Effects of radiation, temperature and humidity on photosynthesis, transpiration and water use efficiency of oilseed rape (Brassica campestris L.)

The net photosynthetic rate (F), transpiration rate (Q) and water use efficiency (F/Q) of oilseed rape (Brassica campestris L. cv. Span) was studied under a range of atmospheric conditions by gas exchange techniques. The plants were at the full bloom/pod initiation stage of development at the time of measurement. The environmental conditions consisted of various levels of photosynthetically active radiation (100 to 2800 (μmol m-2 s-1 PAK: 400-700 nm), air temperature (10 to 42°C) and vapour pressure deficit (0.7 to 2.1 kPa VPD). The peak values ofF were recorded at 1600 μmol m-2 s-1 PAR, 20°C air temperature and 1.2 kPa VPD of air in the chamber.
Q increased with increasing PAR, air temperature and VPD. However, theF/Q remained high and almost constant from 600 to 1600 μmol m-2 s-1 PAR, but declined at the low and high photon flux densities.F/Q decreased progressively with increase in air temperature and VPD of air in the chamber.

Peroxidase activity and isoenzyme patterns in wheat during ontogenesis

Protein content, total and specific peroxidase activity and isoperoxidase patterns were determined in crude protein preparations from individual parts of field-grown wheat (Triticum aestivum L., cv. Jubilar). Protein content in roots, leaves, and stalks increased at the beginning of ontogenesis and then decreased from 6th, 9th, and 10th development phase (according to Feekes), respectively. Steady increase of the protein content in the ears was observed.
Highest peroxidase activity was found in the roots; it diminished from the onset of ontogenesis till maturity of the plants. In the leaves and stalks a slight decrease of peroxidase activity till the 10th development phase and then an increase till maturity was found. The ears exhibited a gradual increase of peroxidase activity. The course of specific peroxidase activity was found to be very similar to that of total activity.
Isoperoxidase patterns did not change significantly. In the leaves, a decrease of activity of C4 and C5 isoperoxidases was recorded. In the stalks, C l isoenzyme emerged at the end of ontogenesis. A gradual increase of A1 and A5 isoperoxidase intensity took place both in the leaves and stalks.

Tissue specificity of assimilation and transport of nitrogen in the root ofZea mays L. seedlings. III. Interaction of radial and longitudinal transport

Roots obtained from 4-day maize seedlings were allowed to absorb uniformly labelled14C-suorose only with their middle part while the apical and basal parts did not contact the medium. The amino acids and their amides produced in the cortex at the expense of nitrates and the14C-sucrose carbon moved radially to the stele where they were selectively distributed in the polar directions of transport. "Primary" amino acids moved predominantly in the apical direction while amides and basic amino acids were transported mainly in the opposite direction. When the nutrient medium was devoid of nitrogen sources, the amino acids entering the stele from the cortex were transported mostly to the root tip, indicating a significant role of young tissues in consuming organic nitrogen compounds when they are in short supply in the root.

Comparison of dependence of growth and sexual reproduction ofChlamydomonas geitleri on temperature and irradiance

Dependence of the growth of this alga culture on temperature displays a wide interval of temperatures (from 4 °C to 30 °C) and the dependence of the sexual reproduction only a narrow one (from 18 °C to 25 °C). The optimum temperatures for both the processes are not so different (round 23 °C). Dependence of both the processes on irradiance displays a wide interval of irradiances in both eases (from 4 W mt-2 to 80 W mt-2 PhAR and higher). The optimum irradiances for these processes are very different (approx. 50 W mt-2 PhAR for the growth and approx. 10 W mt-2 PhAR for the sexual reproduction) under the experimental conditions used.

Acid-soluble chromosomal proteins in maize root and callus cells and after rhizogenesis induction in callus tissues

Callus tissues originating fromZea mays root meristem, induced for rhizogenesis callus, meristematic and differentiated maize root cells for isolation of nuclei and acid-soluble chromosomal proteins were used. Cytological investigations proved that rhizogenesis begins with the formation of meristematic centres, followed by root differentiation about 5-12 days after the treatment with α-naphtalene acetic acid (NAA). When applying electrophoresis in 15% polyacrylamide gel, differences between the electrophoretic profiles of acid-soluble chromosomal proteins, isolated from root cells and from callus tissues, were established. The main differences concern histone H1 and probably H4. There are no differences between electrophoretic patterns of acid-soluble chromosomal proteins of nonorganized callus and callus induced for rhizogenesis. The possible explanation of these results is discussed.

