Heterogeneity of stomatal density in the second wheat leaf

The stomatal density on abaxial epidermis of the second wheat(Triticum aestivum L.) leaf increased step by step from the leaf base to its tip. On adaxial epidermis, this increasing tendency was generally maintained, but close to both the base and the apex two maxima were determined. In the direction perpendicular to leaf length, maximum stomatal density was close to the edges, and minimum one was in the central part of the leaf blade.

Effect of nacl- salinity on metabolism of proline in salt- sensitive and salt- resistant cultivars of rice

The effect of NaCl at sublethal concentration was observed on germinating seeds of salt-sensitive and -resistant rice cultivars with respect to the level of proline regulatory enzymes and the growth of seedlings on different days of early germination period. The two enzymes of proline biosynthesis and catabolism, Δ-pyrroline-5-carboxylate reductase and L-proline dehydrogenase, were taken into consideration to observe the effects of 100 mM NaCl on their activities in both rice cultivars. The activity of Δ-pyrroline-5-carboxylate reductase in salt-resistant cultivar was increased twice after 5 d in 100 mM NaCl. Simultaneously, the activity of L-proline dehydrogenase was decreased significantly. High activities of Δ-pyrroline-5-carboxylate reductase may be regarded as a biological marker for screening the sensitive and resistant cultivars of rice seed under NaCl-salinity.

Activity and isoenzyme composition of peroxidase and esterase in fertile and male-sterile lines of tomatoes (Lycopersicon esculentum Mill.)

A comparative study of the isoenzyme patterns of esterase and peroxidase and overall peroxidase activity in stamens of male-sterile (MS) lines of Pearson ms-35 and P ms-35aa and of the respective male-fertile (MF) tomato plants have been conducted. The study has been made at two stages of stamens development - tetrad and pollen. Higher activities of the esterase isoenzymes in the MF stamens than that of MS in both ontogeny stages have been found. The slow moving esterase isoenzymes both of the MF and the MS stamens are the major isoenzymes in the early stage and are connected with tapetum development while the fast moving esterase isoenzymes are connected with pollen formation in the later ontogeny stage. Overall peroxidase levels in the MS stamens were higher than those of MF. The peroxidase patterns of the MS lines are also characterized by the greater number of isoenzymes and also the presence of specific isoenzymes, the contrast between the MF and the MS stamens being more strongly expressed at the later stage of development. A strong similitude between esterase and peroxidase patterns behaviour in both MS lines has been found.

Photoperiodic induction of flowering in green and photobleachedChenopodium rubrum L. ecotype 184 - a short- day plant

Chenopodium rubrum L. ecotype 184 is a qualitative short-day plant with critical length of the night of eight hours that must be exceeded in order to flower: Five days after sowing, the plants were exposed to a various number of inductive cycles (14/10 h of däy/night cycle) to test the optimal photoperiodic conditions for flowering. In our experimental conditions the plants flowered with high percentage after more than four received inductive cycles, but there was no flowering below that. The plants grown on the herbicide Norflurazon (photobleached plants) showed different photoperiodic characteristics. There was negligible flowering of photobleached plants in the same experimental conditions as for the green ones.

Hormonal regulation of flowering and fruit development: Effect of gibberellic acid and ethrel on fruit setting and development ofMomordica charantia L.

Fruit setting and development in a monoecious cucurbit,Momordica charantia L. could be regulated by the external application of gibberellin (GA3) and ethrel. Both GA3 and ethrel in lower concentrations promoted female flower production as well as fruit setting and development. Both growth regulators improved the quality of theMomordica fruit by increasing length, breath and biomass of the fruits as well as by increasing the content of total sugar of the fruit.

Effect of essential oils from some higher plants against fungi causing damping-off disease

A screening of leaves of 25 taxa of angiosperms was made for their volatile toxicity against damping-off fungi. The volatile substances fromHyptis suaveolens andOcimum canum were toxic againstPythium aphanidermatum, P. debaryanum andRhizoctonia solani. The fungitoxicity of the leaves persisted for 15 d of storage. The volatile substances from the leaves ofO. canum were thermostable, while those fromH. suaveolens were thermolabile. The essential oils exhibited strong potency against the pathogens tested, non-phytotoxic nature to the host plants and superiority over commonly used synthetic fungicidesAgrosan G.N. andCaptan. The findings indicate the possibility to use these essential oils as potential natural fungicides in management of damping-off pathogens.

