Photosynthetic Traits in Wheat Grown under Decreased and Increased CO2 Concentration, and after Transfer to Natural CO2 concentration

Wheat plants were grown from sowing to day 18 in 26-dm3 chambers at three different CO2 concentrations: 150 (-CO2), 350 (C, control), 800 (+CO2) μmol mol-1. Afterwards, plants of the three variants were grown at the same natural CO2 concentration. Plant characteristics were measured just before the transfer (0 days after CO2 treatment, DAT), and at 5 - 8 DAT on the 1st leaf, and at 12 - 22 DAT on the 4th leaf. Decreased or increased CO2 concentrations caused acclimations which persisted after transplantation to natural CO2 concentration. At 5 - 8 DAT, stomatal density, stomatal conductance (gs), CO2 saturated net photosynthetic rate (PNsat0), radiation saturated net photosynthetic rate (PNsat1), and carboxylation efficiency (τ) were higher in -CO2 plants and lower in +CO2 plants than in C plants. As compared with C plants, the photochemical efficiency (α) was lower in -CO2 and higher in -CO2 plants, however, chlorophyll (Chl) a, Chl b, Chl a-b and carotenoid contents were lower in both -CO2 and +CO2 plants. On the 4th leaf, which emerged on plant after finishing CO2 treatments, at 12 - 22 DAT, no differences in stomatal density and g, between treatments were observed. In -CO2 plants, pigment content and PNsat0 were higher, α was lower, and PNsat1 and τ were not different from C plants. In contrast, in +CO2 plants, pigment content, PNsat1 and τ were lower, and PNsat0 and α were unchanged. Leaf area, dry mass, and tiller development increased in +CO2 plants and decreased in -CO2 plants. In the interval between 8 and 22 DAT, lower net assimilation rate in +CO2 than in -CO2 plants was observed.

Proline, Ornithine, Arginine and Glutamic Acid Contents in Detached Rice Leaves

The effects of water stress on the contents of proline, ornithine, arginine and glutamic acid in detached rice leaves were examined. In water stressed leaves, the content of proline was elevated to a content approximately 8-, 14- and 17-fold higher than in control leaves after 4, 8 and 12 h, respectively. We also observed that omithine and arginine contents were much higher under water stress than in control leaves. However, the content of glutamic acid in water stressed leaves was higher after 4 and 8 h and lower after 12 h than that in control leaves.

Induction of Oxidative Stress and Antioxidant Activity by Hydrogen Peroxide Treatment in Tolerant and Susceptible Wheat Genotypes

We induced an oxidative stress by means of exogenous hydrogen peroxide in two wheat genotypes, C 306 (tolerant to water stress) and Hira (susceptible to water stress), and investigated oxidative injury and changes in antioxidant enzymes activity. H2O2 treatment caused chlorophyll degradation, lipid peroxidation, decreased membrane stability and activity of nitrate reductase. Hydrogen peroxide increased the activity of antioxidant enzymes, glutathione reductase and catalase. These effects increased with increasing H2O2 concentrations. However, no change was observed in the activity of superoxide dismutase and proline accumulation.

Nodule Functioning in Trifoliate and Pentafoliate Mungbean Genotypes as Influenced by Salinity

Two genotypes of mungbean differing in the leaf shape, K-851 (trifoliate) and a mutant (pentafoliate), were exposed to salinity (0, 2.5, 5, 10 dS m-1) for 3, 6 and 9 d at flowering stage (40 - 45 d after sowing) to see the effect of salinity on nodule functioning. In both the genotypes, osmotic potential (ψs) and relative water content (RWC) of nodules decreased significantly, and proline content increased with increasing the salinity. The ψs of nodules was more decreased in the mutant than in genotype K-851, while reverse was true for RWC and proline accumulation. A remarkable increase in ethylene evolution was noticed from nodulated roots with the increasing level and duration of salinity and was much higher in K-851. A sharp decline in leghemoglobin content and acetylene reduction assay (ARA) of the nodules was observed with the salinity and this decrease was more marked in K-851 than the mutant. N content declined while Na+/K+ ratio and Cl- content increased significantly. The mutant maintained better N status but lower Na+/K+ ratio and Cl- content in nodules than K-851. Nodule dry matter also declined with salinity and the decrease was more expressed in K-851. Thus the functioning of nodules in the mutant was better than in the genotype K-851 under stress conditions.

