Comparison of esterase isoenzyme patterns in seeds of someAllium species and in cultivars ofAllium cepa LVěra Hadačová, Eva Klozová, E. Fedtsch.,A. altaicum (Pall.) Reyse,A. Cristophii Trautv.,A. fistulosum L.,A. jajlae Vved.,A. Karsianum Fom.,A. nutans L.,A. porrum L. cv. Gigant,A. praemixtum Vved.,A. pskemense Vved.,A. ramosum L.,A.rotundum L.,A. schoenoprasum L.,A. stipitatum Regel. The cultivars differ in their isoenzyme patterns, the cultivar Kaštická stands apart from all the other cultivars, probably due to the high alkalinity of its seed extract. The examined species, arranged according to their mutual similarity of isoenzyme patterns, form several groups corresponding to individual sections of the genus. Our results corroborate the recognizing of the sectionCepa and the subsectionPhyllodolon. The maintaining ofA. jajlae andA. rotundum as well described species fits better with our results, than degrading them to subspecies ofA. scorodoprason. |

Kutáček, Eva KlozováBiologia plantarum 23:220, 1981 | DOI: 10.1007/BF02894892 In the seeds ofAllium altaicun (Pall.)Reyse a set of enzymes was found, metabolizing choline esters, composed of active choline esterases and choline acetyltransferase. Choline esterase cleaving acetylcholine occurs in five isoenzymes. The enzyme preparation hydrolyses strongly acetylthiocholine and sinapine, but weakly butyrylthiocholine (20%) in comparison with acetylthiocholine. The hydrolysis of the substrates mentioned is inhibited by physostigmine and neostigmine, but it is not inhibited by the specific inhibitor of acetylcholine esterase (BW 284 C51). In addition to hydrolytic activity a strong catalytic activity of choline acetyltransferase was also observed during the synthesis of sinapine from sinapic acid and choline. The detection of the mentioned enzymes in some representatives of theAllium genus indicates that choline esterases are more widely distributed in monocotyledons than previously assumed. |
Dependence of the gametogenesis induction, zygote formation and their germination on the culture density of the homothallic algaChlamydomonas geitleri EttlJ. NečasBiologia plantarum 23:278, 1981 | DOI: 10.1007/BF02895365 The induction of gametogenesis has its beginning in the most diluted cell suspension after the transfer of the cells to a nitrogen-less medium. Here, the highest percentage frequencies of zygotes are formed within the same period of time in comparison with the less diluted cell suspensions. The zygotes formed in the mostly diluted cell suspensions mature relatively very slowly and germinate very irregularly. The induction of gametogenesis retards in the denser cultures probably due to the strongest homeostatic forces trying to return the cell population to its initial stage. In our experiments up to now, the effects of the changing irradiance of the cells inside the suspension were not separated from the effects of the changing number of the cells in the given volume of the culture. Neither were the ratios of distilled water to the amount of the zygotes, nor to the vegetative cells constant. Thus, it is necessary to consider the mentioned effects as the result of an interaction of both factors under consideration. Each of the cell populations behaved as an autonomous whole. Also the populations, starting after the transfer to a nitrogen-less medium of the same culture density, may sometimes differ significantly in the observed characteristic features at various phases of their growth. The discontinued supply of nitrogen obviously causes a change of the cell metabolism in favour of the nitrogen-less substances, especially in the more diluted cell suspension. This work completes and explains some earlier results obtained from the study of the life cycle inChlamydomonas geitleri. |
Gibberellic acid- and salicylic acid-caused formation of new proteins associated with extension growth and flowering ofImpatiens balsaminaSurinder Kumar, K. K. A new protein band with Rm 0.47 seems to be associated with floral bud initiation as it develops within 1-3 days in the stem as well as in the leaves of plants exposed to inductive treatments regardless of whether the induction is caused by 8-h photoperiods or by treatment with GA3 of SA under 24-h photoperiods. Another band with Rm 0.23 developed only in the stem of water-as well as GA3- or SA-treated plants under 8-h photoperiods. It may possibly be associated with extension growth. |