The localization of nonspecific esterase and cholinesterase activity in germinating pollen and in pollen tube ofVicia faba L. The effect of actinomycin D and cycloheximide

Two types of esterases, nonspecific esterase and cholin esterase have been distinguished in germinating pollen grains and in pollen tubes ofVicia faba using cytochemical methods. The localization of each of them was different. In the hydrated non-germinating pollen grain the nonspecific esterase was present in the cytoplasm and in the intine. The cholinesterase was localized mainly in the sexine and on the outside of the plasma membrane. A particularly large agglomeration of this enzyme was found in the aperture. During germination both types of extracellular esterases were released into the medium. In the pollen tube the activity of the enzymes studied was connected with the wall of its tip. The localization of both types of esterases in the germinating pollen grain and the growing tube were independent of actinomycin D. Cycloheximid prevented the cholinesterase localization at the pollen tube tip.

Microtubule- like structures in developing wheat-leaf chloroplasts

The effect of CdCl2 in a concentration range 0.01-10.0 g m-3 on the growth ofChlorella vulgaris under synchronous cultivation conditions was determined. The general biological activity, the growth multiplication factor, the cell size and shape and intracellular arrangement showed disturbances of synchronization that depended on Cd2+ concentration. The highest inhibition of all mentioned parameters was observed when Cd2+ was administered after the second hour of synchronous cultivation, whereas the administration after 6 or 8 h did not induce any significant effect.

Effect of iso-osmotic levels of salts and PEG-6000 on enzymes in germinating pea seeds

Effects of iso-osmotic levels of salts (NaCl, CaCl2, Na2SO4) and PEG-6000 on the activity of hydrolytic and nitrogen assimilatory enzymes in pea embryo axis and coty ledon were studied. The activity of nitrate reductase and nitrite reductase in embryo axis and cotyledon and the activity of protease and α-amylase in cotyledon decreased with decreasing medium osmotic potential as compared to control at all the stages of seedling growth. The activity of protease and amylase increases with increasing levels of stress in embryo axis. Sodium chloride induced, stress had more deleterious effects on the activity of nitrate reductase, nitrite reductase and αamylase followed by other salts and PEG-6000. On the other hand, CaCl2 induced salt stress was more depressive for protease activity. The maximum increase in the activity of protease and amylase was observed in embryo axis at higher concentration of salts and PEG-6000.

The temperature gradient as the driving force of water and solutions flow in the root

The examination of thermoosmosis in the maize primary root tissue has shown that under the rising temperature gradient (gradT) all flows are declining, which continue also under the gradT levelling between compartments. The permeability coefficients are declining similarly as flows. The reflection coefficient during the rising gradT declines, but it rises during the gradT levelling between compartments. These phenomena depend on the plant age and concentration of the bathing solution.

The effects of gibberellic acid on photosynthetic pigments and oxygen evolution inChlamydomonas andAnacystis

Gibberellic acid (GA3) generally increased the contents of chlorophyll but not carotenoid in bothChlamydomonas reinhardii andAnacystis nidulans grown under continuous irradiation. The photosynthetic oxygen evolution of the algae was also affected by GA3 except for the high (100 μM) concentration of GA3.

Growth and pigment content of wheat as influenced by the combined effects of salinity and growth regulators

A field experiment was conducted to investigate the effects of presoaking the wheat grains (Triticum aestivum L.) in different levels of salinity (33 or 66 mM) and in growth regulators (indolyl-3-acetic acid, IAA at SO g m-3, gibberellic acid, GA3 at 100 g m-3, or kinetin at 100 g m-3) on the shoot growth and pigment content of the developing wheat flag leaf. Salinity at 33 or 66 mM led to an insignificant increase in the fresh and dry masses as well as in the shoot diameter and shoot length, but it attenuated the flag leaf area. In the majority of cases, salinity increased the chlorophyll (Chla, Chlb) and carotenoid contents as well as the number of chloroplasts per a mesophyll cell. The growth in the wheat shoot of the saline-treated plants was, in general, stimulated in response to presoaking the grains in kinetin or GA3. On the other hand, IAA + salinity led to a negligible effect on the growth in the wheat plants particularly at the early stages of growth. The presoaking of grains in NaCl at 33 mM + IAA or 66 mM + kinetin induced a marked increase in the pigment content of the wheat flag leaf particularly at the early stages of growth. The interaction between salinity and phytohormones increased the number of chloroplasts; kinetin was the most effective.

