Choliae esterases and choline acetyltransferase in the seeds ofAllium altaicum (Pall.) Reyse

In the seeds ofAllium altaicun (Pall.)Reyse a set of enzymes was found, metabolizing choline esters, composed of active choline esterases and choline acetyltransferase. Choline esterase cleaving acetylcholine occurs in five isoenzymes. The enzyme preparation hydrolyses strongly acetylthiocholine and sinapine, but weakly butyrylthiocholine (20%) in comparison with acetylthiocholine. The hydrolysis of the substrates mentioned is inhibited by physostigmine and neostigmine, but it is not inhibited by the specific inhibitor of acetylcholine esterase (BW 284 C51). In addition to hydrolytic activity a strong catalytic activity of choline acetyltransferase was also observed during the synthesis of sinapine from sinapic acid and choline. The detection of the mentioned enzymes in some representatives of theAllium genus indicates that choline esterases are more widely distributed in monocotyledons than previously assumed.

Dependence of the gametogenesis induction, zygote formation and their germination on the culture density of the homothallic algaChlamydomonas geitleri Ettl

The induction of gametogenesis has its beginning in the most diluted cell suspension after the transfer of the cells to a nitrogen-less medium. Here, the highest percentage frequencies of zygotes are formed within the same period of time in comparison with the less diluted cell suspensions. The zygotes formed in the mostly diluted cell suspensions mature relatively very slowly and germinate very irregularly. The induction of gametogenesis retards in the denser cultures probably due to the strongest homeostatic forces trying to return the cell population to its initial stage. In our experiments up to now, the effects of the changing irradiance of the cells inside the suspension were not separated from the effects of the changing number of the cells in the given volume of the culture. Neither were the ratios of distilled water to the amount of the zygotes, nor to the vegetative cells constant. Thus, it is necessary to consider the mentioned effects as the result of an interaction of both factors under consideration. Each of the cell populations behaved as an autonomous whole. Also the populations, starting after the transfer to a nitrogen-less medium of the same culture density, may sometimes differ significantly in the observed characteristic features at various phases of their growth. The discontinued supply of nitrogen obviously causes a change of the cell metabolism in favour of the nitrogen-less substances, especially in the more diluted cell suspension. This work completes and explains some earlier results obtained from the study of the life cycle inChlamydomonas geitleri.

Gibberellic acid- and salicylic acid-caused formation of new proteins associated with extension growth and flowering ofImpatiens balsamina

GA3 as well as SA increase the protein content of the stem and the leaves at 1 day under both 8- and 24-h photoperiods. A new protein band with Rm 0.47 seems to be associated with floral bud initiation as it develops within 1-3 days in the stem as well as in the leaves of plants exposed to inductive treatments regardless of whether the induction is caused by 8-h photoperiods or by treatment with GA3 of SA under 24-h photoperiods. Another band with Rm 0.23 developed only in the stem of water-as well as GA3- or SA-treated plants under 8-h photoperiods. It may possibly be associated with extension growth.

Visual estimation of leaf water stress inMercurialis perennis L.

Leaf inclination of three upper leaf pairs of a plant was used as a visual parameter for estimation of leaf (plant) water status inM. perennis. Negative correlation was found between leaf angle and leaf water saturation deficit (WSD). Large angles (between 130 and 80-90°) indicated WSD below approx. 12%, narrow angles expressed negative water balance of the plant and indicated usually strong or moderate water stress. The correlation may be expressed by two regression lines differing in slope. Some differences were observed between three leaf pairs investigated: At the same water deficits, leaves of the first pair (from apex) were inclined to a less degree than leaves of the other two leaf pairs. Leaves of the third pair wilted the most rapidly, therefore their WSD were highest at moderate and strong water stress of the plant. The approximation method tested is suitable for judgment of the plant water status inM. ptrennis and may be used in ecological investigations in forest ecosystems.

Cytogenetic effect of radiation products of cytosine: Parabanic and oxaluric acids

The two final products of radiolytic degradation of cytosinei.e. parabanic and oxaluric acids were investigated as regards their cytogenetic effects. Broad bean (Vicia faba L.) root meristem was used as experimental material. While the oxaluric acid was not able to induce chromosomal aberrations to a greater extent, the parabanic acid acts as a clastogenic compound. When applied on a resting broad bean root meristem at concentration from 10-3 to 10-2 M it induces chromosome and chromatid aberrations, especially breakages. Their frequency reaches 9-12% at the highest tested concentration. The same concentrations of the parabanic acid increases mitotic index of the first cell generation of primary root.

An easy estimation of tomato root parameters based on calcium absorption

Using tomato as a test plant Ca absorption by the plant and characteristics of root growth in the nutrient solution has been reported. It has been shown that the amount of calcium present in the tomato shoot serves as a good index of the rate of root growth in the solution as well as in the soil. The flux of Ca, F and K into the roots of field grown tomatoes have been calculated based on such estimations of root parameters.