Lack of end-product control of nitrite reductase level in pea roots

The effects of various ammonium salts and amino acids on nitrite reductase (NIR) induction in isolated pea roots cultured in media containing nitrate or nitrite and either exogenous sucrose or no sugar were investigated. Thg aim of these investigations was to determine if the NIR level is subject to end-product control. The results showed that even though some ammonium salts and casamino acids can depress NIR level under certain conditions this inhibition cannot be interpreted in terms of direct end-product inhibition of NIR synthesis because their effects were dependent on the character (anion) and toxicity of the respective ammonium salt, on the presence of exogenous sucrose in the induction medium, and on the inducer of NIR. NH4HCO3 inhibited NIR induction at those concentrations which were toxic to the roots, ammonium phosphates hampered NIR induction only in roots exposed to nitrite in media containing sucrose, while casamino acids slightly depressed NIR induction only in roots exposed to nitrate and exogenous sucrose. The results further show that the basal (noninduced) NIR level changes little even under strongly toxic conditions.

Multiple shoot formation from almond embryos

Stimulation of multiple shoot formation from almond embryos using 6-benzylaminopurine was studied. Maximum shoot numbers and shoot growth were obtained with 2.5 μM in the growth medium. Continuous multiple shoot formation from cut shoots was also achieved.

Effect of some nitrogenous salts on nitrogen transfer and protease activity in germinatingZea mays L. seeds

Maize seeds were allowed to germinate in the presence of different nitrogenous salts for 72 h. Changes in the ethanol soluble and insoluble nitrogen were studied in the embryo and in the endosperm. Supply of Ca(NC3)2 enhanced germination and protease activity in the endosperm resulting in greater solubilisation of protein to soluble nitrogen in the seeds. NH4NO3 and (NH4)2SO4 were less effective as compared to Ca(NO3)2. Cycloheximide inhibited germination and protease activity.
Pretreatment also resulted in increase in growth, soluble and insoluble nitrogen, and nitrate reductase activity in the primary leaves. Ca(NO3)2 was more effective than NH4NO3 and (NH4)2SO4.

Responses of the epidermal diffusive conductance (gep) to irradiance (I) during ontogeny of primary bean leaves or during their wilting were followed. Effects ofI, leaf age and leaf water potential (Ψw) as well as interactive effects (I × leaf age andI × Ψw) ongep were statistically significant.

Electrolyte leakage in relation to viral and abiotic stresses inducing necrosis in cowpea leaves

Different types of stress, such as hypersensitive reaction to viruses or necrotic response induced by chemical and mechanical injuries, caused similar patterns in early electrolyte leakage from discs of cowpea leaves. The electrolyte leakage, suggestive of permeability alterations, always occurred in advance of cell death. Altered permeability can therefore be considered as an aspecific response to stress and a marker of necrogenesis. No correlation was found between permeability alterations and the mechanism for virus localization, the main characteristic of the hypersensitive reaction, because tobacco rattle virus induced similar alterations in either incompatible cowpea or compatible tobacco plants. Early Ca2+ losses occurred following viral and abiotic necrogenic stresses. The external supply of Ca2+ to cowpea during the course of the hypersensitivity to viruses failed to effect pathogenesis, assessed as the number and size of the resulting necrotic local lesions, and the release of electrolyte leakage from the infected leaves. However, the external supply of Ca2+ counteracted the negative effect of EDTA, a chelating agent, on the resistance of cowpea leaves to cell-to-cell spread of viruses, suggesting that a Ca2+-deficiency may weaken the mechanism of defense of the cells against the localized viral infection.

Selective inhibition by arabinosylcytosine of oligo(U)-containing RNA synthesis in germinating wheat embryos

Arabinosylcytosine (araC) was found to influence the pattern of RNA labelling in early wheat embryos. Although the rate of uridine-14C incorporation into total RNA remained unchanged, specific radioactivity of an oligo(U)-containing subclass of nonpolyadenylated RNA was markedly lower (50%) in araC-treated (1.5 mM, 1 h) than in control embryos. The inhibitory effect was stronger for nuclear than for cytoplasmic subpopulation of these RNA species. h labelling of all other RNA fractions, including bulk nonpolyadenylated RNA as well as poly(A)-containing RNA of both nuclear and cytoplasmic origins, did not respond to the presence of araC.

Analysis of specificities in the movement of cytoplasmic clusters in the cell ofAcetabularia mediterranea

The movement of cytoplasmic clusters of high density was studied in the stalk ofAcetabularia mediterranea using time-lapse microcinematography during regeneration. The kinetics of expansion and contraction of cytoplasmic clusters is described in acropetal and basipetal directions.

The inhibition and stimulation of DNA synthesis in shoot apices ofChenopodium rubrum L. during photoperiodic induction of flowering

Three short-day inductive cycles bring about inhibition followed by transitional enhancement of growth, not only in roots and leaves but also in different zones of shoot apical meristem, as shown by measurement of DNA synthesis using3H-thymidine autoradiography. The first inductive cycle resulted in marked inhibition of the cells of the central zone (CZ), rib meristem (RM), and peripheral zone (PZ). Subsequent enhancement of DNA synthesis occurs in RM during the second inductive cycle, but in CZ only in the third cycle. The growth activation in PZ is counteracted by decrease in apical dominance which results in further inhibition of leaf primordia and increases in bud primordia. In plants induced only by one cycle, which later reverse the vegetative pattern of growth and differentiation, increased DNA synthesis in RM and CZ was not observed. The significance of inhibitory and stimulatory processes in particular zones of the shoot apex is discussed considering flower morphogenesis.