Photocontrol of biological processes in monocotyledonous plants after relaxation of phytochrome system

Effects of a short-term pre-irradiation of etiolated wheat seedlings by radiation of various wavelengths and various irradiances on the rates of photophospho-rylation, Hill reaction, and chlorophyll accumulation were studied. The curves, constructed for the dependence of inhibitory effect in different processes on pre-irradiance doses, have a saturation level corresponding to photostationary state of the phytochrome system and a pre-threshold site where the inhibition is absent. For an interpretation of the experimental data we suggest a general model that includes both the threshold concept of Mohr and the presence of an additional control mechanism not sensitive to far-red radiation. According to the model, the phytochrome-controlled biological effect is pro-portional to the time during which the level of active phytochrome is higher than the threshold one and may be analysed according to the inhibition degree. The difference in control by red (RR) and green radiation (GR) pretreatments is attributed to the difference of the phytochrome photoconversion rate under RR and GR.

Effect of temperature onin vitro pollen germination in pigeonpea

Pollen of pigeonpea(Cajanus cajan (L.) Millsp.) cultivars H-77-216 and ICPL-151 were cultivatedin vitro at six different temperatures (12, 17, 22, 27, 32, 37 °C). Pollen of cv. H-77-216 started to germinate at 17 °C whereas the pollen of cv. ICPL-151 at 22 °C, the optimal temperatures were 22 and 27 °C, respectively. Pollen germination at different temperatures was found to be positively correlated with the tube length. Per cent pollen bursting increased with rising temperature. The indeterminate cv. H-77-216 showed a wide range of suitable temperatures (17 - 27 °C) for pollen germination while the determinate cv. ICPL-151 had optimum at 27 °C

The effect of Lead on early stages ofPhaseolus vulgaris L. growthin vitro conditions

Lead chloride (10-5 M) inhibited the growth of the main root, the duration of development, the number and growth of lateral roots, primary and trifoliate leaves, and also the mitotic index in root apical meristems. Lead strongly inhibited root growth rate, mainly by reducing the number of dividing cells. Other mechanisms of this inhibition are discussed.

Leucine aminopeptidase and ferricyanide reductase activities in radish microsomes

A NADH-ferricyanide reductase activity found in radish microsomes isolated from germinated seeds has been shown to be stimulated by pCMB and pCMBS which are both strong nactivators of many plant proteolytic enzymes. In the same preparation a leucine aminopeptidase was found while endoprotease and carboxypeptidase activities were not detected using exogenous substrates. The aminopeptidase, highly active at the same optimal pH-condition of FeCN reductase, was stimulated by CoCl2 and non-polar detergents (Triton X-100 and Brij 35). It was inhibited by sulphydryl reagents. By gel filtration of microsomal detergent extract two peaks of activity were separated: red I coeluted with LeuAPase and red II, free of aminopeptidase. Red I, a protein, was inhibited by sulphydral reagents and stimulated by duroquinone. Red II, stimulated by pCMB, is not a protein because of the small size and the noninfluence of heating treatment on catalytic activity.

An immunochemical testing of pathophysiological reactions of several PSTVinfected tomato (Lycopersicon esculentum L.) cultivars