Specifity of Genetic Determination of Content of Photosynthetic Pigments in Triticale

Lines of winter hexaploid Triticale and their F1 and F2 hybrids differing in morphological structure, pigment contents, photosynthetic productivity, and grain crops were studied. F1 hybrids received by crossing of Triticale lines contrasting in pigment contents showed in some cases a heterosis effect for chlorophyll (Chl) content per unit leaf area. Variation analysis demonstrated a polygenic control of Triticale pigment contents, and different rate of increase in F2 generation. We found maternal type of heritability of Chl b content and Chl content in light-harvesting complex of photosystem 2.

Influence of Cadmium and Zinc on Growth and Photosynthesis of Bacopa monniera Cultivated in vitro

This study evaluates the impact of cadmium and zinc interaction on the amount of soluble proteins, CO2 fixati stomatal conductance and intercellular CO2 contents in regenerants of B. monniera. The regenerants were grown 16 weeks on MS medium containing cadmium and zinc in various concentrations. Cadmium decreased the stom conductance, photosynthetic rate and root growth but increased the protein content. Additional supply of zinc in medium reduced the adverse effects of cadmium on these parameters.

Molecular Characterization of a Hybrid Zone between Orchis mascula and O. pauciflora in Southern Italy

A small population (16 individuals) of Orchis × colemanii Cortesi, a natural hybrid between O. mascula (L.) L. and O. pauciflora Ten. was sampled from a locality near Sassano (National Park of Cilento, province of Salerno, Southern Italy) in order to assess its genetic structure. Filter hybridizations of O. × colemanii, O. mascula and O. pauciflora DNAs against a PCR-amplified ribosomal fragment indicated that all the hybrid individuals in study have the ribosomal repeats of both parents, even if not always combined in equal proportions, and that no specimen morphologically identified as O. mascula or O. pauciflora possessed detectable rDNA of the other species. The majority of the hybrid specimens (12 individuals) showed a preponderance of O. mascula rDNA, three had approximately equal proportions of parental repeats and one had a preponderance of O. pauciflora rDNA. As the parental species have different amounts of rDNA, as inferred from dot blot hybridization, it has been deduced that the 12 specimens may represent F1 or successive, presumably non-recombinant progenies, while the last four specimens may represent either F2 genotypes or backcrosses with O. pauciflora.

Differential Effect of Cd2+ and Ni2+ on Amino Acid Metabolism in Soybean Seedlings

In 10-d-old soybean seedlings, the growth of roots and shoots was significantly inhibited at 50 and 100 μM and more Cd2+, respectively, and by 50 μM or more Ni2+. Although total protein content of roots exposed to 200 μM Cd2+ or Ni2+ was similarly decreased compared to the control, the activity of nitrate reductase was much more inhibited by Cd2+. Ni2+-treatment (200 μM) induced an accumulation of all free amino acids in roots associated with a decrease in alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities reflecting the accumulation of both alanine and aspartic acid, respectively. Cd2+-treatment (200 μM) decreased the amount of all free amino acids. In addition, cysteine which is the main amino acid consisting the phytochelatin complexes constituted about 17.5 % of total free amino acids. The activities of both ALT and AST in Cd2+-treated roots were higher than in Ni2+-treated roots suggesting higher conversion of alanine and aspartate to pyruvate and oxaloacetate. Primary leaves excised from either Cd2+ or Ni2+-treated seedlings showed similar pattern of enzyme activities as roots.

Primer Design and Optimization for RAPD Analysis of Nepenthes

Primers with higher G+C content produced better random amplified polymorphic DNA (RAPD) profiles in Nepenthes. The occurrence of clustered G's and C's in the center of the primer seemed also to influence the banding patterns. It was also observed that for certain polymerases, the use of different buffers other than that recommended by the manufacturer provided a better amplification profile for Nepenthes.