Visual estimation of leaf water stress inMercurialis perennis L.P. EliášBiologia plantarum 23:456-461, 1981 | DOI: 10.1007/BF02880596 Leaf inclination of three upper leaf pairs of a plant was used as a visual parameter for estimation of leaf (plant) water status inM. perennis. Negative correlation was found between leaf angle and leaf water saturation deficit (WSD). Large angles (between 130 and 80-90°) indicated WSD below approx. 12%, narrow angles expressed negative water balance of the plant and indicated usually strong or moderate water stress. The correlation may be expressed by two regression lines differing in slope. Some differences were observed between three leaf pairs investigated: At the same water deficits, leaves of the first pair (from apex) were inclined to a less degree than leaves of the other two leaf pairs. Leaves of the third pair wilted the most rapidly, therefore their WSD were highest at moderate and strong water stress of the plant. The approximation method tested is suitable for judgment of the plant water status inM. ptrennis and may be used in ecological investigations in forest ecosystems. |
Cytogenetic effect of radiation products of cytosine: Parabanic and oxaluric acidsK. Pešina, M. SosnaBiologia plantarum 23:46, 1981 | DOI: 10.1007/BF02909210 The two final products of radiolytic degradation of cytosinei.e. parabanic and oxaluric acids were investigated as regards their cytogenetic effects. Broad bean (Vicia faba L.) root meristem was used as experimental material. While the oxaluric acid was not able to induce chromosomal aberrations to a greater extent, the parabanic acid acts as a clastogenic compound. When applied on a resting broad bean root meristem at concentration from 10-3 to 10-2 M it induces chromosome and chromatid aberrations, especially breakages. Their frequency reaches 9-12% at the highest tested concentration. The same concentrations of the parabanic acid increases mitotic index of the first cell generation of primary root. |

The infection did not influence86Rb+ uptake per plant in the course of a 14-day-period following inoculation, however it did affect its distribution differentially in the S- and the R-plants.86Rb+ uptake was significantly decreased due to the infection in the S-plants on the day 49 after the inoculation (a 4-h-exposure to86Rb+), with the iona also being more slowly translocated to the shoots in diseased S-plants than in diseased R-plants. Likely factors causing these effects and their relationship to alfalfa resistance to the bacterial wilt are discussed. |
Study of selected characteristics of theChlamydomonas reinhardii strains with altered sensitivity to UV radiationD. Vlček, Svetlana Podstavková, Eva MiadokováBiologia plantarum 23:427-433, 1981 | DOI: 10.1007/BF02880591 Selected characteristics and streptomycin resistance were studied in a UV radiation sensitive (UVS1) and a UV radiation resistant (UVR1) strains, and the data were compared with results obtained with an original type strain. A partial prolongation of the cell cycle in the UVR1 strain as compared with the original type strain could be observed in studying cell volume growth, cell numbers, DNA, RNA and protein synthesis during the synchronous cycle. Under these conditions, the UVS1 strain behaved as a temperature sensitive cell cycle mutant. In inducing streptomycin resistant mutants, the highest frequencies in various doses were recorded in the UVS1 strain. |

The other three legumes were considerably affected by salt treatments, and N2-ase activity was significantly reduced by salinity. |
The role of auxin stored in scots pine trunk during spring activation of cambial activityStefania EgierszdorffBiologia plantarum 23:110-115, 1981 | DOI: 10.1007/BF02878415 In a 9-year-old pine girdled during the winter cambial activity was observed below the girdle in the next spring. This indicates that cambial activity was initiated without auxin produced in the spring by buds. The auxin produced in apical shoots successively flows down the stem, where as a result of periodic restriction in transport it remains over the winter till the next year. This auxin of apical origin but locally stored over the winter in the stem is responsible for the activation of cambium before the new flow of auxin produced in the apical meristems arrives. Calculations based on seasonal changes in auxin levels can explain both, earlier spring activation of cambium in the crown and the temporary cambial divisions below the girdle, without assumption of direct auxin synthesis in the lateral meristems. |

The metabolic changes could not be correlated with the yield behaviour of the cultivars under a drought condition in the field. |