Alfalfa (Medicago sativa; L.) plants susceptible (S) and resistant (R) to the bacterial wilt were fedvia roots with a nutrient solution labelled with86Rb+, at different times after inoculating them withCorynebacterium insidiosum (McCull.) H. L. Jens. The infection did not influence86Rb+ uptake per plant in the course of a 14-day-period following inoculation, however it did affect its distribution differentially in the S- and the R-plants.86Rb+ uptake was significantly decreased due to the infection in the S-plants on the day 49 after the inoculation (a 4-h-exposure to86Rb+), with the iona also being more slowly translocated to the shoots in diseased S-plants than in diseased R-plants. Likely factors causing these effects and their relationship to alfalfa resistance to the bacterial wilt are discussed.

Study of selected characteristics of theChlamydomonas reinhardii strains with altered sensitivity to UV radiation

Selected characteristics and streptomycin resistance were studied in a UV radiation sensitive (UVS1) and a UV radiation resistant (UVR1) strains, and the data were compared with results obtained with an original type strain. A partial prolongation of the cell cycle in the UVR1 strain as compared with the original type strain could be observed in studying cell volume growth, cell numbers, DNA, RNA and protein synthesis during the synchronous cycle. Under these conditions, the UVS1 strain behaved as a temperature sensitive cell cycle mutant. In inducing streptomycin resistant mutants, the highest frequencies in various doses were recorded in the UVS1 strain.

Changes in dry weight and mineral composition of some oil producing plants over a range of salinity stresses

Using water culture technique, some experiments have been performed to investigate the effect of 60 days salinization treatments (0.0-100 meq 1-1 NaCl) on dry weight and on the content of some nutrient elements (Na, K, Ca, Mg, P, N) in castor bean, sunflower and flax plants. In general the content of sodium increased progressively with the rise of salinity level. The relatively low and moderate salinization levels (20 and 40 meq I-1 NaCl) resulted in a promotion rather than inhibition of the dry weight and in the content of most of the investigated elements in the different organs of the test plants. However with the rise of salinization level from 60 to 100 meq l-1, the dry weight and the content of these nutrient elements were mostly reduced.

The influence of light on the cytokinin content ofAmaranthus seedlings

The influence of light, particularly blue and red light, on the cytokinin content of seedlings ofAmaranthus caudatus was studied. Cytokinin content was determined by two different bio-assays (amaranthin accumulation byAmaranthus seedlings and mtrate-reductase activity ofAgrostemma embryos). In both bio-assays similar results were obtained. Oytokinin content is increased, especially by blue light. It is suggested that especially blue light promotes amaranthin accumulation inAmaranthus seedlingsvia the increase of cytokinin content of tissues. The results support our hypothesis on cytokinin action.

The localization and the isoenzymes of α- and β-Galactosidases in root tips

The localization was studied of α- and β-galactosidases in frozen sections of Ca-formol fixed root tips using simultaneous azocoupling reaction. In all species studied (Allium cepa,Cucurbita maxima, Lupinus albus, Pisum sativum, Vicia faba, Zea mays) positive results were obtained, the localization being ubiquitous (according to localization typology given here).
InVicia faba andZea mays the isoenzymes of α- and β-galactosidases were revealed by means of acrylamide gel electrophoresis, using authors' modification of Reisfeld method, in whole root tips, particular growth zones and separately in cortex and central cylinder. No differences were observed comparing stele and cortex. Whereas characteristic isoenzyme patterns were found in individual growth zones in maize, no differences appeared in broad bean.
A comparison was made of thein situ localization and of the isoenzyme patterns of α- and β-galactosidases with α- and β-glucosidases. In the case of galactosidases, positive results appear with both α- and β-galactoside. The rising of pH to neutrality leads to considerable decrease in the activity of both galactosidases.

Effect of different concentrations of 5-fluordeoxyuridine on mitosis and chromosomes of barley root meristems

Excised barley embryos were cultivated in a liquid complete medium for 48 h and then 5-fluordeoxyuridine was added. The concentrations of 5-fluordeoxyuridine differed in the range of 6 orders of magnitude (10-8 to 10-3 M). All concentrations, except 10-8 M led to a total inhibition of mitosis during 24 h. 5-fluordeoxyuridine induced chromosomal aberrations of the non-exchange type in all concentrations used. The frequencies of induced breaks increased during the interval of 12 h in whích they were followed and, in samples fixed at later intervals after the beginning of the treatment, there was a tendency to clustering of the induced fragments in some mitotic figures. The most striking feature of the effects oliowed is the relatively small dependence on the concentration used.