In vitro Growth and Leaf Composition of Grapevine Cultivars as Affected by Sodium Chloride

In vitro proliferated shoot culture of six grape genotypes (Vitis vinifera L.) were screened for tolerance to NaCl (0 to 200 mM). The cv. Perlette was found to tolerate 175 mM NaCl followed by cvs. Pusa Seedless and Beauty Seedless 150 mM NaCl. Na, K, Cl, Ca and Mg content increased upto 100 mM NaCl in most of the genotypes. Total sugar and proline content of stem tissue gradually increased under NaCl stress while leaf chlorophyll a+b content declined. Studies suggest that the in vitro screening procedure can be used for ranking the grape genotypes for salinity tolerance.

Relationship between Calcium and Pyruvate Kinase

Tobacco plants (Nicotiana tabacum L. cv. Sevilla) were grown under controlled conditions. The leaf content of Ca2+, Mg2+ and K+, and the activity of the pyruvate kinase were analyzed. The increased application of Ca2+ diminished the content of K+ and Mg2+ in leaves, and decreased the activity of pyruvate kinase. Taking into account these results, we suggest the pyruvate kinase activity as an bioindicator of the contents of the Ca2+, Mg2+ and K+.

Effect of Increasing Concentrations of Lead and Cadmium on Cucumber Seedlings

The effect of lead and cadmium on biomass accumulation of cucumber seedlings (Cucumis sativus L.) as well as the contents of abscisic acid (ABA), free proline and soluble proteins in leaves were studied. Seedlings were subjected to lead nitrate or cadmium bromide in low concentrations (1 - 5 µM) for 1, 4 or 7 d, and then to the action of the same substances in high concentrations (500 - 1000 µM). The pretreatments of the seedlings with heavy metals in low concentrations enabled them to tolerate the subsequent high concentrations of cadmium and lead without injury. The plant responses to heavy metal treatment were accompanied by the accumulation of ABA, free proline and soluble proteins in leaf tissues.

Leaf Structure of Tobacco In Vitro Grown Plantlets as Affected by Saccharose and Irradiance

Tobacco plantlets were cultured in vitro under high (200 µmol m-2 s-1) or low (60 µmol m-2 s-1) irradiance with or without saccharose in the medium. Light microscopy and image analysis were used to evaluate the effect of these culture conditions on leaf anatomy. Addition of saccharose resulted in thicker leaves (all leaf layers) and larger mesophyll cells under both growth irradiances. Various irradiance affected leaf anatomy differently when plantlets had been cultivated in presence or absence of saccharose in the medium. While under high irradiance in presence of saccharose leaf thickness and number of chloroplasts per cell section were increased, plantlets grown under high irradiance in absence of saccharose had thinner leaves and less chloroplasts per cell section. The changes were more pronounced in palisade parenchyma layer.

Dynamics of Seed Protein Biosynthesis in Two Soybean Genotypes Differing in Drought Susceptibility

The dynamics of seed storage protein biosynthesis was studied under field conditions during two vegetative seasons. Two soybean (Glycine max L. Merr.) genotypes were examined: BOSA (drought tolerant) and L 121 (drought susceptible). Seed samples were taken from plants at three stages of seed maturation (50 and 70 d after flowering, and at full maturity). The earlier synthesis of the β-subunit of the 7S protein occurred in the drought susceptible cultivar. We have not found such differences in the synthesis of the α- and α'-subunits of the 7S protein. Our results did not confirm significant genotypic differences in protein composition of the mature seeds between the cultivars studied, but have pointed out to the differences in the dynamics of protein biosynthesis during seed maturation and desiccation.

Karyotype Analysis of Hypericum Perforatum L.

A karyotype study was made on Hypericum perforatum using plants differentiated in vitro with different ploidy level. The chromosomes of this species are small, morphologically similar, median and submedian. In the basic chromosome set the most distinguishable is chromosome number 1 which was subjected to detail analysis. It was found that there are two types of this chromosome which contribute differentially in diploid, triploid and tetraploid plants.