Cellular differentiation and nitrogenase activity in the cyanobacteriumAnabaena

Nitrogenase activity at periods of differentiation of heterocysts and akinetes was assayed by the acetylene reduction technique. There was no nitrogenase activity in ammoniumgrown, non-heterocystousAnabaena sp.; the activity appeared only after a lag-phase of about 17 - 21 h after the ammonium-grown culture had been transferred to medium free of combined nitrogen. This activity started appearing as the proheterocysts were developing to mature heterocysts. Maximum nitrogenase activity was attained with exponential phase of culture and mature heterocysts. This activity gradually decreased with the differentiation of akinetes. Only insignificant nitrogenase activity was observed in old cultures in which most cells had matured into akinetes.

A case of early dissolution of the microsporocyte callose wall in male-sterile (CMS) sweet pepper (Capsicum annuum L.)

On squash preparations of anthers from pollen fertile and sterile plants of sweet pepper (Capsicum annuum L. cv. Severka) callose envelopes of microsporocytes, stained specifically with resorcin blue, were investigated microscopically. During normal course of microsporogenesis in fertile plants the envelopes remained intact up to the stage of microspore tetrads. Then callose begins to dissolve, and that from individual microspores towards the envelope periphery. In sterile analogues of the same cultivar the callose breakdown occurred precociously, usually in the course of the second, but sometimes as early as the first meiotic division of PMCs. Having completed meiosis sporadic microsporocytes formed microspore tetrads. Most PMCs contained an undivided four-nucleate protoplast rimmed with a narrow or wider unstained zone of dissolved callose. In certain cases more condensed callose septa pointing to the furrows on the surface of the PMC protoplast were well-observable in this lytic zone, as a residuum of normal mechanism of tetradogenesis.

Comparative studies on the electrophoretic patterns of acid-soluble chromosomal proteins duringZea mays early stages of embryo germination and root cell differentiation

Acid-soluble chromosomal proteins were extracted from purified nuclei, isolated from 3 - 4 h, 12 -14 h and 24 -26 h maize embryos, as well as from nuclei isolated from meris-tematic, elongating and differentiated cells, from 2 and 3-day-oldZea mays seedlings primary roots.
Using polyacrylamide gel electrophoresis (urea-acetic acid in the cylindrical gels and slab-SDS-electrophoresis), it was established that germination and root cell differentiation induced changes in some nuclear acid-soluble chromosomal proteins, especially in histone H1.
The results of proteins belonging to the high-mobility group proteins (HMG) and some other acid-soluble proteins with unknown nature for the plants, based on the electrophoretic mobility, were discussed.

Effects of toxic concentrations of natrium fluoride on growth and enzyme activities of rice (Oryza sativa L.) and jute (Corchorus olitorius L.) seedlings

Germination was increasingly inhibited at concentrations above 5 and 20 mM and stopped altogether at 80 and 50 mM NaF in rice and jute respectively, whereas the inhibition of seedling growth began at much lower concentrations. Of the enzymes, RNase activity was increased by fluoride, while α-amylase, protease, phytase and ATPase activities exhibited distinct inhibition from the control.

A Simple model of the leaf daily water potential dynamics of some forest tree species

A simple model was developed to characterize the daily water potential dynamics (Ψx of sun and shade leaves of three forest tree species (Quercus cerris, Acer campestre andCarpinus betulus) under anticyclonic weather types. Input data used for this-model were the vapour pressure deficit (d) and the soil moisture content (w.). The model is usable for the calculation of the actual Ψx-values with a probable error 0.18 -0.28 MPa and limits the maximum and minimum Ψx-values which may occur with the particular tree species. The model makes it possible to establish for each species the regime, determined byd andw, at which the water potential of the leaves reacts most sensitively to the changes of the environmental parameters.

Effect of GA3 and AMPs on the activities and electrophoretic patterns of acid and alkaline phosphatases in relation to flowering ofImpatiens balsamina L.

GA3, cyclic AMP as well as 3'-AMP and 5'-AMP induced the formation of floral buds inImpatiens balsamina under strictly non-inductive photoperiods. While photoperiods and treatments with GA3 or AMPs did not much affect acid phosphatase activity, AMPs increased the activity of alkaline phosphatase both in the stem and the leaves under both photoperiods. The phosphatase activity of the water- and GA3-treated plants under inductive photoperiods was higher than that of the plants of the respective treatments under non-inductive photoperiods. GA3 as well as all the three AMPs induced both in the stem and the leaves the formation of new isoenzymes of both these enzymes under both photoperiods.
