylvestris. Tissue expression pattern analysis showed that TmHMGS expressed in needles and stems at similar level, but no expression could be detected in roots. Expression of TmHMGS was all induced by under different elicitors such as silver nitrate, ammonium ceric sulphate and methyl jasmonate, revealed that TmHMGS was an elicitor-responsive gene.

Anion effects on the structural organization of spinach thylakoid membranes

Changes in the conformation of spinach thylakoid membranes were monitored in 5-doxyl stearic acid (SAL)-treated thylakoid membranes in the presence of various anions (Cl-, Br-, I-, NO2-, SO42-, PO43-). The presence of anions made the thylakoid membrane more fluid. The extent of change in membrane fluidity differed with different anion and was reversible.

Agrobacterium rhizogenes-mediated transformation of Brassica oleracea var. sabauda and B. oleracea var. capitata

Agrobacterium rhizogenes A4M70GUS-mediated transformation of Savoy cabbage (Brassica oleracea L. var. sabauda) and two local lines of cabbage (B. oleracea L. var. capitata) was obtained using hypocotyl and cotyledon explants. The percentage of explants which formed roots was very high in all genotypes: 92.3 % in Savoy Gg-1, 64.4 % in cabbage P22I5, and 87.2 % in P34I5. Spontaneous shoot regeneration of excised root cultures grown on the hormone-free medium occurred in all three genotypes. In cabbage lines P22I5 and P34I5 shoot regeneration was higher (9.3 and 2.6 % respectively) than in Savoy cabbage Gg-1 (1.3 %). Transgenic nature of hairy root-derived plants was evaluated by GUS histological test and PCR analysis. All the tested cabbage shoots were GUS positive whilst in a Savoy cabbage GUS expression was registered only in 55 % of tested clones. PCR analysis demonstrated the presence of the GUS gene in regenerated shoot clones and in T1 progeny.

Water relations in Norway spruce trees growing at ambient and elevated CO2 concentrations

Water relations were studied in Norway spruce [Picea abies (L.) Karst.] trees grown at ambient (AC, 350 μmol mol-1) and elevated (EC, 700 μmol mol-1) CO2 concentrations under temperate water stress. The results suggested that both crown position and variability in atmospheric CO2 concentration are responsible for different patterns of crown water relations. Mean hourly sap flux density (FSA) showed higher values in upper crown position in comparison with the whole crown in both AC and EC treatments. Mean soil-to-leaf hydraulic conductance (GTsa) was 1.4 times higher for the upper crown than that calculated across the whole crown for the trees in AC. However, GTsa did not vary significantly with crown position in EC trees, suggesting that elevated CO2 may mitigate differences in hydraulic supply for different canopy layers. The trees in EC treatment exhibited significantly higher values of FSA measured on the whole crown level and slightly higher soil water content compared to AC treatment, suggesting more economical use of soil water and therefore an advantage under water-limited conditions.

Gender-specific responses of Piper betle L. to low temperature stress: changes in chlorophyllase activity

Gender based differences in response to low temperature stress in leaf chlorophyll (Chl), and carotenoids (Car) contents and chlorophyllase (Chlase) activity were monitored in male (Kapoori Vellaikodi and Madras Pan Kapoori) and female (Bangla Mahoba, Desi Bangla and Kaker) betel vine landraces. Although female plants contained nearly two fold more Chl than male counterparts, the low temperature induced Chl loss was comparable, however, male plants showed higher Chl a/b ratio than females. Chlase activity increased due to cold stress in all the landraces. Male plants always showed higher activities of Chlase, which may be one of the reasons for the rather low Chl contents in male plants.

Shoot and root culture of Hypericum perforatum L. transformed with Agrobacterium rhizogenes A4M70GUS

Hairy root cultures of Hypericum perforatum were obtained following inoculation of aseptically germinated seedlings with A. rhizogenes strain A4M70GUS. Effect of sucrose on the growth and biomass production of hairy root cultures was investigated. Hairy root cultures spontaneously regenerated shoots buds from which a number of shoot culture clones was established. Transformed shoot cultures exhibited good shoot multiplication, elongation and rooting on a hormone-free woody plant medium. Plants regenerated from hairy roots were similar in appearance to the normal, nontransformed plants.