Diurnal rhythms of certain sesquiterpenes in wild camomile (Matricaria chamomilla L.)

A study was made of the changes in the the content of camomile essential oil and its components [(-)-α-bisabolol, (-)-α-bisabololoxide A, (-)-α-bisabololoxide B and spathulenol, and chamazulene] in dependence on the time of day under field conditions. The essential oil, distilled immediately after sampling, was analysed by means of gas chromatography at 3-h intervals in the course of 51 h. A 24-h period rhythm of (-)-α-bisabolol with maximum content in the scotoperiod, was demonstrated in 1976 using periodogram and cosinor tests. The other substances investigated exhibit a rhythm with a 12-h period. The latter test did not reveal any rhythm in the total content of essential oil.

Effect of decapitation on morphogenesis of stem and spike in various wheat species

Decapitation induced an additional formation of secondary shoots and anomalous spikes in all the species. The moan numbers of nodes, spikelets per spike, seeds per spikelet and spike, and the mean length of the stem and spike were reduced on secondary shoots of decapitated plants, while the mean and peak numbers of flowers per spikelet and the peak number of seeds per spikelet increased. The increase in the number of flowers per spikelet was the most striking on spike base; the seeds regularly occurred even in spikelets with an expressively increased number of flowers. The post-decapitation changes of the spike could be well expressed quantitatively according to the increased mean number of the flowers per one seed. Morphological ohanges in anomalous spikes of all the wheat species resemble phylogenetic reversions described in literature. Moreover, the peak numbers of flowers and seeds per spikelet were recorded in 52 varieties belonging to 21 wheat species. As compared with the decapitation trial, the greatest variability and the greatest differences between the speoies were also reoorded in the tetraploid group, and the smallest variability and differences between the species in the diploid group. We suppose that the striking morphological differences in post-decapitation spikes take place because the apical dominance was interrupted before differentiation of the recent form had been controlled in meristems on the decapitated stem base. Ancestral forms were morphologically realized with the help of an assimilating part of the decapitated stem.

Effects of mannitol induced water stress on the ribosomes of intact leaves of Azuki bean seedlings

Sucrose gradient analyses were made on free ribosomes extracted from Azuki bean seedling leaves subjected to mannitol induced water stress. Comparisons were made of the effect of duration of stress and the effect of urea on the free ribosomes. The capacities of the ribosomal fractions to incorporate amino-acids were also studied. Water stress reduced total free ribosomes and proportion of polysomes in the ribosomes as well as the capacity of the ribosomes to incorporate amino-acids into proteins. Polysomes which were broken down by the water stress also appeared to be more susceptible to denaturation and synthesized different proteins than polysomes which remained in the leaves after water stress.

Natural infection of sugar beet with tomato bushy stunt virus

A virus transmissible toChenopodium quinoa was isolated from leaves of sugar beet showing large chlorotic ring spots and line pattern. The virus was serologically unrelated to tobacco necrosis virus and tomato black ring virus or to its beet ringspot strain either. A positive result was obtained with antiserum against tomato bushy stunt virus. Reactions of herbaceous indicators and properties of the virus in crude sap were in accordance with the serological diagnosis. A survey of natural hosts of tomato bushy stunt virus demonstrated recently by the authors is given.

The effect of indole-3-Acetic acid on ethylene formation in wheat seedlings

Isoperoxidase B 1 isolated from winter wheat (Triticum aestivum L., cv. Jubilar) seedlings was shown to catalyze ethylene formation from α-keto, γ-methylmercaptobutyric acid (KMBA). In the presence of Mn2+, indole-3-acetic acid (IAA), andp-coumaric acid, the kinetics by isoperoxidase B 1 catalyzed conversion of KMBA into ethylene and other products was similar to that of IAA oxidation. The reaction rate was therefore controlled by IAA through its electrondonating properties.
Exogenous IAA induced ethylene formation in the segments of etiolated wheat coleoptiles. IAA-induced ethylene production was enhanced by L-methionine and mitomycin C. Aminoethoxy-analogue of rhizobitoxine, ferulic acid, sodium benzoate, cycloheximide and actinomyoin D exhibited significant inhibitory effects. These data indicate that the overall reaction mechanism in coleoptile segments involves RNA and protein synthesis.
The site of IAA action is not specific; 2,4-dichlorophenoxyacetic, α-naphthylacetic and indole-3-butyric acids, respectively, possessed comparable inductive effect as IAA. Indole-3-propionic acid, indole, L-tryptophan and glucobrassicin had only low inductive efficiency, and moreover indole and L-tryptophan slowed down IAA-induced ethylene formation.

The stomatal density, dimension and conductances to water vapour diffusion of sevenHevea species were studied. It was found that these parameters were significantly different between the species. However, no distinct relationship between stomatal density or dimension and stomatal conductances could be discerned.