The Development and a Cytogenetic Study of Monosomics of Gossypium Hirsutum L.

Monosomics of cotton (Gossypium hirsutum L.) were obtained by irradiation of pollen by γ-rays and by irradiation of seeds by thermal neutrons. Many monosomics were derived directly from irradiation, but a number of monosomics were also recovered in the progeny of plants with translocations and of desynaptic plants. Only 28 primary monosomics showed normal pairing at metaphase-1 of meiosis. The others formec rare trivalents or additional univalents. Partial desynapsis was detected in some monosomics. The pollen fertility levels of monosomics are presented. New morphological characters were detected among the monosome plants of cotton.

Nodule Structure and Functioning in Chickpea (Cicer Arietinum) as Affected by Salt Stress

Cicer arietinum L. plants raised in sand culture under natural light were subjected to salinity stress induced by mixture of NaCl, CaCl2, MgCl2 and MgSO4 (40, 60 or 80 meq dm-3). Acetylene reduction activity (ARA) of nodules, leghemoglobin content and nodule structure were followed 55, 75 and 85 d after sowing. ARA declined significantly under salt treatments and the lowest ARA was observed at day 85 after sowing. Decrease in ARA was consistent with decreased nodule leghemoglobin content. The leghemoglobin content of control plants decreased by 50 % at day 85 indicating senescence of nodules. This senescence was further accelerated by salt treatment after which the leghemoglobin content fell to negligible levels. The structural changes associated with salt stress were mainly reduction in size of the nodules, decreased meristematic zone, reduced number and degradation of symbiosomes, reduced intercellalar spaces and deposition of electron dense material in the intercellular spaces in the cortex of nodules.

Linkage Mapping of DNA Markers Generated with Specific and Non-Specific Gene Primers in Soybean

Polymerase chain reaction (PCR) has been used extensively in the construction of linkage maps for many cultivated crops including soybean, [Glycine max (L.) Merr]. In this study, four sets of oligonucleotide primer pairs of known genes (pearl millet Adh 1, nodule specific proline-rich protein, Drosophila homeobox, heat shock protein), several different combinations from kits A, D, E, and J of arbitrary primers and five primer pairs of soybean simple sequence repeats of varying length (Satt 9, Satt 20, Satt 42, Satt 64, and Satt 30) were utilized in PCR to identify molecular markers which were then used to construct a genetic linkage map. DNA for the PCR reactions was isolated from 65 recombinant inbred soybean lines resulting from crossing PI 290,136 and BARC-2 (Rj 4 ), followed by self-pollination for seven generations without selection. Mapmaker 3.0, a computer package, was used for construction of the linkage map. A total of 43 polymorphic markers were identified; 30 markers were linked and distributed among 5 linkage groups while 13 markers were unlinked. Arbitrary primers revealed more polymorphisms than specific primers. A combination of arbitrary primers A5 and A18 revealed the maximum number of polymorphic bands. Five observed linkage groups can be expanded in future soybean research by using additional markers.

Amelioration of NaCl Stress by Arginine in Rice Seedlings: Changes in Endogenous Polyamines

Effects of NaCl (0.1 - 0.2 M) alone or in combination with 1 mM arginine on growth and endogenous polyamine (PA) content have been observed in two cultivars of rice differing in NaCl stress tolerance. The germination, seedlings fresh mass and water content decreased with increase in salinity in both the cultivars. This inhibition was partially alleviated by application of arginine. Cv. CSR-27 exhibited relatively better germination than cv. Bas-370 at different salinities. Total PA content increased in both the cultivars under NaCl stress alone and in combination with arginine. Putrescine to spermidine and spermine ratio was higher in NaCl-treated seedlings being more in cv. Bas-370 as compared to cv. CSR-27 and the ratio reversed to almost control level when arginine was applied along with NaCl.