Effects of medium composition and culture duration on in vitro morphogenesis of sweet potato

In vitro morphogenesis of sweet potato (Ipomoea batatas) shoot explants after cultures in callus initiation medium (CIM) with two sucrose contents and plant regeneration medium (PRM) with three growth regulator combinations for different durations was studied. After 4 weeks, explants on 5 % sucrose CIM had significantly more shoots but similar or lower root fresh mass and callus fresh mass than those on 3 % sucrose CIM subsequent to transfer for 6 weeks on all three PRM. Cultures transferred to growth regulator-free PRM after 4 and 12 weeks on 5 % sucrose CIM formed plants through organogenesis and embryogenesis, respectively. Embryogenic cultures from 4 weeks on CIM + 10 weeks on callus proliferation medium when transferred to PRM without growth regulator for 4 and 8 weeks produced multiple embryos in the prior and both embryos and shoot buds in the later.

Thidiazuron stimulates adventitious shoot regeneration in different safflower explants

Adventitious shoot regeneration from root, hypocotyl, cotyledon and primary leaf explants of safflower (Carthamus tinctorius L.) was studied. Shoot regeneration was promoted by benzyladenine (BA) + naphthaleneacetic acid (NAA), BA + indole-3-butyric acid (IBA), kinetin + NAA and thidiazuron (TDZ) + NAA incorporated in Murashige and Skoog (MS) basal medium. High frequency of shoot regeneration and high number of shoots per regenerating explant were obtained on a wide range of TDZ + NAA combinations. Proliferated shoots were elongated in MS + 0.5 mg dm-3 kinetin and well-developed shoots were rooted in half strength MS + 0.5 mg dm-3 NAA. Rooted shoots were successfully acclimatized and established in soil.

Stomatal characteristics during micropropagation of Wrightia tomentosa

A deviation from usually found characteristics of stomata in Wrightia tomentosa was noted during in vitro propagation. Increase in stomatal frequency in leaves of plants grown in vitro was observed with 29.4 % malformed stomata. The stomata were spherical, wide open, did not close in detached leaves even after 3 h. The leaves exhibited 93.4 % total water loss during 3-h period. Stomatal frequency, percentage of malformed stomata and rate of water loss declined in subsequent rooting phase. Nevertheless, for high survival rate plantlets were hardened under gradually decreasing air humidity either in partially opened glass bottles containing Soilrite™ moistened with 1/4 Murashige and Skoog nutrients or in pots covered with polyethylene bags. The stomatal characteristics of hardened plants were comparable to seedlings. Survival rate was more than 95 %.

Histological characterization of in vitro adventitious organogenesis in Citrus sinensis

The adventitious bud development was induced in epicotyl segments of Valencia sweet orange (Citrus sinensis L. Osbeck). Seeds were cultured in vitro for three weeks in the dark, followed by one week at a 16-h photoperiod. Epicotyl segments were cultured horizontally for the induction of organogenesis in Murashige and Tucker (1969, MT) culture medium supplemented with 1.0 mg dm-3 benzylaminopurine. Samples were observed by light and scanning electron microscopy from day zero to day 25, when buds were well grown. It was shown that the adventitious buds originated directly from the cambial region on the cut ends of the explants.

Influence of CCC, putrescine and gellam gum concentration on gynogenic embryo induction in Allium cepa

The induction of haploid plants by in vitro gynogenesis is a promising practice in onion breeding. In order to increase the frequency of embryo regeneration and haploid plant production in Valcatorce INTA, Cobriza INTA and Navidena INTA cultivars, putrescine and CCC were used, either as a component of the culture media or by spraying or injecting them to the umbels. Additionally, two concentration of gellam gum were tested. A higher number of gynogenic embryos was achieved by using 7 g dm-3 gellam gum, and this number was not affected by the addition of putrescine to the media. CCC sprayed at the umbels significantly increased the gynogenic embryo rate, which was more than three times higher than the control. Cobriza INTA showed the highest induced embryo rate (4.76 %).