Several tomato cultivars were infected with a severe strain of PSTV and a pathophysiological reaction was characterized by means of enzyme-linked immunosorbent assay with serum containing IgGs to disease-associated host-specific leaf proteins. A strong expression of disease-associated symptoms (stunting, epinasty, leaf blade malformation and rugosity) and strong immunochemical reaction was found for cultivars Bizon, Linia, Revermun and Rutgers. The immunochemical assay revealed appearence of a major antigenic protein having a molecular mass of about 70 kD in these cultivars. The immunochemical reaction with disease-associated proteins reached a maximum four weeks after inoculation of PSTV. A weak immunochemical reaction was observed, if proteins from cvs. Sonato, Harzfeuer and Karlik were analyzed. Cvs. Sonato and Harzfeuer did not show characteristic symptoms such as stunting, leaf blade malformation and rugosity. Except for stunting, the same was true for cv. Karlik. On the other hand, no significant difference in PSTV accumulation was observed among the cultivars analyzed.
Reciprocal hybrids obtained by crossing cvs. Revermun and Karlik showed strong symptoms of the disease (Revermun-type) and increased activity of nuclease due to PSTV infection. On the contrary, an immunochemical analysis revealed a low level of the disease-associated antigens in these hybrid tomatoes, suggesting rather recessive genetic background determining their expression during PSTV-caused pathogenesis.

Anatomical characteristics of the uppermost internode of winter wheat genoypes differing in stem length

Twenty one winter wheat(Triticum aestivum L.) genotypes classified as (1) short, (2) medium or (3) tall were grown in field trials. The following parameters have been measured on the main stem: stem length, peduncle length, dry mass and conducting system parameters including number of vascular bundles and their total and phloem area on peduncle cross section, ear yield and its components. Regardless of differences in peduncle length among the genotypes groups differing in stem length, the peduncle relative length varied only between 36 and 38 % of the total stem length. Nearly identical was also the peduncle specific dry mass reaching 7.4 to 7.6 mg (dry mass) per cm (peduncle length).
The highest number of vascular bundles as well as the largest bundle and phloem area per peduncle cross section were found in the group of short cultivars. There was a positive correlation between the anatomical parameters and kernel number or dry mass per spike. The relationships need not be valid with each genotype because the final ear productivity is subjected to climatic variation well after anthesis when the formation of the stem anatomy has been by far accomplished.

Influence of Abscisic Acid on K+ absorption by leaf tissues ofSolanum tuberosum L. (cv. Bintje). Relation with calcium fluxes and with calmodulin

Previous results with potato leaf tissues revealed that a treatment with ABA (10-4 M) induced an increase of K+ uptake (Suleimanet al., 1990a).
In this study, we investigate the relationship between increased K+ uptake, Ca2+ fluxes and calmodulin by treating potato leaf discs with Ca2+ channel blockers (La3+, verapamil and nifedipine) and with calmodulin inhibitors (chlorpromazine, W7: N-(6-aminohexyl)-5-chloro-1-naphthalene-sulfonamide, and compound 4880).
We found : a) all these inhibitors decreased K+ uptake in the same ratio in tissues treated or not with ABA; b) a synergistic effect between ABA and the Ca2+ ionophore A 23187 on K+ uptake.
The results suggest that the hormone may act on K+ uptake like a Ca2+ agonist, in accord with HUDDART's hypothesis.

Interactive effects of exogenous growth regulators and artificial acid rain on cytokinins and gibberellins in maple seedlings

Vegetation experiments were conducted under greenhouse conditions in which long-term effects of simulated acid rain on the shoots, and short-term effects of acidified root medium on the roots of two maple species (Acer pseudoplatanus L. andAcer negundo L.) were studied. Both alternatives of the acidification stress were studied with respect to exogenous applications of auxins and gibberellins. Changes in seedling growth caused by acid rain were not marked, but changes in the levels of endogenous growth regulators were detected, both the long-term exposure ofAcer pseudoplatanus L. seedling shoots to acid rain and the short-term acidification of root environment resulted in enhanced cytokinin activity and in decreased gibberellin activity. But when A.pseudoplatanus L. seedlings were pretreated with gibberellins, their activity was further increased by increased acidity.Acer negundo L. seedlings responded to the acidification of their root environment with enhanced cytokinin activity only when pretreated with exogenous auxins. The differences in the responses ofA. pseudoplatanus L. and A.negundo L. to the acidification can be explained in terms of their distinct physiological properties, including changes recorded in chlorophyll content. The results obtained show that a pretreatment of maple seedlings with exogenous growth regulators can modify the effects of increased acidity in the environment on their physiological status as characterized by the level of endogenous cytokinins and gibberellins.