Pigment Diverse Mutants of Pseudomonas sp.: Inhibition of Fungal Growth and Stimulation of Growth of Cicer arietinum

A Pseudomonas strain MRS16 inhibited growth of different pathogenic fungi (Aspergillus sp., Fusarium oxysporum, Pythium aphanidermatum and Rhizoctonia solani) in vitro. Larger inhibition zones were obtained on nutrient agar and King's B media compared to potato dextrose agar and pigment production media. Mutants altered in production of fluorescent pigment were derived by nitrosoguanidine mutagenesis. The pigment overproducer mutant MRS16M-1 was more inhibitory whereas nonproducer mutant MRS16M-5 was less inhibitory than parent strain on nutrient agar medium. Addition of iron (100 µM FeCl3) in the medium decreased inhibition of fungal growth, suggesting the involvement of siderophores and other antifungal secondary metabolites. Seed bacterization of two cultivars of chickpea (Cicer arietinum cvs. H8618 and C235) differing in susceptibility to wilt caused initial root and shoot stunting at 5 d of growth followed by proliferation of secondary root growth at 10 d. Coinoculation of chickpea with Pseudomonas strain MRS16 or mutants and Rhizobium sp. Cicer strain Ca181 enhanced nodulation, nitrogen fixation and plant dry mass as compared to single inoculation with Rhizobium strain under sterile conditions.

Effects of Some Growth Regulators on Young Iron Deficient Maize Plants

Young maize plants, grown hydroponically, were supplied with 1/10 the optimal amount of iron (0.75 mg dm-3). Foliar treatments with solutions, containing N6-benzyladenine (BA), indole-3-acetic acid (IAA) or (2-chloroethyl)-trimethylammoniumchloride (CCC) were conducted after chlorosis had been well manifested. Changes in growth, chlorophyll content, rate of photosynthesis, catalase and peroxidase activities in leaves, and the contents of Fe, Cu, Zn, Mn, and P in leaves were recorded. Growth regulators improved (CCC, IAA) or aggravated (BA) the physiological state of chlorotic plants. Their effect might be explained by changes in Fe transport towards the leaves, by increased efficiency of Fe utilization, and by effects on plant metabolism not involving Fe.

Serologically-Related Anionic Peroxidases from Petunia and Cucumber can Substitute Flavonoid Antioxidants

In this study we have investigated whether naturally occurring flavonoid-deficient mutant Red Star of Petunia hybrida is capable of metabolizing H2O2 by invoking other antioxidant enzyme system. We demonstrated that reduced flower pigmentation due to a reduction in the chalcone synthase mRNA expression results in strong H2O2 accumulation accompanied by the induction of a specific set of anionic peroxidase (PRX), serologically-related to main cucumber srPRX. We found correlation between rate of H2O2 accumulation and qualitative, as well as quantitative changes in the srPRX expression which seems to be determined by flower phenotype. In detached flower buds cultured in vitro both abscisic acid and anther extirpation prevented anthocyanin pigmentation, and thus flavonoid biosynthesis, resulting in a marked accumulation of immunoprecipitable srPRX. In contrast, pigmented flowers cultivated under the same conditions did not accumulate corresponding srPRX. The results suggest that a specific set of anionic PRX can substitute for the absence of flavonoid antioxidants.

Leaf Pubescence, Water Relations and Chlorophyll Fluorescence in Two Subspecies of Lotus Creticus L.

The objective of the present study was to compare the plant morphology, water relations and photochemical efficiency of photosystem 2 in two wild. Mediterranean species Lotus creticus creticus and Lotus creticus cytisoides. L. creticus creticus showed higher density of trichomes and stomatal density on the adaxial leaf surface than L. creticus cytisoides, whereas L. creticus cytisoides showed higher stomatal density in abaxial surface than L. creticus creticus. These morphological traits promoted clear differences in leaf surface water retention and leaf reflectance. Leaf water potential and photochemical efficiency were lower in L. creticus cytisoides than in L. creticus creticus.

Transfer of Cesium from the Xylem to the Phloem in the Stem of Wheat

Steam-girdling experiments with detached wheat shoots showed that cesium was eliminated from the xylem sap and loaded into the phloem during acropetal transport. This transfer is important for the accumulation of cesium (especially also of the radiopollutants 134Cs and 137Cs) in maturing wheat grains.