Micropropagation of Coleus blumei from nodal segments and shoot tips

A rapid and highly-effective method for micropropagation from nodal segment and shoot tip explants was established for Coleus blumei Benth. Nodal segments and shoot tips were inoculated on MS medium containing 0.7 % agar, 3 % commercial sugar, and different combinations of 6-benzyladenine (BA) with indole-3-acetic acid (IAA), indole-3-butyric acid (IBA) or α-naphthaleneacetic acid (NAA). Hundred percent shoot induction from both explants was achieved on the medium containing BA (2 mg dm-3) and NAA (1 mg dm-3). Shoot tips were proved to be the better explant in comparison to nodal segments in having high rate of shoot induction and more number of shoots. The same media conditions were found suitable for shoot multiplication. Multiplied shoots rooted best on MS medium supplemented with IBA (2 mg dm-3). Micropropagated plants were successfully established in soil after hardening, with 100 % survival rate.

Morphological, histochemical and ultrastructural indicators of maize and barley leaf senescence

In this study we report on morphological and histochemical indicators of maize and barley leaf senescence. We determined how the traits such as distribution of stomata and hairs, presence of epicuticular wax, staining of tissues with toluidine blue, change with leaf age and within the leaf blade. We identified regions of young, non-mature leaves as exhibiting juvenile phase, regions with features typical for mature and fully differentiated leaves-as an adult phase and regions with traits of age damage as a senescing phase. Ultrastructural analysis of these regions of leaves gives a clear picture of the time development of the senescence process. Appearance of morphological and histological indicators of senescence in certain regions of leaf is correlated with ultrastructural changes of mesophyll cells of the same regions. We have thus found a relatively simple method of estimating the stage of senescence both in maize and barley.

Generation of white mold disease-resistant sunflower plants expressing human lysozyme gene

Sunflower plants were transformed via co-cultivation of previously bombarded hypocotyl explants with Agrobacterium tumefaciens harboring the plasmid pNGL that contains the human lysozyme gene. The transformed shoots were selected using kanamycin and regenerated plants were analyzed using histochemical β-glucuronidase assay. Southern, Western and Northern blot analyses indicated the transfer, expression and stable integration of the foreign DNA into the sunflower genome. Resistance against the phytopathogenic fungus Sclerotinia sclerotiorum, which causes white mold disease, was confirmed using a phytopathogenic test and microscopic observation of the infection process.

The influence of ammoniates on 14CO2 assimilation in flax

A 1 μM solution of ammoniates [ZnCu(NH3)n]2+(CO3)2- was inserted into a cut shoot of flax with the transpiration stream of water. Analysis of the 14C content after 14CO2 assimilation by the shoot showed that ammoniates increased radioactive label contents in the tissues (especially in the young leaves and stem). In the leaves the higher sucrose to hexoses ratio, an increased radioactivity of glycerate and malate and decreased incorporation of 14C into oligosaccharides and pigments were observed. These effects were more pronounced in the young leaves. Spraying of plants with 20 mM solution resulted in an increase of plant height and leaf number.

Water transport in maize roots under the influence of mercuric chloride and water stress: a role of water channels

The influence of inhibitor of water channels, HgCl2, on water diffusion in maize (Zea mays L.) seedling roots was investigated with the pulsed nuclear magnetic resonance (NMR) method. Blocking of water channels decreased the water permeability of cell membranes by 1.5 - 2 times. This effect of HgCl2 was exhibited only in the roots of seedlings grown in a nutrient solution containing Ca2+ and was reversed with Hg-scavenging agent β-mercaptoethanol. Subsequent incubation of Ca2+-deficient roots in the nutrient solution with Ca2+ recovered the sensitivity to HgCl2. The water stress decreased water diffusion rates similarly to HgCl2 and the effects of water stress and HgCl2 were not additive. The obtained data demonstrate the possibilities of the pulsed NMR method for study of the transmembrane water exchange in vivo in connection with water channel functioning.