Control plants, grown at 25 °C were compared from the aspects of uptake capacity and leakage with plants incubated in nutrient solutions cooled to 15 °C and 5 °C, respectively. Short time intervals as well as 1-7 d exposure to cooling were used. Already after 1 h exposure at 5 °C and 5 h cooling at 15 °C and at 5 °C, considerable changes were manifested in anion uptake and leakage. |
Changes in the anatomical structure of the first two leaves of barley caused by the absence of nitrogen or phosphorus in the nutrient mediumJ. Pazourek, L. NátrBiologia plantarum 23:296, 1981 | DOI: 10.1007/BF02895371 The absence of nitrogen or phosphorus in the nutrient medium affected quantitative anatomical characteristics of the first two leaves of spring barley: nitrogen deficiency brought about changes in both the leaf size and characteristics of stomata, whereas the effect of phosphorus absence was insignificant. N deficiency had an opposite effect on certain characteristics connected with volume ratios of tissues than that of P. The most marked finding is the decrease in the leaf volume and specific volume, and in specific volumes of most tissues at N deficiency and their increase at P deficiency. On the other hand, the number of stomata per unit leaf or photosynthetic tissue volume increased as the result of N deficiency and decreased at P deficiency. |

Effect of different disintegration techniques and media on yield and appearance of isolated nucleiJ. VlasákBiologia plantarum 23:406-413, 1981 | DOI: 10.1007/BF02880585 Cell nuclei have been released from various plant tissues (barley leaves, roots and embryos, tobacco leaves and tissue cultures,Vicia faba roots,Arabidopsis thaliana leaves) by several homogenization methods and the optimal method was established for each tissue. The effect of the composition of isolation medium on the yield and appearance of isolated nuclei was also studied. Longer incubation withn-octanol increases the yield considerably in most cases. Low concentrations of osmoticum increase the yield and their adverse effect on the integrity of nuclei is of little significance. Gum arabic has a favourable effect on nuclei isolation from roots only. |

MehtaBiologia plantarum 23:91-97, 1981 | DOI: 10.1007/BF02878410 Histochemical localization of total proteins, histories, nucleic acids, ascorbic acid and polysaccharides in the developing axillary bud ofMurraya koenigii and its vascular relationship with the main axis are investigated. All the above variable metabolites are richly distributed throughout the bud development. The shell zone is indicative of poor distribution of these metabolites. Histochemical tests prove that the axillary bud is metabolically very active. The initiation of the axillary bud is in the axil of the 2nd leaf where two cells of 2nd tunica layer become prominent and undergo periclinal divisions to give rise to the bud. The bud maintains tunica-corpus organization throughout its development. The cells of the shell zone at the base of the bud differentiate into pith meristem of the bud. The axillary bud has two bud traces which are associated with vascular strands that are not the traces of the subtending leaf. |
Investigations on the causes of low susceptibility of scentless mayweed to 2,4-DDaniela Chodová, Eva Bergmannová, L. TaimrBiologia plantarum 23:161, 1981 | DOI: 10.1007/BF02894877 White mustard (Sinapis alba L.) plants, susceptible to 2,4-D, and scentless mayweed (Tripleurospermum maritimum L.) plants, resistant to 2,4-D, markedly differ in the distribution and metabolism of 2,4-D. When 2,4-D-14C was applied onto the leaf, radioactivity was found in mustard after 8 days in the entire plant, whereas in scentless mayweed radioactivity occurred mainly in the leaf onto which it was applied and in another one or two leaves, with the roots showing only traces of radioactivity. The two plant species differed both in the character and in the number and amount of metabolites detected in aqueous and ether fractions. The metabolite with Rf 0.63 -0.65, soluble in ethylether (39% of the total radioactivity applied), and that with Rf 0.35 -0.36 (23% of the total radioactivity) were present exclusively in the roots of scentless mayweed plants. The Rf values established indicate that the former metabolite may be a conjugate with the amino acids alanine and valine and the latter a hydroxylated derivate of 2,4-D. In the shoots of both plants, 2,4-D-14C was metabolized to identical metabolites with Rf 0.59 -0.61, which occurred in mustard plants only in trace amounts. Free 2,4-D-14C occurred in the shoots of both plants and in considerable amounts also in mustard roots; it could not be demonstrated in the roots of scentless mayweed. The two species did not differ in the uptake rate or in the amount of absorbed 2,4-D-14C. |
